Abstract:The metastatic status of sentinel lymph nodes (SLNs) might be the most important prognostic factor in breast cancer. In this paper, we report to our knowledge the first study of 99mTc-rituximab as a radiotracer for imaging of SLNs using lymphoscintigraphy in both preoperative and intraoperative breast cancer patients. Methods: 99mTcrituximab was designed as an SLN tracer targeting the CD20 antigen, which expresses extensively in LNs. A retrospective study was performed on 2,317 patients with primary breast cancer who underwent lymphoscintigraphy and sentinel lymph node biopsy (SLNB). Before imaging, all patients were administered a preoperative peritumoral injection of 37 MBq of 99mTc-rituximab. Results: 99mTc-rituximab was synthesized in both high radiolabeling yield and high radiochemical purity (.95%), with molecular integrity and immune activity well maintained. The initial study of 100 breast cancer patients showed that the success rate of SLN lymphoscintigraphy by injection of 99mTc-rituximab, as compared with SLNB, was 100%, and the sensitivity, specificity, accuracy, and false negative rate were 97.4%, 100%, 98.0%, and 2.60%, respectively. Of the following 2,217 patients studied, the success rate of lymphoscintigraphy and SLNB was 98.8% and 99.9%, and the average number of SLN was 1.78 (range, 1–10) and 2.85 (range, 1–15). Age was an independent predictor of the number of SLNs identified by lymphoscintigraphy and intraoperative handheld γ-probe (P , 0.05), and other factors—such as sex, imaging time, primary tumor site, histopathologic subtype, clinical T stage, and immunochemistry—were not (P. 0.05). However, the SLN metastatic rates were different in patients with different histopathologic subtype, clinical T stage, and immunochemistry (P , 0.05). Conclusion: Here we report the first study of the new radiotracer 99mTc-rituximab for breast cancer lymphoscintigraphy. This tracer showed great feasibility, safety, and effectiveness for SLN mapping in breast cancer patients.

[Preliminary Study of New Imaging Agent, 99Mtc-Rituximab, for Sentinel Lymph Node Biopsy of Primary Breast Cancer].

Abstract:OBJECTIVE:To study the feasibility and effects of new imaging agent, 99mTc-Rituximab, for sentinel lymph nodes biopsy (SLNB) of primary breast cancer.METHODS:SLNB were performed in eighty-five primary breast cancer patients using 99mTc-Rituximab combined with patent blue. Metastases in sentinel lymph nodes were detected with routine pathologic and immunohistochemistry method.RESULTS:The successful rate of SLNB using combining method of 99mTc-Rituximab and patent blue was 96% (82/85). Thirty cases of SLN (37%, 30/82) were metastasis positive, including twenty-four positive cases by HE staining and six by immunohistochemistry method. SLN was the only metastasis lymph nodes in 18/30 cases. One case has false negative SLN metastasis. The sensitivity and accuracy of SLNB were 97% (30/31) and 99% (81/82). The specificity was 100% (51/51). The false negative rate was 3% (1/30) and the negative predictive value was 98% (51/52). The positive predictive value was 100% (31/31). Internal mammary sentinel lymph node lymphoscintigraphy was positive in eleven cases but all of them were confirmed metastases negative by pathologic examination.CONCLUSIONS:99mTc-Rituximab, as a new imaging agent, can keep SLN imaging durable and can make SLNB more convenient. Through primary clinical manifestation, higher sensitivity and accuracy could obtained by combining method of 99mTc-Rituximab and patent blue for SLNB of primary breast cancer.
99Mtc-It-rituximab: an Effective Radiotracer for Identifying Sentinel Lymph Nodes in Breast Cancer

Abstract:1647 Objectives: To characterize the effects of 99mTc-IT-rituximab for locating SLN, and tested its feasibility as a lymphoscintigraphic agent in breast cancer patients. Methods: Rituximab was radiolabeled with 99mTc by way of the bifunctional chelator 2-iminothiolane (2-IT). The immunological integrity of 99mTc-IT-rituximab was evaluated by SDS-PAGE, ELISA and flow-cytometric analysis (FCA). The biological property and safety limitation of 99mTc-IT-rituximab was studied according to China Pharmacopoeia. Eighty-five breast cancer cases, including one with bilateral breast cancer, were studied. Lymphoscintigraphy was performed after peritumor and intradermal injection of 99mTc-IT-rituximab, and was followed by high-count images of the SLN. Eighty-five cases with breast cancer underwent sentinel lymph node biopsy (SLNB) directed by vital blue dye mapping and the gamma probe. All SLNs were examined by H&E staining and immunohistochemistry method. Results: 99mTc-IT-rituximab was proved to have the similar immunological character with rituximab by SDS-PAGE, ELISA and FCA. 99mTc-IT-rituximab was free of bacteria, pyogen free and toxicity. 99mTc-IT-rituximab was mainly excreted by kidney. High kidney uptake was found for 99mTc-IT-rituximab at 1 h, its uptake at each time point was (14.01±0.61)%/g, (9.81±0.61) %/g, (5.64±0.77) %/g and (3.51±0.48) %/g. Other tissues exhibited less than 1 percent of the injected dose at all time points. In ten patients with breast cancer, the results of dynamic SLN mapping show that SLN could be seen within a wide range from 30 min to 24 h postinjection 99mTc-IT-rituximab. The higher order nodes had not been seen all the time. Draining lymphatic basins and nodes were imaged by lymphoscintigraphy in 82 cases of eighty-five. The SLN was identified in 96.5 % (82/85) of patients when both blue dye and intraoperative gamma probe were used. Thirty cases of SLN (36.6%) were metastasis positive, including 24 positive cases by H&E staining and 6 by immunohistochemistry method. SLN was the only metastasis lymph node in 18 out of 30 cases. One case has negative SLN metastasis. The sensitivity and accuracy of SLNB were 96.8% (30/31) and 98.8% (81/82). The specificity was 100% (51/51). The false negative rate was 3.3 (1/31) and the negative predictive value was 98.1 (51/52). The positive predictive value was 100% (31/31). Conclusions: 99mTc-IT-Rituximab appears to be potential and useful for clinical SLN study.

Abstract:To develop a new imaging agent, (99m)Tc-Rituximab, for sentinel lymphoscinti-graphy and investigate its feasibility in detecting the sentinel lymph node (SLN) for breast cancer patient.(99m)Tc-Rituximab was prepared by introducing (99m)Tc directly into the mclecule of Rituximab, thus a new kind of lymphoscintigraphy agent ((99m)Tc-Rituximab) was produced. Five Bal b/c mice and ten breast cancer patients underwent dynamic SLN mapping procedures using (99m)Tc-Rituximab. Axillary lymph node status in breast cancer patients were evaluated with the fusion images, which was processed by computer software. With ROI technique, SLN/background ratio and the rate of SLN uptake were calculated and the dynamic curve of SLN uptake was drawn. The other 61 breast carcinoma patients underwent routine lymphatic mapping. Sentinel lymph node biopsy (SLNB) and axillary lymph node dissection (ALND) were both performed in all 71 patients. Fresh SLNs and all axillary nodes were sent for pathological examination using hematoxylin and eosin (H&E) staining.(99m)Tc-Rituximab, with a yield of about 95.0%, can show SLN status in both mice and patients. The fusion image showed SLNs clearly, even the depth of SLNs in the axilla. The results of dynamic SLN mapping showed visible SLN starting from 10 min, up to 16 h after injecting tracer, however, secondary lymph nodes were not detectable at all. In mice, the ratio of SLN/background was 2.1, the rate of uptake was about 3.0% to 3.5%, and the equation of the curve was produced as: y = 0.036x + 2.7875, r(2) = 0.9787. For breast cancer patients, the uptake rate was different from case to case, but the cure seemed to be consistent with logarithm. The result of SLNB and ALND showed that the sensitivity and accuracy of SLNB was both 100.0%. The average number of SLNs found in each case was 2.6. Of nineteen breast cancer patients (26.8%) with lymph node metastasis, 11 were found to only have SLN metastasis.(99m)Tc-Rituximab, as a new tracer to mapping sentinel lymph node for breast cancer patient, is able to show the status of sentinel lymph node reliably. The fusing image is easy to obtain and be interpreted by the clinician.
Sentinel Lymph Node Biopsy with a Novel Sentinel Lymphoscintigraphy Agent 99Mtc-Rituximab for Breast Cancer

Abstract:Objective To study the feasibility and reliability of the sentinel lymph node(SLN)imaging and biopsy with a new imaging agent 99mTc-Rituximab in 102 breast cancer patients(one suffered bilateral breast cancers).Methods The sentinel lymph node imaging was taken after peritumoral and subdermal injection of 99mTc-Rituximab guided with ultrasound,and was fused with each digital photo.After the sentinel lymph node biopsy(SLNB),routine surgery of breast and axillary lymph node dissection(ALND)was performed.Fresh SLNs and all axillary nodes were examined by pathology.Results There were total 103 times SLN imaging and SLNB in 102 patients,and the success rate of the SLN imaging was 100%.Total 177 SLNs(the average was 1.72)were showed in the images,which distributed in the axilla,the internal mammary and the breast region.The success rate of SLNB was 99.03%,and total 266 SLNs(the average was 2.58)were identified.Pathological examination showed that there were 76 times of metastatic SLN and the total number of metastatic SLN were 110.The sensitivity,specificity and accuracy of SLNB were 96.20%,100% and 97.09%,respectively;and the false negative rate of SLNB was 3.80%.Conclusion Using 99mTc-Rituximab as the imaging agent,the sentinel lymph node imaging and biopsy can obtain high success rate,high sensitivity and accuracy,and low false negative rate,so this novel sentinel lymphoscintigraphy agent will have a good future in clinical practice.
The Efficacy of 99Mtc-Rituximab As a Tracer for Sentinel Lymph Node Biopsy in Cutaneous Melanoma Patients

Abstract:Background: The sentinel lymph node (SLN) status is a vital prognostic factor for malignant melanoma (MM) patients. There is increasing evidence that a radioactive agent, rather than its combination with blue dye, is sufficient for a SLN biopsy (SLNB). Thus, we discussed the efficacy of Tc-99m-rituximab as a tracer in MM patients. Methods: A total of 502 consecutive patients with MM who underwent SLNB were enrolled in this study. All participants were peritumorally injected with Tc-99m-rituximab before imaging, and scanned with singlephoton emission computed tomography-computed tomography (SPECT-CT) to detect the number and location of the SLN. A gamma detection probe was employed to detect radioactive SLNs in operation. Follow up was conducted to observe whether nodal or distant recurrence occurred. Results: The SLNs were successfully imaged via SPECT-CT and harvested from all 502 participants. No drainage tube was indwelled and 32 (6.3%) participants experienced the following complications: seroma (n=26, 5.2%), wound infections or lymphangitis (n=6, 1.2%), sensory nerve injuries (n= 4, 0.8%). There were 380 patients who were diagnosed as SLN-negative and 122 (24.2%) were SLN-positive. A total of 85 SLN-positive patients received complete lymph node dissection, and 28 (32.9%) had additional positive lymph nodes. During a median follow-up of 24 months, 28 participants were found to have a false negative (FN) SLN. The FN rate was 18.7%. A higher T stage was a predictive factor for FN [odds ratio (OR) 1.77; P<0.05]. There was no significant difference in the positive or FN rate between the acral and cutaneous groups. Conclusions: The radiopharmaceutical Tc-99m-rituximab could be employed as a simple and safe tracer in acral and cutaneous melanoma SLN biopsies.

But the SLN metastasis rates were different in patients with different pathologic type and clinical stage(x2=14.134, 29.184, all P <0.05). Abstract:To evaluate the efficiency of a novel lymph node radiotracer 99mTc-rituximab in sentinel lymph node (SLN) lymphoscintigraphy and SLN biopsy (SLNB), and the influence of SLNB results on the prognosis of cutaneous malignant melanoma (CMM) patients. A retrospective study was performed on 533 patients with CMM who underwent lymphoscintigraphy and SLNB. All patients received a preoperative peritumoral injection of 11.1–18.5 MBq of 99mTc-rituximab 0.5 to 1 h before lymphoscintigraphy and SLNB. Technetium-99m-Labeled Rituximab For Use As A Specific Tracer Of Sentinel Lymph Node Biopsy: A Translational Research Study

Abstract:Purpose: We aimed to develop and translate a CD20-antigen-targeted radiopharmaceutical, Technetium-99 m-labeled (Tc-99m) rituximab, for sentinel lymph node (SLN) detection.Methods: Tc-99m-rituximab was synthesized and tested for stability in human serum. The binding affinity to CD20 was evaluated in Raji cells by flow cytometric analysis. Biodistribution and sentinel node mapping were carried out in bal b/c mice. Eighty-five patients with breast cancer participated in this study. Dynamic sentinel lymphoscintigraphy was first assessed in 12 patients before planar lymphoscintigraphy was assessed in a larger cohort. All patients underwent sentinel lymph node biopsy (SLNB), followed by axillary lymph node dissection.Results: The cell-binding study showed that 99mTc-rituximab possessed compatible affinity to human CD20. In the mechanism study, Tc-99m-labeled anti-mouse CD20 monoclonal antibodies could bind to mouse CD20 and accumulate in the SLN with 2.62 +/- 1.25 % of the percentage of injected activity, which could be blocked by excessive unlabeled antibody. Low uptake of non-sentinel nodes and fast clearance from the injection site were observed in the mice. Sentinel nodes were identified in 82 of 85 breast cancer patients (96.5%) by lymphoscintigraphy and SLNB. The sensitivity, specificity, and accuracy were 96.8% (30/31), 100% (51/51), and 98.8% (81/82), respectively.Conclusion: Tc-99m-rituximab, specifically binding to CD20, met most of the requirements of an ideal sentinel mapping agent for use in clinical settings.
Sentinel Lymph Node Imaging with a Novel Radiotracer: Preparation of 99Tcm-It-rituximab and Experimental Study

Abstract:Objective To investigate the located ability of a novel sentinel lymphoscintigraphy agent, ~ 99 Tc~m-IT-Rituximab, in detecting the sentinel lymph node (SLN). Methods ~ 99 Tc~m-Rituximab was prepared indirectly via 2-iminothiolane (2-IT) conjugation and labeled with techenium-99m. The locating ability of ~ 99 Tc~m-IT-Rituximab was evaluated with biological distribution results in mice, and the influenced factors on the imaging by the injecting dosage and volume were also observed in mice. Results ~ 99 Tc~m-IT-Rituximab, with a yield of about 90%, were intact. The results of the dynamic SLN mapping showed that SLN could be seen within a wide range from 30 min to 24 h after injecting, and higher order nodes had not been visualized all the time. The results of the biodistribution showed that SLNs could be identified by the imaging agent, ~ 99 Tc~m-IT-Rituximab, successfully, the rate of uptake in 24 h was about 4.49%, while the uptake of higher echelon lymph node was close to background. SLN imaging would be influenced by the dosage and volume of radiotracer, which could both result in the decrease of SLN uptake and the increase of higher order nodes uptake. Conclusion The labeling method is simple and safe. ~ 99 Tc~m-IT-Rituximab appears to be potential and useful for clinical SLN study.

Abstract:1862 Objectives: To prepare 99mTc-IT-rituximab, a novel sentinel lymphoscintigraphy agent, and evaluate its ability in detecting the sentinel lymph node. The influenced factors on the imaging by the injecting dosage and volume were also observed in mice. Results: Optimal conditions for modifying Rituximab involved a 10:1 molar ratio of 2-IT : protein in 4°C, the number of apparent thiol produced were 2.1 per IT-Rituximab. 99mTc-IT-rituximabwas stable for 24 h at 37°C in human serum. 99mTc-IT-rituximab was proved to have the similar immunological character with rituximab by SDS-PAGE, ELISA and FCA. Its binding rate of B lymphoma cell line Raji cell is 69.4%. After subcutaneous injection into the foot pads of mice, 99mTc-IT-rituximab showed high accumulationn the knee. The mean %ID of the popliteal lymph node at 1, 4, 8, and 24 h was (2.33±0.13)%, (2.72±0.12)%, (3.71±0.28)%, and (4.49±0.43)%, respectively. The mean %ID of NSLN was less than 0.2% at each time point. With increasing the injected dose or volume the %ID of SLN would decrease, while the %ID of NSLN would increase gradually. 99mTc-IT-rituximab exhibited faster injection site clearance than did 99mTc-SC and 99mTc-DX, %ID of the rear paws was (22.14±2.08)% at 24 h after administration. In mice, the results of dynamic SLN mapping show that SLN could be seen within a wide range from 30 min to 24 h postinjection. The higher order nodes had not been seen all the time. 99mTc-IT-rituximab was mainly excreted by kidney, other tissues exhibited less than 1 percent of the injected dose at all time points. For radiocolloid, the higher echelon node extraction was significantly higher. The 99mTc-SC and 99mTc-DX %ID of the second-order node was (4.93±0.37) % and (8.54±0.78) % at 2 h. The uptake of second echelon for 99mTc-HSA was also higher than 99mTc-IT-rituximab. Conclusions: 99mTc-IT-Rituximab, as a tracer to mapping SLN, could predict the status of SLNs reliably.

Abstract:Objective To explore the use of 99m Tc-rituximab tracer injection for internal mammary sentinel lymph node (IM-SLN) detection in patients with primary breast cancer. Methods This prospective observational study enrolled female patients with primary breast cancer between September 2017 and June 2022 at Fujian Provincial Hospital. The participants were divided into the peritumoral group (two subcutaneous injection points on the surface of the tumor), two-site group (injections into the glands at 6 and 12 o’clock around the areola area), and four-site group (injections into the gland at 3, 6, 9, and 12 o’clock around the areola area). The outcomes were the detection rates of the IM-SLNs and axillary sentinel lymph nodes (A-SLNs). Results Finally, 133 patients were enrolled, including 53 in the peritumoral group, 60 in the two-site group, and 20 in the four-site group. The detection rate of the IM-SLNs in the peritumoral group (9.4% [5/53]) was significantly lower than in the two-site (61.7% [37/60], P<0.001) and four-site (50.0% [10/20], P<0.001) groups. The detection rates of A-SLNs among the three groups were comparable (P=0.436). Conclusion The two-site or four-site intra-gland injection of 99m Tc-rituximab tracer might achieve a higher detection rate of IM-SLNs and a comparable detection rate of A-SLNs compared with the peritumoral method. The location of the primary focus has no impact on the detection rate of the IM-SLNs.

Abstract:Objective:To investigate the feasibility of detecting the sentinel lymph node(SLN) and its clinical significance in colon cancer.Methods:SLNs of 26 cases with colon cancer were mapped by staining method.The SLNs and all regional lymph nodes were searched in the sample after radical excision,and then examined metastases of lymph nodes by routine hematoxylin and eosin(HE) staining.When the lymph nodes were negative by HE stain,the immunohistchemical staining was used to detect the micrometastases of SLNs and regional lymph nodes.Results: A total of 65(14%) SLNs among all regional lymph nodes were inspected in the 26 cases.Metastases was diagnosed in 9 cases and totally 17 SLNs by HE staining.The metastases rate of SLN was 35% in cases and 26% in SLNs.Metastases rate was 6% in the regional lymph nodes except for the SLNs.The metastases rate was higher in SLNs than in regional lymph nodes(P0.01).Metastases rate was 54% in SLNs by immunohistchemical staining.Conclusion: The SLN mapping of patients with colon cancer can predict the metastases state of the regional lymph nodes and afterwards pathologic examination SLNs may improve the staging system for colon cancer and also assist treatment programs.

Abstract:To evaluate the biological property and safety limitation of a specific sentinel lymphoscintigraphy imaging agent,99Tcm-rituximab,99Tcm-rituximab were analyzed for chemical and radiochemical purity,via SDS-PAGE and ITLC-SG.The immunoreactivity were detected via ELISA and flow cytometry.The safety limitation and biodistribution were tested according to the Chinese Pharmacopeia.The results showed that 99Tcm-rituximab,with a labeling yield of 90%~95%,was intact,and processed similar immunoreactivity of rituximab.The final product was bacteria and pyogen free,and was safe to the body.According to the data of biodistribution,most of agent was excreted by the kiney,and the radio uptake decreased from(14.01±0.61)%ID/g at 1 h to(3.51±0.48)%ID/g at 24 h.The liver also uptook the agent.No accumulation was found in the normal tissue.99Tcm-rituximab is stable and contains a good immunoreacivity and can be effectively used for clinical study.

Abstract:Objective:To evaluate the accuracy and feasibility of sentinel lymph node biopsy (SLNB) marked by 99Tcm-IT-Ritux-imab and to discuss the clinical value of the method in diagnosis and treatment of cutaneous melanoma. Methods:A total of 67 patients with cutaneous malignant melanoma received 99Tcm-IT-Rituximab-tagged SLNB from March 2008 to March 2012. Lymphoscintigra-phy was conducted 30 min to 1 h after intra-dermal injection of 99Tcm-IT-Rituximab. Subsequently, the surgery of SLNB was carried out using gamma probe. The detection and positive rates of SLNB were counted. The relationship between the status and the clinical features of the sentinel lymph node (SLN) was analyzed, such as the T stage, ulceration, age, gender, and location. The influence of SLN status on overall survival (OS) and disease-free survival (DFS) was evaluated. Results:SLNs were detected in all the 67 patients by SPECT and gamma detector, with detection rate of 100%. Fifteen patients had SLN metastasis, and the positive rate was 22.4%. Chi-square indicates that SLN metastasis is associated with age, T stage, and ulceration (P<0.05). A total of 63 patients were followed up for 24-69 months, and the median follow up time was 43 months. Kaplan-Meier survival analysis shows that both OS and DFS in the SLN-negative group are better than those in the SLN-positive group (OS:93.9%vs. 57.1%, P<0.01;DFS:79.6%versus 28.6%, P<0.01). Cox-regression multiple factors analysis suggests that both SLN status and T stage are independent factors that affect the DFS of malignant melanoma. Conclusion:SLNB assisted by 99Tcm-IT-Rituximab can well reflect the state of lymph node metastasis and is es-sential for accurate staging, prognosis judging, and treatment guiding. Its operation procedure is simple with high accuracy, and the im-aging status is stable. Therefore, it is convenient and feasible as a means of SLNB.

Abstract:Objective To investigate labelling method of a novel sentinel lymphoscintigraphy agent ~(99)Tc~m-Rituximab,and to evaluate its ability in detection of sentinel lymph node(SLN).Methods Ritux- imab was prepared indirectly via 2-iminothiolane(2-IT)conjugatinn and labeled with ~(99)Tc~m.The final prod- uct ~(99)Tc~m-IT-Rituximab was analysed for numbers of free sulfhydryl group,chemical and radiochemical puri- ty by sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE)and instant thin-layer chroma- tography-silica gel(ITLC-SG)and also the immuno-reactivity by ELISA method.The localization ability of ~(99)Tc~m-IT-Rituximab and ~(99)Tc~m-sulfur colloid(SC)was compared with biological distribution in mice.Also, 10 cases of breast cancer underwent dynamic SLN mapping.Results Optimal conditions for labelling Rit- uximab was a 10:1 molar ratio of 2-IT:Rituximah in 4 ℃,the nunxber of—SH group per mole of Ritux- imab was 2.1.~(99)Tc~m-IT-Rituximab,with a yield of about 90%,was intact,and it kept full immuno reactiv- ity.The binding rate with B lymphoma cell line Raji cell was 69.4%.Dynamic SLN mapping in mice showed that SLN visualized in 30 min to 24 h after injection,and it could visualize clearly as early as 2 h. The secondary lymph nodes were not seen even at 24 h.Generally,the localization ability of ~(99)Tc~m-IT-Rit- uximab was superior to ~(99)Tc~m-SC.The uptake of ~(99)Tc~m-IT-Riluximab in 24 h was 4.49%,with no uptake in secondary and tertiary lymph nodes.For the clinical result,~(99)Tc~m-IT-Rituximab could identify SLN success- fully in all 10 cases.Conclusions The labelling method is simple with high yield rate.~(99)Tc~m-IT-Ritux- imab,as a tracer to mapping SLN,demonstrate its good localization ability.Therefore it would be a poten- tial novel SLN imaging agent.

Abstract:Objective To investigate the accuracy and feasibility of 99Tcm-rituximab SLNB in cutaneous malignant melanoma (CMM) and its influence on survival.Methods A total of 89 patients (48 males,41 females,average age (51.62±14.35) years) with CMM underwent SLN planar imaging 0.5-1.0 h after intradermal injection of 99Tcm-rituximab around primary lesions from June 2008 to November 2014.Based on the image,the SLNs were identified by a gamma probe and removed.The resected SLNs were examined pathologically with both HE and immunohistochemical staining.The results of SLN imaging and SLNB were analyzed retrospectively,and their OS and DFS were followed up.The relationship between the results and the clinical characteristics (age,gender,primary tumor site,primary tumor thickness,ulceration)were analyzed and the influence of SLN status (with or without metastasis) on OS and DFS was evaluated.Data were analyzed with one-way analysis of variance,two-sample t test,x2 test and Cox regression.Results The success rate of SLN imaging and biopsy was 100％ (89/89).There were 207 SLNs detected by lymphoscintigraphy and 228 SLNs by operation.Thirty-three metastatic SLN in 21 cases were confirmed by pathology.The numbers of SLN detected by lymphoscintigraphy were statistically significant among groups with different primary tumor sites (1.51±1.44,1.38±0.65,2.20±1.79;F=3.660,P＜0.05).SLN numbers detected by SLNB were statistically significant among groups with different primary tumor sites (2.85±1.74,1.54±0.78,1.20±1.10;F=5.504,P＜0.05) and between groups with or without ulceration (2.59±1.96,2.54±1.46;t=2.588,P＜0.05).SLN metastatic ratios were statistically significant only in patients with different primary tumor thickness (x2 =18.544,P＜0.05).Median follow-up time was 31 months.The rate of OS was 95.51％ (85/89),and the rate of DFS was 77.53％ (68/89).Univariate analysis found that DFS was affected by SLN status (with or without metastasis) and primary tumor thickness,but no statistical significance was found in Cox-regression.Conclusion 99Tcm-rituximab may be a reliable tracer in SLNB and a helpful tool for staging and prognosis of malignant melanoma.
