Abstract:Abstract The realization of automated chemical experiments by robots unveiled the prelude to an artificial intelligence (AI) laboratory. Several AI-based systems or robots with specific chemical skills have been demonstrated, but conducting all-round scientific research remains challenging. Here, we present an all-round AI-Chemist equipped with scientific data intelligence that is capable of performing basic tasks generally required in chemical research. Based on a service platform, the AI-Chemist is able to automatically read the literatures from a cloud database and propose experimental plans accordingly. It can control a mobile robot in-house or online to automatically execute the complete experimental process on 14 workstations, including synthesis, characterization and performance tests. The experimental data can be simultaneously analysed by the computational brain of the AI-Chemist through machine learning and Bayesian optimization, allowing a new hypothesis for the next iteration to be proposed. The competence of the AI-Chemist has been scrutinized by three different chemical tasks. In the future, the more advanced all-round AI-Chemists equipped with scientific data intelligence may cause changes to the landscape of the chemical laboratory.

Abstract:Recent years witness rapid progress made in artificial intelligent (AI), data-driven chemistry research. An automated experimental platform with functions of synthesis, characterization and testing could transform a synthesis plan into characterization and testing data of real products, which can then be used to re-optimize synthesis, thus close the loop of materials design. Herein, we reported the design, framework and hardware/software systems of a robotic AI-chemist platform that integrates chemical synthesis, sample characterization and performance testing functions. The robotic AI-chemist was instructed to carry out photocatalysis experiments, which demonstrates its utility to replace a human chemist in real experimental operations.

Abstract:The successful integration of large language models (LLMs) into laboratory workflows has demonstrated robust capabilities in natural language processing, autonomous task execution, and collaborative problem-solving. This offers an exciting opportunity to realize the dream of autonomous chemical research on demand. Here, we report a robotic AI chemist powered by a hierarchical multi-agent system, ChemAgents, based on an on-board Llama-3-70B LLM, capable of executing complex, multi-step experiments with minimal human intervention. It operates through a Task Manager agent that interacts with human researchers and coordinates four role-specific agents—Literature Reader, Experiment Designer, Computation Performer, and Robot Operator—each leveraging one of four foundational resources: a comprehensive Literature Database, an extensive Protocol Library, a versatile Model Library, and a state-of-the-art Automated Lab. We demonstrate its versatility and efficacy through six experimental tasks of varying complexity, ranging from straightforward synthesis and characterization to more complex exploration and screening of experimental parameters, culminating in the discovery and optimization of functional materials. Our multi-agent-driven robotic AI chemist showcases the potential of on-demand autonomous chemical research to drive unprecedented efficiencies, accelerate discovery, and democratize access to advanced experimental capabilities across academic disciplines and industries.

Abstract:Recent AI research plots a promising future of automatic chemical reactions within the chemistry society. This study proposes Chemist-X, a transformative AI agent that automates the reaction condition recommendation (RCR) task in chemical synthesis with retrieval-augmented generation (RAG) technology. To emulate expert chemists' strategies when solving RCR tasks, Chemist-X utilizes advanced RAG schemes to interrogate online molecular databases and distill critical data from the latest literature database. Further, the agent leverages state-of-the-art computer-aided design (CAD) tools with a large language model (LLM) supervised programming interface. With the ability to utilize updated chemical knowledge and CAD tools, our agent significantly outperforms conventional synthesis AIs confined to the fixed knowledge within its training data. Chemist-X considerably reduces chemists' workload and allows them to focus on more fundamental and creative problems, thereby bringing closer computational techniques and chemical research and making a remarkable leap toward harnessing AI's full capabilities in scientific discovery.

Abstract:Large language models (LLMs) have demonstrated outstanding capabilities in general problem-solving and been shown to improve productivity in certain domains. Thanks to their flexibility, recent work has leveraged them for diverse scientific applications, ranging from predictive modeling, scientific Q&A, and even as autonomous agents towards automation in chemistry. The democratization of high-quality chemistry education faces several challenges, including heterogeneity among sub-fields, limited access to personalized guidance, and an uneven distribution of resources. Additionally, hands-on laboratory experiments, a crucial component of chemistry education, are difficult to scale due to inherent safety risks that necessitate close supervision. We propose that LLMs can help overcome these obstacles by providing scalable solutions that tailor educational content to individual needs, enhancing the overall learning experience. In this perspective, we discuss how LLMs can catalyze chemistry education across multiple dimensions, from preparing and delivering lectures and tackling guidance in both wet lab and computational experiments, to re-thinking evaluation methodologies in the classroom. We also discuss some potential risks of this technology, such as the possibility of generating inaccurate or biased content, and emphasize the need for further development to ensure the successful integration of LLMs in the chemistry classroom.

ChatGPT Chemistry Assistant for Text Mining and Prediction of MOF Synthesis

Abstract:We use prompt engineering to guide ChatGPT in the automation of text mining of metal-organic frameworks (MOFs) synthesis conditions from diverse formats and styles of the scientific literature. This effectively mitigates ChatGPT's tendency to hallucinate information -- an issue that previously made the use of Large Language Models (LLMs) in scientific fields challenging. Our approach involves the development of a workflow implementing three different processes for text mining, programmed by ChatGPT itself. All of them enable parsing, searching, filtering, classification, summarization, and data unification with different tradeoffs between labor, speed, and accuracy. We deploy this system to extract 26,257 distinct synthesis parameters pertaining to approximately 800 MOFs sourced from peer-reviewed research articles. This process incorporates our ChemPrompt Engineering strategy to instruct ChatGPT in text mining, resulting in impressive precision, recall, and F1 scores of 90-99%. Furthermore, with the dataset built by text mining, we constructed a machine-learning model with over 86% accuracy in predicting MOF experimental crystallization outcomes and preliminarily identifying important factors in MOF crystallization. We also developed a reliable data-grounded MOF chatbot to answer questions on chemical reactions and synthesis procedures. Given that the process of using ChatGPT reliably mines and tabulates diverse MOF synthesis information in a unified format, while using only narrative language requiring no coding expertise, we anticipate that our ChatGPT Chemistry Assistant will be very useful across various other chemistry sub-disciplines.

ChemSafetyBench: Benchmarking LLM Safety on Chemistry Domain

Abstract:The advancement and extensive application of large language models (LLMs) have been remarkable, including their use in scientific research assistance. However, these models often generate scientifically incorrect or unsafe responses, and in some cases, they may encourage users to engage in dangerous behavior. To address this issue in the field of chemistry, we introduce ChemSafetyBench, a benchmark designed to evaluate the accuracy and safety of LLM responses. ChemSafetyBench encompasses three key tasks: querying chemical properties, assessing the legality of chemical uses, and describing synthesis methods, each requiring increasingly deeper chemical knowledge. Our dataset has more than 30K samples across various chemical materials. We incorporate handcrafted templates and advanced jailbreaking scenarios to enhance task diversity. Our automated evaluation framework thoroughly assesses the safety, accuracy, and appropriateness of LLM responses. Extensive experiments with state-of-the-art LLMs reveal notable strengths and critical vulnerabilities, underscoring the need for robust safety measures. ChemSafetyBench aims to be a pivotal tool in developing safer AI technologies in chemistry. Chemistry3D: Robotic Interaction Benchmark for Chemistry Experiments

Abstract:The advent of simulation engines has revolutionized learning and operational efficiency for robots, offering cost-effective and swift pipelines. However, the lack of a universal simulation platform tailored for chemical scenarios impedes progress in robotic manipulation and visualization of reaction processes. Addressing this void, we present Chemistry3D, an innovative toolkit that integrates extensive chemical and robotic knowledge. Chemistry3D not only enables robots to perform chemical experiments but also provides real-time visualization of temperature, color, and pH changes during reactions. Built on the NVIDIA Omniverse platform, Chemistry3D offers interfaces for robot operation, visual inspection, and liquid flow control, facilitating the simulation of special objects such as liquids and transparent entities. Leveraging this toolkit, we have devised RL tasks, object detection, and robot operation scenarios. Additionally, to discern disparities between the rendering engine and the real world, we conducted transparent object detection experiments using Sim2Real, validating the toolkit's exceptional simulation performance. Abstract:This work utilizes collected and organized instructional data from the field of chemical science to fine-tune mainstream open-source large language models. To objectively evaluate the performance of the fine-tuned models, we have developed an automated scoring system specifically for the chemistry domain, ensuring the accuracy and reliability of the evaluation results. Building on this foundation, we have designed an innovative chemical intelligent assistant system. This system employs the fine-tuned Mistral Nemo model as one of its primary models and features a mechanism for flexibly invoking various advanced models. This design fully considers the rapid iteration characteristics of large language models, ensuring that the system can continuously leverage the latest and most powerful AI capabilities.A major highlight of this system is its deep integration of professional knowledge and requirements from the chemistry field. By incorporating specialized functions such as molecular visualization, SMILES string processing, and chemical literature retrieval, the system significantly enhances its practical value in chemical research and applications. More notably, the system possesses autonomous evolution capabilities. Through carefully designed mechanisms for knowledge accumulation, skill acquisition, performance evaluation, and group collaboration, the system can continuously optimize its professional abilities and interaction quality. This dynamic adaptive feature enables the system to evolve autonomously, breaking through the inherent static limitations of traditional AI systems.

Abstract:Many of the greatest challenges facing society today likely have molecular solutions that await discovery. However, the process of identifying and manufacturing such molecules has remained slow and highly specialist dependent. Interfacing the fields of artificial intelligence (AI) and synthetic organic chemistry has the potential to powerfully address both limitations. The Molecule Maker Lab Institute (MMLI) brings together a team of chemists, engineers, and AI‐experts from the University of Illinois Urbana‐Champaign (UIUC), Pennsylvania State University, and the Rochester Institute of Technology, with the goal of accelerating the discovery, synthesis and manufacture of complex organic molecules. Advanced AI and machine learning (ML) methods are deployed in four key thrusts: (1) AI‐enabled synthesis planning, (2) AI‐enabled catalyst development, (3) AI‐enabled molecule manufacturing, and (4) AI‐enabled molecule discovery. The MMLI's new AI‐enabled synthesis platform integrates chemical and enzymatic catalysis with literature mining and ML to predict the best way to make new molecules with desirable biological and material properties. The MMLI is transforming chemical synthesis and generating use‐inspired AI advances. Simultaneously, the MMLI is also acting as a training ground for the next generation of scientists with combined expertise in chemistry and AI. Outreach efforts aimed toward high school students and the public are being used to show how AI‐enabled tools can help to make chemical synthesis accessible to nonexperts.

Abstract:Machine learning (ML) is currently transforming the field of chemistry by offering unparalleled efficiency in addressing complex challenges. Despite the progress made, a notable gap persists in the availability of user-friendly tools tailored to chemical problems involving small and sparse datasets. Here, we introduce PythiaCHEM, an ML toolkit designed to develop data-driven predictive ML models. It enables the use of various descriptors and ML frameworks for regression and classification tasks in an automated, flexible, and accessible manner through Jupyter Notebooks, making it easy to customize for specific tasks. We showcase the capabilities and versatility of PythiaCHEM in two distinct chemistry tasks: first, the evaluation of the transmembrane chloride anion transport activity of synthetic anion transporters, and second, the prediction of enantioselectivity in the Strecker synthesis of a-amino acids. Our results highlight the utility of PythiaCHEM as a powerful and user-friendly framework for developing predictive ML models applicable in different domains of chemistry.

ORGANA: A Robotic Assistant for Automated Chemistry Experimentation and Characterization

Abstract:Chemistry experimentation is often resource- and labor-intensive. Despite the many benefits incurred by the integration of advanced and special-purpose lab equipment, many aspects of experimentation are still manually conducted by chemists, for example, polishing an electrode in electrochemistry experiments. Traditional lab automation infrastructure faces challenges when it comes to flexibly adapting to new chemistry experiments. To address this issue, we propose a human-friendly and flexible robotic system, ORGANA, that automates a diverse set of chemistry experiments. It is capable of interacting with chemists in the lab through natural language, using Large Language Models (LLMs). ORGANA keeps scientists informed by providing timely reports that incorporate statistical analyses. Additionally, it actively engages with users when necessary for disambiguation or troubleshooting. ORGANA can reason over user input to derive experiment goals, and plan long sequences of both high-level tasks and low-level robot actions while using feedback from the visual perception of the environment. It also supports scheduling and parallel execution for experiments that require resource allocation and coordination between multiple robots and experiment stations. We show that ORGANA successfully conducts a diverse set of chemistry experiments, including solubility assessment, pH measurement, recrystallization, and electrochemistry experiments. For the latter, we show that ORGANA robustly executes a long-horizon plan, comprising 19 steps executed in parallel, to characterize the electrochemical properties of quinone derivatives, a class of molecules used in rechargeable flow batteries. Abstract:Motivation: Natural language is poised to become a key medium for human-machine interactions in the era of large language models. In the field of biochemistry, tasks such as property prediction and molecule mining are critically important yet technically challenging. Bridging molecular expressions in natural language and chemical language can significantly enhance the interpretability and ease of these tasks. Moreover, it can integrate chemical knowledge from various sources, leading to a deeper understanding of molecules. Results: Recognizing these advantages, we introduce the concept of conversational molecular design, a novel task that utilizes natural language to describe and edit target molecules. To better accomplish this task, we develop ChatMol, a knowledgeable and versatile generative pre-trained model. This model is enhanced by incorporating experimental property information, molecular spatial knowledge, and the associations between natural and chemical languages. Several typical solutions including large language models (e.g., ChatGPT) are evaluated, proving the challenge of conversational molecular design and the effectiveness of our knowledge enhancement approach. Case observations and analysis offer insights and directions for further exploration of natural-language interaction in molecular discovery. Abstract:We leveraged the power of ChatGPT and Bayesian optimization in the development of a multi-AI-driven system, backed by seven large language model-based assistants and equipped with machine learning algorithms, that seamlessly orchestrates a multitude of research aspects in a chemistry laboratory (termed the ChatGPT Research Group). Our approach accelerated the discovery of optimal microwave synthesis conditions, enhancing the crystallinity of MOF-321, MOF-322, and COF-323 and achieving the desired porosity and water capacity. In this system, human researchers gained assistance from these diverse AI collaborators, each with a unique role within the laboratory environment, spanning strategy planning, literature search, coding, robotic operation, labware design, safety inspection, and data analysis. Such a comprehensive approach enables a single researcher working in concert with AI to achieve productivity levels analogous to those of an entire traditional scientific team. Furthermore, by reducing human biases in screening experimental conditions and deftly balancing the exploration and exploitation of synthesis parameters, our Bayesian search approach precisely zeroed in on optimal synthesis conditions from a pool of 6 million within a significantly shortened time scale. This work serves as a compelling proof of concept for an AI-driven revolution in the chemistry laboratory, painting a future where AI becomes an efficient collaborator, liberating us from routine tasks to focus on pushing the boundaries of innovation.

Abstract:Conversational agents are a recent trend in human-computer interaction, deployed in multidisciplinary applications to assist the users. In this paper, we introduce "Atreya", an interactive bot for chemistry enthusiasts, researchers, and students to study the ChEMBL database. Atreya is hosted by Telegram, a popular cloud-based instant messaging application. This user-friendly bot queries the ChEMBL database, retrieves the drug details for a particular disease, targets associated with that drug, etc. This paper explores the potential of using a conversational agent to assist chemistry students and chemical scientist in complex information seeking process.

Abstract:Artificial intelligence (AI) has played an increasingly important role in chemical research. However, most models currently used in chemistry are specialist models that require training and tuning for specific tasks. A more generic and efficient solution would be an AI model that could address many tasks and support free-form dialogue in the broad field of chemistry. In its utmost form, such a generalist AI chemist could be referred to as Chemical General Intelligence. Large language models (LLMs) have recently logged tremendous success in the general domain of natural language processing, showing emerging task generalization and free-form dialogue capabilities. However, domain knowledge of chemistry is largely missing when training general-domain LLMs. The lack of such knowledge greatly hinders the performance of generalist LLMs in the field of chemistry. To this end, we develop ChemDFM, a pioneering LLM for chemistry trained on 34B tokens from chemical literature and textbooks, and fine-tuned using 2.7M instructions. As a result, it can understand and reason with chemical knowledge in free-form dialogue. Quantitative evaluations show that ChemDFM significantly surpasses most representative open-source LLMs. It outperforms GPT-4 on a great portion of chemical tasks, despite the substantial size difference. Integrating Machine Learning and Color Chemistry: Developing a High-School Curriculum toward Real-World Problem-Solving

Abstract:Artificial intelligence (AI) is rapidly transforming our world, making it imperative to educate the next generation about both the potential benefits and challenges associated with AI. This study presents a cross-disciplinary curriculum that connects AI and chemistry disciplines in the high school classroom. Particularly, we leverage machine learning (ML), an important and simple application of AI to instruct students to build an ML-based virtual pH meter for high-precision pH read-outs. We used a "codeless" and free ML neural network building software - Orange, along with a simple chemical topic of pH to show the connection between AI and chemistry for high-schoolers who might have rudimentary backgrounds in both disciplines. The goal of this curriculum is to promote student interest and drive in the analytical chemistry domain and offer insights into how the interconnection between chemistry and ML can benefit high-school students in science learning. The activity involves students using pH strips to measure the pH of various solutions with local relevancy and then building an ML neural network model to predict the pH value based on color changes of pH strips. The integrated curriculum increased student interest in chemistry and ML and demonstrated the relevance of science to their daily lives and global issues. This approach is transformative in developing a broad spectrum of integration topics between chemistry and ML and understanding their global impacts.
Structured Chemistry Reasoning with Large Language Models

Abstract:Large Language Models (LLMs) excel in diverse areas, yet struggle with complex scientific reasoning, especially in the field of chemistry. Different from the simple chemistry tasks (e.g., molecule classification) addressed in previous studies, complex chemistry problems require not only vast knowledge and precise calculation, but also compositional reasoning about rich dynamic interactions of different concepts (e.g., temperature changes). Our study shows that even advanced LLMs, like GPT-4, can fail easily in different ways. Interestingly, the errors often stem not from a lack of domain knowledge within the LLMs, but rather from the absence of an effective reasoning structure that guides the LLMs to elicit the right knowledge, incorporate the knowledge in step-by-step reasoning, and iteratively refine results for further improved quality. On this basis, we introduce StructChem, a simple yet effective prompting strategy that offers the desired guidance and substantially boosts the LLMs' chemical reasoning capability. Testing across four chemistry areas -- quantum chemistry, mechanics, physical chemistry, and kinetics -- StructChem substantially enhances GPT-4's performance, with up to 30\% peak improvement. Our analysis also underscores the unique difficulties of precise grounded reasoning in science with LLMs, highlighting a need for more research in this area. Abstract:The field of natural language processing (NLP) has witnessed a transformative shift with the emergence of large language models (LLMs), revolutionizing various language tasks and applications, and the integration of LLM into specialized domains enhances their capabilities for domain-specific applications. Notably, NLP has made significant strides in organic chemistry, particularly in predicting synthetic tasks, paving the way for the development of LLMs tailored to the organic chemistry field. In this work, we introduce SynAsk, a comprehensive organic chemistry domain-specific LLM platform developed by AIChemEco Inc. By finetuning an LLM with domain-specific data and integrating it with a chain of thought approach, SynAsk seamlessly accesses our knowledge base and advanced chemistry tools in a question-and-answer format. This includes functionalities such as a basic chemistry knowledge base, molecular information retrieval, reaction performance prediction, retrosynthesis prediction, chemical literature acquisition, and more. This novel methodology synergizes fine-tuning techniques with external resource integration, resulting in an organic chemistry-specific model poised to facilitate research and discovery in the field. Abstract:The new era with prosperous artificial intelligence (AI) and robotics technology is reshaping the materials discovery process in a more radical fashion. Here we present authentic intelligent robotics for chemistry (AIR-Chem), integrated with technological innovations in the AI and robotics fields, functionalized with modules including gradient descent-based optimization frameworks, multiple external field modulations, a real-time computer vision (CV) system, and automated guided vehicle (AGV) parts. AIR-Chem is portable and remotely controllable by cloud computing. AIR-Chem can learn the parametric procedures for given targets and carry on laboratory operations in standalone mode, with high reproducibility, precision, and availability for knowledge regeneration. Moreover, an improved nucleation theory of size focusing on inorganic perovskite quantum dots (IPQDs) is theoretically proposed and experimentally testified to by AIR-Chem. This work aims to boost the process of an unmanned chemistry laboratory from the synthesis of chemical materials to the analysis of physical chemical properties, and it provides a vivid demonstration for future chemistry reshaped by AI and robotics technology.
