Abstract:Aquaporins are channel proteins which transport water across cell membranes. We show that the bread wheat aquaporin gene TaTIP2;2 maps to the long arm of chromosome 7b and that its product localizes to the endomembrane system. The gene is expressed constitutively in both the root and the leaf, and is down-regulated by salinity and drought stress. Salinity stress induced an increased level of C-methylation within the CNG trinucleotides in the TaTIP2;2 promoter region. The heterologous expression of TaTIP2;2 in Arabidopsis thaliana compromised its drought and salinity tolerance, suggesting that TaTIP2;2 may be a negative regulator of abiotic stress. The proline content of transgenic A. thaliana plants fell, consistent with the down-regulation of P5CS1, while the expression of SOS1, SOS2, SOS3, CBF3 and DREB2A, which are all stress tolerance-related genes acting in an ABA-independent fashion, was also down-regulated. The supply of exogenous ABA had little effect either on TaTIP2;2 expression in wheat or on the phenotype of transgenic A. thaliana. The expression level of the ABA signalling genes ABI1, ABI2 and ABF3 remained unaltered in the transgenic A. thaliana plants. Thus TaTIP2;2 probably regulates the response to stress via an ABA-independent pathway(s).
Proteomic analysis reveals the molecular mechanism underlying the cold acclimation and freezing tolerance of wheat (Triticum aestivum L.)

Abstract:Cold acclimation (CA) is an important evolutionary adaptive mechanism for wheat freezing resistence. To clarify the molecular basis of wheat CA and freezing tolerance, the effects of CA (4 °C) and non-CA (20 °C) treatments and freezing stress (-5 °C) on the proteins in the wheat crown were characterized via an iTRAQ-based proteomic analysis. A total of 669 differentially accumulated proteins (DAPs) were identified after the CA, of which seven were also DAPs in the CA plants exposed to freezing stress. Additionally, the 15 DAPs in the CA group and the 23 DAPs in the non-CA group after the freezing treatment differed substantially. Functional analyses indicated that CA enhanced freezing tolerance by regulating proteins involved in signal transduction, carbohydrate metabolism, stress and defense responses, and phenylpropanoid biosynthesis. An integrated transcriptomic, proteomic, and metabolomic analysis revealed significant changes in various components of the glutathione metabolic pathway. The overexpression and silencing of Wdhn13 in Arabidopsis and wheat resulted in increased tolerance and sensitivity to freezing stress, respectively, suggesting Wdhn13 promotes freezing tolerance. Overall, our study offers insights into the regulatory network underlying the CA and freezing tolerance of wheat, which may be useful for elucidating wheat freezing resistance.

The Wheat Aquaporin Gene TaAQP7 Confers Tolerance to Cold Stress in Transgenic Tobacco.

Abstract:Aquaporin proteins (AQPs) have been shown to be involved in abiotic stress responses. However, the precise role of AQPs, especially in response to cold stress, is not understood in wheat (Triticum aestivum). In the present study, quantitative real time polymerase chain reaction (qRT-PCR) analysis revealed that TaAQP7 expression increased in leaves, but decreased in roots after cold treatment. Expression of TaAQP7 in tobacco plants resulted in increased root elongation and better growth compared with wild-type (WT) plants under cold stress. Moreover, after cold treatment, the transgenic tobacco lines exhibited higher chlorophyll contents, lower levels of malondialdehyde (MDA), and less ion leakage (IL) than WT plants. Thus, expression of TaAQP7 enhanced cold stress tolerance in transgenic tobacco. Taken together, our results suggest that TaAQP7 confers cold stress tolerance by relieving membrane damage in the transgenic plants.
Identification of genetic loci and candidate genes underlying freezing tolerance in wheat seedlings

Abstract:Ten stable loci for freezing tolerance (FT) in wheat were detected by genome-wide association analysis. The putative candidate gene TaRPM1-7BL underlying the major locus QFT.ahau-7B.2 was identified and validated. Frost damage restricts wheat growth, development, and geographical distribution. However, the genetic mechanism of freezing tolerance (FT) remains unclear. Here, we evaluated FT phenotypes of 245 wheat varieties and lines, and genotyped them using a Wheat 90 K array. The association analysis showed that ten stable loci were significantly associated with FT (P < 1 × 10-4), and explained 6.45-26.33% of the phenotypic variation. In particular, the major locus QFT.ahau-7B.2 was consistently related to all nine sets of FT phenotypic data. Based on five cleaved amplified polymorphic sequence (CAPS) markers closely linked to QFT.ahau-7B.2, we narrowed down the target region to the 570.67-571.16 Mb interval (0.49 Mb) on chromosome 7B, in which four candidate genes were annotated. Of these, only TaRPM1-7BL exhibited consistent differential expression after low temperature treatment between freezing-tolerant and freezing-sensitive varieties. The results of cloning and whole-exome capture sequencing indicated that there were two main haplotypes for TaRPM1-7BL, including freezing-tolerant Hap1 and freezing-sensitive Hap2. Based on the representative SNP (+1956, A/G), leading to an amino acid change in the NBS domain, a CAPS marker (CAPS-TaRPM1-7BL) was developed and validated in 431 wheat varieties (including the above 245 materials) and 318 F2 lines derived from the cross of 'Annong 9267' (freezing-tolerant) × 'Yumai 9' (freezing-sensitive). Subsequently, the TaRPM1-7BL gene was silenced in 'Yumai 9' by virus-induced gene silencing (VIGS), and these silenced wheat seedlings exhibited enhanced FT phenotypes, suggesting that TaRPM1-7BL negatively regulates FT. These findings are valuable for understanding the complex genetic basis of FT in wheat.
Quantitative trait loci and candidate genes associated with freezing tolerance of winter triticale (× Triticosecale Wittmack)

Abstract:Freezing tolerance of triticale is a major trait contributing to its winter hardiness. The identification of genomic regions - quantitative trait loci (QTL) and molecular markers associated with freezing tolerance in winter hexaploid triticale - was the aim of this study. For that purpose, a new genetic linkage map was developed for the population of 92 doubled haploid lines derived from 'Hewo' × 'Magnat' F1 hybrid. Those lines, together with parents were subjected to freezing tolerance test three times during two winter seasons. Plants were grown and cold-hardened under natural fall/winter conditions and then subjected to freezing in controlled conditions. Freezing tolerance was assessed as the plants recovery (REC), the electrolyte leakage (EL) from leaves and chlorophyll fluorescence parameters (JIP) after freezing. Three consistent QTL for several fluorescence parameters, electrolyte leakage, and the percentage of the survived plants were identified with composite interval mapping (CIM) and single marker analysis (SMA). The first locus Qfr.hm-7A.1 explained 9% of variation of both electrolyte leakage and plants recovery after freezing. Two QTL explaining up to 12% of variation in plants recovery and shared by selected chlorophyll fluorescence parameters were found on 4R and 5R chromosomes. Finally, main locus Qchl.hm-5A.1 was detected for chlorophyll fluorescence parameters that explained up to 19.6% of phenotypic variation. The co-located QTL on chromosomes 7A.1, 4R and 5R, clearly indicated physiological and genetic relationship of the plant survival after freezing with the ability to maintain optimal photochemical activity of the photosystem II and preservation of the cell membranes integrity. The genes located in silico within the identified QTL include those encoding BTR1-like protein, transmembrane helix proteins like potassium channel, and phosphoric ester hydrolase involved in response to osmotic stress as well as proteins involved in the regulation of the gene expression, chloroplast RNA processing, and pyrimidine salvage pathway. Additionally, our results confirm that the JIP test is a valuable tool to evaluate freezing tolerance of triticale under unstable winter environments.
Genomic Regions Associated with Tolerance to Freezing Stress and Snow Mold in Winter Wheat

Abstract:Plants grown through the winter are subject to selective pressures that vary with each year's unique conditions, necessitating tolerance of numerous abiotic and biotic stress factors. The objective of this study was to identify molecular markers in winter wheat (Triticum aestivum L.) associated with tolerance of two of these stresses, freezing temperatures and snow mold-a fungal disease complex active under snow cover. A population of 155 F2:5 recombinant inbred lines from a cross between soft white wheat cultivars "Finch" and "Eltan" was evaluated for snow mold tolerance in the field, and for freezing tolerance under controlled conditions. A total of 663 molecular markers was used to construct a genetic linkage map and identify marker-trait associations. One quantitative trait locus (QTL) associated with both freezing and snow mold tolerance was identified on chromosome 5A. A second, distinct, QTL associated with freezing tolerance also was found on 5A, and a third on 4B. A second QTL associated with snow mold tolerance was identified on chromosome 6B. The QTL on 5A associated with both traits was closely linked with the Fr-A2 (Frost-Resistance A2) locus; its significant association with both traits may have resulted from pleiotropic effects, or from greater low temperature tolerance enabling the plants to better defend against snow mold pathogens. The QTL on 4B associated with freezing tolerance, and the QTL on 6B associated with snow mold tolerance have not been reported previously, and may be useful in the identification of sources of tolerance for these traits.
Genome-Wide Identification and Analysis of bZIP Gene Family and Resistance of TaABI5 (TabZIP96) under Freezing Stress in Wheat (Triticum aestivum)

Abstract:The basic leucine zipper (bZIP) regulates plant growth and responds to stress as a key transcription factor of the Abscisic acid (ABA) signaling pathway. In this study, TabZIP genes were identified in wheat and the gene structure, physicochemical properties, cis-acting elements, and gene collinearity were analyzed. RNA-Seq and qRT-PCR analysis showed that ABA and abiotic stress induced most TabZIP genes expression. The ectopic expression of TaABI5 up-regulated the expression of several cold-responsive genes in Arabidopsis. Physiological indexes of seedlings of different lines under freezing stress showed that TaABI5 enhanced the freezing tolerance of plants. Subcellular localization showed that TaABI5 is localized in the nucleus. Furthermore, TaABI5 physically interacted with cold-resistant transcription factor TaICE1 in yeast two-hybrid system. In conclusion, this study identified and analyzed members of the TabZIP gene family in wheat. It proved for the first time that the gene TaABI5 affected the cold tolerance of transgenic plants and was convenient for us to understand the cold resistance molecular mechanism of TaABI5. These results will provide a new inspiration for further study on improving plant abiotic stress resistance.

Abstract:Late spring frost is an important meteorological factor threatening the safe production of winter wheat in China. The young ear is the most vulnerable organ of the wheat plant to spring frost. To gain an insight into the mechanisms underpinning young wheat ears’ tolerance to freezing, we performed a comparative proteome analysis of wheat varieties Xumai33 (XM33, freezing-sensitive) and Jimai22 (JM22, freezing-tolerant) under normal and freezing conditions using label-free quantitative proteomic techniques during the anther connective tissue formation phase (ACFP). Under freezing stress, 392 and 103 differently expressed proteins (DEPs) were identified in the young ears of XM33 and JM22, respectively, and among these, 30 proteins were common in both varieties. A functional characterization analysis revealed that these DEPs were associated with antioxidant capacity, cell wall modification, protein folding, dehydration response, and plant–pathogen interactions. The young ears of JM22 showed significantly higher expression levels of antioxidant enzymes, heat shock proteins, and dehydrin under normal conditions compared to those of XM33, which might help to prepare the young ears of JM22 for freezing stress. Our results lead to new insights into understanding the mechanisms in young wheat ears’ response to freezing stress and provide pivotal potential candidate proteins required for improving young wheat ears’ tolerance to spring frost.

Abstract:Plants that are tolerant of extreme low temperatures, and plants that are tolerant of desiccation, face similar physiological challenges, as low temperatures often result in extracellular ice formation resulting in desiccation of cells. Desiccation due to either low temperatures or water loss leads to severe mechanical as well as oxidative stress, and plants tolerant to either low temperatures or desiccation utilize similar mechanisms to cope with the extreme stress. Key mechanisms to cope with the mechanical stressors associated with cellular crowding include accumulation of both non-reducing sugars as well as LEA proteins. Key mechanisms to cope with oxidative stress include increases in (or the ability to maintain) antioxidant capacity, as well as increased usage of thermal energy dissipation. During both stressors major reductions in photochemical efficiency are observed that correlate with dark retention of the xanthophyll pigments zeaxanthin and antheraxanthin, suggesting sustained forms of thermal energy dissipation persist in these extreme stress scenarios to cope with the increased excitation pressure. The focus of this review is to highlight the shared physiological mechanisms used in both stress scenarios, with a focus on the mechanisms of photoprotection.
Winter Hardiness Physiological Response with Dehydration in Winter Wheat

Abstract:Freezing stress presents a severe threat for winter wheat (Triticum aestivum L.) during overwintering. Dehydration plays a significant role in plant cold-hardiness. In this study, dehydration-related traits were investigated between highly and tender freezing-tolerant winter wheat cultivars during the overwintering period. Accompanied by a decrease in temperature, the water content in crowns of highly freezing-tolerant winter wheat was significantly lower compared to that of tender freezing-tolerant winter wheat (control). The ratio of free and bound water content had slight changes in highly freezing-tolerant winter wheat, though there were wide fluctuations in the control. The transcript levels of dehydration-related genes were more expressed in the highly freezing-tolerant winter wheat than those in the control under freezing stress. The plant growth also showed significant differences between the two winter wheat cultivars. Based on these results, the study proposed that the highly freezing-tolerant winter wheat produces higher expression of dehydrins under freezing stress, causing additional dehydration in the tissue to avoid cell death caused by the formation of ice crystals. Furthermore, winter hardy cultivar also reduced the percentage of free water content that inhibited plant growth, and regulated the water composition for plant survival under freezing stress.
Overexpression of TaMYB4 Confers Freezing Tolerance in Arabidopsis thaliana

Abstract:Freezing stress is one of the main factors limiting the growth and yield of wheat. In this study, we found that TaMYB4 expression was significantly upregulated in the tillering nodes of the strong cold-resistant winter wheat variety Dongnongdongmai1 (Dn1) under freezing stress. Weighted gene co-expression network analysis, qRT-PCR and protein-DNA interaction experiments demonstrated that monodehydroascorbate reductase (TaMDHAR) is a direct target of TaMYB4. The results showed that overexpression of TaMYB4 enhanced the freezing tolerance of transgenic Arabidopsis. In TaMYB4 overexpression lines (OE-TaMYB4), AtMDHAR2 expression was upregulated and ascorbate-glutathione (AsA-GSH) cycle operation was enhanced. In addition, the expression of cold stress marker genes such as AtCBF1, AtCBF2, AtCBF3, AtCOR15A, AtCOR47, AtKIN1 and AtRD29A in OE-TaMYB4 lines was significantly upregulated. Therefore, TaMYB4 may increase freezing tolerance as a transcription factor (TF) in Arabidopsis through the AsA-GSH cycle and DREB/CBF signaling pathway. This study provides a potential gene for molecular breeding against freezing stress.
Cold Stress Triggers Freezing Tolerance in Wheat (triticum Aestivum L.) Via Hormone Regulation and Transcription of Related Genes

Abstract:Low temperatures limit the geographic distribution and yield of plants. Hormones play an important role in coordinating the growth and development of plants and their tolerance to low temperatures. However, the mechanisms by which hormones affect plant resistance to extreme cold stress in the natural environment are still unclear.In this study, two winter wheat varieties with different cold resistances, Dn1 and J22, were used to conduct targeted plant hormone metabolome analysis on the tillering nodes of winter wheat at 5 degrees C, -10 degrees C and -25 degrees C using an LC-ESI-MS/MS system. We screened 39 hormones from 88 plant hormone metabolites and constructed a partial regulatory network of auxin, jasmonic acid and cytokinin.GO analysis and enrichment of KEGG pathways in different metabolites showed that the 'plant hormone signal transduction' pathway was the most common. Our study showed that extreme low temperature increased the most levels of auxin, cytokinin and salicylic acid, and decreased levels of jasmonic acid and abscisic acid, and that levels of auxin, jasmonic acid and cytokinin in Dn1 were higher than those in J22. These changes in hormone levels were associated with changes in gene expression in synthesis, catabolism, transport and signal transduction pathways. These results differ from the previous hormone regulation mechanisms, which were mostly obtained at 4 degrees C.Our results provide a basis for further understanding the molecular mechanisms by which plant endogenous hormones regulate plant freezing stress tolerance.
Abscisic Acid Enhances Tolerance to Spring Freeze Stress and Regulates the Expression of Ascorbate–glutathione Biosynthesis-Related Genes and Stress-Responsive Genes in Common Wheat

Abstract:The present study showed that exogenous abscisic acid (ABA) pre-treatment remarkably increases the freeze tolerance of common wheat ( Triticum aestivum L.) seedlings at the anther connective tissue formation phase (ACFP) and tetrad developmental phase. After 3-day freeze stress, ABA pre-treatment significantly increases the content of soluble sugar, soluble protein, and proline, activities of superoxide dismutase (SOD) and peroxidase (POD) and decreases hydrogen peroxide (H 2 O 2 ) and malondialdehyde (MDA) levels. Under freeze stress conditions, ABA treatment at the ACFP and tetrad developmental phase resulted in a marked increase in leaf ascorbic acid (ASA) and glutathione (GSH) levels. Temporal and spatial expression patterns of eight genes encoding ASA–GSH synthesis-related enzymes and eight stress-responsive genes were measured using quantitative real-time PCR (qRT-PCR). The results showed that ABA temporally regulated the transcript levels of these genes. Moreover, these genes were differentially expressed in ABA-treated wheat seedlings between the ACFP and tetrad developmental phase during freeze stress. These results implied that exogenous ABA increases the levels of GSH and ASA in freeze stressed wheat seedlings in time- and developmental phase-specific manners. Furthermore, we found that the ABA-independent and ABA-dependent pathways were not completely independent in this study and that ABA-independent stress-responsive genes were also induced by exogenous ABA in at least one stress exposure time point. Comparison of our results with the findings of previous studies showed that plant hormones induce an increase in GSH and ASA levels, possibly by differentially regulating the expression levels of genes encoding ASA–GSH synthesis enzymes. Our results provide the first insights on the molecular mechanism of ABA-induced spring freeze tolerance in wheat.
Overexpression of TaMYC2 confers freeze tolerance by ICE-CBF-COR module in Arabidopsis thaliana

Abstract:Dongnongdongmai No.1 (Dn1) is one of the few winter wheat varieties that can successfully overwinter at temperatures as low as -25°C or even lower. To date, few researches were carried to identify the freeze tolerance genes in Dn1 and applied them to improve plant resistance to extreme low temperatures. The basic helix-loop-helix (bHLH) transcription factor MYC2 is a master regulator in JA signaling, which has been reported to involve in responses to mild cold stress (2°C and 7°C). We hypothesized that MYC2 might be part of the regulatory network responsible for the tolerance of Dn1 to extreme freezing temperatures. In this study, we showed that wheat MYC2 ( TaMYC2 ) was induced under both extreme low temperature (-10°C and-25°C) and JA treatments. The ICE-CBF-COR transcriptional cascade, an evolutionary conserved cold resistance pathway downstream of MYC2, was also activated in extreme low temperatures. We further showed that overexpression of any of the MYC2 genes from Dn1 TaMYC2A, B, D in Arabidopsis led to enhanced freeze tolerance. The TaMYC2 overexpression lines had less electrolyte leakage and lower malondialdehyde (MDA) content, and an increase in proline content, an increases antioxidant defences, and the enhanced expression of ICE-CBF-COR module under the freezing temperature. We further verified that TaMYC2 might function through physical interaction with TaICE41 and TaJAZ7, and that TaJAZ7 physically interacts with TaICE41. These results elucidate the molecular mechanism by which TaMYC2 regulates cold tolerance and lay the foundation for future studies to improve cold tolerance in plants.

Abstract:Wheat (Triticumaestivum L.) is a major cereal crop grown across a wide range of environments, but its productivity around the world is challenged by various biotic and abiotic factors. Wheat landraces from around the world are a source of unexploited genetic diversity that can be essential for modern wheat-breeding programs in search of resistance to abiotic stresses like freezing tolerance. This genetic diversity study of 553 winter wheat landraces based on single-nucleotide polymorphisms (SNPs) revealed separate clusters of landraces related to the latitude of origin. Linkage block analysis revealed genomic regions with specific alleles skewed towards landraces from higher latitudes, suggesting that migration to higher latitudes resulted in the fixing of specific alleles. Electrolyte leakage was used to measure the tolerance of freezing to −14 °C, −16 °C, and −18 °C of 192 landraces. There was a significant negative correlation between latitude and electrolyte leakage, with an R2 value of 0.14, (p < 0.0001), in a regression analysis indicating greater freezing tolerance in landraces from higher latitudes. Genome-wide association studies identified regions in chromosomes 4A and 6A associated with higher latitudes and freezing tolerance, respectively. Landraces with freezing tolerance may be useful in developing new germplasm as novel sources of greater cold hardiness.

Abstract:Abstract In plant cells, phosphatidylglycerol (PG) in the chloroplast has a characteristic trans‐∆3‐hexadecenoic acid (t16:1) at the sn‐2 position. The t16:1 content in wheat leaf tissues decreases during cold treatment, but the significance of this fatty acid compositional change and the underlying biochemical mechanism remains poorly understood. Using a large collection of wheat cultivars displaying a varying capacity of freezing tolerance, we show for the first time under field conditions that this low temperature induced t16:1 change is associated with winter hardiness. To explore the metabolic mechanism responsible for the reduction of t16:1, we performed detailed lipid analysis and comparative transcriptome study with four selected wheat lines under cold acclimation. Our results show that wheat leaf tissues experience a gradual decrease in chloroplast lipid pathway activity during cold acclimation and that the decline in chloroplast lipid synthesis manifests itself in the decrease of t16:1 in PG. Comparative RNA‐seq analyses with leaf tissues further reveal concerted transcriptome shifts indicating a rebalancing of chloroplast and cytosolic lipid synthesis during cold acclimation. Our study, thus, provides mechanistic understanding on chloroplast lipid adjustments as a “molecular ideotype” and the t16:1 change as a specific metabolite marker for screening freezing tolerance in wheat.
Overexpression of the Wheat Trehalose 6-Phosphate Synthase 11 Gene Enhances Cold Tolerance in Arabidopsis Thaliana.

Abstract:Low temperature is a key stress factor for the growth and development of wheat (Triticum aestivum L.), and glycometabolism plays an important role in plant cold tolerance. Our previous study identified trehalose 6-phosphate synthase 11 gene (TaTPS11), which had a significantly different expression pattern between a high freezing-tolerant wheat cultivar and a low freezing-tolerant wheat cultivar. In this study, TaTPS11 was isolated from a winter-hardy wheat cultivar (D1) and overexpressed in Arabidopsis thaliana to study its effect on cold tolerance in plants. Transgenic plants expressing TaTPS11 had lower sucrose content, higher starch content, and higher activity of key enzyme (sucrose phosphate synthase, sucrose synthase, and invertase) involved in sucrose metabolism. In addition, the expression level of sucrose non-fermenting 1-related kinase 1 (SnRK1), which catalyzes the sucrose in plants, increased in the TaTPS11-overexpressed plants. These results indicated that heterologous expression of TaTPS11 influenced carbohydrate metabolism in Arabidopsis plants. The resultant plants had a significantly higher survival rate after - 5 degrees C treatment for 2 h and exhibited enhanced cold tolerance without unfavorable phenotypes compared to wild-type. Our findings indicated that manipulation of TaTPS11 improved cold tolerance in plants and TaTPS11 had potential values in wheat cold-tolerance breeding.

Abstract:Freezing injury is a common abiotic stress in alpine regions. γ-Aminobutyric acid (GABA) is a ubiquitous non-protein amino acid that plays an active role in plant stress resistance. Metabonomic results have shown that the GABA content in winter wheat (Triticum aestivum L.) “Dongnongdongmai 1” (Dn1) significantly changes with decrease in temperature. To clarify the relationship between GABA metabolism and low-temperature stress in winter wheat Dn1, we analyzed the expression of TaGAD and TaGABA-T in the tillering node under natural cooling conditions in the field. Additionally, we constructed plant overexpression vectors and introduced them into Arabidopsis thaliana to obtain stable genetic T3 plants, which were then analyzed under low-temperature stress (–10°C). The expression of TaGAD and TaGABA-T in winter wheat Dn1 gradually increased with decrease in temperature and significantly increased with a temperature decreased from 0 to –10°C. TaGAD expression was highest at –25°C, and the expression abundance of TaGABA-T was close to –10 at –25°C. Moreover, the T3 generation of A. thaliana overexpressing TaGAD and TaGABA-T showed stronger cold resistance than the wild-type. After low-temperature stress, the relative expression in the overexpression lines significantly increased and the relative conductivity and malondialdehyde content decreased compared to those in the wild-type; however, TaGAD-overexpressing lines were better than TaGABA-T-overexpressing lines. These results indicate that the GABA pathway can positively respond to low-temperature stress and that the overexpression of TaGAD is better than that of TaGABA-T for enhancing cold resistance.

Abstract:‘Dongnong Dongmai1 (Dn1)’ is a cultivar of wheat ( Triticum aestivum L.) which may endure‒30°C in field with a reviving rate of 85%. ABA may enhance plant resistance. Aiming to reveal Dn1 coldresistant mechanism after exogenous ABA (exo-ABA) treated (control: non-ABA treated), some transcription factors (TFs) were selected and detected. Seedlings were prayed with exo-ABA at three-leaf stage, subsequently their tilling nodes were respectively sampled when it was at 5, 0,–10 and–25°C in field. Six TF genes ( TabZIP1 , TaWABI5 , TaMYB1 , TaMYB80 , TaNAC2 and TaWRKY80 ) were selected and their expression at different low temperature was detected, the results showed that: in non-ABA treated tilling nodes, the expression of six TF genes all reached their peak level at–10°C; in exo-ABA treated tilling nodes, the expression of five TF genes ( TabZIP1 , TaWABI5 , TaMYB80 , TaNAC2 and TaWRKY80 ) reached their peak level at–25°C, and particularly the expression level of TabZIP1 was the highest in five TFs. In order to further defined the biological function of TabZIP1 , the physical and chemical properties, protein secondary structure and the evolutionary tree was analyzed and predicted by some bioinformatics software or online program; TabZIP1 was localized in nuclear of tobacco ( Nicotiana tabacum ), which was observed by laser scanning confocal microscope (LSCM). This study initially exposed some TFs in Dn1 were upregulated to respond to the coldstress after exo-ABA treated, which indicated that a serious cold-responsive genes could be transcripted with the TFs assistance, and the results provided new theoretical basis to reveal the cold-reponsive mechanism of winter wheat.

Abstract:Abiotic stress experienced by autumn-sown crops during winter is of great economic importance as it can have a severe negative impact on yield. In this study, we investigated the genetic architecture of winter hardiness and frost tolerance in triticale. To this end, we used a large mapping population of 647 DH lines phenotyped for both traits in combination with genome-wide marker data. Employing multiple-line cross QTL mapping, we identified nine main effect QTL for winter hardiness and frost tolerance of which six were overlapping between both traits. Three major QTL were identified on chromosomes 5A, 1B and 5R. In addition, an epistasis scan revealed the contribution of epistasis to the genetic architecture of winter hardiness and frost tolerance in triticale. Taken together, our results show that winter hardiness and frost tolerance are complex traits that can be improved by phenotypic selection, but also that genomic approaches hold potential for a knowledge-based improvement of these important traits in elite triticale germplasm.
