Abstract:This Feature Article summarizes the design and synthesis of supramolecular hydrogels based on nanofibers of self-assembled small bioactive molecules. The hydrogels show responses to molecular recognition and find applications in wound healing, toxin removal, and drug release. We also described our recent results of using an enzyme or enzymatic switch to trigger or regulate the self-assembly of small molecules for the generation of nanofibers and the subsequent hydrogelation. The results in this Feature Article clearly indicate that supramolecular hydrogels, as an expression of the self-assembly of molecules in water, promise a broad range of biomaterials and therapeutics.

Abstract:Dipeptides stereoisomers and regioisomers composed of norleucine (Nle) and phenylalanine (Phe) self-assemble into hydrogels in physiological conditions that are suitable for cell culture. The supramolecular behavior, however, differs as the packing modes comprise amphipathic layers, or water channels, whose diameter is defined by either four or six dipeptide molecules. A variety of spectroscopy, microscopy, and synchrotron-radiation based techniques unveil fine details of intermolecular interactions that pinpoint the relationship between chemical structure and ability to form supramolecular architectures that define soft biomaterials.

Abstract:Supramolecular hydrogels bound by hydrogen bonding, host-guest, hydrophobic, and other non-covalent interactions are among the most attractive biomaterials available. Supramolecular hydrogels have attracted extensive attention due to their inherent dynamic reversibility, self-healing, stimuli-response, excellent biocompatibility, and near-physiological environment. However, the inherent contradiction between non-covalent interactions and mechanical strength makes the practical application of supramolecular hydrogels a great challenge. This review describes the mechanical strength of hydrogels mediated by supramolecular interactions, and focuses on the potential strategies for enhancing the mechanical strength of supramolecular hydrogels and illustrates their applications in related fields, such as flexible electronic sensors, wound dressings, and three-dimensional (3D) scaffolds. Finally, the current problems and future research prospects of supramolecular hydrogels are discussed. This review is expected to provide insights that will motivate more advanced research on supramolecular hydrogels.

Abstract:Supramolecular-based nanofibers, which successfully combine the unique properties of supramolecular interactions with the advantages of nanofibrous structure, are widely used in a variety of biomedical applications such as controlled drug delivery. Compared with traditional polymer nanofibers, supramolecular-based nanofibers can overcome the bottleneck of sensitivity because of the non-covalent binding modes, and therefore match the requirements of rapid and reversible response to the external stimuli. In addition, supramolecular-based nanofibers can achieve extra controllable and dynamic responsive (e.g. pH, temperature) functions in different environments. In this review, we retrospected and summarized the recent development of supramolecular-based nanofibers, focusing particularly on electrospun supramolecular nanofibers, while also touching on the advances of directly self-assembled supramolecular nanofibers without the use of electrospinning. Furthermore, we discussed the potential biomedical applications of supramolecular nanofibers. Finally, this review was concluded by elaborating upon individual reflection on the current situation, forecasting the future trend of this promising material.

Abstract:Hydrogels are of intense recent interest in connection with biomedical applications ranging from 3-D cell cultures and stem cell differentiation to regenerative medicine, controlled drug delivery and tissue engineering. This prototypical form of soft matter has many emerging material science applications outside the medical field. The physical processes underlying this type of solidification are incompletely understood and this limits design efforts aimed at optimizing these materials for applications. We address this general problem by applying multiple techniques (e.g., NMR, dynamic light scattering, small angle neutron scattering, rheological measurements) to the case of a peptide derivative hydrogelator (molecule 1, NapFFKYp) over a broad range of concentration and temperature to characterize both the formation of individual nanofibers and the fiber network. We believe that a better understanding of the hierarchical self-assembly process and control over the final morphology of this kind of material should have broad significance for biological and medicinal applications utilizing hydrogels.

Abstract:With the increased research on supramolecular hydrogels, many spectroscopic, diffraction, microscopic, and rheological techniques have been employed to better understand and characterize the material properties of these hydrogels. Specifically, spectroscopic methods are used to characterize the structure of supramolecular hydrogels on the atomic and molecular scales. Diffraction techniques rely on measurements of crystallinity and help in analyzing the structure of supramolecular hydrogels, whereas microscopy allows researchers to inspect these hydrogels at high resolution and acquire a deeper understanding of the morphology and structure of the materials. Furthermore, mechanical characterization is also important for the application of supramolecular hydrogels in different fields. This can be achieved through atomic force microscopy measurements where a probe interacts with the surface of the material. Additionally, rheological characterization can investigate the stiffness as well as the shear-thinning and self-healing properties of the hydrogels. Further, mechanical and surface characterization can be performed by micro-rheology, dynamic light scattering, and tribology methods, among others. In this review, we highlight state-of-the-art techniques for these different characterization methods, focusing on examples where they have been applied to supramolecular hydrogels, and we also provide future directions for research on the various strategies used to analyze this promising type of material.

Abstract:In an effort to study natural fiber formation, such as, e.g., spider silk, we present a model, which is capable of forming biomimetic fibrillar nanostructure from a hydrogel micellar network. The latter consists of interacting atomic groups which form cores of micelles, and of flexible chains forming the shells of the micelles. Micelles are connected in a compact network by linearly stretched chains. The structural elements of the network can be transformed during deformation from micellar into fibrillary type and their evolution is found to depend significantly on strain rate. Our model suggests a set of conditions suitable for the formation of nanostructured fibrillar network. It demonstrates that a fibrillar structure is only formed upon sufficiently fast stretching while, in contrast, the micellar gel structure is preserved, if the material is pulled slowly. We illustrate this key aspect by a minimalistic model of only four chains as part of the whole network, which provides a detailed view on the mechanism of fibril formation. We conclude that such a simplified structure has similar functionality and is probably responsible for the formation of nano-structured molecular fibrils in natural materials.

Abstract:The recognition-directed host-guest interaction is recognized as a valuable tool for creating supramolecular polymers. Functional hydrogels constructed through the dynamic and reversible host-guest complexation are endowed with a great many appealing features, such as superior self-healing, injectability, flexibility, stimuli-responsiveness and biocompatibility, which are crucial for biological and medicinal applications. With numerous topological structures and host-guest combinations established previously, recent breakthroughs in this area mostly focus on further improvement and fine-tuning of various properties for practical utilizations. The current contribution provides a comprehensive overview of the latest developments in host-guest supramolecular hydrogels, with a particular emphasis on the innovative molecular-level design strategies and hydrogel formation methodologies targeting at a wide range of active biomedical domains, including drug delivery, 3D printing, wound healing, tissue engineering, artificial actuators, biosensors, etc. Furthermore, a brief conclusion and discussion on the steps forward to bring these smart hydrogels to clinical practice is also presented.
Uncovering the relationship between microstructure and mechanical properties in polymer–nanoparticle hydrogels through fluorescent and super-resolution optical microscopy

Abstract:Characterization and understanding the structure of hydrogel networks is usually accomplished through measuring mechanical properties and relating them to theoretical models of the expected network structure. However, several classes of hydrogels are comprised of more complex network architectures, such as non-covalent molecular cross-links and secondary macromolecular interactions, which deviate from ideal polymer networks. In the case of polymer–nanoparticle (PNP) hydrogels, the hydrogel network is formed through polymer–nanoparticle interactions or nanoparticle–nanoparticle interactions, which imbues them with the observed bulk viscoelasticity. While these materials are often used as injectable hydrogels for the delivery of therapeutics and 3D printing, the origin of their dynamic mechanical properties remains poorly understood, which can be addressed through the development of characterization techniques that are able to resolve the microstructure of the networks. Here, we used total internal reflection fluorescence (TIRF) microscopy and direct stochastic optical reconstruction microscopy (dSTORM) to investigate the nanoparticle distribution within PNP hydrogels to relate microstructure to macroscopic properties in this class of materials. We formulated PNP hydrogels with varying α-cyclodextrin (αCD) concentrations and employed dye-labelled nanoparticles for imaging. At low concentrations of αCD, we observed a homogeneous, network-like distribution of the nanoparticles with minimal aggregation, while at higher concentrations aggregates of increased size formed within the network indicative of increased nanoparticle–nanoparticle interactions. This work lays the foundation for advanced microscopy techniques to be employed to fill the gap between understanding of the molecular behavior, changes in topology, and macroscopic properties in hydrogel characterization.

Abstract:The main drawback regarding hydrogels utilization in biomedical engineering is represented by their weak mechanical properties and it is often addressed either by using nanospecies as fillers or by generating secondary networks that strengthen the material. Our study validates a new synthesis route of obtaining double-network nanostructured materials making use of the functional groups of an adsorbed glyco-protein on carbon-based nanospecies. Following a simple procedure, the decorated nanospecies were embedded in a synthetic polymer and subsequently formed a secondary hydrogen-bond based network through incubation in natural polyphenol solutions of various concentrations. The natural macromolecule's adsorption was evaluated through FTIR and DLS measurements. When compared to the single-network nanostructured materials, the double-network hydrogels exhibited lower affinity to aqueous media and improved ability to dissipate energy upon mechanical stress, depending on the amount of the employed natural crosslinker. As a result of generating the second network through incubation in polyphenol aqueous solution, the surface storage and loss moduli exceed the values of the bulk ones, as indicated by rheology and nanoindentation tests. The properties of the double-network hydrogels can also be tailored towards more hydrophilic surfaces with increased roughness through the control of the supplementary crosslinker amount. Considering the studied physical-chemical properties, and especially their mechanical characteristics, the obtained materials hold great promise for biomedical applications, particularly for the fabrication of scaffolds for wound dressing, soft or low load bearing tissue regeneration and repair.

Abstract:Exploration on hydrogel fibres concerning about smart based application in the medical sector has stimulated great interests for the last couple of years due to its wide range of purposes that include actuators, artificial adhesives, transplantable tissue organs, cell scaffolds, cell therapeutics, wound healing, cartilage or bone regeneration. Nevertheless, recently hydrogel fibre based biomaterials have drawn great concentration for use in a wide variety of biomedical applications like the sustained release of drugs. This is due to the fact that, hydrogel fibers are biocompatible and their similarity about physical properties is in relation with natural tissue. This review article prescribes about the application of hydrogels with diversified prospects in tissue engineering, wound care dressings, soft tissue recovery and plastic surgery. As the products of hydrogels are composed with a group of polymeric materials, the hydrophilic network structure makes them competent for holding an immense quantity of water in their three-dimensional polymer network structure. A wide-ranging application of these products in modern industrial and environmental areas has already taken into account to be of prime importance. Inevitably, natural hydrogels right is now gradually replaced by synthetic types due to their larger amount of water absorption capacity, durability alongside with wide ranges of raw chemical resources.

Abstract:Due to the similarity to extracellular matrix, hydrogels have been widely used in biomedical field such as drug delivery, tissue engineering and in vitro cell culture. Deoxyribonucleic acid (DNA) is known for the clear chemical composition, designable sequence and unique structural rigidity, which makes DNA an ideal material for construction of hydrogels. Based on our research, this review summarizes recent developments in DNA supramolecular hydrogels. Firstly, the construction strategies including the pure and hybrid DNA supramolecular hydrogels are introduced. While the construction of pure DNA supramolecular hydrogels is mainly based on base pairing or physical entanglement, covalent or non-covalent doping is also demonstrated to play important roles in preparing the hybrid hydrogels. Then, the physical properties of the DNA supramolecular hydrogels are discussed, such as the excellent permeability, tunable mechanical strength and dynamic properties. Next, the functionalization strategies are also summarized, including sequence design, chemical modification and network interpenetration. Furthermore, the applications of DNA supramolecular hydrogels in biomedicine are introduced, focusing on three-dimensional cell culture and printing, tissue engineering, immunotherapy and drug delivery. Finally, some challenges and future perspectives in the development of DNA supramolecular hydrogels are discussed. It is expected that with the flourishing development, DNA supramolecular hydrogels will have more applications. [GRAPHICS] .

Abstract:DNA-based supramolecular hydrogels are important and promising biomaterials for various applications due to their inherent biocompatibility and tunable physicochemical properties. The three-dimensional supramolecular matrix of DNA formed by non-covalently dynamic cross-linking provides exceptional adaptability, self-healing, injectable and responsive properties for hydrogels. In addition, DNA hydrogels are also ideal bio-scaffold materials owing to their tissue-like mechanics and intrinsic biological functions. Technically, DNA can assemble into supramolecular networks by pure complementary base pairing; it can also be combined with other building blocks to construct hybrid hydrogels. This review focuses on the development and construction strategies of DNA hydrogels. Assembly and synthesis methods, diverse responsiveness and biomedical applications are summarized. Finally, the challenges and prospects of DNA-based supramolecular hydrogels are discussed.

Abstract:Hydrogels have gained a lot of attention with their widespread use in different industrial applications. The versatility in the synthesis and the nature of the precursor reactants allow for a varying range of hydrogels with different mechanical and rheological properties. Understanding of the rheological behavior and the relationship between the chemical structure and the resulting properties is crucial, and is the focus of this review. Specifically, we include detailed discussion on the correlation between the rheological characteristics of hydrogels and their possible applications. Different rheological tests such as time, temperature and frequency sweep, among others, are described and the results of those tests are reported. The most prevalent applications of hydrogels are also discussed.

Abstract:Hydrogels have emerged as versatile biomaterials with remarkable applications in biomedicine and tissue engineering. Here, we present an overview of recent and ongoing research in Italy, focusing on extracellular matrix-derived, natural, and synthetic hydrogels specifically applied to biomedicine and tissue engineering. The analyzed studies highlight the versatile nature and wide range of applicability of hydrogel-based studies. Attention is also given to the integration of hydrogels within bioreactor systems, specialized devices used in biological studies to culture cells under controlled conditions, enhancing their potential for regenerative medicine, drug discovery, and drug delivery. Despite the abundance of literature on this subject, a comprehensive overview of Italian contributions to the field of hydrogels-based biomedical research is still missing and is thus our focus for this review. Consolidating a diverse range of studies, the Italian scientific community presents a complete landscape for hydrogel use, shaping the future directions of biomaterials research. This review aspires to serve as a guide and map for Italian researchers interested in the development and use of hydrogels in biomedicine.

Abstract:<p>The biomedical applications of physically entangled polymeric hydrogels are generally limited due to their weak mechanical properties, rapid swelling and dissolution in physiologically relevant environment. Chemical crosslinking helps stabilizing hydrogel structure and enhancing mechanical properties, thereby allowing a higher stability in phisiological environment. In this context, it is known that the mechanical properties of the hydrogel are affected by both the molecular weight (MW) of the starting polymer and the concentration of the crosslinker. Here, our aim was to assess the influence of polymer molecular weight and concentration in the precursor solution on the mechanical features of the final hydrogel and their influence on cells-material interaction. In detail, 3D synthetic matrices based on poly(ethylene glycol) diacrylate (PEGDA) at two molecular weights (PEG 700 and PEG 3400) and at three different concentrations (10, 20, 40 w/v %), which were photopolymerized using darocour as an initiator, were studied. Then, infrared and swelling analyses, along with a comprehensive mechanical characterization of the obtained hydrogels (i.e. oscillatory shear and confined compression tests), were performed. Finally, to evaluate the influence of the mechanical features on the biological behaviour, the hydrogels were characterized in terms of cell adhesion percentage and viability after functionalizing the substrates with RGD peptide at three different concentrations. Results have demonstrated that both the Young's modulus (E) in compression and the elastic modulus (G') in shear of the hydrogels increase with increasing polymer precursor concentration. E decreased as MW increased, and the differences are more relevant for more concentrated hydrogels. On the contrary, G' appears to increase with increasing PEGDA MW and in particular for the lowest polymer precursor concentration. The biological results have demonstrated that cells cultured for longer times seem to prefer PEG 3400 hydrogels with a larger mesh size structure that posses higher viscoelastic properties in shear.</p>

Abstract:In this review we intend to provide a relatively comprehensive summary of the work of supramolecular hydrogelators after 2004 and to put emphasis particularly on the applications of supramolecular hydrogels/hydrogelators as molecular biomaterials. After a brief introduction of methods for generating supramolecular hydrogels, we discuss supramolecular hydrogelators on the basis of their categories, such as small organic molecules, coordination complexes, peptides, nucleobases, and saccharides. Following molecular design, we focus on various potential applications of supramolecular hydrogels as molecular biomaterials, classified by their applications in cell cultures, tissue engineering, cell behavior, imaging, and unique applications of hydrogelators. Particularly, we discuss the applications of supramolecular hydrogelators after they form supramolecular assemblies but prior to reaching the critical gelation concentration because this subject is less explored but may hold equally great promise for helping address fundamental questions about the mechanisms or the consequences of the self-assembly of molecules, including low molecular weight ones. Finally, we provide a perspective on supramolecular hydrogelators. We hope that this review will serve as an updated introduction and reference for researchers who are interested in exploring supramolecular hydrogelators as molecular biomaterials for addressing the societal needs at various frontiers.
