Abstract:Two new morphinane alkaloids, 1-hydroxy-10-oxo-sinomenine (1) and 4,5-epoxy-14-hydroxy sinomenine N-oxide (2), have been isolated from the stems of Sinomenium acutum. Their structures were established by various spectral analyses, especially 2D NMR experiments. The structure of 2 was confirmed by single crystal X-ray diffraction. The absolute configurations of 1 and 2 were deduced by comparison of CD spectra with the known alkaloid sinomenine (3). Compound 1 was tested for DPPH inhibition and gave IC50 of 27.9M. Compound 2 was tested for neuroprotective effect and showed significant activity against -amyloid25-35-induced oxidative injury (*P0.05) at 10M in PC-12 cells.

Abstract:AIM: To study chemical constituents of Sinomenium acutum and their cell-protective effects. METHODS: Chemical constituents were isolated by various column chromatographic techniques and their structures were elucidated by spectroscopic methods. RESULTS: Three alkaloids, N-trans-feruloylmethoxytyramine (1), gusanlung C (2), and sinoacutine (3) were obtained from the plant stems. CONCLUSION: Alkaloids 1 and 2 have been first isolated from the plant, and both 1 and 2 show protective effects on cell injury induced by hydrogen peroxide in PC12 cells.

Abstract:Sinomenium acutum stem is widely used to treat rheumatoid arthritis, gout, ankylosing spondylitis and other diseases in China. However, its metabolism in vivo is still unclear. In this study, UPLC-Q-TOF/MS was used to analyze the main components and their metabolites in rats after oral administration of Sinomenium acutum stem extract. A total of 41 compounds were identified from the ethanol extract of Sinomenium acutum stem based on the established database and the reference substance; a total of 25 prototype components and 107 metabolites (74 phase I metabolites and 33 phase II metabolites) were speculated and identified in the plasma, urine, bile and feces of rats administered. The metabolic pathways included hydroxylation, demethylation, dehydrogenation, glucuronidation and acetylation. In conclusion, this study revealed the metabolism of Sinomenium acutum stem in vivo , which may provide a better basis for the study of Sinomenium acutum stem and provide useful chemical information on the material basis and pharmacological mechanism of drug efficay.

Abstract:Two pyrrolo[2,1-a]isoquinolines (1 and 2) and three pyrrole alkaloids (3−5), including three new ones, named sinopyrines A−C (1−3), were isolated from the 95% EtOH extract of the stems and rhizomes of Sinomenium acutum (Thumb.) Rehd. et Wils. The structures of the new compounds were elucidated on the basis of spectroscopic data. This is the first report of pyrrole-bearing natural compounds from the family Menispermaceae.

Abstract:An alkaloid with novel skeleton, sinoscrewtine (1), has been isolated from the roots of Sinomenium acutum. Its structure was established by spectral analysis and X-ray crystallographic study, and its possible biosynthetic pathway was delivered. In vitro experiments, 1 showed weak injurious effects against H2O2/Aβ25–35 induced oxidative injury in PC-12 cells and DPPH radical scavenging activity with IC50 of 32.6μM.

Abstract:Alkaloids, the principal constituents in the caulis of Sinomenium acutum, have gained an increasing interest over the past decades since they are widely employed as a clinical treatment for rheumatoid arthritis. In the present study, an integrated characterization strategy by combining mass defect filtering-based structure classification (MDFSC) and diagnostic fragment-ion-based extension (DFIBE) was firstly proposed for rapid profiling of alkaloids in Sinomenii Caulis (SC) via ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry (UPLC-QTOF/MS). The rectangular MDFSC window could more accurately screen the target alkaloids of different types, and the DFIBE could facilitate the acquisition of characteristic fragment ions for structural elucidation of alkaloids. High-performance liquid chromatography (HPLC) fingerprints with principal component analysis (PCA) and hierarchical clustering analysis (HCA) was established for identifying the chemical markers and simultaneous determination of sinomenine, magnoflorine, menisperine, stepharanine and ehydrodiscretine. A total of 91 alkaloids, including 82 targeted ones (26 morphinans, 22 aporphines, 20 protoberberines and 14 benzylisoquinolines) were unambiguously identified or tentatively characterized in SC, and 14 of them were reported for the first time. Sinomenine and magnoflorine could be selected as chemical markers to evaluate the quality of SC from different localities. In conclusion, the proposed method provided a potential approach for chemical profiling and holistic quality control of herbal medicines.

Abstract:A phytochemical investigation on the stems and rhizomes ofSinomenium acutum led to the isolation of 19 compounds,including three phenanthrenes,six anthraquinones,and ten phenolic derivatives.The structures of the isolated compounds were elucidated by analysis of the MS and NMR spectroscopic data and comparison with the literature.Compounds 1 and 2 are two new natural products,and their 1H and 13C NMR spectral data were fully assigned for the first time,with the help of2D NMR.All of the isolates were obtained from genus Sinomenium for the first time and the phenanthrene and anthraquinone are the structure type first reported from this genus plants.

Abstract:Two new morphinane alkaloid dimers, 2,2'-disinomenine (1) and 7',8'-dihydro-1,1'-disinomenine (2), and known 1, 1'-disinomenine (3), were isolated from ethanol extracts of stems of Sinomenium acutum: Their structures were elucidated on the basis of spectroscopic methods. The absolute configuration of alkaloids 1-3 was determined by direct comparison of their CD spectra with the known alkaloid sinomenine. The isolated alkaloids were tested for cytotoxicity against A549, P388, and HeLa cell lines, and 1 and 3 showed weak inhibition against A549 and Hela cells.

Abstract:Sinomendine (1), a good receptor antagonist of angiotensin II and rare oxoaporphine alkaloid isolated from traditional Chinese drug Sinomenium acutum (Thunb.) Rehd. et Wils, was synthesized from a bromobenzylidenetetrahydroisoquinolineenamide precursor (6). Photocyclization of this enamide led to a protected aporphine (7), which could be converted into oxoaporphine (9) and (+/-)-sinomendine (1) by Fremy's salt oxidation followed an available Grignard reaction.

Abstract:Objective To extract total alkaloids of Sinomenium acutum by Soxhlet extractor and analyze them by gas chromatography-mass spectrometry(GC-MS).Methods The Sinomenium acutum was extracted by 90% alcohol,acidified by hydrochloric acid,alkalinized by ammonia water,and then extracted by chloroform.Fifteen peaks were separated and identified by GC-MS.Results The relative content of sinomenine,ambelline and stepharine was up to 80.18%,3.44% and 3.05%,respectively.The compounds were quantitatively determined by normalization method,and the identified compounds represented 95.1% of total GC peak area.Conclusion The results provide important foundation to further study and exploit Sinomenium acutum.

Abstract:Eleven previously undescribed alkaloids, named sinometumines A-K, along with three known alkaloids, were isolated from the rhizomes of Sinomenium acutum . The chemical structures of these unreported compounds were established using extensive spectroscopic methods (IR, UV, HRESIMS, and NMR), and their absolute configurations were determined by single crystal X-ray diffraction analyses and calculated circular dichroism spectroscopy (ECD). Sinometumine D was the first aporphine-type derived alkaloid inner salt with a rearranged dibenzofuran ring backbone. Sinometumine E was a rare protoberberine-type alkaloid with a complex 6/6/6/6/6/6 hexacyclic skeleton. This was the first report of alkaloids with these two skeletons isolated from S. acutum . All isolates were evaluated for their inhibitory activities against indoleamine 2,3-dioxygenase 1 (IDO1) and tryptophan 2,3-dioxygenase (TDO). Lysicamine possessed noteworthy inhibitory activities as IDO1/TDO dual inhibitors with IC 50 values of 6.22 ± 0.26 μ M and 23.76 ± 2.93 μ M, respectively, and liriodenine revealed moderate dual inhibition with IC 50 values of 31.65 ± 4.44 μ M and 15.64 ± 0.26 μ M. The intermolecular interactions and binding modes between lysicamine and IDO1/TDO were elaborated by molecular docking studies.

Abstract:Objective:To investigate the chemical constituents from Sinomenium acutum.Methods: The compounds were isolated and purified by chromatography on silica gel and Sephadex LH-20 column and recrystallization.Their structures were elucidated on the basis of physicochemical properties and spectral analysis.Results:Five triterpenoids were isolated and identified as lupeol(1),lupenone (2),glutinol(3),glutinone(4),acetyl-oleanolic acid(5).Conclusion:These compounds were ob- tained for the first time from this plant.

Abstract:Sinomenium acutum (Thunb.) Rehd. et Wils is widely used in the treatment of rheumatoid arthritis, with its alkaloid compound sinomenine (SIN) being renowned for its significant anti‐inflammatory properties. However, despite its widespread application, the in vivo anti‐inflammatory mechanisms and metabolic pathways of SIN remain incompletely understood. This study established a rapid and reliable method based on an ultra‐high‐performance liquid chromatography method coupled with Quadrupole‐Exactive Orbitrap mass spectrometry and molecular docking to identify and characterize SIN and 69 metabolites in rat plasma, urine, and feces, revealing primary metabolic pathways of hydroxylation, demethylation, sulfation, and glucuronidation. Molecular docking results revealed that phase I reactions, including dedimethylation, demethylation, dehydrogenation, and dihydroxylation, along with their composite reactions, were pivotal in influencing SIN's in vivo anti‐inflammatory activity. M28, M36, and M59 are potentially the most anti‐inflammatory active metabolites of SIN in vivo. This comprehensive analysis unveils SIN's metabolic pathways, offering insights into its biological processes and suggesting a novel approach for exploring active drug constituents. These findings pave the way for further understanding SIN's anti‐inflammatory mechanisms, contributing significantly to the development of new therapeutic strategies.

Abstract:Sinomenine (SIN), an alkaloid derived from the traditional Chinese medicine, Caulis Sinomenii, has been used as an anti-inflammatory drug in China for over 30 years. With the continuous increase in research on the pharmacological mechanism of SIN, it has been found that, in addition to the typical rheumatoid arthritis (RA) treatment, SIN can be used as a potentially effective therapeutic drug for anti-tumour, anti-renal, and anti-nervous system diseases. By reviewing a large amount of literature and conducting a summary analysis of the literature pertaining to the pharmacological mechanism of SIN, we completed a review that focused on SIN, found that the current research is insufficient, and offered an outlook for future SIN development. We hope that this review will increase the public understanding of the pharmacological mechanisms of SIN, discover SIN research trial shortcomings, and promote the effective treatment of immune diseases, inflammation, and other related diseases.

Discovery of chemical markers for improving the quality and safety control of Sinomenium acutum stem by the simultaneous determination of multiple alkaloids using UHPLC-QQQ-MS/MS

Abstract:Abstract Sinomenium acutum stem is a popular traditional Chinese medicine used to treat bone and joint diseases. Sinomenine is considered the only chemical marker for the quality control of S. acutum stem in mainstream pharmacopeias. However, higenamine in S. acutum stem is a novel stimulant that was banned by the World Anti-Doping Agency in 2017. Therefore, enhancing the quality and safety control of S. acutum stem to avoid potential safety risks is of utmost importance. In this study, a fast, sensitive, precise, and accurate method for the simultaneous determination of 11 alkaloids in S. acutum stem by ultrahigh-performance liquid chromatography coupled with triple quadrupole tandem mass spectrometry (UHPLC-QQQ-MS/MS) was established. This method successfully analyzed thirty-five batches of S. acutum stem samples. The average contents of sinomenine, magnoflorine, coclaurine, acutumine, higenamine, sinoacutine, palmatine, magnocurarine, columbamine, 8-oxypalmatine, and jatrorrhizine were 24.9 mg/g, 6.35 mg/g, 435 μg/g, 435 μg/g, 288 μg/g, 44.4 μg/g, 22.5 μg/g, 21.1 μg/g, 15.8 μg/g, 9.30 μg/g, and 8.75 μg/g, respectively. Multivariate analysis, including principal component analysis (PCA), orthogonal partial least square method-discriminant analysis (OPLS-DA), and hierarchical cluster analysis (HCA), were performed to characterize the importance and differences among these alkaloids in S. acutum stem samples. As a result, sinomenine, magnoflorine, coclaurine, acutumine, and higenamine are proposed as chemical markers for quality control. Higenamine and coclaurine are also recommended as chemical markers for safety control. This report provides five alkaloids that can be used as chemical markers for improving the quality and safety control of S. acutum stem. It also alerts athletes to avoid the risks associated with consuming S. acutum stem.

Abstract:Six new alkaloids (1-6) and six known alkaloids (7-12) were obtained from the stems of Sinomenium acutum. Among them, compounds 1-3 and 6 were four N-oxide alkaloids. The structures and absolute configurations of these new alkaloids were elucidated through comprehensive data of 1D and 2D NMR, HRESIMS and ECD spectra. All isolated compounds were evaluated in vitro for their inhibitory activities against nitric oxide (NO) production and inhibitory effects on AChE. Among them, the sinomenine N-oxide (9) was the most potent NO production inhibitor, with an IC50 value of 23.04 μM.

Abstract:Sinomenine is an isoquinoline-type alkaloid found in Sinomenium acutum (Thunb.) Rehd. et Wils and S. acutum (Thunb.) Rehd. et Wils var. cinereum Rehd. et Wils. When used as a medicine, this compound exhibits anti-inflammatory properties; however, sinomenine's use as a medication is limited by side effects, a short half-life, and low efficacy. Owing to these limits, attempts have been made to synthesize sinomenine derivatives with enhanced efficacy. In this study, the anti-inflammatory effects of novel sinomenine derivatives (S1a-S1f) were examined on the basis of lipopolysaccharide-induced inflammatory factor expression in Raw264.7 cells, dimethylbenzene-induced ear oedema, and Evan's blue leakage in mice, and carrageenan-induced paw oedema in rats. Compared with sinomenine, the derivatives significantly inhibited the expression of the inflammatory factors IL-1β and IL-6 at the transcriptional and translational levels. Topical application of 3.250mg/kg of the derivatives also alleviated ear oedema. Compared with the vehicle, the derivatives significantly inhibited carrageenan-induced rat paw oedema after 6h. Among the derivatives, S1a exhibited the most potent anti-inflammatory activity. S1a also significantly increased the sinomenine-induced inhibition of Evan's blue leakage. Thus, S1a may elicit the strongest anti-inflammatory effects of the tested compounds. Based on these results, further development of this compound may be warranted.

Abstract:Abstract: In this manuscript, the resource distribution, pharmacological activity, pharma-cokinetics of sinomenine and the structure, synthesis, biological activity and mechanism of sinomenine derivatives reported from 2000 to December 2023 were reviewed. The lit-erature was retrieved through Web of Science, PubMed, Science Direct, SciFiner Scholar and other websites. Sinomenine belongs to isoquinoline alkaloids and was extracted from the Chinese herb Sinomenium acutum root. In Asian countries such as China and Japan, it is commonly prescribed as a treatment for rheumatoid arthritis. In addition, sinomenine also has sedative, analgesic, anti-inflammatory, immunosuppressive, neuroprotective, an-ti-drug dependence, anti-tumor and other biological activities. Sinomenine limited its ap-plication prospects because of its large dosage, poor epidermal permeability and short half-life. To overcome these defects, new sinomenine derivatives have been synthesized. Based on the comprehensive analysis of relevant literature at home and abroad, this paper reviews the recent progress in the study of sinomenine's pharmacological effects and structural modifications. Future research on sinomenine will focus on improving its thera-peutic effect, and developing new drug preparations and structural modifications. It is hoped that this review will help to better understand the research progress of sinomenine and provide constructive suggestions for further research of sinomenine.

Abstract:AIM: To study the alkaloids from the roots of Aconitum sinomontanum of Ranunculaceae. METHODS: The alkaloids were isolated using silica gel chromatography (column and chromatodron), and the structures were elucidated on the basis of IR, MS, 1HNMR, 13CNMR, 1H-1H COSY, HMQC and HMBC analysis. RESULTS: Twenty seven compounds have been isolated from the total alkaloids, two of which are sinomontanines D (1) and E (2). CONCLUSION: Sinomontanines D (1) and E (2) are new bisnorditerpenoid alkaloids.
