P. serrulata), also known as Chinese photinia, is native to China, Taiwan, Japan, the Philippines, Indonesia, and India. P. serratifolia is a widely cultivated ornamental plant worldwide. In China, P. serratifolia is not only a popular ornamental plant, but is also used for medicinal purposes, such as anthelmintics and purgatives. In October 2017, ∼20% of P. serratifolia leaves were observed to have leaf spots in a community garden in Qingdao, China. The diseased leaves had oval or irregularly shaped brown lesions. The lesions gradually expanded and coalesced, and most large lesions were perforated. Small pieces of tissue taken from the edge of actively growing lesions were surface sterilized in 70% alcohol for 1 min, rinsed twice in sterile distilled water, and then placed on potato dextrose agar (PDA) at 25°C in the dark. The resulting fungal colonies were white, displayed circular growth, undulate edges, and the back had a pale luteous color. After 11 days of incubation, the fungus produced abundant black acervuli over the entire plate. The fungal conidia were fusiform and had five cells (17 to 22× 5 to 8.5 μm). The apical and basal cells were hyaline, the second cell from the base was pale brown, whereas the third and fourth cells were brown. In addition, one basal appendage (3 to 6 μm) and two to four apical appendages (16 to 28 μm) were observed. Based on the colony and conidial morphology, the isolate was identified as Pestalotiopsis clavispora (G.F. Atk.) Steyaert, which has also been designated Neopestalotiopsis clavispora (Maharachchikumbura et al. 2014). The internal transcribed spacer region of rDNA and a β-tubulin gene were amplified from DNA extracted from two single spore isolates using the primers ITS1/ITS4 and BT2a/BT2b (GenBank accession nos. MG673517 and MG673518) (Maharachchikumbura et al. 2012). These sequences had 99% nucleotide identity with the corresponding sequences (HM999902.1, KX721073.1) of the reference strains of P. clavispora in GenBank. In order to perform pathogenicity tests, conidia of three isolates harvested from 10-day-old cultures were suspended in sterile distilled water with the final concentration of 107 conidia/ml. Fifteen microliters of the conidial suspension was placed on the right side of detached P. serratifolia leaves (20 per isolate) previously sterilized with 70% alcohol, while 15 μl of sterile distilled water was placed on the left side as a control treatment. Inoculated leaves were incubated at 25°C and 90% relative humidity with a 12-h photoperiod for 4 days and then 25°C and 20% relative humidity with a 12-h photoperiod for 1 day. The experiment was performed a total of three times. After 5 days, the conidial suspension caused leaf spots similar to those previously observed on infected leaves, whereas the control did not. The fungus was reisolated from the symptomatic tissues, fulfilling Koch’s postulates. To our knowledge, this is the first report that P. clavispora causes leaf spots on P. serratifolia in China and other countries in the world.References:Maharachchikumbura, S. S. N., et al. 2012.
