Abstract:Synchrotron‐based micro‐X‐ray fluorescence (μXRF) elemental images of the inflorescences of the nickel hyperaccumulator Noccaea praecox. Noccaea praecox and Noccaea caerulescens (Brassicaceae) are nickel and zinc hyperaccumulators, native to Europe. To date, most studies have focused on metal accumulation in the leaves, whereas the distribution of metals in the inflorescences of hyperaccumulator plants remains largely unexplored, but of great interest in the context of adaptation to fertility and (insect) pollination. Samples of N. praecox from an ultramafic site and N. caerulescens from an industrially contaminated site were used for synchrotron‐based micro‐X‐ray fluorescence (μXRF) analysis. The results showed that nickel and zinc in the flowers of N. praecox are mainly distributed in the receptacle, ovary, and anthers, but at different concentrations. Similar results were found in N. caerulescens, with the greatest accumulation in the receptacle and ovary, especially in the walls, however at lower levels in the anthers. Although the leaves of N. praecox and N. caerulescens are the main deposition sites for nickel and zinc, significant concentrations of these elements were deposited in the flowers, especially in the pistils and anthers, indicating possible negative effects on fertility and pollinator species.

Abstract:Nicotianamine is a methionine derivative involved in iron homeostasis, able to bind various other metals in vitro. To investigate its role in vivo, we expressed a nicotianamine synthase cDNA (TcNAS1) isolated from the polymetallic hyperaccumulator Thlaspi caerulescens in Arabidopsis thaliana. Transgenic plants expressing TcNAS1 over-accumulated NA, up to 100-fold more than wild type plants. Furthermore, increased NA levels in different transgenic lines were quantitatively correlated with increased nickel tolerance. The tolerance to nickel is expressed at the cellular level in protoplast experiments and is associated with an increased NA content. We have also shown that the most NA-over accumulating line showed a high tolerance to nickel and a significant Ni accumulation in the leaves when grown on nickel-contaminated soil. Our results highlight a new potential role for nicotianamine in heavy metal tolerance at the cellular but also at the whole plant level, easily transposable to a non-tolerant non-hyperaccumulator species. These results open new perspectives for the modulation of nicotianamine content in plants for phytoremediation.

Abstract:X‐ray fluorescence spectroscopy analysis of Noccaea herbarium specimens deposited at the National Museum of Natural History of France revealed hyperaccumulators of Ni and Zn, with the highest concentrations of Ni in Noccaea cappadocica and Zn in Noccaea caerulescens subsp. caerulescens. X‐ray fluorescence spectroscopy (XRF) is a relatively new method for non‐destructive elemental analysis of herbarium material that meets the scientific interest in being capable of discovering new hyperaccumulator plant species. Since the genus Noccaea (Brassicaceae family) is known to be one of the most numerous in term of the hyperaccumulator plant species it contains, especially those that hyperaccumulate Ni, the herbarium material available worldwide represents a great resource for expanding our knowledge of their elemental profiles. In this first systematic XRF scanning of herbarium specimens of the genus Noccaea, a total of 794 specimens from the collection of the National Museum of Natural History in Paris (MNHN) were analyzed, and the raw values obtained were corrected using regression formulas against inductively coupled plasma atomic emission spectroscopy data. Hyperaccumulation of Ni was detected in 90 specimens covering 21 taxa, with Ni concentrations reaching up to 48,700 mg kg−1 in Noccaea cappadocica, an ultramaficophyte from Syria. Zinc concentrations above the hyperaccumulation threshold were found in 210 specimens covering 23 taxa, most of which belonged to different subspecies of Noccaea caerulescens, with the highest concentration reaching up to 56,200 mg kg−1 in N. caerulescens subsp. caerulescens. Although the accumulation of Ni and Zn is contrasting in most of the specimens studied, in 10 specimens, predominantly belonging to N. caerulescens, simultaneous hyperaccumulation of Ni and Zn was found. This study also revealed previously unknown hyperaccumulation of Ni in several Noccaea taxa, as well as a simultaneous hyperaccumulation of Ni and Zn that needs to be confirmed by further experimental and field studies.

Abstract:Multi-elements hyperaccumulators are of high scientific interest to be applied in remediation of mix-contaminated soils. Sedum alfredii is a well-known Cd/Zn co-hyperaccumulator with high Pb and Cu tolerance. This study investigated the ability of the hyperaccumulating ecotype (HE) S. alfredii to tolerate and accumulate Ni. Differed from the non-hyperaccumulating ecotype (NHE), HE plants grew healthy after 50 μM Ni exposure for 4 weeks. The HE plants translocated up to 40 % Ni to the shoots under high Ni stress and accumulated >3000 and 200 mg kg-1 Ni in roots and shoots, respectively. Micro-XRF image showed that Ni was highly restricted within the HE stem and leaf vascular bundles, especially the xylem tissues. The HE roots were of high Ni tolerance, showing much less pronounced Ni-induced phytotoxicity as compared with the NHEs. Ni-induced O2- was observed in the apoplastic part of HE root cells, but both Ni and the induced O2- were highly accumulated in the sensitive zone (root cap, meristem, and cylinder) of NHE roots. These results suggest that although low Ni mobility out of vascular tissues limits the metal accumulation in stems and leaves, HE S. alfredii is highly tolerant towards Ni stress by metal homeostasis in root cells.
Expression of the ZNT1 Zinc Transporter from the Metal Hyperaccumulator Noccaea caerulescens Confers Enhanced Zinc and Cadmium Tolerance and Accumulation to Arabidopsis thaliana

Abstract:Prompt regulation of transition metal transporters is crucial for plant zinc homeostasis. NcZNT1 is one of such transporters, found in the metal hyperaccumulator Brassicaceae species Noccaea caerulescens. It is orthologous to AtZIP4 from Arabidopsis thaliana, an important actor in Zn homeostasis. We examined if the NcZNT1 function contributes to the metal hyperaccumulation of N. caerulescens. NcZNT1 was found to be a plasma-membrane located metal transporter. Constitutive overexpression of NcZNT1 in A. thaliana conferred enhanced tolerance to exposure to excess Zn and Cd supply, as well as increased accumulation of Zn and Cd and induction of the Fe deficiency response, when compared to non-transformed wild-type plants. Promoters of both genes were induced by Zn deficiency in roots and shoots of A. thaliana. In A. thaliana, the AtZIP4 and NcZNT1 promoters were mainly active in cortex, endodermis and pericycle cells under Zn deficient conditions. In N. caerulescens, the promoters were active in the same tissues, though the activity of the NcZNT1 promoter was higher and not limited to Zn deficient conditions. Common cis elements were identified in both promoters by 5' deletion analysis. These correspond to the previously determined Zinc Deficiency Responsive Elements found in A. thaliana to interact with two redundantly acting transcription factors, bZIP19 and bZIP23, controlling the Zn deficiency response. In conclusion, these results suggest that NcZNT1 is an important factor in contributing to Zn and Cd hyperaccumulation in N. caerulescens. Differences in cis- and trans-regulators are likely to account for the differences in expression between A. thaliana and N. caerulescens. The high, constitutive NcZNT1 expression in the stele of N. caerulescens roots implicates its involvement in long distance root-to-shoot metal transport by maintaining a Zn/Cd influx into cells responsible for xylem loading.

Abstract:Changes in soil rhizosphere properties after growing the Cd hyperaccumulator Noccaea caerulescens and the Ni hyperaccumulator Thlaspi goesingense were investigated. Dissolved organic carbon content increased in the rhizosphere, but there were no significant changes in the solution concentrations of Cd and Ni. Concentrations of these metals extracted by NH4Cl and EDTA decreased in the rhizosphere, as did DGT-measured concentrations, indicating a depletion of labile metal in the solid phase. The results could be explained by the increased DOC in the rhizosphere maintaining a higher proportion of the labile metal in solution through complexation, with the overall depletion of metals only manifest in the solid phase. The DGT induced fluxes in soils and sediments (DIFS) model was used to provide key kinetic information on soil processes and labile pool size. These data showed that the more limited metal supply in the rhizosphere after the growth of hyperaccumulators was due to both depletion of the solid phase pool and a lower rate constant of supply from solid phase to solution. The effect on the rate constant, which could be rationalized by the plant sequentially accessing and consuming the more labile pools of metal, was most marked for Cd, which had the highest accumulation factors.
Nitrate Transporter 1.1 Alleviates Lead Toxicity in Arabidopsis by Preventing Rhizosphere Acidification

Abstract:Identification of the mechanisms that control lead (Pb) concentration in plants is a prerequisite for minimizing dietary uptake of Pb from contaminated crops. This study examines how nitrate uptake by roots affects Pb uptake and reveals a new resistance strategy for plants to cope with Pb contamination. We investigated the interaction between nitrate transporter (NRT)-mediated NO3- uptake and exposure to Pb in Arabidopsis using NRT-related mutants. Exposure to Pb specifically stimulated NRT1.1-mediated nitrate uptake. Loss of function of NRT1.1 in nrt1.1-knockout mutants resulted in greater Pb toxicity and higher Pb accumulation in nitrate-sufficient growth medium, whereas no difference was seen between wild-type plants and null-mutants for NRT1.2, NRT2.1, NRT2.2, NRT2.4, and NRT2.5. These results indicate that only NRT1.1-mediated NO3- uptake alleviated Pb toxicity in the plants. Further examination indicated that rhizosphere acidification, which favors Pb entry to roots by increasing its availability, is prevented when NRT1.1 is functional and both NO3- and NH4+ are present in the medium.

Abstract:NRT1.1 is a dual-affinity nitrate (NO3(-)) transporter involved in both high- and low-affinity NO3(-) uptake in Arabidopsis plants. In a recent study, we showed that, under cadmium (Cd) exposure, blocking the NRT1.1-mediated NO3(-) uptake reduces Cd entry into roots, thus lowing Cd levels in plants and improving plant growth. In addition, we also found that the Cd levels in edible parts of 11 Chinese cabbage (Brassica rapa L. ssp. pekinensis) cultivars correlated well with the NO3(-) uptake rates of their roots. These results suggested that the NO3(-) uptake of roots negatively regulate Cd uptake. Modification of NO3(-) uptake in crops by modulating NO3(-) uptake pathway might provide a biological engineering approach to reducing Cd accumulation in edible organs, thus improving food safety.

Abstract:A survey of herbarium specimens from ultramafic (serpentine) soils in Guatemala, using x‐ray fluorescence analysis, has revealed hyperaccumulation of nickel in several species, not previously known to accumulate metals. The figure shows the geographic distribution of Ni‐hyperaccumulating plants in Arachnothryx linguiformis (Rubiaceae), and the frequency distribution of Ni concentrations in specimens of this shrub. Until 2019, there were no published reports of trace element hyperaccumulation in the plants of Central America or Mexico. Recent reports, based mostly on measurement of herbarium specimens using x‐ray fluorescence (XRF) spectroscopy, have begun to identify hyperaccumulators of nickel (Ni) in this region. The most extensive deposits of Ni‐rich ultramafic rocks and soils in Central America occur along the Polochic‐Motagua fault system in eastern Guatemala. This study focused on discovery and characterization of new hyperaccumulators in Guatemala, starting with surveys of herbarium specimens, followed by field studies to understand the ultramafic soils of the region and the ecology of hyperaccumulators that occur there. Initial XRF screening at the Missouri Botanical Garden Herbarium, along with two herbaria in Guatemala, identified four previously unreported Ni hyperaccumulators—Arachnothryx linguiformis, Arachnothryx buddleioides, Chionanthus panamensis, and Orthion guatemalense. Field studies in Guatemala characterized the communities in which these species occur, some of which include multiple hyperaccumulator species growing in close proximity. In two taxa that showed phenotypic variation in foliar Ni concentration, there was no statistically significant correlation between Ni concentrations in leaves and either the total or DTPA‐extractable Ni in soils, suggesting that other genetic or environmental variables may influence hyperaccumulation.

Abstract:Hyperaccumulators within the Noccaea genus possess many promising genetic and metabolic adaptations that could be potentially exploited to support phytoremediation efforts and/or crop improvement and biofortification. Although hyperaccumulation is very common in this genus, individual species display specific traits as they can accumulate different elements (e.g., zinc, cadmium, and/or nickel). Moreover, there appears to be some populational variability with natural selection increasing the metal tolerance in metallicolous populations. Therefore, employing robust methods, such as integrated analysis of the transcriptome and metabolome, is crucial for uncovering pivotal candidate genes and pathways orchestrating the response to metal stress in Noccaea hyperaccumulators. Our study highlights several species-specific traits linked to the detoxification of metals and metal-induced oxidative stress in hyperaccumulating N. praecox when compared to a closely related model species, N. caerulescens, when grown in the field. Transcriptome analysis revealed distinct differences between the three studied natural Noccaea populations. Notably, we observed several pathways frequently connected to metal stress, i.e., glutathione metabolism, phenylpropanoid biosynthesis, and flavonoid biosynthesis, which were enriched. These differences were observed despite the relative evolutionary closeness of studied species, which emphasizes the importance of further expanding our knowledge on hyperaccumulators if we want to exploit their mechanisms for phytoremediation efforts or food quality improvements.

Identifying the Specific Root Microbiome of the Hyperaccumulator Noccaea brachypetala Growing in Non-metalliferous Soils

Abstract:Noccaea brachypetala is a close relative of Noccaea caerulescens, a model plant species used in metal hyperaccumulation studies. In a previous survey in the Catalan Pyrenees, we found two occidental and two oriental N. brachypetala populations growing on non-metalliferous soils, with accumulated high concentrations of Cd and Zn. Our hypothesis was that the microbiome companion of the plant roots may influence the ability of these plants to absorb metals. We performed high-throughput sequencing of the bacterial and fungal communities in the rhizosphere soil and rhizoplane fractions. The rhizobiomes and shoot ionomes of N. brachypetala plants were analyzed along with those from other non-hyperaccumulator Brassicaceae species found at the same sampling locations. The analyses revealed that microbiome richness and relative abundance tended to increase in N. brachypetala plants compared to non-hyperaccumulator species, regardless of plant location. We confirmed that the root compartment is a key factor in describing the community composition linked to the cohabiting Brassicaceae species, and the rhizoplane fraction contained the specific and rare taxa associated with each species. N. brachypetala plants harbored a similar relative abundance of fungi compared to the other plant hosts, but there was a notable reduction in some specific taxa. Additionally, we observed an enrichment in the hyperaccumulator rhizoplane of previously described metal-tolerant bacteria and bacteria involved in nitrogen cycling. The bacteria involved in the nitrogen cycle could contribute indirectly to the hyperaccumulator phenotype by improving soil quality and fertility. Our results indicate that N. brachypetala captures a particular prokaryotic community from the soil. This particular prokaryotic community may benefit the extraction of metal ions and/or improve plant nutrition. Our research identified satellite groups associated with the root niche of a hyperaccumulator plant that may assist in improving biological strategies in heavy metal remediation.
Interaction of hyperaccumulating plants with Zn and Cd nanoparticles

Abstract:Metal hyperaccumulating plant species are an interesting example of natural selection and environmental adaptation but they may also be useful to developing new technologies of environmental monitoring and remediation. Noccaea caerulescens and Arabidopsis halleri are both Brassicaceae and are known metal hyperaccumulators. This study evaluated tolerance, uptake and translocation of zinc sulfide quantum dots by N. cearulescens and cadmium sulfide quantum dots by A. halleri in direct comparison with the non-hyperaccumulator, genetically similar T. perfoliatum and A. thaliana. Growth media were supplied with two different concentrations of metal in either salt (ZnSO4 and CdSO4) or nanoscale form (ZnS QDs and CdS QDs). After 30 days of exposure, the concentration of metals in the soil, roots and leaves was determined. Uptake and localization of the metal in both nanoscale and non-nanoscale form inside plant tissues was investigated by Environmental Scanning Electron Microscopy (ESEM) equipped with an X-ray probe. Specifically, the hyperaccumulators in comparison with the non-hyperaccumulators accumulate ionic and nanoscale Zn and Cd in the aerial parts with a BCF ratio of 45.9 for Zn ion, 49.6 for nanoscale Zn, 2.64 for Cd ion and 2.54 for nanoscale Cd. Results obtained with a differential extraction analytical procedure also showed that a significant fraction of nanoscale metals remained inside the plants in a form compatible with the retention of at least a partial initial structure. The molecular consequences of the hyperaccumulation of nanoscale materials are discussed considering data obtained with hyperaccumulation of ionic metal. This is the first report of conventional hyperaccumulating plants demonstrating an ability to hyperaccumulate also engineered nanomaterials (ENMs) and suggests a potential novel strategy for not only understanding plant-nanomaterial interactions but also for potential biomonitoring in the environment to avoid their entering into the food chains.
Inhibition of Nitrate Transporter 1.1-Controlled Nitrate Uptake Reduces Cadmium Uptake in Arabidopsis.

Abstract:Identification of mechanisms that decrease cadmium accumulation in plants is a prerequisite for minimizing dietary uptake of cadmium from contaminated crops. Here, we show that cadmium inhibits nitrate transporter 1.1 (NRT1.1)-mediated nitrate (NO3 (-)) uptake in Arabidopsis (Arabidopsis thaliana) and impairs NO3 (-) homeostasis in roots. In NO3 (-)-containing medium, loss of NRT1.1 function in nrt1.1 mutants leads to decreased levels of cadmium and several other metals in both roots and shoots and results in better biomass production in the presence of cadmium, whereas in NO3 (-)-free medium, no difference is seen between nrt1.1 mutants and wild-type plants. These results suggest that inhibition of NRT1.1 activity reduces cadmium uptake, thus enhancing cadmium tolerance in an NO3 (-) uptake-dependent manner. Furthermore, using a treatment rotation system allowing synchronous uptake of NO3 (-) and nutrient cations and asynchronous uptake of cadmium, the nrt1.1 mutants had similar cadmium levels to wild-type plants but lower levels of nutrient metals, whereas the opposite effect was seen using treatment rotation allowing synchronous uptake of NO3 (-) and cadmium and asynchronous uptake of nutrient cations. We conclude that, although inhibition of NRT1.1-mediated NO3 (-) uptake by cadmium might have negative effects on nitrogen nutrition in plants, it has a positive effect on cadmium detoxification by reducing cadmium entry into roots. NRT1.1 may regulate the uptake of cadmium and other cations by a common mechanism.

Abstract:This work set out to test the hypothesis that uptake of metals by hyperaccumulator (HA) plants is more likely to be diffusion limited than uptake by nonhyperaccumulator (NHA) plants. Two circumneutral soils, with different contents of organic matter (0.8 and 5.8%), were amended with Cd (0.5 to 5 mg kg-1) and Ni (10 to 100 mg kg-1). A Cd HA plant, Thlaspi caerulescens, was grown in pots containing the Cd amended soils, and a Ni HA, Thlaspi goesingense, was grown in pots containing the Ni amended soils. A NHA plant of the same family, Thlaspi arvense, was grown in the same soils. Metals were measured in both roots and shoots of all plants. Concentrations of Cd and Ni were measured in soil solution and using the technique of diffusive gradients in thin-films (DGT). The dependencies of metal measured by DDT, [M](DGT), and in soil solution, [M](ss), on the amended metal concentration, [M]add, were consistent with fast supply of Cd but a slower rate of release of Ni from solid phase to solution at lower [Ni](add). Detailed consideration of the dependence of Ni and Cd in shoots and roots on [M](add), [M](ss), [MIDST allowed assessment of the supply mechanism. The weight of evidence suggested that diffusion limitation applies for uptake of Cd by both HA and NHA plants and for uptake of Ni by the HA. However, uptake of Ni by the NHA is not limited by diffusion and the biotic ligand model is probably appropriate.

Abstract:The uptake of trace elements by wild herbaceous plants in a multiple trace element-contaminated site was investigated. The bioaccumulation factor (BF) of trace elements was markedly variable among the different plant species. On average, the BF for various trace elements was in the following decreasing order: Zn > Cu > Mn > Ni > As > Pb > Cr. The translocation factor among the investigated plant species was also considerably variable and showed the following decreasing order: Mn > Zn > Ni > Cu > Cr > As > Pb. Several hyperaccumulating plants were identified: Artemisia vulgaris for As, Mn and Zn, Phalaris arundinacea for Mn and Ni, Heracleum sphondylium for Cr and Zn, and Bistorta officinalis for Mn and Zn. The marked accumulation of trace elements in the plant tissue suggests that the site may not be suitable for urban agricultural production. The plant tissue-borne trace elements could affect microbial activities and consequently interfere with the ecosystem functioning in the affected areas.

Abstract:Plant tolerance to nickel (Ni) toxicity depends on plant differences for uptake and distribution within tissues. Differences among and within species for Ni tolerance/accumulation might be used to identify or develop plants for remediation of high Ni soil conditions. Solution culture experiments were conducted under controlled conditions to determine influx (IN) into roots, transport (TR) from roots to shoots, and accumulation of Ni in four plant species grown at different Ni levels. White clover (Trifolium repens L.) had high dry matter (DM) at high Ni levels because of its low IN and TR of Ni. At Ni levels <60 mu M, maize (Zea mays L.) had high DM probably because of its low TR even though it had high IN of Ni. Ryegrass (Lolium perenne L.) was sensitive to Ni toxicity because of its high IN and TR of Ni. The sensitivity of cabbage (Brassica oleracea var. capitata L.) to Ni toxicity was correlated with high TR even though it had low IN of Ni. Nickel accumulation in shoots was relatively high for cabbage and low for maize. Maize averaged similar to 60-fold less Ni in shoots than cabbage and similar to 10-fold less than ryegrass when plants were grown with <120 mu M Ni. Plant tolerance to Ni toxicity was related to low IN of Ni, and especially to TR of Ni. Selecting or developing genotypes with low TR of Ni might improve plant tolerance to moderate Ni toxicity and reduce the flow of Ni from contaminated soils to shoot organs.
The discovery and global distribution of hyperaccumulator plants: A personal account

Abstract:This article is a review of work carried out by the author and others during the last 48 years, relating to the discovery of plant species that exhibit hyperaccumulation of metallic elements. There is a particular focus on species that accumulate nickel from ultramafic soils. An instance of extreme accumulation of Zn by a plant species was found in the 19th century, and observations of unusually high concentrations of other elements (Cu, Co, Ni, Cd, Pb, As, Se) were recorded between the 1920s and 1970s. Of relevance to the study of ultramafic areas and their floras is the occurrence of extreme Ni accumulation. The term "hyperaccumulator" was introduced in 1976 to signify a species exhibiting a concentration that can be hundreds or thousands of times greater than that usually found in plants on most common soils. Concentration criteria that allow a species to be regarded as a hyperaccumulator have been defined and refined from time to time. The following account details my own involvement in the discovery of new examples of metal hyperaccumulation, with particular emphasis on Ni accumulation by plants of ultramafic floras worldwide. The interest in hyperaccumulators has prompted investigations into different aspects of the biota of metalliferous soils. The search for new examples of hyperaccumulation continues, aided in part by non‐destructive X‐ray fluorescence scanning of herbarium specimens, which previously served as a resource providing small fragments for sensitive but destructive analysis. Identification of species as hyperaccumulators must be supported by further field exploration to understand the factors governing the level of metal uptake, and to stimulate further work on plant systematics, biochemistry, genetics, and molecular biology. Applications to phytoremediation and agromining are worth pursuing in some cases. Conservation issues are important because many hyperaccumulators are rare and restricted in their distribution.

Abstract:AbstractNickel hyperaccumulator plants have been the focus of considerable research because of their unique ecophysiological characteristics that can be exploited in phytomining technology. Comparatively little research has focussed on the soil chemistry of tropical nickel hyperaccumulator plants to date. This study aimed to elucidate whether the soil chemistry associated with nickel hyperaccumulator plants has distinctive characteristics that could be indicative of specific edaphic requirements. The soil chemistry associated with 18 different nickel hyperaccumulator plant species occurring in Sabah (Malaysia) was compared with local ultramafic soils where nickel hyperaccumulator plants were absent. The results showed that nickel hyperaccumulators in the study area were restricted to circum-neutral soils with relatively high phytoavailable calcium, magnesium and nickel concentrations. There appeared to be a ‘threshold response’ for the presence of nickel hyperaccumulator plants at >20 μg g−1 carboxylic-extractable nickel or >630 μg g−1 total nickel, and >pH 6.3 thereby delimiting their edaphic range. Two (not mutually exclusive) hypotheses were proposed to explain nickel hyperaccumulation on these soils: (1) hyperaccumulators excrete large amounts of root exudates thereby increasing nickel phytoavailability through intense rhizosphere mineral weathering; and (2) hyperaccumulators have extremely high nickel uptake efficiency thereby severely depleting nickel and stimulating re-supply of Ni from diffusion from labile Ni pools. It was concluded that since there was an association with soils with highly labile nickel pools, the available evidence primarily supports hypothesis (2).

Abstract:Soil contamination by heavy metals like Ni was originally restricted to metalliferous soils but in modern time it has become a general problem due to increasing anthropogenic activities. Because of the characteristics of cost-effectiveness, environmental friendliness, and fewer side-effects, the development of plant based remediation technologies for the cleanup of Ni-contaminated soils has attracted much attention. Nickel is an essential micronutrient, but is toxic to plants at excessive levels. Some plant species can accumulate Ni in the shoots at a high concentration, these plants are called hyperaccumulators. In the past two decades, researchers have endeavored to understand the physiological and molecular mechanisms of Ni uptake, transport, and detoxification in the Ni-hyperaccumulator plants. This is the basis of creating ideal plants for phytoremediation through cell and genetic engineering technologies, which may subsequently improve phytoremediation efficiency for decontaminating Ni-contaminated soils. Both rhizosphere microorganisms and endophytes can play a role in phytoremediation. Optimizing plant and soil management practices, particularly the correction of soil pH and additions of amendments of exogenous chelates and fertilizers, can also enhance phytoremediation of Ni-contaminated soils. The primary objective of this review is to discuss the recent progresses in basic and applied research relevant to phytoremediation of Ni-contaminated soils.

Abstract:Plant natural resistance-associated macrophage protein (NRAMP) plays an important role in maintaining intracellular metal homeostasis and coping with environmental heavy metal stress. Until now, studies on NRAMP in tobacco have been limited. In this study, NtNRAMP1 was cloned from tobacco cultivar TN90, and the highest expression level was observed in the roots, which was strongly induced by Fe deficiency. Heterologously expressed NtNRAMP1 significantly increased the Cd sensitivity of the yeast Δycf1 mutant. Three overexpressed NtNRAMP1 lines were generated to reveal the biofunction of NtNRAMP1. In the soil pot experiments under natural conditions, the contents of Fe and total chlorophyll were increased in the leaves of transgenic tobacco compared with the WT. To reveal the characteristics of NtNRAMP1 in metal transport, transgenic plants were cultured in hydroponic solution with 50 μM Cd and 200 μM Fe. Compared with the WT, the Cd concentrations in transgenic plants increased by 1.26-2.02-fold in the roots. Interestingly, the Cd content in the shoots of transgenic plants was slightly reduced compared with that of the WT. Overexpression of NtNRAMP1 did not promote Fe uptake from the external environment into the roots but enhanced the transfer of Fe from the roots to shoots. Additionally, Fe overload in the leaves of transgenic tobacco resulted in increased levels of MDA and H<sub>2</sub>O<sub>2</sub> while Fe toxicity may be relieved by POD. In conclusion, overexpression of NtNRAMP1 in tobacco could promote Cd uptake and Fe transport from the roots to shoots while disturbing Fe homeostasis in the leaves of transgenic tobacco.
