Abstract:Sucrose non-fermenting 1-related protein kinase 2 (SnRK2) family members play crucial roles in plant abiotic stress response. However, the precise mechanism underlying the function of SnRKs has not been thoroughly elucidated in plants. In this research, a novel SnRK2 gene, TaSnRK2.9 was cloned and characterized from common wheat. The expression of TaSnRK2.9 was upregulated by polyethylene glycol (PEG), NaCl, H2O2, abscisic acid (ABA), methyl jasmonate (MeJA), and ethrel treatments. TaSnRK2.9 was mainly expressed in wheat young root, stamen, pistil, and lemma. Overexpressing TaSnRK2.9 in transgenic tobacco enhanced plants' tolerance to drought and salt stresses both in young seedlings and mature plants with improved survival rate, seed germination rate, and root length. Physiological analyses suggest that TaSnRK2.9 improved antioxidant system such as superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and glutathione (GSH) to reduce the H2O2 content under drought or salt stress. Additionally, TaSnRK2.9 overexpression plants had elevated ABA content, implying that the function of TaSnRK2.9 may be ABA-dependent. Moreover, TaSnRK2.9 increased the expression of some ROS-related, ABA-related, and stress-response genes under osmotic or salt treatment. TaSnRK2.9 could interact with NtABF2 in yeast two-hybrid assay, and increased the expression of NtABF2 under mannitol or NaCl treatment in transgenic tobacco plants. In conclusion, overexpression of TaSnRK2.9 in tobacco conferred plants tolerance to drought and salt stresses through enhanced ROS scavenging ability, ABA-dependent signal transduction, and specific SnRK-ABF interaction.

Abstract:Drought stress is a severe threat to plants. Genes that respond to drought stress are essential for plant growth and development. General control nonderepressible 2 (GCN2) encodes a protein kinase that responds to various biotic and abiotic stresses. However, the mechanism of GCN2 in plant drought tolerance remains unclear. In the present study, the promoters of NtGCN2 from Nicotiana tabacum K326, which contained a drought-responsive Cis-acting element MYB that can be activated by drought stress, were cloned. Furthermore, the drought tolerance function of NtGCN2 was investigated using NtGCN2-overexpressed transgenic tobacco plants. NtGCN2-overexpressed transgenic plants were more tolerant to drought stress than wild-type (WT) plants. The transgenic tobacco plants exhibited higher proline and abscisic acid (ABA) contents, antioxidant enzyme activities, leaf relative water content, and expression levels of genes encoding key antioxidant enzymes and proline synthase, but lower levels of malondialdehyde and reactive oxygen species, and reduced stomatal apertures, stomatal densities, and stomatal opening rates compared to WT plants under drought stress. These results indicated that overexpression of NtGCN2 conferred drought tolerance in transgenic tobacco plants. RNA-seq analysis showed that overexpression of NtGCN2 responded to drought stress by regulating the expression of genes related to proline synthesis and catabolism, abscisic acid synthesis and catabolism, antioxidant enzymes, and ion channels in guard cells. These results showed that NtGCN2 might regulate drought tolerance by regulating proline accumulation, reactive oxygen species (ROS) scavenging, and stomatal closure in tobacco and may have the potential for application in the genetic modification of crop drought tolerance.
Overexpression of Suaeda Salsa SsNHX1 Gene Enhanced Salt and Drought Tolerance of Transgenic Tobacco

Abstract:Tobacco (Nicotiana tobacum L.) is an important economic and model plant. Salt and drought are two important en-vironmental factors that are harmful to plant growth, development, production and quality of tobacco. In addition to conven-tional salt and drought prevention measures, genetic engineering of plants has also been proven to be effective. In order to improve salt and drought tolerance of tobacco, Na+/H+ anti-porter gene SsNHX1was cloned from Suaeda salsa salt-tolerant plant and overexpressed in tobacco byagrobacterium-mediated genetic transformation. The differences in salt tolerance and drought resistance were compared by determining phenotypic and physiological indexes of wild and transgenic tobacco L1 and L5. Phenotypic analysis showed that salt tolerance ofSsNHX1transgenic tobacco lines L1 and L5 were significantly higher than that of the wild type. This was evident from the vigorously growth and uninhibited root elongation under salt stress con-dition. The overexpressed transgenic lines of tobacco accumulated more Na+and K+in the both leaves and roots and with faster rate of increase of Na+ and slower decreasing rate of K+. The lines also maintained significantly higher contents of relative leaf water and chlorophyll, but lower malondialdehyde contents and relative conductivities. The results indicated that overexpres-sion of SsNHX1 gene apparently promoted compartmentalization of Na+from vacuolar cells into vacuoles and improved salt tolerance of transgenic plants of tobacco. On the other hand,SsNHX1transgenic tobacco plants showed significant enhance-ment of drought tolerance than the wild type and restored normal growth after rehydration. Under drought stress, the contents of malondialdehyde and relative conductivities of transgenic lines were lower than those of the wild type, while the relative water and chlorophyll contents of leaves were maintained. The results suggested that under drought stress, overexpressed SsNHX1 in tobacco reduced damage to cell membrane by reducing osmotic potential of cells, maintaining relative water and chlorophyll content of leaves, and finally improved drought resistance of tobacco.

Abstract:Aquaporin (AQP) proteins have been shown to transport water and other small molecules through biological membranes, which is crucial for plants to combat stress caused by drought. However, the precise role of AQPs in drought stress response is not completely understood in plants. In this study, a PIP2 subgroup gene AQP, designated as TaAQP7, was cloned and characterized from wheat. Expression of TaAQP7-GFP fusion protein revealed its localization in the plasma membrane. TaAQP7 exhibited high water channel activity in Xenopus laevis oocytes and TaAQP7 transcript was induced by dehydration, and treatments with polyethylene glycol (PEG), abscisic acid (ABA) and H2O2. Further, TaAQP7 was upregulated after PEG treatment and was blocked by inhibitors of ABA biosynthesis, implying that ABA signaling was involved in the upregulation of TaAQP7 after PEG treatment. Overexpression of TaAQP7 increased drought tolerance in tobacco. The transgenic tobacco lines had lower levels of malondialdehyde (MDA) and H2O2, and less ion leakage (IL), but higher relative water content (RWC) and superoxide dismutase (SOD) and catalase (CAT) activities when compared with the wild type (WT) under drought stress. Taken together, our results show that TaAQP7 confers drought stress tolerance in transgenic tobacco by increasing the ability to retain water, reduce ROS accumulation and membrane damage, and enhance the activities of antioxidants.
Overexpression of the Transcription Factor NtNAC2 Confers Drought Tolerance in Tobacco

Abstract:NAC proteins constitute one of the largest families of plant-specific transcription factors and play an important role in biological processes, including plant development and phytohormone homeostasis, and in responses to various environmental stresses. In this study, we isolated an NAC group A gene (named NtNAC2) from Nicotiana tabacum L. Quantitative RT-PCR (qRT-PCR) analysis indicated that NtNAC2 was significantly upregulated under drought stress, which implied NtNAC2 was important in tobacco under such conditions. Overexpression of NtNAC2 in tobacco plants exhibited enhanced drought tolerance by means of improved seedling growth. Under drought stress, organic osmoprotectants were significantly accumulated in these plants. Additionally, the activities of antioxidant defense enzymes, like superoxide dismutase (SOD) and peroxidase (POD), which could effectively scavenge accumulated reactive oxygen species (ROS), increased in NtNAC2-overexpression transgenic tobacco plants compared with wild-type plants. The net photosynthetic rate was also significantly increased in NtNAC2-overexpression transgenic lines compared with wild-type plants, and the content of malondialdehyde (MDA) and proline was lower in NtNAC2-overexpression transgenic lines than that in wild-type plants (P < 0.01). Furthermore, the expression of NtWRKY28, a drought resistance gene, was significantly increased and the δ-OAT gene was downregulated in NtNAC2-overexpression plants relative to wild-type plants. Taken together, these results indicated that NtNAC2 functions as a positive regulator of drought stress tolerance. This study provides a basis for further study of drought resistance conferred by the NtNAC2 gene.
Drought Resistance of Transgenic Tobacco with Sugarcane△1-Pyrroline-5-carboxylate Synthase (sop5cs )Gene

Abstract:In order to analyze the drought resistance of sugarcane △ 1 -pyrroline-5-carboxylate syn-thase gene (SoP 5CS ),SoP 5CS was transformed into tobacco using Agrobacterium -mediated leaf discs method.Four transgenic tobacco lines with low copy numbers were screened for drought stress experiment.The results showed that both the relative expression levels of SoP 5CS and its encoded protein were the lowest in T9 transgenic tobacco line under drought stress.Compared with T9 ,the relative expression level of SoP 5CS in T1 ,T4 and T6 lines were 9.02-fold,3.93-fold and 8.51-fold while the protein level of SoP5CS in T1 ,T4 and T6 lines was 2.22-fold,3.91-fold and 1.36-fold,respectively.Results of further analyses showed that the contents of proline and ABA in the four transgenic lines were all significantly higher than those in the control.The contents of proline and ABA in T1 and T6 lines were higher than those in the other two lines.The MDA content in the control was significantly higher than that of the transgenic plants.The accumulation of MDA was the lowest in T1 line.The P5CR and P5CS activities were different between the control and transgenic plants.The activities of both enzymes in T1 and T4 lines were nearly the same as those in the control,significantly higher than those in T6 and T9 lines.The CAT activity was the highest in T6 line,which in the other transgenic lines significantly higher than that of the control.SOD activity in all the transgenic lines was significantly higher than that in the control, with which in T4 and T6 lines being relatively higher.The relative water content in the leaves of the transgenic lines was significantly higher than that in the control,while the chlorophyll reduction was lower.The result of RA comprehensive analysis showed that the drought resistance of T6 line was the best,followed by the T1 line,indicating that drought resistance of SoP 5CS gene is strong.

Abstract:Key message The expression of BdWRKY36 was upregulated by drought treatment. BdWRKY36 -overexpressing transgenic tobacco increased drought tolerance by controlling ROS homeostasis and regulating transcription of stress related genes. Abstract WRKY transcription factor plays important roles in plant growth, development and stress response. However, the function of group IIe WRKYs is less known. In this study, we cloned and characterized a gene of group IIe WRKY , designated as BdWRKY36 , from Brachypodium distachyon . Transient expression of BdWRKY36 in onion epidermal cell suggested its localization in the nucleus. Transactivation assay revealed that the C-terminal region, instead of full length BdWRKY36, had transcriptional activity. BdWRKY36 expression was upregulated by drought. Overexpression of BdWRKY36 in transgenic tobacco plants resulted in enhanced tolerance to drought stress. Physiological–biochemical indices analyses showed that BdWRKY36 -overexpressing tobacco lines had lesser ion leakage (IL) and reactive oxygen species (ROS) accumulation, but higher contents of chlorophyll, relative water content (RWC) and activities of antioxidant enzyme than that in control plants under drought condition. Meanwhile expression levels of some ROS-scavenging and stress-responsive genes were upregulated in BdWRKY36 -overexpressing tobacco lines under drought stress. These results demonstrate that BdWRKY36 functions as a positive regulator of drought stress response by controlling ROS homeostasis and regulating transcription of stress related genes.

Abstract:Low temperature stress affects the yield and quality of crops. To adapt to abiotic stress, plant tubulin families play a crucial role to increase cell cold stability. In the present study, our aim was to develop transgenic tobacco by using a cold-responsive SoTUA gene isolated from Saccharum spp. hybrid. Based on fluorescent microscope observation and TUA gene expression analysis in different tissues, TUA gene expression was found mainly in veins. Moreover, a significant increase of TUA transcripts and protein under cold stress was detected by quantificational real-time polymerase chain reaction (qRT-PCR) and western blot in the transgenic plants. Results of peroxidase (POD), superoxide dismutase (SOD) and catalase (CAT) activities, and proline, soluble sugar and protein contents indicated oxidative damage is less than wild type (WT). Overexpression of SoTUA gene has improved the cold stress tolerance ability in tobacco.

Evolutionary Analysis of StSnRK2 Family Genes and Their Overexpression in Transgenic Tobacco Improve Drought Tolerance

Abstract:Sucrose non-ferment 1-related protein kinase 2 (SnRK2) is a highly conserved protein kinase in plants that plays an important role in regulating plant response to drought stress. Although it has been reported in some plants, the evolutionary relationship of potato SnRK2s and their function in drought resistance have not been systematically analyzed. In this study, molecular characteristic analysis showed that 8 StSnRK2s were distributed on six chromosomes, coding proteins were divided into three subgroups, and StSnRK2s clustered in the same subgroup had similar conserved motifs and domains. In addition, StSnRK2 has a wide range of replication events in some species, making it closer to dicots in the process of evolution. In addition, the average nonsynonymous substitution rate/synonymous substitution rate (Ka/Ks) value of SnRK2s in monocots was higher than that of dicots. The codon usage index showed that SnRK2s prefer to use cytosine 3 (C3s), guanine 3 (G3s) and GC content (GC3s) in monocots, whereas thymine 3 (T3s) and adenine 3 (A3s) are preferred in dicots. Furthermore, stress response analysis showed that the expression of StSnRK2s under different degrees of drought stress significantly correlated with one or more stress-related physiological indices, such as proline and malondialdehyde (MDA) content, superoxide dismutase (SOD) and catalase (CAT) activity, ion leakage (IL) etc. The drought resistance of StSnRK2 transgenic plants was determined to occur in the order of StSnRK2.1/2.8 > StSnRK2.2/2.5 > StSnRK2.4/2.6 > StSnRK2.3 > StSnRK2.7, was attributed to not only lower IL but also higher proline, soluble sugar contents and stress-related genes in transgenic plants compared to wild type (WT). In conclusion, this study provides useful insights into the evolution and function of StSnRK2s and lays a foundation for further study on the molecular mechanism of StSnRK2s regulating potato drought resistance.
TaASR1, a Transcription Factor Gene in Wheat, Confers Drought Stress Tolerance in Transgenic Tobacco

Abstract:Abscisic acid (ABA)-, stress-, and ripening-induced (ASR) proteins are reported to be involved in abiotic stresses. However, it is not known whether ASR genes confer drought stress tolerance by utilizing the antioxidant system. In this study, a wheat ASR gene, TaASR1, was cloned and characterized. TaASR1 transcripts increased after treatments with PEG6000, ABA and H(2)O(2). Overexpression of TaASR1 in tobacco resulted in increased drought/osmotic tolerance, which was demonstrated that transgenic lines had lesser malondialdehyde (MDA), ion leakage (IL) and reactive oxygen species (ROS), but higher relative water content (RWC) and superoxide dismutase (SOD) and catalase (CAT) activities than wild type (WT) under drought stress. Overexpression of TaASR1 in tobacco also enhanced the expression of ROS-related and stress-responsive genes under osmotic stress. In addition, transgenic lines exhibited improved tolerance to oxidative stress by retaining more effective antioxidant system. Finally, TaASR1 was localized in the cell nucleus and functioned as a transcriptional activator. Taken together, our results showed that TaASR1 functions as a positive factor under drought/osmotic stress, involved in the regulation of ROS homeostasis by activating antioxidant system and transcription of stress-associated genes.
Overexpression of SoACLA-1 Gene Confers Drought Tolerance Improvement in Sugarcane

Abstract:Drought is one of the most severe stresses which limit sugarcane production in China. ATP citrate lyase (ACL) is a major enzyme responsible for the production of acetyl-CoA in cytoplasm and plays an important role in plant metabolism and stress response. In this study, sugarcane ACL gene SoACLA-1 was cloned. The plant overexpression vector of SoACLA-1 was built and transformed into sugarcane calli by Agrobacterium -mediated transformation, and PCR analysis confirmed that SoACLA-1 gene had been stably present in the T0, T1, and T2 generations of the transgenic sugarcane. In order to evaluate the drought resistance of the transgenic lines and verify the function of SoACLA-1 gene in the transgenic sugarcane, T1 generation of the SoACLA-1 transgenic sugarcane lines was used as the material to investigate the physiological and biochemical characteristics at 0 day, 3 days, 6 days, and 9 days after water stress and rewatering for 3 days. Comprehensive evaluation of four indicators (chlorophyll, malondialdehyde, proline, soluble sugar) related to drought resistance was done with membership fuzzy function method. The results showed that the drought resistance of five transgenic sugarcane lines from strong to weak, in turn, was RT2 > RT4 > RT3 > RT1 > WT, and the recovery ability after drought, in turn, was RT1 > RT2 > RT4 > RT3 > WT. The T2 generation of the SoACLA-1 transgenic sugarcane lines was used to analyze the physiological and biochemical changes and the expression of drought-related genes under water stress. The results showed that the transgenic sugarcane lines were more tolerant to drought as compared with the wild-type plants. Our findings indicated that SoACLA-1 gene plays an important role as a positive factor in response to water stress, and overexpression of SoACLA-1 can enhance drought tolerance in transgenic sugarcane plants.

Abstract:Drought is one of the very important growth-limiting factors for sugarcane production in China. Δ1-pyrroline-5-carboxylate synthase (P5CS) is the rate-limiting enzyme in proline synthesis, and it plays an important role in plant response to drought stress. In this study, sugarcane P5CS (SoP5CS) gene was cloned and submitted in GenBank with the accession number KJ546350. The gene SoP5CS was 2151 bp in length, encoding 716 amino acids with a predicted molecular weight of 77.73 kDa and an isoelectric point of 6.14. The qRT-PCR analysis showed that the expression of SoP5CS was higher in leaf than in root and stalk, and strongly induced under ABA, PEG, NaCl and 4 °C treatments. The plant excessive expression vector of SoP5CS was built and transformed into sugarcane calli by Agrobacterium-mediated transformation method, and transgenic sugarcane plants were obtained. Under drought stress, some transgenic lines with overexpressed SoP5CS showed significantly higher in SoP5CS expression level, proline accumulation, abscisic acid content, superoxide dismutase activity and relative water content but lower MDA content and chlorophyll (SPAD value) decrease as compared with the WT plants. Moreover, all the transgenic plants increased drought stress tolerance. Our findings indicated that SoP5CS plays an important role in response to abiotic stresses and overexpression of SoP5CS can improve drought tolerance in transgenic sugarcane plants.
Overexpression of CsSnRK2.5 increases tolerance to drought stress in transgenic Arabidopsis

Abstract:Drought is a major factor limiting crop productivity and quality. Sucrose non-fermenting-1 (SNF1)-related protein kinase 2s (SnRK2s) play critical roles in plant abiotic stress responses, especially in drought stress. However, knowledge regarding the functional roles of SnRK2s in drought stress and their underlying mechanisms is relatively limited in tea plant. In this study, CsSnRK2.5, a PEG 6000- and ABA-induced SnRK2 gene from tea plant, was overexpressed in Arabidopsis to investigate its potential function in drought stress response. The results showed that overexpression of CsSnRK2.5 resulted in enhanced drought tolerance, as indicated by an amelioration of the changes in various physiological indexes, including a decreased rate of water loss and decreased accumulation of ROS and MDA. In addition, CsSnRK2.5 overexpression conferred hypersensitivity to exogenous ABA, and transgenic plants exhibited improved ABA-mediated stomatal closure compared to WT plants. Moreover, the expression of some stress response genes, including AtRAB18 and AtRD29b, was more strongly induced in transgenic plants than in the WT when subjected to ABA and drought treatments. Taken together, our results indicate that CsSnRK2.5 is a positive regulator of ABA-regulated drought stress responses.
Overexpression of Sugarcane SoTUA Gene Enhances Cold Tolerance in Transgenic Sugarcane

Abstract:Alpha (alpha)-tubulin (TUA) is a primary unit of cytoskeleton microtubules. In recent years, more frequent frosts have caused considerable yield loss in sugarcane (Saccharum spp. hybrid) production. Temperature is one of the most influential abiotic factors affecting the microtubule polymerization state in plants. In this research, we developed overexpressing SoTUA transgenic sugarcane lines from the cold-susceptible cultivar ROC22. The transgene was confirmed by polymerase chain reaction (PCR) and quantificational real-time polymerase chain reaction (qRT-PCR) analysis, while the relative expression of SoTUA protein was verified by Western blot. Compared with the nontransformed control plants (wild type [WT]), the transgenic lines showed higher relative protein expression than the WT at 25 degrees C, which indicates the overexpression of SoTUA protein. The contents of soluble protein and sugar and the activity of peroxidase (POD) were higher while the content of maleicdialdehyde was lower in the transgenic plants than the WT plants under the chilling treatment. Meanwhile, the expression levels of cold-defense related genes (P5CS, WRKY, Cu/Zn-SOD, and POD1) were higher in the transgenic lines than in the WT. These results suggest that the SoTUA protein plays an important role in protecting plants during chilling stress. This positive effect is not affected by transgenic plant generations.
Over-Expression of a Maize N-Acetylglutamate Kinase Gene (zmnagk) Improves Drought Tolerance in Tobacco.

Abstract:Water deficit is a key limiting factor that affects the growth, development and productivity of crops. It is vital to understand the mechanisms by which plants respond to drought stress. Here an N-acetylglutamate kinase gene, ZmNAGK, was cloned from maize (Zea mays). ZmNAGK was expressed at high levels in maize leaves and at lower levels in root, stem, female flower and male flower. The expression of ZmNAGK was significantly induced by PEG, NaCl, ABA, brassinosteroid and H2O2. The ectopic expression of ZmNAGK in tobacco resulted in higher tolerance to drought compared to plants transformed with empty vector. Further physiological analysis revealed that overexpression of ZmNAGK could enhance the activities of antioxidant defense enzymes, and decrease malondialdehyde content and leakage of electrolyte in tobacco under drought stress. Moreover, the ZmNAGK transgenic tobacco accumulated more arginine and nitric oxide (NO) than control plants under drought stress. In addition, the ZmNAGK transgenic tobaccos activated drought responses faster than vector-transformed plants. These results indicate that ZmNAGK can play a vital role in enhancing drought tolerance by likely affecting the arginine and NO accumulation, and ZmNAGK could be involved in different strategies in response to drought stress.
Overexpression of Sugarcane ScDIR Genes Enhances Drought Tolerance in Nicotiana Benthamiana

Abstract:Dirigent proteins (DIRs) are known to function in lignin biogenesis and to be involved in stress resistance in plants. However, the sugarcane DIRs have not been functionally characterized. In this study, we investigated the DIR−protein−encoding genes in Saccharum spp. (ScDIR) by screening collections of sugarcane databases, monitoring the responses of these genes to drought stress by real−time quantitative PCR, and identifying their heterologous expression in tobacco. Of the 64 ScDIRs identified, four belonging to the DIR−b/d (ScDIR5 and ScDIR11) and DIR−c (ScDIR7 and ScDIR40) subfamilies showed a significant transcriptional response when subjected to drought stress. ScDIR5, ScDIR7, and ScDIR11 are localized in the cell membrane, whereas ScDIR40 is found in the cell wall. The overexpression of these ScDIR genes in tobacco generally increased the drought tolerance of the transgenic lines, with ScDIR7 conferring the highest degree of drought tolerance. The characterization of the physiological and biochemical indicators (superoxide dismutase, catalase, malondialdehyde, and H2O2) confirmed that the ScDIR−overexpressing lines outperformed the wild type. These results demonstrated that specific ScDIRs in sugarcane respond and contribute to tolerance of drought stress, shedding light on potential means of improving drought tolerance in this crop.
Overexpression of Rice NAC Gene SNAC1 Improves Drought and Salt Tolerance by Enhancing Root Development and Reducing Transpiration Rate in Transgenic Cotton

Abstract:The SNAC1 gene belongs to the stress-related NAC superfamily of transcription factors. It was identified from rice and overexpressed in cotton cultivar YZ1 by Agrobacterium tumefaciens-mediated transformation. SNAC1-overexpressing cotton plants showed more vigorous growth, especially in terms of root development, than the wild-type plants in the presence of 250 mM NaCl under hydroponic growth conditions. The content of proline was enhanced but the MDA content was decreased in the transgenic cotton seedlings under drought and salt treatments compared to the wild-type. Furthermore, SNAC1-overexpressing cotton plants also displayed significantly improved tolerance to both drought and salt stresses in the greenhouse. The performances of the SNAC1-overexpressing lines under drought and salt stress were significantly better than those of the wild-type in terms of the boll number. During the drought and salt treatments, the transpiration rate of transgenic plants significantly decreased in comparison to the wild-type, but the photosynthesis rate maintained the same at the flowering stage in the transgenic plants. These results suggested that overexpression of SNAC1 improve more tolerance to drought and salt in cotton through enhanced root development and reduced transpiration rates.

Drought Tolerance Evaluation of Tobacco Plants Transformed with Different Set of Genes under Laboratory and Field Conditions

Abstract:Although extensive studies have demonstrated that many drought-responsive genes confer drought tolerance to plants, comparisons of the drought tolerance capabilities conferred by different genes under various natural conditions have seldom been reported. We evaluated and compared the effects of two sets of transgenes, the drought-responsive genes (AtDREB1B and AtCBL1) and the root architecture-regulated genes (iaaM and AtCKX), on drought tolerance in Nicotiana tabacum plants subjected to different conditions. The expression of AtCKX3 driven by a root-specific promoter PYK10 (designated hereafter as P10; P10:AtCKX3), 35S:AtCKX3, or P10:iaaM promoted root growth and development. Compared to plants harboring P10:AtCKX3, 35S:AtCKX3, P10:iaaM, or the empty vector, those carrying 35S:AtDREB1B, 35S:AtCBL1, or 35S:iaaM exhibited increased drought tolerance under laboratory-controlled conditions. Conversely, in field conditions, plants transformed with 35S:AtDREB1, 35S:AtCBL1, or 35S:iaaM were sensitive to drought stress. Under field conditions, drought stress dramatically reduced the growth and seed production of plants harboring 35S:AtDREB1B, 35S:AtCBL1, 35S:iaaM, or the empty vector, whereas it had little effect on plants carrying P10:AtCKX3, 35S:AtCKX3, or P10:iaaM. This study demonstrates that a plant’s tolerance to drought stress changes with environmental conditions, and our results indicates that manipulating the expression of genes that control root architecture may be important for engineering plants with improved drought tolerance in natural conditions.

Abstract:Water stress is by far the leading environmental stress limiting crop yields worldwide. Genetic engineering techniques hold great promise for developing crop cultivars with high tolerance to water stress. In this study, the Brassica oleracea var. acephala BoRS1 gene was transferred into tobacco through Agrobacterium-mediated leaf disc transformation. The transgenic status and transgene expression of the transgenic plants was confirmed by polymerase chain reaction (PCR) analysis, Southern hybridization and semi-quantitative one step RT-PCR analysis respectively. Subsequently, the growth status under water stress, and physiological responses to water stress of transgenic tobacco were studied. The results showed that the transgenic plants exhibited better growth status under water stress condition compared to the untransformed control plants. In physiological assessment of water tolerance, transgenic plants showed more dry matter accumulation and maintained significantly higher levels of leaf chlorophyll content along with increasing levels of water stress than the untransformed control plants. This study shows that BoRS1 is a candidate gene in the engineering of crops for enhanced water stress tolerance.
Analysis of drought and salt-alkali tolerance in tobacco by overexpressing WRKY39 gene from Populus trichocarpa

Abstract:WRKY is one of the largest families of transcription factors in plants. It not only regulates plant growth and development but also participates in the regulation of plant defense against biological and abiotic stresses. In this study, research was aimed to overexpress WRKY39 gene of P. trichocarpa (PtWRKY39) and to identify its important role played in drought and saline-alkali tolerance in tobacco model plant. Under the control of CaMV35S promoter, the overexpression of PtWRKY39 gene was increased to more than 10 times in T3 generation of transgenic tobacco plant. The drought resistance and saline-alkali tolerance were evidenced in overexpressed PtWRKY39 transgenic lines at germination/seedling stage. The overall germination rate, fresh weight, and chlorophyll contents of overexpressed lines were significantly higher while the level of malondialdehyde was significantly lower in PtWRKY39 transgenic lines than that of wild type (WT) lines. The content of H2O2 in leaves was detected by the 3, 3-Diaminobenzidine method showed that the overexpression of PtWRKY39 gene could reduce the accumulation of ROS (mainly H2O2) and enhance salt-alkali tolerance. Phenotypic analysis at 7-leaf pot transgenic seedlings stage treated with the saline-alkali soil extract and salt NaCl under root irrigation stress, revealed growth of the transgenic line was significantly higher than that of WT. This work concludes that overexpression of PtWRKY39 gene can improve the regulation of drought resistance and saline-alkali tolerance of transgenic plants during seed germination and vegetative growth.
