Abstract:Electrospinning is a very effective method to fabricate nanofibers. This technique has been achieved more attention due to its cheap cost, simple device and obtaining nanofibers similar to extracellular matrix.Silk fibroin(SF) is a natural protein with good biodegradability and low immunogenicity, Therefore, it can be widely applied in tissue engineering. In this paper, we summarize the research of electrospun silk fibrion nanofibers in regenerative medicine. Furthermore, the future research work of SF nanofibers was suggested.
Integration of mesenchymal stem cell sheet and bFGF-loaded fibrin gel in knitted PLGA scaffolds favorable for tendon repair

Abstract:Tendon injuries are common and require a long time to heal, and are particularly associated with some adverse problems such as adhesion and rupture. Herein, we aim to develop new bioactive scaffolds endowed with stem cell sheets and growth factors to enable cell migration and proliferation favorable for tendon regeneration in situ. An exogenous basic fibroblast growth factor (bFGF)-loaded fibrin gel was firstly incorporated into the porous network of knitted poly(lactide-co-glycolide) (PLGA) scaffolds and then sheets of mesenchymal stem cells (MSCs) were also integrated into the scaffolds. It was shown that the pores in the knitted PLGA scaffold were readily filled with a complex network of fibrin fiber gel and the fibrin fibers were beneficial for the controlled release of bFGF over a long time period. After transplantation in a critical-size Achilles tendon defect model (7 mm) in the rat right hindlimb, gross observation revealed no immunologic incompatibility or rejection derived from the scaffold systems. It was observed that the MSC sheets contributed directly to tendon regeneration, and exerted an environment-modifying effect on the injuries in situ, consistent with the beneficial effect of bFGF. It was interesting that the knitted PLGA-fibrin gel scaffolds loaded with MSC sheets and bFGF showed the highest expression of tendon-related gene markers and outstanding repair efficacy, including appreciable biomechanical strength and native-like histological microstructures. Therefore, the integration of MSC sheets and bFGF into PLGA/bFGF-fibrin gel scaffolds may stimulate the proliferation and tenogenic differentiation of MSCs in situ and synergistically enhance the injured tendon reconstruction.
Dual-layer Aligned-Random Nanofibrous Scaffolds for Improving Gradient Microstructure of Tendon-to-bone Healing in a Rabbit Extra-Articular Model

Abstract:Background: Tendon/ligament injuries are common sports injuries. Clinically, the repair of a ruptured tendon or ligament to its bony insertion is needed, but the enthesis structure is not well reestablished following surgical repair. Herein, we fabricated dual-layer aligned-random scaffold (ARS) by electrospinning and aimed to investigate the effect of the scaffold on tendonto-bone healing in vivo. Materials and methods: The random and dual-layer aligned-random silk fbroin poly(L-lactic acid-co-e-caprolactone) (P(LLA-CL)) nanofibrous scaffolds were successfully fabricated by electrospinning methods. Ninety New Zealand white rabbits were randomly divided into three groups (random scaffold [RS], ARS, and control groups), and they were subjected to surgery to establish an extra-articular tendon-to-bone healing model with autologous Achilles tendon. Results: Histological assessment showed that the ARS significantly increased the area of metachromasia, decreased the interface width, and improved collagen maturation and organization at the tendon bone interface compared with the RS and control groups. Microcomputed tomography analysis showed that the bone tunnel area of RS and ARS groups was significantly smaller than those of the control group. Real-time polymerise chain reaction showed that BMP-2 and osteopontin expression levels of the tissue at the interface between the bone and graft in the RS and ARS groups were higher than those of the control group at 6 weeks. Collagen I expression level of the ARS group was significantly higher than those of the RS and control groups at 6 and 12 weeks. Moreover, the ARS groups had a better ultimate load-to-failure and stiffness than the RS and control groups. Conclusion: ARS could effectively augment the tendon-to-bone integration and improve gradient microstructure in a rabbit extra-articular model by inducing the new bone formation, increasing the area of fibrocartilage, and improving collagen organization and maturation. The dual-layer aligned-random silk fibroin/P(LLA-CL) nanofibrous scaffold is proved to be a promising biomaterial for tendon-to-bone healing.

Abstract:The unique amino acid composition and spatial structure of silk fibroin make it an excellent biomaterial with perfect biocompatibility and biodegradability.Silk fibroin nanofibers and composite fibroin nanofibers prepared via high voltage electrostatic spinning technology are conferred further superior property for application as biomedical materials.This review systematically introduces advances in application and modification of the electrospun silk fibroin nanofibers and composite fibroin nanofibers in bone tissue engineering,in repair of cartilage,wound,nerve and blood vessel,and in controlled release of drugs in recent years,based on which key innovations for future development of electrospun silk fibroin nanofiber materials are briefly discussed.
Preparation of Silk Fibroin Nanofiber Scaffold and Its Application in Tendon-Bone Healing and Sports Rehabilitation

Abstract:ABSTRACT In this work, polylactic acid/polycaprolactone/silk fibroin (PLA/PCL/SF) nanofiber scaffolds with different mass ratios were prepared by electrospinning technology. The morphology and structure of the nanofiber scaffold were characterized with the scanning electron microscope (SEM), and its porosity and adsorption were tested using the Fourier transform infrared spectrometer (FTIR). 24 New Zealand white rabbits were rolled into control group ( n = 8, with autologous tendons) and experimental group ( n = 16) randomly. The PLA/PCL/SF nanofiber scaffolds were adopted to wrap autologous tendons to establish extra-articular models. Tendon-bone healing was evaluated six weeks after surgery through histological and biomechanical tests, and the related gene expressions in tissue cells were detected. It turned out that mass ratio of PLA/PCL and SF components had a considerable impact on the morphology of the nanofiber scaffold. The surface of nanofiber with a mass ratio of 3:1 was distributed with dense pores. As the content of SF increased, the porosity and adsorption of the nanofiber scaffold gradually decreased. Moreover, the experimental results suggested that the addition of SF improved the hydrophilicity of PLA/PCL/SF scaffold, which was beneficial to the adhesion and proliferation of NIH/3T3 (a mouse embryonic fibroblast cell line established by the National Institutes of Health (NIH)). In addition, histological observation results showed that the width of the tendon-bone interface (TBI) of rabbits in control group was still relatively large at the 6th week after the surgery, with poor healing effect and disordered collagen arrangement. The widths of the TBI of the material group and rehabilitation group were substantially narrower relative to that in control group, and the collagen was arranged regularly. It was suggested that the healing effects between tendon and bone in material group and rehabilitation group were accelerated, and the effect in the rehabilitation group was superior to that of material group, indicating that rehabilitation exercise could organize the negative effects of training in postoperative rehabilitation training and promote the healing between tendons and bones.

Abstract:In the realm of skeletal muscle tissue engineering, anisotropic materials that emulate natural tissues show substantial promise. Electrospun scaffolds, mimicking the fibrillar structure of the extracellular matrix, are commonly employed but often fall short in achieving optimal alignment and mechanical strength. Silk fibroin has emerged as a versatile material in tissue engineering, valued for its biocompatibility, mechanical robustness, and biodegradability. However, conventional electrospinning methods of SF result in randomly oriented fibers, limiting their efficacy. In this work, we developed a straightforward method to fabricate directional tissue scaffolds using silk fibroin. By integrating a magnetic field collecting device and incorporating Fe3O4 nanoparticles into the spinning solution, we successfully produced well-aligned silk nanofiber scaffolds. These aligned fibers not only improved scaffold orientation and mechanical properties but also exhibited magnetic responsiveness. The aligned SF scaffolds effectively guided the adhesion, proliferation, and differentiation of mesenchymal stem cells along the fiber direction. Cultured on these scaffolds, myoblast C2C12 cells demonstrated oriented growth, highlighting the potential of aligned SF fibers in advancing skeletal muscle engineering for biomedical applications.

Abstract:In this study, electrospinning was used to fabricate silk-fibroin (SF)-based mats, which served as substrates for the culturing of rat Schwann cells. Microscopic observation and physical parameter measurements revealed that the electrospun SF mats had a nanofibrous structure with favorable physical properties. Fourier transform infrared analysis provided chemical characterization of the molecular confirmation of the SF proteins in the mats. The morphology and immunocytochemistry showed that the mats supported the survival and growth of the cultured Schwann cells, and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide analysis indicated that the electrospun SF mat extract had no cytotoxic effects on Schwann cell proliferation. Collectively, all of the results suggest that the electrospun SF mats might become a candidate scaffold for tissue-engineered nerve grafts to promote peripheral nerve regeneration. Abstract:This study developed a bioactive knitted silk-collagen sponge scaffold by incorporation of exogenous SDF-1 alpha, to enable selective migration and homing of cells for in situ tendon regeneration. With in vitro studies, it was observed that CXCR4 gene expression and migration of bone mesenchymal stromal cells and hypo-dermal fibroblasts were more sensitive to exogenous SDF-1 alpha, while expression of tendon repair gene markers by hypo-dermal fibroblasts and Achilles tendon fibroblasts were more sensitive to exogenous SDF-1 alpha. With a rat Achilles tendon injury model, exogenous SDF-1 alpha was shown to reduce infiltration of inflammatory cells and enhance migration of fibroblast-like cells into the scaffold at 4 days and 1 week post-surgery. After 4 weeks, SDF-1 alpha treated tendon had increased expression of tendon repair gene markers and endogenous SDF-1 alpha, exhibited more physiological microstructures with larger diameter collagen fibrils, and had better biomechanical properties than the control group. Hence, our bioactive scaffold improved efficacy of tendon regeneration by increasing the recruitment of fibroblast-like cells, enhancing local endogenous SDF-1 alpha and tendon extracellular matrix production, and decreasing accumulation of inflammatory cells. Incorporation of SDF-1 alpha within a knitted silk-collagen sponge scaffold can therefore be a practical application for tendon tissue engineering.

Abstract:Silk fibroin (SF)-derived silkworms represent a type of highly biocompatible biomaterial for tissue engineering. We have previously investigated biocompatibility of SF with neural cells isolated from the central nervous system or peripheral nerve system in vitro, and also developed a SF-based nerve graft conduit or tissue-engineered nerve grafts by introducing bone marrow mesenchymal stem cells, as support cells, into SF-based scaffold and evaluated the outcomes of peripheral nerve repair in a rat model. As an extension of the previous study, the electrospun technique was performed here to fabricate SF-based neural scaffold inserted with silk fibres for bridging a 30-mm-long sciatic nerve gap in dogs. Assessments including functional, histological and morphometrical analyses were applied 12 months after surgery. All the results indicated that the SF-based neural scaffold group achieved satisfactory regenerative outcomes, which were close to those achieved by autologous nerve grafts as the golden-standard for peripheral nerve repair. Overall, our results raise a potential possibility for the translation of SF-based electrospun neural scaffolds as an alternative to nerve autografts into the clinic.

Abstract:The objective of the present study was to perform an in vivo assessment of a novel silk-collagen scaffold for anterior cruciate ligament (ACL) reconstruction. First, a silk-collagen scaffold was fabricated by combining sericin-extracted knitted silk fibroin mesh and type I collagen to mimic the components of the ligament. Scaffolds were electron-beam sterilized and rolled up to replace the ACL in 20 rabbits in the scaffold group, and autologous semitendinosus tendons were used to reconstruct the ACL in the autograft control group. At 4 and 16 weeks after surgery, grafts were retrieved and analyzed for neoligament regeneration and tendon-bone healing. To evaluate neoligament regeneration, H&E and immunohistochemical staining was performed, and to assess tendon-bone healing, micro-CT, biomechanical test, H&E and Russell-Movat pentachrome staining were performed. Cell infiltration increased over time in the scaffold group, and abundant fibroblast-like cells were found in the core of the scaffold graft at 16 weeks postoperatively. Tenascin-C was strongly positive in newly regenerated tissue at 4 and 16 weeks postoperatively in the scaffold group, similar to observations in the autograft group. Compared with the autograft group, tendon-bone healing was better in the scaffold group with trabecular bone growth into the scaffold. The results indicate that the silk-collagen scaffold has considerable potential for clinical application.

Abstract:Tissue engineering is an ideal therapeutic strategy for the development of functional tendon replacement tissue for tendon repair in the clinic. Currently, the synergistic roles of mechano-chemical factors and the mechanisms involved in tendon repair and regeneration are not fully understood. In this study, we developed a three-dimensional (3D) culture system based on a silicone chamber and collagen sponge scaffold that can deliver cyclic mechanical stretch and biochemical stimulation to bone marrow-derived mesenchymal stem cells (BMSCs) seeded on the scaffold. We found that the combined stimulation of cyclic stretch and transforming growth factor beta 1 (TGF-β1) treatment not only increased cell viability but also synergistically promoted the differentiation of BMSCs into tenocytes in a 3D culture environment. Meanwhile, the combined stimulation increased the Young’s modulus of the BMSC-collagen sponge constructs by reducing the porosity of the scaffold compared to the non-treated constructs. Furthermore, a rat Achilles tendon in situ repair experiment showed that enhanced tendon regeneration was achieved using the BMSC-collagen sponge construct combined with cyclic stretch and TGF-β1, as confirmed by Achilles functional index (AFI) measurement, morphological observation, histological analysis, and mechanical testing. These results suggest that this approach could offer a practical benefit in tendon healing and future tendon tissue engineering.
Silk-Based Scaffold for Ligament Tissue Engineering

Abstract:In recent years, silk has been increasingly studied as the scaffold for ligament tissue engineering due to the biocompatibility, slow degradability, and remarkable mechanical properties. Braided silk scaffold modified with short polypeptide also significantly increases collagen synthesis on it. To increase cell attachment and tissue infiltration, the braided scaffold can be incorporated with silk-gelatin microsponges. A novel silk cable-reinforced gelatin/silk fibroin hybrid scaffold was fabricated, which, apart from providing proper mechanical strength and enlarged surface area, also supported the proliferation and differentiation of MSCs on it. The knitted silk mesh is another important silk-based scaffold for its excellent mechanical properties and good nutrients transport. To prevent cells from leaking out of scaffold after seeding, freeze-dried silk microsponges were incorporated into the macro pores of knitted scaffold. In vitro culture demonstrated that MSCs on scaffolds proliferated vigorously and produced abundant collagen. The transcription levels of ligament-specific genes (collagen I, collagen III, and tenascin-C) also increased significantly with time. The comparison of MSCs and fibroblasts as cell sources for ligament tissue engineering demonstrated that MSC was the most suitable candidate for its vigorous proliferation and ECM production. The MSCs/knitted scaffolds were implanted into rabbits to regenerate ACL in vivo. After 24 weeks, histology observation showed that MSCs were distributed throughout the regenerated ligament and exhibited fibroblast morphology. The key ligament ECM components including collagen I, collagen III, and tenascin-C were produced prominently. Furthermore, direct ligament-bone insertion with typical four zones (bone, mineralized fibrocartilage, fibrocartilage, ligament) was reconstructed, which resembled the native structures of ACL-bone insertion. The tensile strength of regenerated ligament also met the mechanical requirements of daily activities. In conclusion, the results imply that silk scaffold has great potentials in future clinical applications.
Carbon Fiber-Mediated Electrospinning Scaffolds Can Conduct Electricity for Repairing Defective Tendon

Abstract:Partial or complete rupture of the tendon can damage the collagen structure, resulting in the disruption of the electrical signal pathway. It is a great challenge to reconstruct the original electrical signal pathway of the tendon and promote the regeneration and functional recovery of defective tendon. In this study, carbon fiber-mediated electrospinning scaffolds were fabricated by wrapping conductive, high-strength, loose single-bundle carbon fibers with nanofiber membranes. Due to the presence of nanofiber membranes, the maximum tensile force of the scaffolds was 2.4 times higher than that of carbon fibers, while providing excellent temporal and spatial prerequisites for tenocytes to adapt to electrical stimulation to accelerate proliferation and expression. The diameter of the carbon fiber monofilaments used in this study was 5.07 ± 1.20 μm, which matched the diameter of tendon collagen, allowing for quickly establishing the connection between the tendon tissue and the scaffold, and better promoting the recovery of the electrical signal pathway. In a rabbit Achilles tendon defect repair model, the carbon fiber-mediated electrospinning scaffold was almost filled with collagen fibers compared to a nonconductive polyethylene glycol terephthalate scaffold. Transcriptome sequencing revealed that fibromodulin and tenomodulin expression were upregulated, and their related proteoglycans and glycosaminoglycan binding proteins pathways were enhanced, which could regulate the TGF-β signaling pathway and optimize the extracellular matrix assembly, thus promoting tendon repair. Therefore, the scaffold in this study makes up for the shortage of conductive scaffolds for repairing tendon defects, revealing the potential impact of conductivity on the signaling pathway of tendon repair and providing a new approach for future clinical studies.
Biomimetic Triphasic Silk Fibroin Scaffolds Seeded with Tendon-Derived Stem Cells for Tendon-Bone Junction Regeneration.

Abstract:The regeneration of tendon and bone junctions (TBJs), a fibrocartilage transition zone between tendons and bones, is a challenge due to the special triphasic structure. In our study, a silk fibroin (SF)-based triphasic scaffold consisting of aligned type I collagen (Col I), transforming growth factor beta (TGF-beta), and hydroxyapatite (HA) was fabricated to mimic the compositional gradient feature of the native tendon-bone architecture. Rat tendon-derived stem cells (rTDSCs) were loaded on the triphasic SF scaffold, and the high cell viability suggested that the scaffold presents good biocompatibility. Meanwhile, increased expressions of tenogenic-, chondrogenic-, and osteogenic-related genes in the TBJs were observed. The in vivo studies of the rTDSC-seeded scaffold in a rat TBJ rupture model showed tendon tissue regeneration with a clear transition zone within 8 weeks of implantation. These results indicated that the biomimetic triphasic SF scaffolds seeded with rTDSCs have great potential to be applied in TBJ regeneration. The regeneration of tendon and bone junction (TBJ), a fibrocartilage transition zone between tendons and bones, is a challenge due to the special triphasic structure. Abstract:The tendon-bone junction (TBJ), a critical transitional zone where tendons and bones connect, is particularly prone to injury due to the forces from muscle contractions and skeletal movements. Once tendon-bone injuries occur, the complex original tissue structure is difficult to restore, increasing the risk of re-tear. In this study, we initially established a rabbit model of tendon-bone injury and treated it using either interference screw or suture anchor. Biomechanical testing demonstrated the maximum tension and strength of TBJ with interference screw fixation were superior. However, histologic and immunohistochemical results showed more tissue regeneration and expression of cartilage markers at the site of injury with suture anchor fixation. Moreover, Gelatin Methacryloyl encapsulated with exosomes from mesenchymal stem cell (GelMA-exosomes) were prepared, showing a consistent and stable exosome release characteristic. The combined application of GelMA-exosomes with either interference screws or suture anchors further enhanced the healing of tendon-bone injuries, which may be achieved by promoting cellular proliferation as well as regulating the decreased expression of local pro-inflammatory factors IL-1β, IL-6 and TNF-α and increased expression of anti-inflammatory factors IL-10 and TGF-β. This provides a viable therapeutic strategy to enhance tendon-bone healing.

Abstract:The repair of tendon to bone junction (TBJ) remains a tremendous challenge in tissue engineering due to the complicated structure, components, mechanical properties, and cell types. In order to reconstruct the tissue and restore its functionality, biomedical scaffolds with hierarchical and gradient structures have been fabricated by various strategies. In recent decades, electrospinning has become one of the most popular methods in fabricating TBJ scaffolds due to easy fabrication, high porosity, and ECM-like nano-scale structure. However, mechanical properties are the pain point of electrospun biomedical scaffolds. Traditional textile technology can be exploited to compensate for this weakness, which will be deeply discussed here. This review will start with a brief introduction to the structure and function of the native TBJ tissue and a short overview of electrospinning technology. Then, different electrospun biomedical scaffolds for TBJ repair will be summarized and compared. Furthermore, some advanced technologies and modification methods in fabricating functionalized electrospun TBJ scaffolds are discussed. In the end, current challenges and solutions are being proposed, which would provide instruction for the research of electrospun textile TBJ scaffolds.Graphical Abstract

Synergic Combination Of Collagen Matrix With Knitted Silk Scaffold Regenerated Ligament With More Native Microstructure In Rabbit Model

Abstract:A number of researches on ligament and tendon tissue engineering scaffold have been carried out. However, a scaffold which simultaneously possesses optimal strength, porous structure and biocompatible microenvironment is yet to be developed. This study aims to design a new practical ligament scaffold by synergic incorporation of silk material, knitting structure and collagen matrix.Physical properties and biocompatibility of the knitted silk-collagen sponge scaffolds were evaluated in vitro and in mice model. Their efficacy for in situ ligament tissue engineering was investigated in New Zealand White rabbit model with a 6-mm gap defect of the medial collateral ligament (MCL). The histology, microstructure, biochemical composition, matrix genes expression and strength of the ligament repair of different groups were evaluated at 2, 4 and 12 weeks.In vitro data show that cells on collagen matrix expressed higher collagen I and decorin genes at 3 and 5 days than that on silk substrate. The study in mice model demonstrated that silk scaffold caused little inflammatory reaction after subcutaneous implantation, the strength of scaffolds decreased to 25.9% of original value at 3 month and increased to 33.5% at 6 month. In the MCL repair study, silk-collagen treated MCLs had more connective bands of collagen matrix formed and more collagen deposition compare to those in silk treated MCLs at 2 4 and 12 weeks. Moreover, silk-collagen treated MCLs possessed better mechanical property, larger diameter of collagen fibrils and stronger scaffold-ligament interface healing at 12weeks.These results proved that the knitted silk-collagen sponge scaffold possessed optimal internal space, biocompatible environment and mechanical strength. It could improve the structure and function of ligament repair by regulating ligament matrix genes expression and collagen fibril assembly. The findings suggested that knitted silk-collagen sponge scaffold is a promising candidate scaffold for practical ligament and tendon tissue engineering.
Structure and ingredient-based biomimetic scaffolds combining with autologous bone marrow-derived mesenchymal stem cell sheets for bone-tendon healing

Abstract:Tendon attaches to bone across a robust fibrocartilaginous tissue termed the bone-tendon interface (BTI), commonly injured in the field of sports medicine and orthopedics with poor prognosis. So far, there is still a lack of effective clinical interventions to achieve functional healing post BTI injury. However, tissue-engineering may be a promising treatment strategy. In this study, a gradient book-type triphasic (bone-fibrocartilage-tendon) scaffold is fabricated based on the heterogeneous structure and ingredient of BTI. After decellularization, the scaffold exhibits no residual cells, while the characteristic extracellular matrix of the original bone, fibrocartilage and tendon is well preserved. Meanwhile, the bone, fibrocartilage and tendon regions of the acellular scaffold are superior in osteogenic, chondrogenic and tenogenic inducibility, respectively. Furthermore, autologous bone marrow mesenchymal stem cell (BMSC) sheets (CS) combined with the acellular scaffolds is transplanted into the lesion site of a rabbit BTI injury model to investigate the therapeutic effects. Our results show that the CS modified scaffold not only successfully achieves triple biomimetic of BTI in structure, ingredient and cell distribution, but also effectively accelerates bone-tendon (B-T) healing. In general, this work demonstrates book-type acellular triphasic scaffold combined with autologous BMSCs sheets is a promising graft for repairing BTI injury.

Abstract:The regeneration of tendon-bone interface tissue has become a topic of great interest in recent years. However, the complex nature of this interface has posed challenges in finding suitable solutions. Tissue engineering, with its potential to improve clinical outcomes and play a crucial role in musculoskeletal function, has been increasingly explored for tendon-bone interface regeneration. This review focuses on the research advancements of electrospinning technology in interface tissue engineering. By utilizing electrospinning, researchers have been able to fabricate scaffolds with tailored properties to promote the regeneration and integration of tendon and bone tissues. The review discusses the unique structure and function of the tendon-bone interface, the mechanisms involved in its healing, and the limitations currently faced in achieving successful regeneration. Additionally, it highlights the potential of electrospinning technology in scaffold fabrication and its role in facilitating the development of functional and integrated tendon-bone interface tissues. Overall, this review provides valuable insights into the application of electrospinning technology for tendon-bone interface tissue engineering, emphasizing its significance in addressing the challenges associated with regeneration in this complex interface.
