Abstract:OBJECTIVE:To investigate the association that the polymorphisms of interleukin 10 gene (IL10) promoter region are related to the susceptibility and clinical phenotypes of hepatitis B virus (HBV) in Chinese Han population. METHODS:With polymerase chain reaction with restriction fragment length polymorphism (PCR-RFLP) method, the single nucleotide polymorphisms (SNP) of the promoter region of IL10 gene at position -1082G/A, -819T/C, -592A/C were detected in 231 patients with chronic hepatitis B, 165 individuals spontaneously recovered from HBV infection and 135 normal controls. RESULTS:No significant difference was found in frequencies of genotypes and alleles of IL10 gene promoter region at position -1082G/A, -819T/C, -592A/C among normal controls, individuals spontaneously recovering from HBV infection and patients with chronic hepatitis B (P>0.05), also between patients with HBV infection with HBV-DNA<1x10(3)copies/mL and those with HBV-DNA> or =1 x 10(3)copies/mL (P>0.05). However, frequencies of TT genotype at position -819T/C and AA genotype at position -592A/C in chronic hepatitis B were significantly higher than that in asymptomatic HBV carriers (P<0.05). CONCLUSION:It is possible that genetic polymorphisms of IL10 promoter region are not associated with both susceptibility of HBV infection and HBV-DNA replication after infected HBV in Chinese Han population. However, the polymorphisms of the promoter region IL10 at position -819T/C and -592A/C are related to inflammatory reaction to liver of the patients with HBV infection.
Study on the Association of IL-10-592 Polymorphism with Susceptibility to Hepatitis B Viral Infection in Han, Yi and Yao Ethnic Groups in Guizhou Province

Abstract:OBJECTIVE:To investigate the association of IL-10 gene promoter polymorphism with susceptibility to hepatitis B viral infection in Han, Yi and Yao ethnic groups from Guizhou province.METHODS:Five hundred volunteers from Guizhou province were selected to undertake PCR-RFLP for detection of IL-10 gene promoter -592 polymorphism.RESULTS:The genotypic distributions of IL-10-592 were 32.53%-51.43% (AA), 40.74%-54.82% (AC), 5.79%-18.52% (CC) whereas the allelic frequencies were 59.94%-72.38% for the A allele, and 27.62%-40.06% for the C allele in Han, Yi and Yao ethnic from Guizhou. The distributions of allele and genotype frequencies of IL-10-592 were statistically different between Yao ethnic in Libo and Yi ethnic in Qianxi, Yao ethnic in Libo and Han ethnic in Libo,Yi ethnic in Qianxi and Yi ethnic in Weining,Yi ethnic in Weining and Han ethnic in Libo (P < 0.05). IL-10-592 polymorphism was associated with HBV infection in Yi ethnic in Qianxi and the whole population.CONCLUSION:IL-10-592 gene polymorphisms influenced the susceptibility to HBV infection in Han, Yao, Yi sub-populations in Guizhou. Result of the study suggested that IL-10-592 gene polymorphisms might serve as a risk factor to HBV infection.
Interleukin-10 Promoter Polymorphisms in Patients with Hepatitis B Virus Infection or Hepatocellular Carcinoma in Chinese Han Ethnic Population.

Abstract:BACKGROUND:Since single nucleotide polymorphisms (SNPs) can serve as gene markers, polymorphism profiles may help scientists to identify the full collection of genes that contribute to the development of complex diseases such as cancer. The distribution of interleukin-10 (IL-10) promoter polymorphisms in Chinese Han ethnic patients with hepatitis B virus (HBV) infection and hepatocellular carcinoma (HCC) was investigated in this study.METHODS:The polymorphisms of IL-10 promoter region were detected by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and sequencing. Sixty-six health controls, 42 patients with HBV infection, 30 HCC patients, and cell line SMMC-7721 were examined this way.RESULTS:Polymorphisms of T/C or T/N on -872 site occurred frequently in Han ethnic population. Polymorphisms were detected in HBV and HCC patients and cell line SMMC-7721. The hotspot among the polymorphisms was inserting base A between -1058 and -1057.CONCLUSION:Polymorphisms of IL-10 promoter in HBV and HCC patients may be associated with HBV infection and HCC development.

Abstract:To investigate whether the polymorphisms in TNF-alphagene, IL-10 gene and IL-6 gene were associated with the outcomes of chronic HBV infection.The genotypes at the position -238 and -308 of TNF-alphagene, -1082, -819 and -592 of IL-10 gene, -174 of IL-6 gene in the promoter region were detected in 62 chronic hepatitis B patients, 60 asymptomatic carriers and 63 heath controls by polymerase chain reaction-sequence specific primer assay.The frequency of G/G genotype at the position -238 in the promoter region of TNF-alphagene increased significantly in hepatitis B patients, in comparison with that in controls (83.9% versus 66.7%, P<0.05), whereas the frequency of G/A genotype at the same position decreased significantly in hepatitis B patients comparing with that in controls (16.1% versus 33.3%, P<0.05). Between chronic hepatitis B patients, asymptomatic carriers and heath controls, the genotypes frequency at the position -308 of TNF-alphagene, -1082, -819 and -592 of IL-10 gene, -174 of IL-6 gene in the promoter region showed no significant differences (P>0.05).The presence of G/A genotype at the position -238 may contribute to virus elimination, whereas the presence of G/G genotype at the position -238 may lead to virus persistent infection. There were no association between IL-10 gene and IL-6 gene polymorphisms and chronic HBV infection.

Abstract:Summary. Recent reports indicated that high levels of interleukin 10 (IL‐10) contribute to the monocytes paralysis and poor clinical outcome in acute liver failure (ALF). Polymorphisms in the promoter region of IL‐10 affect IL‐10 production and confer susceptibility to inflammatory diseases. The aim of this study was to determine the possible association of the three polymorphisms (A‐1082G, T‐819C, A‐592C) in the IL‐10 gene promoter with the susceptibility to hepatitis B virus (HBV)‐related ALF in a Chinese population. The IL‐10 gene promoter polymorphisms were genotyped in 414 unrelated healthy blood donors, 367 asymptomatic HBV carriers and 345 HBV‐related ALF patients. Functional analyses were conducted to verify the biological significances of the associated genetic variations. The allele frequencies of IL‐10 −592C and −819C were significantly higher in HBV‐related ALF patients than in blood donors and asymptomatic HBV carriers. Logistic regression analysis and stratification analysis with adjustment for age and sex indicated that the polymorphisms of A‐592C and T‐819C were associated with susceptibility to HBV‐related ALF ( P = 6.9 × 10 −7 ), and the ‐1082A‐819C‐592C haplotype in the IL‐10 gene promoter were associated with an increased susceptibility to ALF in HBV carriers (dominant model, P = 0.0002, odds ratio = 1.60, 95% CI 1.25–2.07). Functional analyses showed that the A‐592C polymorphism is a nuclear proteins binding site, and the disease susceptible −592C allele had a higher transcription activity compared with −592A allele. This study emphasizes the importance of IL‐10 in the pathophysiology of HBV‐related ALF on the population level.

Abstract:Objective To investigate the association of IL-10 592 and ESR1 T29 C gene polymorphism with the susceptibility to hepatitis B virus in Yi minority of Guizhou.Methods Allelic frequencies of IL-10 592 and ESR1 T29 C were identified by PCR-RFLP.Results IL-10 592 polymorphism was associated with the susceptibility to HBV in Yi minority.There wasno association between ESR1 T29 C gene polymorphism and the susceptibility to hepatitis B virus in Yi minority.Conclusion IL-10 592 polymorphisms influence the susceptibility to HBV infection in Yi minority of Guizhou.The study suggests that IL-10 592 polymorphism may be a dangerous factor of HBV infection.

Abstract:OBJECTIVE:To investigate the association between interleukin-10 (IL-10) gene promoter microsatellite polymorphisms and the susceptibility to hepatitis B virus infection in Han, Yi and Yao ethnicities in GuiZhou province. METHODS:500 volunteers were selected from Guizhou province. Allelic frequency of IL-10.G and IL-10.R loci was identified by short tandom repeat polymerase chain reaction. The relativity between allelic frequency and HBV infection was analyzed. RESULTS:Genotype data from H-W analysis on all the IL-10 polymorphisms indicated that it was a random distribution. Very high HBV infection rates were found in the native ethnic minorities of Guizhou province. The overall HBV infection rate among the total population was 67.00%, with the HBV infection rates of Yi nationality in Weining, Yi nationality in Qianxi, Yao nationality in Libo and Han nationality in Libo as 51.85%, 42.86%, 79.52% and 84.30%, respectively. The polymorphisms distribution of IL-10.G and IL-10.R were statistically different among the ethnic groups (P < 0.05). The polymorphisms distribution of IL-10.R had no significant difference between HBV infection group and non-infection group, as well as among HBV natural removal group and non-infected group in all the ethnic groups. The frequency of IL-10.G 459 bp (19CA) was significantly higher in non-infection group than in the infected group (P < 0.05). The frequency of IL-10.G 471 bp (25CA) was significantly higher in the non-infection group than in the HBV natural removal group (P < 0.05). The polymorphisms distribution of IL-10.G did not show significant difference between the HBV infection group and the HBV natural removal group in all the ethnic groups. We did not find any differences in allelic and genotypic frequencies of IL-10.G between infection group and non-infection group in Yi nationality in Weining, and Yao nationality in Libo (P > 0.05), as well as HBV natural removal group and non-infected group (P > 0.05). CONCLUSION:The polymorphisms distribution of IL-10.R and IL-10.G did not show significant difference in Yi, Yao and Han ethnics population living in Guizhou province. IL-10.G seemed to influence the susceptibility of HBV infection in Han, Yao and Yi ethnics population of Guizhou province.

Abstract:Objectives To investigate the relation between the polyphmorphism of the sites-1082,-592 and-819 in the promoter region of interleukin(IL)-10 gene and susceptibility to hepatitis B virus(HBV) infection.Methods Using ,including interleukin-10,polymorphism,hepatitis B virus and corresponding Chinese words were used to retrieve from the literature,the articles concerning the relationship between interleukin(IL)-10 gene promoter polymorphisms and the susceptibility to HBV infection,published from Jan.1985 to Dec.2007,and those presented in the database CNKI,Medline and ISI.A total of 3 034 cases and 1 289 controls were obtained.Results The presence of AA and AG genotypes at the site-1082 of IL-10 gene promoter region showed no association with the risk of HBV infection(P>0.05),while the presence of GG genotype was significantly associated with a decreased risk for HBV infection(P<0.01).Furthermore,the frequencies of allele A and allele G showed no association with HBV infection(P>0.05).For the site-592,the presence of AA,AC and CC genotypes and the frequencies of alleles A and C showed no relation with the risk of HBV infection(P>0.05).Regarding the site-819,the presence of TT genotype showed no association with the risk of HBV infection(P>0.05).However,the presence of TC genotype was significantly associated with an increased risk of HBV infection(P<0.01).In contrast,the presence of CC genotype was significantly associated with a decreased risk of HBV infection(P<0.01).In addition,alleles T and C were not associated with the risk of HBV infection(P>0.05).Conclusions The presence of GG genotype at the site-1082 and CC genotype at the site-819 are significantly associated with decreased risk of HBV infection,while the presence of TC genotype at the site-819 is associated with an increased risk of HBV infection.The presence of other genotypes and the frequencies of all the alleles are not associated with the risk of HBV infection.

Abstract:Objective Leukocyte mediated IL-6 (IL-6) plays an important role in chronic viral hepatitis pathogenesis and related liver diseases,We did a large sample-size case-control study and clinical data analysis to find association between IL-6 single nucleotide polymorphism and HBV susceptibility,and to achieve the detection of the body on the expression of IL-6 for the prevention and treatment of HBV infection.Methods Totally 848 HBV patients and 894 healthy controls in Shenzhen were selected and rs1800796 genotypes were determined by TaqMan assays.Results The result showed that rs1800796 polymorphism was associated with susceptibility to HBV infection (P =0.0003,odds ratio (OR) =1.43,the difference was statistically significant.Conclusions The single nucleotide polymorphism of IL-6rs1800796 locus was associated with the susceptibility of HBV infection in Chinese population,and the rs1800796 G allele is a protective gene for HBV infection.

Abstract:BACKGROUND:There is growing evidence suggesting that interleukin-18 (IL-18) plays a crucial role in viral clearance and disease pathogenesis, and that single nucleotide polymorphisms (SNPs) within the gene may influence its production.OBJECTIVES:To investigate the potential association of two polymorphisms ( 137G/C and 607C/A) in the promoter region of the IL-18 gene with the risk of HBV recurrence after liver transplantation (LT) in Han Chinese patients.PATIENTS AND METHODS:IL-18 promoter genotyping was performed by the snapshot technique in 125 patients with HBV-related end-stage liver disease (ESLD) receiving LT in our center from 2004 to 2008.RESULTS:Among the study samples, no significant association between the IL-18 promoter polymorphisms ( 137G/C and 607C/A) or haplotypes and HBV recurrence after LT was found.CONCLUSIONS:The polymorphisms ( 137G/C and 607C/A) in the promoter region of the IL-18 gene may not play a key role in HBV recurrence after LT in Han Chinese population, and may not be suitable predictors for HBV recurrence in clinical practice.

Abstract:BACKGROUND:The influence of interleukin-10 (IL-10) gene promoter polymorphisms on the mode and sequel of HBeAg seroconversion (a favorable event usually) in patients with chronic Hepatitis B virus (HBV) infection has not been clarified. PATIENTS AND METHODS:IL-10 genotyping and haplotype analyses of 340 HBsAg carriers and 100 volunteers with self-limiting HBV infection from southern China, a high prevalent area of HBV were performed according to the single nucleotide polymorphisms in its promoter (-1,082, -819 and -592) using a competitively differentiated PCR. RESULTS:High-producer genotype (GG at -1,082) or haplotype (GCC) was rarely found in patients from southern China (<1%). Intermediate-producer haplotype (ACC) was closely associated with chronic liver disease (P=0.004); compared with this, low-producer genotype (AA at -592) and haplotype (ATA) were closely associated with asymptomatic carriers (P=0.035 and 0.035). Intermediate-producer genotype (AC at -592) and haplotype (ACC) were closely associated with covert seroconversion of HBeAg (P=0.0086 and 0.0013) and progressive sequel after HBeAg seroconversion (P=0.013 and 0.0008), while, low-producer genotype (AA at -592) and haplotype (ATA) were closely associated with overt seroconversion of HBeAg (P=0.0023 and 0.0061) and silent sequel after HBeAg seroconversion (P=0.0009 and 0.001). CONCLUSIONS:IL-10 gene promoter polymorphisms significantly influence the mode and sequel of HBeAg seroconversion in patients with chronic HBV infection.
Interleukin-10 Genotype Correlated to Deficiency Syndrome in Hepatitis B Cirrhosis.

Abstract:Traditional Chinese medicine (TCM) syndrome is an important basis for TCM diagnosis and treatment. As Child-Pugh classification as well as compensation and decompensation phase in liver cirrhosis, it is also an underlying clinical classification. In this paper, we investigated the correlation between single nucleotide polymorphisms (SNPs) of Interleukin-10 (IL-10) and TCM syndromes in patients with hepatitis B cirrhosis (HBC). Samples were obtained from 343 HBC patients in China. Three SNPs of IL-10 (−592A/C, −819C/T, and −1082A/G) were detected with polymerase chain-reaction-ligase detection reaction (PCR-LDR). The result showed the SNP-819C/T was significantly correlated with Deficiency syndrome (P=0.031), but none of the 3 loci showed correlation either with Child-Pugh classification and phase in HBC patients. The logistic regression analysis showed that the Excess syndrome was associated with dizzy and spider nevus, and the Deficiency syndrome was associated with dry eyes, aversion to cold, IL-10-819C/T loci, and IL-10-1082A/G loci. The odds ratio (OR) value at IL-10-819C/T was 4.022. The research results suggested that IL-10-819C/T locus (TC plus CC genotype) is probably a risk factor in the occurrence of Deficiency syndrome in HBC patients.

Abstract:Aim: Interleukin-17 (IL-17) can accelerate the release of many pro-inflammatory cytokines. The purpose of our study was to investigate the potential association between polymorphisms in the IL-17 gene and susceptibility to hepatitis B virus (HBV) infection in the Han Chinese population.Methods: We recruited 596 HBV-infected patients and 612 ethnically matched controls, who were then genotyped for the IL-17A and IL-17F polymorphisms, rs2275913 and rs763780, respectively, by using TaqMan probe-based real-time polymerase chain reaction. The frequencies of the alleles and genotypes in patients and controls were compared by the chi(2) test.Results: Statistically significant differences in genotypic and allelic frequencies were revealed at both polymorphic sites between HBV-positive patients and controls (rs2275913: genotype chi(2) = 37.74, p < 0.001 and allele chi(2) = 22.17, p < 0.001, odds ratio [OR] = 0.654, 95% confidence interval [CI] = 0.548-0.781. rs763780: genotype chi(2) = 19.80, p < 0.001 and allele chi(2) = 18.78, p < 0.001, OR= 0.507, 95% CI = 0.371-0.692).Conclusions: These results indicate that the IL-17A rs2275913 and IL-17F rs763780 polymorphisms are associated with HBV infection in the Han Chinese population. We conclude that possession of the GG genotype and the G allele at rs2275913, and the TT genotype and the T allele at rs763780 might increase the risk of HBV infection. Larger-scale, multiracial studies are necessary to evaluate the role of IL-17 polymorphisms in relation to an enhanced risk of HBV infection.
Relationship Between the -572G/C Polymorphism in IL-6 Gene Promoter and Hepatitis Virus B Infection in Chinese Han Population

Abstract:AIM: To evaluate the association of IL-6 gene promoter polymorphism with the infection of hepatitis B virus (HBV) in Chinese population. METHODS: The genome DNA was obtained from blood samples and three IL-6 promoter polymorphisms (G-174C, G-572C and G-597A) were evaluated using PCR-RFLP in 160 HBV-infected patients and 212 healthy controls. Frequencies of single-nucleotide polymorphisms (SNPs) in the control group were compared with those of the HBV infection. Furthermore, subgroups of chronic carriers were compared among them. For cases and controls, the distribution of gender, age, smoking, drinking and Hardy-Weinberg equilibrium equation of the IL-6 genotype was determined using Chi-square Test; and the diversity of age using Mann-Whitney U-test. The unconditional Logistic regression model was used to analyze the statistical association of genotypes in two groups adjusted for confused factors. RESULTS: No polymorphism at position -174 and -597 was detected in all participants, but significant difference was found in the polymorphism at position -572 between the cases and controls (G/C vs G/G, OR=2.65, P＜0.05; C/C vs G/G, OR=3.31, P＜0.05). Subgroups analysis, revealed the odds ratio value in the less than thirty years old group (G/G vs G/C or C/C, OR=16.92, P＜0.05). CONCLUSION: The data reveal that the polymorphism at position -572 of the IL-6 gene promoter may be associated with the susceptibility to HBV infection; meanwhile, the age is a coordinative risk factor.
Interleukin-10gene Polymorphisms and Hepatocellular Carcinoma Susceptibility: A Meta-Analysis

Abstract:AIM: To assess the association between Interleukin-10 (IL-10) gene IL-10-1082 (G/A), IL-10-592(C/A), IL-10-819 (T/C) polymorphisms and hepatocellular carcinoma (HCC) susceptibility.METHODS: Two investigators independently searched the Medline, Embase, China National Knowledge Infrastructure, and Chinese Biomedicine Database. Summary odds ratios (ORs) and 95% confidence intervals (95% CIs) for IL-10 polymorphisms and HCC were calculated in a fixed-effects model (the Mantel-Haenszel method) and a random-effects model (the DerSimonian and Laird method) when appropriate.RESULTS: This meta-analysis included seven eligible studies, which included 1012 HCC cases and 2308 controls. Overall, IL-10-1082 G/A polymorphism was not associated with the risk of HCC (AA vs AG + GG, OR = 1.11, 95% CI = 0.90-1.37). When stratifying for ethnicity, the results were similar (Asian, OR = 1.12, 95% CI = 0.87-1.44; non-Asian, OR = 1.10, 95% CI = 0.75-1.60). In the overall analysis, the IL-10 polymorphism at position -592 (C/A) was identified as a genetic risk factor for HCC among Asians; patients carrying the IL-10-592*C allele had an increased risk of HCC (OR = 1.29, 95% CI = 1.12-1.49). No association was observed between the IL-10-819 T/C polymorphism and HCC susceptibility (TT vs TC + CC, OR = 1.02, 95% CI = 0.79-1.32).CONCLUSION: This meta-analysis suggests that IL-10-592 A/C polymorphism may be associated with HCC among Asians. IL-10-1082 G/A and IL-10-819 T/C polymorphisms were not detected to be related to the risk for HCC. (C) 2011 Baishideng. All rights reserved.
Associations Between Polymorphisms in IL - 10 Gene and the Risk of Viral Hepatitis: a Meta-Analysis

Abstract:Abstract Background The relationships between polymorphisms in interleukin-10 (IL-10) gene and the risk of viral hepatitis remain inconclusive. Therefore, the authors conducted so far the very first meta-analysis to robustly assess the relationships between polymorphisms in IL-10 gene and the risk of viral hepatitis by integrating the results of previous works. Methods Medline, Embase, Wanfang, VIP and CNKI were searched throughly for eligible studies, and 76 genetic association studies were finally included in this meta-analysis. Results We noticed that rs1800871 (− 819 C/T), rs1800872 (− 592 C/A) and rs1800896 (− 1082 G/A) polymorphisms were all significantly associated with the risk of viral hepatitis in Asians, whereas only rs1800896 (− 1082 G/A) polymorphism was significantly associated with the risk of viral hepatitis in Caucasians. In further analyses by disease subtypes, we noticed that the three investigated polymorphisms were all significantly associated with the risk of both HBV and HCV. Conclusions This meta-analysis demonstrates that rs1800871 (− 819 C/T), rs1800872 (− 592 C/A) and rs1800896 (− 1082 G/A) polymorphisms may influence the risk of viral hepatitis in Asians, while only rs1800896 (− 1082 G/A) polymorphism may influence the risk of viral hepatitis in Caucasians. In further analyses by disease subtypes, we noticed that the three investigated polymorphisms may influence the risk of both HBV and HCV.
IL12 Polymorphisms, HBV Infection and Risk of Hepatocellular Carcinoma in a High‐risk Chinese Population

Abstract:To investigate the association between the potentially functional polymorphisms in IL12A and IL12B, HBV infection and risk of hepatocellular carcinoma in a Chinese population, we genotyped three polymorphisms, rs568408 (3'UTR G>A), rs2243115 (5'UTR T>G) in IL12A and rs3212227 (3'UTR A>C) in IL12B in a case-control study of 869 hepatocellular carcinoma (HCC) cases and 891 cancer-free controls. We found that the IL12A rs568408 GA/AA variant genotypes were associated with a significantly increased risk of HCC (adjusted odds ratio (OR) = 1.53, 95% confidence interval (CI) = 1.17-2.00), compared with the wild-type GG homozygote. In the stratified analyses, the increased risk of HCC associated with rs568408 GA/AA was more evident in patients who were negative for HBsAg (adjusted OR = 1.71, 95% CI = 1.23-2.39). However, no significant associations between IL12A rs2243115 T/G, IL12B rs3212227 A/C and risk of HCC were observed. Our findings indicate that IL12A rs568408 may contribute to the risk of HCC and modify HCC risk associated with HBV infection.
Interleukin-10 Promoter Gene Polymorphisms and Susceptibility to Tuberculosis: A Meta-Analysis

Abstract:OBJECTIVE:As an update to other recent meta-analyses, the purpose of this study was to explore whether interleukin-10 (IL-10) polymorphisms and their haplotypes contribute to tuberculosis (TB) susceptibility.METHODS:We searched for published case-control studies examining IL-10 polymorphisms and TB in PubMed, EMBASE, Cochrane Central Register of Controlled Trials (CENTRAL), Wanfang databases and the Chinese National Knowledge Infrastructure (CNKI). Odds ratios (ORs) with 95% confidence intervals (CIs) were used to calculate the strengths of the associations.RESULTS:A total of 28 studies comprising 8,242 TB patients and 9,666 controls were included in the present study. There were no significant associations between the -1082G/A, -819C/T, and -592A/C polymorphisms and TB in the pooled samples. Subgroup analyses revealed that the -819T allele was associated with an increased TB risk in Asians in all genetic models (T vs. C: OR=1.17, 95% CI=1.05-1.29, P=0.003; TT vs. CC: OR=1.37, 95% CI=1.09-1.72, P=0.006; CT+TT vs. CC: OR=1.33, 95% CI=1.09-1.63, P=0.006; TT vs. CT+CC: OR=1.17, 95% CI=1.02-1.35, P=0.03) and that the -592A/C polymorphism was significantly associated with TB in Europeans under two genetic models (A vs. C: OR=0.77, 95% CI=0.60-0.98, P=0.03; AA vs. CC: OR=0.53, 95% CI=0.30-0.95, P=0.03). Furthermore, the GCC IL-10 promoter haplotype was associated with an increased risk of TB (GCC vs. others: P=1.42, 95% CI=1.02-1.97, P=0.04). Subgroup analyses based on ethnicity revealed that the GCC haplotype was associated with a higher risk of TB in Europeans, whereas the ACC haplotype was associated with a lower TB risk in both Asians and Europeans.CONCLUSIONS:This meta-analysis suggests that the IL-10-819T/C polymorphism is associated with the risk of TB in Asians and that the IL-10-592A/C polymorphism may be a risk factor for TB in Europeans. Furthermore, these data indicate that IL-10 promoter haplotypes play a vital role in the susceptibility to or protection against the development of TB.
