Abstract:To clone new members of a zinc finger protein gene family, a pair of primers was synthesized based on the conserved sequence of Kruppel-type zinc finger genes, and used to perform PCR amplification with human ge-nomic DNA as a template. The sequence analysis of the PCR product identified a novel zinc finger gene fragment, HGZA-1. Using HGZA-1 as a probe to screen cDNA library the full-length cDNA of a novel human zinc finger gene ZNF191 has been cloned. The computer analysis of the cDNA sequence reveals an open reading frame coding potentially for a zinc finger protein with 4 tandemly repeated C2H2 zinc finger motifs at C-terminus. This putative protein consists of 368 aa and has a molecular weight of 43 ku approximately. Hybridization of human/rodent somatic cell hybrid panel and fluorescence in situ hybridization mapped this gene to chromosome 18q12.1. Northern blot analysis revealed that this gene is expressed ubiquitously, while its expression level in liver is much higher than in other tissues.

Abstract:To clone a novel C2H2-type zinc finger cDNA gene,the total RNA isolated from the K562 cells was purified.Methods:The cNDA firs t strand was synthesized fro the total RNA,after reverse transcription,and the c DNA was amplified by add-on PCR.Products were digested with both EcoR I and Bam H I and then were recombined into the pGEM7Zf(+) vector.Final,we measured five t he inserted DNA fragments,and compared homology with nucleotide sequence databas e of GenBank.Results:One of them was a novel cDNA gene,which enc oding product containing five C2H2-type zinc finger domain at least.Therfore.Conclusions:In our laboratory discovered the novel C2H2-type zi nc finger domai n cDNA gene laid a foundation for research of the total sequence and function of the gene.

Abstract:In order to isolate novel genes related to early human embryo development and differentiation, a directional cDNA library was constructed from 3-week-old human embryo. Single-pass DNA sequence analysis was used to sequence 47 randomly picked low-abundance cDNA clones. This approach led us to select a clone, L30, showing significant homology with the telomeric-associated DNA and zinc finger protein genes. It is about 3.8 kb in length and contains an open reading frame of notable length within 5'-region, and a tailing signal of AAUAAA and poly (A+) with 39 A in 3'-region. The gene was transcribed in human embryo by Northern blot hybridization and assigned to human chromosome 12 by in situ hybridization.

Abstract:Zinc finger protein, one of the most important transcription factors, plays an essential role in regulating gene expression. C(2)H(2) type zinc protein with KRAB domain contains two parts, one is C(2)H(2) zinc finger motif which is use to binding to the DNA, while the other is KRAB associated box which mostly performs as a transcription repressor. In this study, we report the cloning and characterization of a novel splice variant of the human ZNF300 gene (ZNF300-B). The ZNF300-B gene cDNA is 2293 bp in length, encoding a putative protein with 619 amino acid residues. ZNF300-B gene is mapped to chromosome 5q32-5q33 with seven exons. Reverse transcription polymerase chain reaction (RT-PCR) analysis showed that ZNF300-B and ZNF-300 were expressed highly in human testis.

Abstract:Zinc-fingers and homeoboxes 1 (ZHX1) is a protein that interacts with the activation domain of the A subunit of nuclear factor-Y. The function of ZHX1, as a transcription factor, was characterized and their domains were mapped. To determine the nuclear localization signal, expression vectors, in which various truncated forms of ZHX1 were fused to the C-terminal of green fluorescence protein (GFP), were transfected into human embryonic kidney (HEK) 293 cells. All GFP–ZHX1 fusion proteins including an arginine-rich region that corresponds to the amino acid sequence between 734 and 768 were localized in the nuclei. A dimerization domain of the ZHX1 was also mapped using protein–protein interaction assays. The homeodomain (HD) 1 consisting of the amino acid sequence between 272 and 432 of ZHX1 was necessary and sufficient for dimerization. Lastly, the transcriptional activity of ZHX1 was examined using a mammalian one-hybrid system. ZHX1, fused to the C-terminal of the GAL4 DNA-binding domain, was co-transfected with luciferase reporter plasmids with or without five copies of the GAL4-binding site into HEK293 cells. The luciferase activity was decreased in both concentration- and GAL4-binding site-dependent manner. The acidic region corresponding to the amino acid sequence between 831 and 873 was a repressor domain and dimerization was prerequisited for full repressor activity.

Abstract:We report here the cloning and characterization of a novel human cytoplasm-distribution zinc finger protein (CDZFP) gene, isolated from human ovary cDNA library, and mapped to 4p12 by searching the UCSC genomic database. The CDZFP cDNA is 1793 base pairs in length and contains an open reading frame (ORF) encoding 236 amino acids. The CDZFP gene consists of 7 exons and encodes a putative zinc finger protein with a transmembrane region and two zinc finger motifs. Subcellular localization demonstrated that CDZFP protein was located in the cytoplasm when overexpressed in Hela cells and northern blot analysis revealed that CDZFP was ubiquitously expressed in 16 human tissues.

Abstract:Through screening a fetal brain cDNA library, a cDNA coding a protein of 332AA was isolated. The protein sequence was searched against prosite library, two C 2H 2 zinc finger motifs were found. The first motif was like a Zn finger (C 2H 2) in RNA binding proteins. Also the protein was homologous to some of the protein arginine N methyltransferases (PRMT) through BLASTp against nr database of the NCBI, it contained no SAM(S Adenosyl L methionine) binding motif which was partial catalytic domain of the PRMT. The function of the novel C 2H 2 zinc finger protein was unknown. In this study, the cDNA microarray was used to check the expression change of the gene in human blood vascular endothelials, which treated with PMA for 6 hours. Interestingly, its expression increased 11 times. This result indicated that the novel C 2H 2 zinc finger gene was one of the immediate early response genes( ERG ) in endothelials.

Abstract:Digital Differential Display(DDD) is a method that a user can apply it to determine the differences between libraries to quantitate the transcript levels statistically. A novel human zinc finger protein gene that contains C2H2 domain was first isolated by using a data mining tool called DDD from the National Center for Biotechnology Information(NCB). The full length cDNA of this transcript was deduced and further confirmed by reverse transcriptase polymerase chain reaction (RT PCR). Total 2 exons of the human gene span a 30 065 bp genomic DNA sequence that was mapped to chromosome 5q23.1 q23.2. The cDNA encodes a novel protein of 364 amino acids with a theoretical molecular weight of 40.3 kDa and isoelectric point of 9.59. Northern blot analyses revealed that ZNF474 mRNA was exclusively expressed in testis and ovary and has one tran script. It could be hypothesized that the ZNF474, functioning as a germ cell specific transcription factor may play important roles in spermatid differentiation and oocyte development.

Abstract:A pair of degenerate primers were designed according to the DNA sequences of the C2H2 zinc finger genes conservative domain and then homologious PCR was performed with the human genomic total DNA as template. The zinc finger fragments thus obtained were used as probes to screen cDNA libraries of human fetal kidney, muscle and marrow and 22 C2H2 zinc finger gene fragments were selected. Among these fragments, 17 were confirmed to be novel zinc finger gene fragments by literature searches in the National Center Biotechnology Information (NCBI) database. Expression patterns of the clones K3-4 and K5-12 selected from kidney cDNA library were analyzed. The results showed that the expression level of 3-4 in kidney is obviously higher than in other tissues and K5-12 is expressed at different levels in 8 tissues.

Abstract:To investigate the zinc finger genes involved in human embryonic development, we constructed a C2H2-ZNF enriched human embryonic cDNA library, from which a novel human gene named hKid3 was identified. The hKid3 cDNA encodes a 554 amino acid protein with an amino-terminal KRAB domain and 11 carboxyl-terminal C2H2 zinc finger motifs. Northern blot analysis indicates that two hKid3 transcripts of 6 and 8.5kb express in human fetal brain and kidney. The 6kb transcript can also be detected in human adult brain, heart, and skeletal muscle while the 8.5kb transcript appears to be embryo-specific. GFP-fused hKid3 protein is localized to nuclei and the ZF domain is necessary and sufficient for nuclear localization. To explore the DNA-binding specificity of hKid3, an oligonucleotide library was selected by GST fusion protein of hKid3 ZF domain, and the consensus core sequence 5′-CCAC-3′ was evaluated by competitive electrophoretic mobility shift assay. Moreover, The KRAB domain of hKid3 exhibits transcription repressor activity when tested in GAL4 fusion protein assay. These results indicate that hKid3 may function as a transcription repressor with regulated expression pattern during human development of brain and kidney.

Abstract:Objective To construct a plasmid containing the functional domain of human Zinc-fingers and homeoboxs 2(ZHX2).Methods A truncated fragment of human ZHX2(242-501AA) was amplified with pcDNA-ZHX2-HA and cloned into pcDNA3.0 to construct expression vector pZXH2(242-501AA).The recombinant plasmids were identified with enzyme digestion and sequencing.Expression vector pZXH2(242-501AA) was transfected into HepG2.2.15 and COS-7cells by Lipofectamine 2000,and expression of fusion proteins was assayed by western blot.Results Recombinant plasmids containing the functional domain(242-501AA) of the human ZHX2 gene were constructed.Enzyme digestion and sequencing analysis showed successful construction of the plasmids.Western blot showed protein expressions of the functional domain of human ZHX2 in HepG2.2.15 and COS-7cells lines.Conclusion The functional domain of the human ZHX2 gene was successfully cloned and expressed,which sets a good basis for further study on the functional mechanism of ZHX2.

Abstract:The human zinc finger protein ZNF191 is a krüppel-like transcription factor, which may be relevant to many diseases such as neuropsychiatric, cardiovascular and liver caner diseases. To elucidate the function of ZNF191 by gene targeting, it is necessary to clone and characterize of the homologous gene in model organisms (mice). The mouse homologous gene (ZF-12) was cloned and sequenced for the first time, the GenBank accession number is AY052495. It contains four exons and three introns; all intronic splice sites exhibited consensus GT/AG sequences. The single nucleotide polymorphisms (SNPs) in exon 2 and the alternative length of 3'-untranslated region (3'-UTR) have been found. The linkage of the ZF-12 gene and the zinc finger protein gene Zfp-35 has been found, so the ZF-12 gene can be localized to B3 to C or beside of chromosome 18. We assessed approximately 1.2 kb of 5'-flanking region of the ZF-12 gene for basal promoter activity. A series of deletion mutants of 5'-flanking region linked to the luciferase gene was constructed. Basal level expression of these constructs was tested in COS-7 cells, NIH3T3 cells and HeLa cells. By measuring luciferase activity, which was transiently expressed in the transfected cells, we found that regulatory elements sufficient for basal expression lie between -762 and +70 bp relative to the transcription start site and that a negative regulatory region lie between -824 and -762 bp. This research provides a basis for further study on ZF-12 by gene targeting.

Abstract:ZNF191 is a new zinc finger gene which has a 1107 bp open reading frame (ORF) and encodes a 368 amino acid protein including a putative DNA-binding domain of four Cys2His2 zinc finger motifs at its C-terminal region. The ZNF191 clDNA is located in the chromosome 18q12.1 region where it is known that a variety of hereditary diseases are related to. Probably, this protein has potential function of stimulating the gene transcription.

Abstract:The zinc finger gene (ZNF) family plays an important role in the regulation of transcription. This study reports the cloning and characterization of a novel human zinc finger protein cDNA (ZNF284L) from fetal brain cDNA library. The ZNF284L cDNA is 2223 bp in length encoding a 593-aa polypeptide. The protein contains a KRAB A+b box and eleven C2H2 type zinc finger motifs. ZNF284L gene is mapped to 19q13.2-19q13.3 with 5 exons, and the expression pattern of ZNF284L gene was also examined by reverse transcription polymerase chain reaction (RT-PCR). The transcripts were detected in the human lung, liver, pancreas, thymus, heart, placenta, spleen, prostate, ovary, small intestine and colon, but in human brain, skeletal muscle, kidney, testis and peripheral blood leukocyte, no expression was detected.

Abstract:The human ZNF268 gene is a novel C2H2 zinc finger gene restrictively expressed in the liver of human embryo. As an initial study on the regulation of its expression, 2.5 kb fragment of the ZNF268 5'-flanking region was cloned from human genome by PCR. This fragment was inserted into pEGFP1 vector and then the recombinant plasmid was transfected into several cell lines by liposomal transfection method. EGFP expression was observed in four cell lines under confocal laser scanning microscope. A series of 5'-deletion fragments from -2456(-)+77 bp to -20(-)+77 bp were inserted into pCAT-Basic vector to construct recombinant reporter plasmids, which were then transfected into Hela cells for deletion analysis. Results of deletion analysis revealed that an important region for transcriptional activity lies between -156 and -20 bp.

Abstract:With the aim of identifying genes involved in early human embryonic development, we have isolated a cDNA clone representing a novel human zinc finger gene ZNF268 from 3 week old human embryo cDNA library using a differential hybridization strategy. The complete cDNA sequence of ZNF268 contained an open reading frame of 2841 nucleotides that encodes a 947 amino acid protein with an N-terminal Krüppel-associated box (KRAB) domain and 24 C-terminal zinc fingers. Northern blot analysis showed that ZNF268 mRNA is mainly expressed in 3–5 week old human embryos suggesting it could play certain roles in the embryogenesis. The gene consists of six exons spanning about 22 kb of genomic DNA. According to the genomic sequence from the HTGS database, the ZNF268 gene is assigned to human chromosome 5.

Abstract:The histone chaperone SET encoded by the SET gene, which is also known as template-activating factor Iß (TAF-Iß), is a multifunctional molecule that is involved in many biological phenomena such as histone binding, nucleosome assembly, chromatin remodelling, replication, transcription and apoptosis. A truncated SET/TAF-Iß ΔN protein that lacked the first 22 residues of the N-terminus but contained the C-terminal acidic domain and an additional His6 tag at the C-terminus was overexpressed in Escherichia coli and crystallized by the hanging-drop vapour-diffusion method using sodium acetate as precipitant at 283 K. The crystals diffracted to 2.7 A resolution and belonged to space group P4(3)2(1)2.

Abstract:The Na + /H + exchangers (NHEs) catalyze the transport of Na + in exchange for H + across membranes in organisms and are required for numerous physiological processes. Here we report the cloning and characterization of a novel human NHEDC1 (Na + /H + exchanger like domain containing 1) gene, which was mapped to human chromosome 4p24. This cDNA is 1859 bp in length, encoding a putative protein of 515 amino acids. The NHEDC1 proteins are highly conserved in mammals including human, mouse, rat, and Macaca fascicularis . One remarkable characteristic of human NHEDC1 gene is that it is exclusively expressed in the testis by RT-PCR analysis. Western blot analysis showed that the molecular weight of NHEDC1 is about 56 KDa.

Abstract:The mouse zinc finger protein ZF-12 gene is homologous to human gene and encodes a protein of 368 amino acids, which contains four tandem C2H2-type zinc finger motifs in the N-terminal and one SCAN domain in the C-terminal. Some recent studies suggest that ZNF191 might be a hepatocarcinogenesis-associated gene. We screened a mouse lambda genomic library with a human ZNF191 cDNA probe and isolated a ZF-12-like gene, named ZF12p (GenBank AY040222). This intronless gene closely resembles ZF-12 but displays several mutations, suggesting that ZF12p represents a ZF-12-related pseudogene. RT-PCR analysis on total RNA from mouse tissue and bioinformatis analysis on promoter region of ZF12p gene, suggest the transcripts of ZF12p may be not synthesized. BLAST on the data of the human genome in the GenBank with ZNF191 cDNA and Southern blotting show there is no any psedogene related to ZNF191 gene in the human genome. The high similarity of ZF12p to ZF-12 might be of considerable importance for mutation and evolution analysis of ZF-12.
