The Role of Circadian Rhythm in the Pathogenesis of Colorectal Cancer

Abstract:The circadian clock controls many cellular processes, some of which are key to tumor suppression, including cell proliferation and apoptosis, DNA repair, metabolism, hormone signaling, and immune function. The association between altered circadian clock function and increased risk of colorectal cancer is increasingly evident from epidemiologic, preclinical, and clinical studies. In this review, we discuss several distinct mechanisms by which altered components of the circadian clock might promote intestinal tumorigenesis. We also review the progress in identifying circadian and clock-based molecular processes that might be targeted to promote the prevention and control of colorectal cancer.
