Abstract:Isonucleosides were synthesized by the reaction of suger epoxide and nucleobase in the presence of K(2)CO(3 )and crown ether. The substitution is regioselective. Some of isonucleoside derivatives showed significant activities of cytotoxcity in HL-60 cells.Oligodeoxynucleotides incorporated with isonucleoside have a increase in stability towardes nuclease S1.
A New Route to 3-Amino Sugars. A Concise Synthesis of L-Daunosamine and D-Ristosamine Derivatives

Abstract:An asymmetric aldol strategy has been developed for the synthesis of L-daunosamine and D-ristosamine derivatives starting from noncarbohydrate precursors. Lithium and boron enolate mediated aldol reactions of 12 with O-TBS lactaldehyde gave non-Evans syn and Evans syn aldol products, respectively, with high selectivity. The chemical efficiency of the lithium enolate reactions were higher than the corresponding reactions with the boron enolates. Curtius rearrangement of lactone acids 23 and 26 gave the corresponding N-BOC amino lactones 30 and 32 in 64% and 62%, respectively, with complete retention of configuration. Lactone 30 was converted by a two-step sequence to N-benzoyldaunosamide 40. The overall yield for the amino sugar 40 was 18% over six steps. Similarly, lactone 32 was converted to N-benzoylristosamide 42 with an overall yield of 18% starting from 12.

Abstract:3,6-Di-O-methanesulfonyl-2,5-anhydro-l-idofuranose dimethyl acetal 9 was reacted with NaN3 in DMF to give 6-azido substituted carbohydrate 10 selectively. A series of 6-amino-isonucleosides 4a–d were synthesized by the reaction of nucleobases with epoxide 13, followed by hydrolysis, reduction, and deprotection, in good yield.

Abstract:Two synthetic strategies were developed to obtain isonucleosides 2a and 2b. Starting from the known compound 4, an extension of one carbon unit at sugar 6-terminal was achieved by Wittig reaction and Stannyl-desulfonylation reaction. After oxidation of the double bond, the isonucleosides with elongated side chain 2a and 2b were synthesized. For the synthesis of isonucleosides containing different bases, an epoxide intermediate approach was developed. Isonucleosides 2a and 2b were synthesized by regioselective epoxide opening reaction of 18 in good yield.

Abstract:A concise synthesis of 3-deoxy-3-nucleobase-2,5-anhydro-d-mannitol 6(a–d) has been achieved, using the deamination of 2-amino-2-deoxy-d-glucose to construct in one step the sugar skeleton with the desired sense of chirality at each asymmetric center. The selective dibenzoylation of 2,5-anhydro-d-mannitol 9 was investigated, and the key epoxide intermediate 13 was obtained in good yield via an intramolecular Mitsunobu reaction. The process of opening of epoxide 13 by nucleobases appeared to be regioselective.

Abstract:The reagent system based on the combined use of Et3SiH/I2 acts as an efficient N-glycosidation promoter for the synthesis of natural and sugar-modified nucleosides. An analysis of reaction stereoselectivity in the absence of C2-positioned stereodirecting groups revealed high selectivity with six-membered substrates, depending on the nucleophilic character of the nucleobase or based on anomerization reactions. The synthetic utility of the Et3SiH/I2-mediated N-glycosidation reaction was highlighted by its use in the synthesis of the investigational drug apricitabine.

Abstract:A versatile and concise synthesis of N-alkylated 1,4-dideoxy-1,4-imino-d-arabinitol and 1,4-dideoxy-1,4-imino-l-xylitol derivatives is described. These were prepared using pseudohemiketal lactams as key intermediates, which in turn were obtained from sucrose. The key intermediates were prepared by a diastereospecific tandem reaction which facilitated the introduction of various substituents on the nitrogen atom of the iminosugars.

Abstract:New C-nucleoside analogues were synthesized via the key intermediate 2-(2′ ,3′ 4′-tri-O-benzoyl-β-D-xylopyranosyl)-furan. The stability of the α- and β-isomer was described. The conversion of the furan ring into pyridazine ring was performed by Clauson-Kaas method to give 3-(β-D-xylopyranosyl)-pyridazine and 3-(β-D-xylopyranosyl-4,5-dihydro-6(1H)-pyridazinone.

Abstract:The octaoligoisonucleotide (isoT)2(isoG)4(isoT)2 (I), consisting of isonucleoside units 6'-O-allyl-4'-deoxy-4'-(nucleobase)-2',5'-anhydro-L-mannitol, was synthesized by the phosphotriester approach in solution phase. Based on CD spectra and capillary electrophoresis, it was confirmed that iso-oligomer I could form a parallel intermolecular G-quadruplex structure. K+, Na+ and Li+ can prompt the formation of G-quartet structures and stabilize them. The effective order of these cations is K+ > Na+ > Li+.

Abstract:Four novel 2-(trimethylsilyl)ethyl glycosides have been synthesized by a short and efficient route starting from d-glucose. Their structures were elucidated by applying high-resolution mass spectra, and one-dimensional and two-dimensional NMR techniques. These glycosides were prepared and used as intermediate building blocks in the scheme developed for oligosaccharide construction.

Abstract:2-Deoxy-4'-thio-D-sugars are potential thiosugar donors for glycosylation reactions with nucleobases to produce various D-deoxy-4'-thionucleosides. However, thio-inversion reactions that produce D-deoxy-4'-thio-D-sugars depend on using a starting material, L-deoxysugar, which is expensive. Herein, we report a unique synthetic methodology to produce 2-deoxy-4'-thio-D-sugars using a D-sugar as an inexpensive starting material. Following our synthetic procedures for carbonyl translocation, D-ribose was transformed to 2-deoxy-L-ribosyl dithioacetal as a key intermediate. Mesylation and triflation provide the corresponding dialkylthio acetals as key precursors. Under thio-inversion reaction with iodide reagents, D-2-deoxy-4'-thioxyloside was successfully produced for the first time.

Abstract:A series of novel 1'-O-alpha-methyl isonucleosides were efficiently and stereoselectively synthesized with 1,2,3,5-tetra-O-acetyl-D-ribofuranose as the starting material. The key steps include regioselective and stereoselective deprotection of the 2,4-dichlorobenzyl group at C2, triflation, and substitution with appropriate nucleobases using cesium carbonate as the base. Removal of the residual 2,4-dichlorobenzyl groups and subsequent transformation afforded the title compounds in 30-37% overall yield. The products represent a new type of isonucleosides with 1'-O-substitution.

Abstract:The ribonucleosides of pyrazolo (5,4-e)-1,2,4-triazine derivatives were synthesized. The ��,��-isomes were distinguished with 1H NMR. ��-isomer was the major product in acetyl protected ribonucleosides. In benzoyl protected ribonucleosides, no ��-isomer was formed. The J H1H2 of cis isomer was bigger than that of trans in five membered ring.

Abstract:An efficient synthesis of D-2'-deoxy-2',2'-difluoro-4'-dihydro-4'-thionucleosides is described. The conformations of D-2'-deoxy-2', 2'-difluoro-4'-dihydro-4'-thiouridine were studied by X-ray crystallography, NMR spectroscopy and molecular modeling in an attempt to explore the roles of the two gem-difluorine atoms in the puckering preferences of the thiosugar ring. No matter which conformation (south or north) the thiosugar adopts, there is always one fluorine in a pseudoaxial position, with the other in a pseudoequitorial position and thus the strong antiperiplanar (ap) effects from C-H and C-C sigma-bonds to sigma*C-F are equal to each other in these two conformers. Therefore, the other weak effects, such as dipole-dipole interactions and electrostatic attractions, become more important for determining the overall conformation of the sugar ring. Based on the results of NMR spectroscopy, high-level quantum computations and molecular dynamic simulations were performed to study the preferred pucker of the thiosugar ring in solution. Our results showed that the strong antiperiplanar preference of C-H and C-C sigma-bonds to sigma*C-F and sigma*C-O seemed to be responsible for the favored S-conformation in solution, and the weak electrostatic attractions between (delta+)C2-F beta(delta-) and (delta+)H6-C6(delta-) may stabilize the preferred structure further, and keep the base moiety in a high anti-rotamer population in solution. In contrast, the packing forces (hydrogen bond OH center dot center dot center dot O=C, dipole-dipole interaction C-F center dot center dot center dot C=O) in the solid state compensated the energetic disadvantage of the relatively less stable N-conformation, and drove the thiouridine to crystallize in the N-conformation. These results, along with the earlier empirical rules regarding proton chemical shifts in carbohydrates and nucleosides, were used to propose a method based on proton chemical shifts for the analysis of the N (sic) S equilibrium of the fluorinated sugar ring.

Abstract:2,5:3,4-Dianhydro-L-talofuranose dimethylacetal 7 was synthesized from D-glucose in 7 steps. A series of 4-deoxy-4-nucleobase-2,5-anhydro-L-mannitols 13-16 were synthesized regioselectively from 7 in good yields. 6-O-p-Tolylsulfonyl-2,5:3,4-dianhydro-L-talofuranose dimethylacetal 8 reacted with uracil or thymine to give the corresponding isonucleosides 17 and 19, but in the case of reaction of 8 with adenine, reformation of tetrahydrofuran ring took place, giving 4-(S)-adenyl-5-(R)-[1'-(R)-hydroxy-2',2'-dimethoxyl] ethyl-2,3-dihydrofuran 21 and 4-deoxy-4-adenyl-2,5:3,6-dianhydro-L-mannofuranose dimethylacetal 22. Abstract:Although several isonuleoside triphosphates can be incorporated into the DNA chain and prolonged in various degrees, none of the corresponding nucleosides exhibited significant inhibitory potency against va-ries of virus in vitro. The active antiretroviral form of a nucleoside analog is its triphosphate anabolite. How-ever, the monophosphorylation catalyzed by cellular kinases step is usually very restrictive. In order to bypass the first phosphorylation step, a series of isonuleoside analogues and their bis (isopropyl) monophosphates were designed and synthesized by selective microwave-aided method. The cell-based assay results showed that bis(isopropyl) isonucleoside monophosphates behaved improved potency against HSV-2 and HIV-1.

Abstract:(Sp)-octyl-8-chloroadenosine 3',5'-cyclophosphate inhibited die growth of tumor cells by regulating cell signaling pathways. Its metabolic compound, 8-Cl-adenosine, also demonstrated the behaviors of inducing differentiation and initiating apoptosis in some tumor cells. Starting from D ( L)-xylose, D-glucose, or 2-amino-2-deoxy-D-glucose, five types of isonucleosides were synthesized. The oligonucleotides containing isonucleosides showed remarkable stability to the degradation by phosphodiesterase and their capacities of hybridizing complementary sequence depended on the structure of the isonucleosides and the linkage mode of the phosphodiester.
