Abstract:The catalytic activity of carbohydrate-derived amino alcohols in the enantioselective aldol reaction of ketones with isatin and its derivatives has been examined for the first time. The carbohydrate-derived amino alcohols 5 were found to be efficient organocatalysts for asymmetric aldol reactions. A variety of isatins were used as substrates and the corresponding aldol products were obtained in high yields (up to 99%) and with moderate enantioselectivities (up to 75%).

Subsequent dehydration of the reaction adducts with a catalytic amount of concentrated H2SO4 in a one-pot fashion readily afforded a series of new optically active naphthopyran derivatives with moderate to good yields (up to 91%) and enantioselectivities (up to 90%).

Abstract:A catalytic, regioselective Friedel-Crafts alkylation of beta-naphthol with allylic alcohols has been developed. This procedure allows for selective α-alkylation of β-naphthol with a p-toluenesulfonic acid catalyst. This transformation demonstrated functionalized naphthol synthesis under mild reaction conditions with high product yields – 20 examples with up to 96% yields. Abstract:New chiral ligands could be obtained by introducing proline moieties and imidazoline moieties to binaphthyl skeletons. The chiral ligands exhibited balanced rigidity and flexibility which could allow the change of the conformations during the reactions on one hand, and could provide sufficient asymmetric induction on the other. The proline moiety could act as a linker connecting the binaphthyl skeletons and the imidazoline moieties as well as a coordinating group for the central metal, and the electronic and steric properties of the imidazoline groups could be carefully fine‐tuned by the use of different substituents. In the presence of Cu(II) catalyst bearing such chiral ligands, aza‐Friedel‐Crafts reaction of 1‐naphthols and electron‐rich phenols with isatin‐derived ketimines provided the desired products with good to excellent yields and up to 99% ee. The reactions showed good scalability, and excellent ee could still be obtained when the reaction was carried out in gram‐scale.

Abstract:The iron-catalyzed highly Markovnikovtype selective and enantioselective hydrosilylation of terminal aliphatic alkenes with good functional group tolerance is developed. This operationally simple protocol uses earth-abundant transition metal catalyst, readily available aliphatic alkenes and hydrosilanes to construct valuable chiral organosilanes with better than 99% ee in most cases. The chiral aliphatic alkan-2-ol and chiral dihydroxysilane as an analogue of ketone could be efficiently synthesized via further derivatization of chiral organosilanes without any racemization.

Abstract:Readily available potassium alkynyltrifluoroborates were used for organocatalytic asymmetric conjugate alkynylation of β-enaminones. The interception of a modified binaphthol catalyst and in situ generated organodifluoroboranes proved important to access functionalized β-alkynyl-β-amino carbonyls and derivatives with improved chemo-reactivity and enantio-induction. Mechanistic studies revealed the impact of molecular sieves on efficiency and stereocontrol. Abstract:Herein, we report an example of a stereoselective γ-addition reaction of trifluoromethyl ketimines to 1-alkynyl ketones mediated by an isothiourea, BTM, under mild conditions, which afforded tetrasubstituted allenes with central chiralities in high yields (up to 94% yield), good enantioselectivities (up to 91% ee), and excellent diastereoselectivities (all >20 : 1 dr). In addition, the BTM-catalyzed γ-addition reaction was successfully applied to the gram-scale reaction, and an unexpected benzopyrrolothiazine derivative was successfully converted, albeit racemic.

The catalysts are recoverable and reusable, and one of the supported catalyst has been used in a flow process allowing the synthesis of 3‐amino‐2‐oxindole derivatives in multigram scale with high yield and enantioselectivity.

Abstract:Catalytic oxidative functionalization of alkynes has emerged as an effective method in synthetic chemistry in recent decades. However, enantioselective transformations via metal carbene intermediates are quite rare due to the lack of robust chiral catalysts, especially in the intermolecular versions. Herein, we report the first asymmetric three-component reaction of commercially available alkynes with nitrones and alcohols, which affords α-alkoxy-β-amino-ketones in good yields with high to excellent enantioselectivity using combined catalysis by an achiral gold complex and a chiral spiro phosphoric acid (CPA). Abstract:Chiral phosphoric acid-catalyzed enantioselective aza-Friedel-Crafts reaction of trifluoromethyl benzoxazinones with pyrroles is reported. Under mild conditions, a range of enantioenriched dihydrobenzoxazinones bearing trifluoromethylated quaternary stereocenters could be obtained in good to excellent yield and ee. A remarkable fluorine effect is observed, and preliminary mechanistic studies combined with theory calculations suggest that triple-hydrogen-bonding interactions hold the transition structure rigidly and allow the bulky substituents of the catalyst to influence the enantioselectivity.

Abstract:Chiral nickel complexes promoted enantioselective reductive alkenylation of a range of conjugated enones, using alkenyl bromides, triflates, and tosylates. The neutral condition was compatible with sensitive groups and azaheteroaryl rings. Importantly, alkenyl groups in products can be readily converted to functionalized alkyl groups via iron-catalyzed reductive hydrofunctionalization. Some examples of asymmetric alkenylation of N-enoyl pyrroles and indoles were also included.

Abstract:The enantioselective organocatalytic conjugate alkenylation of β-substituted alkenyl benzimidazoles afforded β-stereogenic 2-alkyl benzimidazole derivatives in excellent enantioselectivities. Chiral binaphthols were effective catalysts for promoting the nucleophilic addition of bench-stable alkenyl trifluoroborate salts under mild conditions, expanding their applications by utilizing C=N-containing azaarenes as activating groups. The synthetic utility of this strategy is demonstrated by conversions into several useful enantiomerically enriched benzimidazole building blocks.
