Abstract:Micro and nano technologies are of increasing importance in microfluidics devices used for electroporation (electroporation – the permeabilization of the cell membrane with brief high electric field pulses). Electrochemical reactions of electrolysis occur whenever an electric current flows between an electrode and an ionic solution. It can have substantial detrimental effects, both on the cells and solutions during the electroporation. As electrolysis is a surface phenomenon, between electrodes and solution, the extent of electrolysis is increased in micro and nano electroporation over macro-electroporation, because the surface area of the electrodes in micro and nano electroporation is much larger. A possible way to eliminate the electrolytic effect is to develop non-electrolytic microelectroporation by coating the microelectroporation devices with a dielectric insulating layer. In this study, we examine the effect of a dielectric insulating layer on the performance of a singularity microelectroporation device that we have recently designed. Using numerical analysis, we study the effects of various design parameters including, input sinusoidal voltage amplitude and frequency, geometrical configuration and material electrical properties on the electroporation performance of the non-electrolytic microelectroporation device. In the simulation, we used properties of four real dielectric materials and four solutions of interest for microelectroporation. We characterized the effect of various design parameters of relevance to singularity based microelectroporation, on non-electrolytic microelectroporation. Interestingly, we found that the system behaves in some aspects as a filter and in many circumstances saturation of performance is reached. After saturation is reached, changes in parameters will not affect the performance of the device.

Abstract:It was recently shown that electrolysis may play a substantial detrimental role in microfluidic electroporation. To overcome this problem, we have developed a non-electrolytic micro/nano electroporation (NEME) electrode surface, in which the metal electrodes are coated with a dielectric. A COMSOL based numerical scheme was used to simultaneously calculate the excitation frequency and dielectric material properties dependent electric field delivered across the dielectric, fluid flow, electroporation field and Clausius-Mossotti factor for yeast and E. coli cells flowing in a channel flow across a NEME surface. A two-layer model for yeast and a three-layer model for E. coli was used. The numerical analysis shows that in NEME electroporation, the electric fields could induce electroporation and dielectrophoresis simultaneously. The simultaneous occurrence of electroporation and dielectrophoresis gives rise to several interesting phenomena. For example, we found that a certain frequency exists for which an intact yeast cell is drawn to the NEME electrode, and once electroporated, the yeast cell is pushed back in the bulk fluid. The results suggest that developing electroporation technologies that combine, simultaneously, electroporation and dielectrophoresis could lead to new applications. Obviously, this is an early stage numerical study and much more theoretical and experimental research is needed.

Abstract:Membrane electropermeabilization is the observation that the permeability of a cell membrane can be transiently increased when a micro-millisecond external electric field pulse is applied on a cell suspension or on a tissue. Applicative aspects for the transfer of foreign molecules (macromolecules) into the cytoplasm are routinely used. But only a limited knowledge about what is really occurring in the cell and its membranes at the molecular levels is available. This chapter is a critical attempt to report the present state of the art and to point out some of the still open problems. The experimental facts associated to membrane electropermeabilization are firstly reported. They are valid on biological and model systems. Secondly, soft matter approaches give access to the bioelectrochemical description of the thermodynamical constraints supporting the destabilization of simplified models of the biological membrane. It is indeed described as a thin dielectric leaflet, where a molecular transport takes place by electrophoresis and then diffusion. This naïve approach is due to the lack of details on the structural aspects affecting the living systems as shown in a third part. Membranes are part of the cell machinery. The critical property of cells as being an open system from the thermodynamical point of view is almost never present. Computer simulations are now contributing to our knowledge on electropermeabilization. The last part of this chapter is a (very) critical report of all the efforts that have been performed. The final conclusion remains that we still do not know all the details on the reversible structural and dynamical alterations of the cell membrane (and cytoplasm) supporting its electropermeabilization. We have a long way in basic and translational researches to reach a pertinent description.

Abstract:When a certain strong electrical pulse applied across a cell or tissue, the structures of the cell or tissue would be rearranged to cause the permeabilization of the cell membrane, named in early 1980’s “electroporation”[1]. The theoretical and experimental studies of electric field effects on living cells with their bilayer lipid membrane has been studies in 1960’s to 1970’s century [1-6]. During these years, the researches were primarily dealt with reversible and irreversible membrane breakdown in vitro. Based on these research, the first gene transfer by custom-built electroporation chamber on murine cells was performed by Neumann et al. in 1982 [7]. When electric field (E≈0.2V, Usually 0.5-1V) applied across the cell membrane, a significant amount of electrical conductivity can increase on the cell plasma membrane. As a result, this electric field can create primary membrane “nanopores” with minimum 1 nm radius, which can transport small amount of ions such as Na+ and Clthrough this mem‐ brane “nanopores”. The essential features of electroporation included (a) short electric pulse application (b) lipid bilayer charging (c) structural rearrangements within the cell mem‐ brane (d) water-filled membrane structures, which can perforate the membrane (“aqueous pathways” or pores) and (e) increment of molecular and ionic transportation [8]. Abstract:Treatment of biological material by pulsed electric fields is a versatile technique in biotechnology and biomedicine used, for example, in delivering DNA into cells (transfection), ablation of tumors, and food processing. Field exposure is associated with a membrane permeability increase usually ascribed to electroporation, i.e., formation of aqueous membrane pores. Knowledge of the underlying processes at the membrane level is predominantly built on theoretical considerations and molecular dynamics (MD) simulations. However, experimental data needed to monitor these processes with sufficient temporal resolution are scarce. The whole-cell patch-clamp technique was employed to investigate the effect of millisecond pulsed electric fields on DC-3F cells. Cellular membrane permeabilization was monitored by a conductance increase. For the first time, to our knowledge, it could be established experimentally that electroporation consists of two clearly separate processes: a rapid membrane poration (transient electroporation) that occurs while the membrane is depolarized or hyperpolarized to voltages beyond so-called threshold potentials (here, +201 mV and -231 mV, respectively) and is reversible within ∼100 ms after the pulse, and a long-term, or persistent, permeabilization covering the whole voltage range. The latter prevailed after the pulse for at least 40 min, the postpulse time span tested experimentally. With mildly depolarizing or hyperpolarizing pulses just above threshold potentials, the two processes could be separated, since persistent (but not transient) permeabilization required repetitive pulse exposure. Conductance increased stepwise and gradually with depolarizing and hyperpolarizing pulses, respectively. Persistent permeabilization could also be elicited by single depolarizing/hyperpolarizing pulses of very high field strength. Experimental measurements of propidium iodide uptake provided evidence of a real membrane phenomenon, rather than a mere patch-clamp artifact. In short, the response of DC-3F cells to strong pulsed electric fields was separated into a transient electroporation and a persistent permeabilization. The latter dominates postpulse membrane properties but to date has not been addressed by electroporation theory or MD simulations.

Abstract:Electroporation has been widely used in biology, medicine, and the food industry as a means to transport various molecules through the cell membrane. The phenomenon of electroporation is the result of cell membrane damage caused by the application of an electric field. In order to understand more precisely how cells function, we established a dielectric model of a spherical cell and analyzed its characteristics by the finite element method. The effects of altering different electrical parameters were determined. The results showed that the electric field strength was positively related to the transmembrane voltage (TMV) and pore density. There was a minimum electric field strength necessary to induce a critical TMV for the formation of pores. Pulse width also had to be long enough to charge the cell membrane, compared with the normal membrane charging time constant of about 1 mu s. When the pulse width was shorter than the charging time constant, it was necessary to increase pulse frequency to create a high enough TMV. The rise-time of the electric pulse also affected electroporation: a fast rise-time pulse not only allowed penetration of the plasma membrane but also the organelle membrane. With slow rise-time pulse, the organelle was shielded from electroporation. This study defines the response characteristics of electrical parameters on the electric load cell and establishes the specificity of parameters for different purposes.

Abstract:The electropermeabilization of biological cell membranes by the application of an external field occurs whenever an applied field exceeds a threshold value. For fields above this threshold value but less than another critical value, the pores formed in the membrane are transient or reversible. Several mechanisms have been proposed for the formation of these transient pores. Here we examine the local electric fields generated for the configuration of cells embedded in a polycarbonate filter, both in the region in and around the pore. We consider the shear forces created in the membrane due to the gradient of the field along the surface of the membrane, and the interaction of the charged molecules in the membrane with this field. A relationship between the electric field strength and the size of the pore formed is derived.

Abstract:Electroporation-based techniques, i.e. techniques based on the perturbation of the cell membrane through the application of electric pulses, are widely used at present in medicine and biotechnology. However, the electric pulse - cell membrane interaction is not yet completely understood neither explicitly formalized. Here we introduce a Multiphysics (MP) model describing electric pulse - cell membrane interaction consisting on the Poisson equation for the electric field, the Nernst-Planck equations for ion transport (protons, hydroxides, sodium or calcium, and chloride), the Maxwell tensor and mechanical equilibrium equation for membrane deformations (with an explicit discretization of the cell membrane), and the Smoluchowski equation for membrane permeabilization. The MP model predicts that during the application of an electric pulse to a spherical cell an elastic deformation of its membrane takes place affecting the induced transmembrane potential, the pore creation dynamics and the ionic transport. Moreover, the coincidence among maximum membrane deformation, maximum pore aperture, and maximum ion uptake is predicted. Such behavior has been corroborated experimentally by previously published results in red blood and CHO cells as well as in supramolecular lipid vesicles.

Abstract:We demanstrated an cell electroporation system based on Au electrode microfabricated chip. We realized high efficiency electroporation in both standard expression cell(HEK-293A)and hard-to-transfected cell(3T3-L1), with a transfection rate of 90% and 80%, respectively. Abstract:Generally, electroporation of in vitro cells is performed under very high electric fields to overcome the physical barrier of plasma membrane. Since traditional electroporation techniques make use of very high voltages, which is critical to cell viability, this study presents a microfluidic platform able to perform cell membrane electroporation with the application of low voltages (1.5-2 V). The platform is manufactured based on the milling by mean of focused ionic beam, which offers an established approach to fabricate ordered arrays of 3D gold hollow nanoelectrodes protruding from an insulating substrate. The novelty of this fabrication relies on the fact that the nanoelectrodes used for electroporation are simultaneously metallic, hollow and communicate through its nanochannels with an isolated microfluidic chamber beneath the device. Adherent cultured cells on the nanoelectrodes can be electroporated in this platform, and molecules can be selectively delivered only inside the porated cells.

Abstract:Exposure to electrical fields can reversibly increase the electrical conductivity and permeability of a cell membrane, which regulates and directs the exchange of materials and information between the cell and its environment. If cell membranes (or artificial lipid membranes) are exposed to a field pulse of high intensity and short duration (ns to μs), local electrical breakdown occurs in them. This electrical breakdown is associated with a large permeability change in the membrane, which is such that substances or particles (up to the size of genes) which cannot normally permeate through the membrane, are able to traverse the membrane into the cell. The original properties of the membrane are restored within μs to min, depending on the experimental conditions and the membrane properties. Electrical breakdown in the zone of contact between the membranes of cells (or lipid vesicles), which have been made to adhere to each other by the action of weak inhomogeneous alternating electrical fields, leads to fusion of these cells with formation of a single cell having new functional characteristics. The electrical fusion method is very mild, and the yield of fused cells is high. The electrically induced fusion and entrapment of membrane‐impermeable substances and genes in cells provide a new tool for the productions of a wide range of cells with manipulated functions, which could be used (or are being used) for the solution of a number of problems in cell biology, medicine and technology. The application of electrical membrane breakdown to clinical diagnostics, the development of cellular carrier systems for the selective transport of drugs to a site of action within the organism and the potential applications of electrically induced fusion for breeding salt‐tolerant crop plants for converting solar energy into ethanol, for synthesizing natural materials and manipulating genes, are described. The transfer of membrane‐impermeable substances and particles into cells and the fusion of different types of cells can be achieved by effect of an electric field. Potential applications are the selective transport of pharmaċeuticals into organisms and the conversion of CO2 and H2O by sunlight into ethanol.

A molecular insight into the electro-transfer of small molecules through electropores driven by electric fields

Abstract:The transport of chemical compounds across the plasma membrane into the cell is relevant for several biological and medical applications. One of the most efficient techniques to enhance this uptake is reversible electroporation. Nevertheless, the detailed molecular mechanism of transport of chemical species (dyes, drugs, genetic materials, …) following the application of electric pulses is not yet fully elucidated. In the past decade, molecular dynamics (MD) simulations have been conducted to model the effect of pulsed electric fields on membranes, describing several aspects of this phenomenon. Here, we first present a comprehensive review of the results obtained so far modeling the electroporation of lipid membranes, then we extend these findings to study the electrotransfer across lipid bilayers subject to microsecond pulsed electric fields of Tat11, a small hydrophilic charged peptide, and of siRNA. We use in particular a MD simulation protocol that allows to characterize the transport of charged species through stable pores. Unexpectedly, our results show that for an electroporated bilayer subject to transmembrane voltages in the order of 500mV, i.e. consistent with experimental conditions, both Tat11 and siRNA can translocate through nanoelectropores within tens of ns. We discuss these results in comparison to experiments in order to rationalize the mechanism of drug uptake by cells. Abstract:Multivalent membrane‐anchored DNA frameworks are engineered for in situ evaluation of electric field‐induced membrane permeability during reversible electroporation. Fluorescence imaging accurately verifies excellent analytical capability of the engineered MMFNPs. Benefited from multi‐site membrane anchoring and sensitive ATP profiling, differential membrane disturbances are precisely characterized under distinct μsPEF stimulations. Significantly, the membrane‐repairing process is well demonstrated via monitored ATP fluctuations. Electroporation techniques have emerged as attractive tools for intracellular delivery, rendering promising prospects towards clinical therapies. Transient disruption of membrane permeability is the critical process for efficient electroporation‐based cargo delivery. However, smart nanotools for precise characterization of transient membrane changes induced by strong electric pulses are extremely limited. Herein, multivalent membrane‐anchored fluorescent nanoprobes (MMFNPs) that take advantages of flexible functionalization and spatial arrangement of DNA frameworks are developed for in situ evaluation of electric field‐induced membrane permeability during reversible electroporation . Single‐molecule fluorescence imaging techniques are adopted to precisely verify the excellent analytical performance of the engineered MMFNPs. Benefited from tight membrane anchoring and sensitive adenosine triphosphate (ATP) profiling, varying degrees of membrane disturbances are visually exhibited under different intensities of the microsecond pulse electric field (μsPEF). Significantly, the dynamic process of membrane repair during reversible electroporation is well demonstrated via ATP fluctuations monitored by the designed MMFNPs. Furthermore, molecular dynamics (MD) simulations are performed for accurate verification of electroporation‐driven dynamic cargo entry via membrane nanopores. This work provides an avenue for effectively capturing transient fluctuations of membrane permeability under external stimuli, offering valuable guidance for developing efficient and safe electroporation‐driven delivery strategies for clinical diagnosis and therapeutics.

Abstract:Electroporation has been widely used in delivering foreign biomolecules into cells, but there is still much room for improvement, such as cell viability and integrity. In this manuscript, we investigate the distribution and the toxicity of pH changes during electroporation, which significantly decreases cell viability. A localized pH gradient forms between anode and cathode leading to a localized distribution of cell death near the electrodes, especially cathodes. The toxicity of hydroxyl ions is severe and acute due to their effect in the decomposition of phospholipid bilayer membrane. On the other hand, the electric field used for electroporation aggravates the toxicity of hydroxyl because the electropermeabilization of cell membrane makes bilayer structure more loosen and vulnerable. We also investigate the side effects during scaling down the size of electrodes in electroporation microchips. Higher percentage of cells is damaged when the size of electrodes is smaller. At last, we propose an effective strategy to constrain the change of pH by modifying the composition of electroporation buffer. The modified buffer decreases the changes of pH, thus enables high cell viability even when the electric pulse duration exceeds several milliseconds. This ability has potential advantage in some applications that require long-time electric pulse stimulation.
Nanofocused Electric Field for Localized Single Cell Nanoelectroporation with Membrane Reversibility.

Abstract:Despite the significant research in electroporation, high electric field was applied to the whole cells resulted in permeabilizing the membrane of millions of cells without reversibility [1]. To deliver biomolecules through the specific region of the cell membrane with high cell viability and high transfection rate is important for various biological and therapeutic applications.This report presents a new type localized single cell membrane electroporation (LSCMEP), at specific region of the single cell with the application of 800 μs electric pulse. The ITO nano-electrodes with 100nm thickness and 500 nm gap between two electrodes can generate an intense electric field to track biomolecules inside HeLa cell in our studies. This small gap between two nano-electrodes can neglect thermal effect on cell membrane and permit reversible electroporation with high cell viability (90%) and minimum effected electroporation region (0.48 μm). Our approach successfully delivers biomolecules through a specific region of single cell with high transfection rate (82%) and high cell viability. This process, not only generates well-controlled nano-pores allowing rapid recovery of cell membrane, but also it provides a clear optical path potentially tracking of drugs to deliver inside single cell.

Abstract:Reversible electropermeabilization (or electroporation) of cell membranes is a very efficient method for intracellular delivery of xenomolecules, particularly of DNA. In the case of anchorage-dependent cells, however, enzymatic or mechanical detachment from the substratum is required prior to electropulsing. This can damage the plasma membrane and lead to low transfection yields. Here we present an efficient method for in situ electroporation of mammalian cells while they are attached to a solid substratum. For this purpose an electroporation chamber was constructed that housed a cell culture insert with a cell monolayer grown on a porous filter. By real-time monitoring the transmonolayer resistance, the field pulse parameters resulting in transient and reversible permeabilization of cell membranes were determined for two adherent cell lines, which were found to differ markedly in their sensitivity to electropulsing. Based on the transmonolayer resistance data, the pulsing conditions for optimum electrotransfection of two murine cell lines with plasmid DNA could be established in a very short time. The transfection yield and gene expression were significantly higher in cell monolayers facing the cathode compared to those exposed to field pulses of the reverse direction. This might be due to contribution of the electrophoresis to the translocation of the polyanionic plasmid DNA across the electropermeabilized cell membrane. The experimental setup presented here appears to be a promising tool not only for rapid optimization of in situ electrotransfection of anchorage-dependent cells but also for studying the molecular/biophysical mechanisms of the membrane breakdown and resealing.

Theory of electroporation of planar bilayer membranes: predictions of the aqueous area, change in capacitance, and pore-pore separation

Abstract:A large increase in the transmembrane voltage, U(t), of a fluid bilayer membrane is believed to result in the occurrence of temporary aqueous pathways ("pores") across the membrane. The number, size, and evolution dynamics of these pores are expected to be crucial to the transport of water-soluble species ranging from small ions to macromolecules such as proteins and nucleic acids. In this paper we use a transient aqueous pore theory to estimate the fraction of the membrane area, Fw(t), which is temporarily occupied by water-filled pores for short square, exponential, and bipolar square pulses. For short pulses, "reversible electrical breakdown" occurs when the transmembrane voltage reaches about 1 V, and Fw(t) is predicted to rise rapidly, but always to be less than 10(-3). The conductance of a large number of pores causes reversible electrical breakdown and prevents a significantly larger U from being reached. Despite the large dielectric constant of water, for reversible electroporation the associated change in membrane capacitance, delta C, due to the pores is predicted to be small. Moreover, for a flat membrane the minimum value of the mean pore-pore separation is large, about 60 times the minimum pore radius. In flat membranes, pores are predicted to repel, but the opposite is expected for curved cell membranes, allowing the possibility of coalescence in cell membranes. For some moderate values of U, rupture (irreversible electrical breakdown) occurs, as one or more supracritical pores expand to the membrane boundary and the entire membrane area becomes aqueous. In all cases it is found that a quantitative description of electroporation requires that a pore size distribution, rather than a single size pore.
