Abstract:In this work, a novel nanozyme (Cu@Zr) with all-in-one dual enzyme and fluorescence properties is designed by simple self-assembly. A nanozyme cascade sensor with disodium phenyl phosphate (PPDS) as substrate was first established by exploiting the dual enzymatic activities of phosphatase and laccase. Specifically, phosphatase cleaves the P-O bond of PPDS to produce colorless phenol, which is then oxidized by laccase and complexed with the chromogenic agent 4-aminoantipyrine (4-AP) to produce...

What problem does this paper attempt to address?
### Problems the Paper Attempts to Solve This paper aims to develop a novel nanozyme (Cu@Zr) with dual enzyme activities of phosphatase and laccase, as well as fluorescent properties. This nanozyme was designed through a simple self-assembly method, and a biosensor based on a dual-enzyme cascade reaction was established for portable urease analysis. Specifically, the paper addresses the following issues: 1. **Challenges in Urease Detection**: - Urease is widely present in microorganisms, plants, and animals, and is of great significance for disease diagnosis, environmental monitoring, and agricultural production. - Existing urease detection methods (such as fluorescence, colorimetry, surface-enhanced Raman scattering, and electrochemical methods) are usually based on single signal output, making it difficult to achieve rapid and accurate on-site detection. 2. **Portable Urease Detection with Multi-Signal Output**: - A nanozyme (Cu@Zr) with dual enzyme activities (phosphatase and laccase) was developed, and its catalytic activity was used to establish a cascade reaction sensor. - Through the cascade reaction, high-sensitivity colorimetric and fluorescent dual-mode detection of urease was achieved, with detection limits of 3.56 U/L and 1.83 U/L, respectively. 3. **Portable On-Site Detection**: - A portable test strip loaded with Cu@Zr nanozyme was prepared, enabling on-site urease detection through smartphone RGB reading. - This portable test strip demonstrated good accuracy and anti-interference performance in practical applications, suitable for urease detection in environmental and biological samples. ### Main Contributions - **Design and Synthesis of Novel Nanozyme**: A Cu@Zr nanozyme with dual enzyme activities and fluorescent properties was prepared through a self-assembly method. - **Construction of Cascade Reaction Sensor**: Utilizing the dual enzyme activities of Cu@Zr, a cascade reaction sensor based on phosphatase and laccase was established. - **High-Sensitivity Dual-Mode Detection**: High-sensitivity colorimetric and fluorescent dual-mode detection of urease was achieved, with detection limits as low as 3.56 U/L and 1.83 U/L. - **Portable On-Site Detection Tool**: A portable test strip was developed, enabling on-site urease detection through smartphone RGB reading, suitable for the analysis of environmental and biological samples. ### Conclusion This study successfully developed a fluorescent nanozyme (Cu@Zr) with dual enzyme activities of phosphatase and laccase, and achieved high-sensitivity colorimetric and fluorescent dual-mode detection of urease through a cascade reaction sensor. Additionally, a portable test strip was developed for on-site urease detection, providing a convenient and promising method for the analysis of environmental and biological samples.
