Abstract:In this report, originally proposed apratoxin E (30S-7), revised apratoxin E (30R-7), and (30S)/(30R)-oxoapratoxin E (30S)-38/(30R)-38 were efficiently prepared by two synthetic methods. The chiral lactone 10, recycled from the degradation of saponin glycosides, was utilized to prepare the key nonpeptide fragment 9. Our alternative convergent assembly strategy was applied to the divergent synthesis of revised apratoxin E and its three analogues. Moreover, ring-closing metathesis (RCM) was for the first time found to be an efficient strategy for the macrocyclization of apratoxins.

Abstract:Apratoxin A, a cyclodepsipeptide isolated from a marine cyanobacterium, demonstrates potent cytotoxicity against cancer cell lines by a unique mechanism. A lactam analogue of apratoxin A, named as amidapratoxin A, was efficiently synthesized over 22 linear steps in 2.1% overall yield for the first time. The further conformation analysis was conducted by NMR techniques and computer-based molecular modeling. The results showed that the orientation of the benzene ring in tyrosine moiety, the iso-butyl in isoleucine moiety and hydroxyl group in Ahtmna moiety are quite different from that of apratoxin A, which might result in a significant decrease of the cytotoxicity. While further investigation is on the way, these results provide increased understanding of conformation-activity relationship for apratoxin family members which is an important complement to structure-activity relationship.

Abstract:An efficient method for asymmetric synthesis of apratoxin E 2 is described in this report. The chiral lactone 8, recycled from the degradation of saponin glycosides, was utilized to prepare the non-peptide fragment 6. In addition to this "from nature to nature" strategy, olefin cross-metathesis (CM) was applied as an alternative approach for the formation of the double bond. Moreover, pentafluorophenyl diphenylphosphinate was found to be an efficient condensation reagent for the macrocyclization.

Abstract:Asymmetric total synthesis of compound 1, as a proposed molecular structure of a natural product, in 11 steps is described. The inconsistency of the characterization data between our synthesized sample and the natural product prompted us to propose a different molecular structure as compound 2 and accordingly accomplish total synthesis in 9 steps and confirm the structural revision of this natural product. Both total syntheses feature highly regio- and diastereoselective epoxidation, Stille cross-coupling and cross-metathesis.

Abstract:Herein, we report a concise and collective total synthesis of three natural products aranorosinol A, aranorosinol B, and EI-2128-1 and also the known penicimutanolone. A unified strategy was conceived, which divergently accessed all four congeners within 9-11 steps (LLS) from a common intermediate. Bisoxirane-directed 1,2-addition was utilized as a key step to generate the desired configuration of the C8 stereocenter of aranorosinol A and B, 28 years after their first isolation, and these were both assigned as S configurations, while the C4' and C6' configurations of EI-2128-1 were both determined to be R,R-configurations.

Abstract:The intriguing structure of tagetitoxin (1), a long-standing challenge in natural product synthesis, has been the subject of multiple revisions and has been confirmed through total synthesis. The route commences from a renewable furan starting material and features a number of unusual transformations (such as rearrangements, bromocyclization, and P(V)-based phosphate installation) to arrive at the target in 15 steps. As the route was designed to enable access to both enantiomers, the absolute configuration of the natural product could be assigned using a bioassay on (+)-1 and (-)-1.

Abstract:Aaptaminoids are a unique family of marine alkaloids bearing a benzo[ de ][1,6]-naphthyridine core. This work describes the first total synthesis of suberitines A-D ( 1 - 4 ), four typical dimeric natural aaptaminoids, employing a step-saving bidirectional strategy. Key methods applied in the total synthesis include a cationic cascade to construct the bis-isoquinoline(s) with Hendrickson reagent-mediated Friedel-Crafts-type cyclization and eliminative aromatization, and a Bronsted acid-promoted Vilsmeier cyclization to generate the naphthyridine(s). The conditionally tunable PIDA-mediated oxidative dearomatization and subsequent methanolysis or hydrolysis successfully served as a powerful biomimetic tool to elaborate the essential oxygenated functionalities of suberitines A-D ( 1-4 ) in proper solvent-combinations at the final stage of total synthesis. The biomimetic proposal employed in the late-stage redox interchanges of related natural products was eventually supported by the isolation of synthetic intermediate 23a as a natural product from the same natural source. Biological screening revealed that five of the synthetic samples including two natural suberitines and three full-skeleton natural product-like intermediates exhibited low micromolar inhibitory activities against the growth of cancer cell line K562.

Abstract:Cephalotaxus diterpenoids are attractive natural products with intriguing molecular frameworks and promising biological features. As a structurally unusual member, (-)-cephalotanin B possesses an extraordinarily congested heptacyclic skeleton, three lactone units, and nine consecutive stereocenters. Herein, we report an enantioselective total synthesis of (-)-cephalotanin B based on a divergent asymmetric Michael addition reaction, a novel Pauson-Khand/deacyloxylation process discovered in the development of a second-generation stereoselective Pauson-Khand reaction protocol, and an epoxide-opening/elimination/dual-lactonization cascade to construct the challenging propeller-shaped A-B-C ring system as key transformations.

Abstract:Huperserratines A (1) and B (2), two novel macrocyclic Lycopodium alkaloids, possess an aza-12-membered ring. Here, we describe the first total synthesis of these two natural products in 12 steps. Key elements of the synthesis include Suzuki-Miyaura coupling, Hosomi-Sakurai reaction, ring-closing metathesis, dihydroxylation, and Swern oxidation. A gram-scale ring-closing metathesis was pivotal in forging the challenging aza-12-membered ring. This total synthesis would enable future studies of the structure-activity relationship for 1 and 2.
