Abstract:Bacterial infection poses a significant threat to human health, and the emergence of antibiotic-resistant strains has exacerbated the situation. Antimicrobial photodynamic therapy (aPDT) has emerged as a promising antibiotic-free treatment option that employs reactive oxygen species (ROS) to cause oxidative damage to bacteria and surrounding biomolecules for treating microbial infections. This review summarizes the recent progress in the development of organic photosensitizers, including porphyrins, chlorophyll, phenothiazines, xanthenes and aggregation-induced emission photosensitizers, for aPDT. A detailed description of innovative therapeutic strategies that rely on the infection microenvironment or the unique structural properties of bacteria to amplify the therapeutic effects is provided. Moreover, the combination of aPDT with other therapy strategies such as antimicrobial peptide therapy, photothermal therapy (PTT) or gas therapy, is described. Finally, the current challenges and perspectives of organic photosensitizers for clinical antibacterial applications are discussed.

Abstract:Antimicrobial photodynamic therapy (aPDT) has become a potent contender in the fight against microbial infections, especially in the context of the rising antibiotic resistance crisis. Recently, there has been significant interest in polyphenolic natural products as potential photosensitizers (PSs) in aPDT, given their unique chemical structures and inherent antimicrobial properties. Polyphenolic natural products, abundant and readily obtainable from natural sources, are generally regarded as safe and highly compatible with the human body. This comprehensive review focuses on the latest developments and future implications of using natural polyphenols as PSs in aPDT. Paramount polyphenolic compounds, including curcumin, hypericin, quercetin, hypocrellin, celastrol, riboflavin, resveratrol, gallic acid, and aloe emodin, are elaborated upon with respect to their structural characteristics, absorption properties, and antimicrobial effects. Furthermore, the aPDT mechanism, specifically its targeted action on microbial cells and biofilms, is also discussed. Polyphenolic natural products demonstrate immense potential as PSs in aPDT, representing a promising alternate approach to counteract antibiotic-resistant bacteria and biofilm-related infections.

Abstract:The emergence of multi-drug-resistant pathogens threatens the healthcare systems world-wide. Recent advances in phototherapy (PT) approaches mediated by photo-antimicrobials (PAMs) provide new opportunities for the current serious antibiotic resistance. During the PT treatment, reactive oxygen species or heat produced by PAMs would react with the cell membrane, consequently leaking cytoplasm components and effectively eradicating different pathogens like bacteria, fungi, viruses, and even parasites. This Perspective will concentrate on the development of different organic photo-antimicrobials (OPAMs) and their application as practical therapeutic agents into therapy for local infections, wound dressings, and removal of biofilms from medical devices. We also discuss how to design highly efficient OPAMs by modifying the chemical structure or conjugating with a targeting component. Moreover, this Perspective provides a discussion of the general challenges and direction for OPAMs and what further needs to be done. It is hoped that through this overview, OPAMs can prosper and will be more widely used for microbial infections in the future, especially at a time when the global COVID-19 epidemic is getting more serious.

Abstract:Recently, photoactivated sterilization, as a rapid, effective, and antibiotic-free antibacterial method, has attracted increasing attention from researchers. Many outstanding photoresponsive materials, including photocatalysts, photosensitizers (PSs), and photothermal materials, have been developed and applied to microbial inactivation and to treat infectious bacterial diseases. This mini-review aims to provide insights gathered from studying photoresponsive antibacterial materials and systems, including (1) reactive oxygen species (ROS)-based photodynamic therapy (PDT) and (2) hyperthermia-based photothermal therapy (PTT). In addition, potential application fields for these methods, the remaining challenges to using photoresponsive materials for microbial inactivation, perspectives for the future, and prospective research directions are discussed in depth.

Abstract:Microbial infections represent a grave global healthcare challenge, especially in a "post‐antibiotic" era, prompting the development of new antimicrobial technologies. Herein, a systematic overview of recent advances and emerging trends in the design, nanoengineering, and bioapplications of photoactive antimicrobials is provided, highlighting fundamental theories and advanced strategies for creating photo‐antimicrobials with precise structures and forms and application‐specific biofunctionalities. Currently, microbial infections have posed an arduous challenge to global public health, whereas the rise of antibiotic resistance is rendering traditional antibiotic therapies futile, prompting the development of new antimicrobial technologies. Photoactive nanomaterials have thus garnered a thriving interest for disinfection owing to their superior antibacterial efficaciousness, favorable biosafety, and rapidness and spatiotemporal precision in excreting bactericidal actions. The review summarizes recent advances and emerging trends in the design, nanoengineering, and bioapplications of photoactive antimicrobials. It commences by elaborating fundamental theories on bacterial resistance, and antibacterial mechanisms of nanomaterials and phototherapy. Subsequently, the regulation of the antibacterial effectiveness of photoactive nanomaterials is comprehensively discussed, centering on criteria and strategies for tuning photoabsorption spectra, photothermal conversion, and photocatalytic efficiency, alongside tactics for enabling synergistic therapies. This is followed by comparative analyses of techniques and modalities for synthesizing and engineering photoactive nanomaterials with diverse structures, forms, and functionalities. Thereafter, the state‐of‐the‐art applications of phototherapies across various medical sectors are portrayed, and key challenges and opportunities are finally discussed to spur future innovations and translation. This review is envisaged to provide useful guidance for devising and developing nanomaterials‐based photoresponsive antimicrobials with application‐specific materials properties and biological functions.

Abstract:Bacterial infections pose a significant threat to human health and entail substantial economic losses. Due to the broad-spectrum antibacterial effect and low susceptibility to drug resistance, photodynamic therapy (PDT), a nontraditional antibacterial approach, has garnered a lot of attention. In PDT, the selection of photosensitizer (PS) is crucial because it directly affects the efficiency and safety of the treatment. As a versatile fluorophore, the advantages of 4,4-difluoro-4-bora-3a,4a-diaza-s-indacene (BODIPY) used as a PS for antibacterial PDT are mainly reflected in its high quantum yield of singlet oxygen, easy modification, and exceptional photostability. Through strategic chemical modifications of the BODIPY structures, it is possible to enhance their photodynamic antibacterial activity and refine their selectivity for bacterial killing. This review focuses on the application of BODIPY-based PSs for treating bacterial infections. According to the design strategies of photodynamic antibacterial materials incorporating BODIPY, a variety of representative therapeutic agents having emerged in recent years are classified and discussed, aiming to offer insights for future research and development in this field.

Abstract:Antimicrobial photodynamic therapy (aPDT) has become a fundamental tool in modern therapeutics, notably due to the expanding versatility of photosensitizers (PSs) and the numerous possibilities to combine aPDT with other antimicrobial treatments to combat localized infections. After revisiting the basic principles of aPDT, this review first highlights the current state of the art of curative or preventive aPDT applications with relevant clinical trials. In addition, the most recent developments in photochemistry and photophysics as well as advanced carrier systems in the context of aPDT are provided, with a focus on the latest generations of efficient and versatile PSs and the progress towards hybrid-multicomponent systems. In particular, deeper insight into combinatory aPDT approaches is afforded, involving non-radiative or other light-based modalities. Selected aPDT perspectives are outlined, pointing out new strategies to target and treat microorganisms. Finally, the review works out the evolution of the conceptually simple PDT methodology towards a much more sophisticated, integrated, and innovative technology as an important element of potent antimicrobial strategies.

Abstract:With the growing crisis of the availability of effective antimicrobial treatments, the use of phototherapy has gained increasing interest as an alternative to traditional antibiotic therapy. Even though phototherapy is already used in the clinic, there is an emerging interest in new materials with enhanced antimicrobial activity triggered by light. Among the different light-responsive materials, conjugated polymers are emerging candidates with successful application in the field of antimicrobial photodynamic and photothermal therapy. Due to their organic composition, easy processability, tailorability of physicochemical properties, and capability of reactive oxygen species (ROS) and heat generation, their use as photoresponsive agents against bacteria has greatly expanded. Conjugated polymers have been designed to interact with specific bacterial targets, and their chemical composition has been optimized to enhance ROS and/or heat generation and new cationic polymers have been developed for augmented water solubility and increased interaction with negatively charged bacteria. Herein, the use of conjugated polymers in the field of phototherapy is discussed, focusing on the different approaches that have improved their performance against bacteria and their current preclinical safety assessment.

Abstract:Microbial infections continue to pose a significant health challenge, especially with an increase in drug-resistant bacteria. Conventional antibiotic treatments show limited efficacy, prompting researchers to explore alternative treatments. Photodynamic therapy (PDT) has emerged as a promising alternative that uses reactive oxygen species (ROS) to induce oxidative stress, offering the potential for cyclic treatment without fostering new drug resistance mechanisms. The success of PDT relies heavily on the selection of appropriate photosensitizers (PSs). Various nanomaterials are being developed as PSs or carriers to enhance the efficacy of PDT in the antibacterial field. In this comprehensive review, we discuss the four main ROS generated during PDT and outline their corresponding antibacterial mechanisms. Additionally, we highlight the prominent types of nanomaterials used as PSs or carriers in PDT. We analyze the current challenges associated with nanomaterial-based PDT for antibacterial therapy and propose potential strategies for optimizing their applications.

Abstract:Some infectious or malignant diseases such as cancers are seriously threatening the health of human beings all over the world. The commonly used antibiotic therapy cannot effectively treat these diseases within a short time, and also bring about adverse effects such as drug resistance and immune system damage during long-term systemic treatment. Phototherapy is an emerging antibiotic-free strategy to treat these diseases. Upon light irradiation, phototherapeutic agents can generate cytotoxic reactive oxygen species (ROS) or induce a temperature increase, which leads to the death of targeted cells. These two kinds of killing strategies are referred to as photodynamic therapy (PDT) and photothermal therapy (PTT), respectively. So far, many photo-responsive agents have been developed. Among them, the metal-organic framework (MOF) is becoming one of the most promising photo-responsive materials because its structure and chemical compositions can be easily modulated to achieve specific functions. MOFs can have intrinsic photodynamic or photothermal ability under the rational design of MOF construction, or serve as the carrier of therapeutic agents, owing to its tunable porosity. MOFs also provide feasibility for various combined therapies and targeting methods, which improves the efficiency of phototherapy. In this review, we firstly investigated the principles of phototherapy, and comprehensively summarized recent advances of MOF in PDT, PTT and synergistic therapy, from construction to modification. We expect that our demonstration will shed light on the future development of this field, and bring it one step closer to clinical trials.

Abstract:Antimicrobial photodynamic therapy has considerable promise in the fight against bacterial infections. The superior photophysical characteristics of porphyrins have made them effective photosensitizers in the field of phototherapy. Herein, the light-induced antimicrobial effects of three porphyrins with different substituents have been compared. 5,10,15,20-tetrakis(4-hydroxyphenyl) porphyrin (THPP) shows superior photosensitizing activity and antimicrobial ability under irradiation with green light. THPP can also inhibit and destroy mature Staphylococcus aureus biofilms under irradiation. This work provides a reference for the rational design of photosensitizers for application in antimicrobial photodynamic therapy.
Functional nanomaterials as photosensitizers or delivery systems for antibacterial photodynamic therapy

Abstract:Bacterial infection is a global health problem that closely related to various diseases threatening human life. Although antibiotic therapy has been the mainstream treatment method for various bacterial infectious diseases for decades, the increasing emergence of bacterial drug resistance has brought enormous challenges to the application of antibiotics. Therefore, developing novel antibacterial strategies is of great importance. By producing reactive oxygen species (ROS) with photosensitizers (PSs) under light irradiation, antibacterial photodynamic therapy (aPDT) has emerged as a non-invasive and promising approach for treating bacterial infections without causing drug resistance. However, the insufficient therapeutic penetration, poor hydrophilicity, and poor biocompatibility of traditional PSs greatly limit the efficacy of aPDT. Recently, studies have found that nanomaterials with characteristics of favorable photocatalytic activity, surface plasmonic resonance, easy modification, and high drug loading capacity can improve the therapeutic efficacy of aPDT. In this review, we aim to provide a comprehensive understanding of the mechanism of nanomaterials-mediated aPDT and summarize the representative nanomaterials in aPDT, either as PSs or carriers for PSs. In addition, the combination of advanced nanomaterials-mediated aPDT with other therapies, including targeted therapy, gas therapy, and multidrug resistance (MDR) therapy, is reviewed. Also, the concerns and possible solutions of nanomaterials-based aPDT are discussed. Overall, this review may provide theoretical basis and inspiration for the development of nanomaterials-based aPDT.

Abstract:This review summarizes the mechanism of action of nanomaterials as sensitizing agents or sensitizing carriers in antimicrobial photodynamic therapy and antimicrobial sonodynamic therapy (aPDT/aSDT) and their applications in the treatment of infectious diseases, to address the problem of refractory bacterial infections from multiple perspectives and provide ideas for the clinical transformation of nanotherapeutic systems. Antibacterial photodynamic therapy (aPDT) and antibacterial sonodynamic therapy (aSDT) utilize sensitizers (photosensitizers/sonosensitizers) to produce reactive oxygen species (ROS) for antibacterial treatment under the stimulation of light/ultrasound, which have the characteristics of broad‐spectrum antibacterial properties, low drug‐resistance, and effective targeting of infected tissues. Nanomaterials in aPDT/aSDT are primarily used as nano‐sensitizers or nano‐carriers of sensitizers. They enhance the stability and permeability of sensitizers, improve targeting of sensitizers, strengthen photodynamic/sonodynamic properties of sensitizers (modification of sensitizers absorption efficiency and light/ultrasonic response stress by modulation of nanoparticle shape, size, and structure). Also, they improve modifiability of sensitizers (controlling the release rate and time of the sensitizer as needed to optimize the therapeutic effect), enhance programmability and multifunctionality of sensitizers (flexible application of nanotechnology for designing sensitizers with multiple functions, such as drug delivery, targeted therapy, and therapeutic monitoring), and expand possibilities for combination therapies (the nano‐carriers can be loaded with other therapeutic agents, enabling combination therapies). Nanomaterials are expected to further promote the development of aPDT/aSDT and achieve improved antibacterial effects. This review summarizes the progress in nanomaterials in aPDT/aSDT in recent years and based on the current development strategies to provide a theoretical reference for the application of nanomaterials in aPDT/aSDT.

Abstract:Antimicrobial resistance is threatening to overshadow last century’s medical advances. Etiological agents of previously eradicated infectious diseases are now resurgent as multidrug-resistant strains, especially for Gram-negative strains. Finding new therapeutic solutions is a real challenge for our society. In this framework, Photodynamic Antimicrobial ChemoTherapy relies on the generation of toxic reactive oxygen species in the presence of light, oxygen, and a photosensitizer molecule. The use of reactive oxygen species is common for disinfection processes, using chemical agents, such as chlorine and hydrogen peroxide, and as they do not have a specific molecular target, it decreases the potential of tolerance to the antimicrobial treatment. However, light-driven generated reactive species result in an interesting alternative, as reactive species generation can be easily tuned with light irradiation and several PSs are known for their low environmental impact. Over the past few years, this topic has been thoroughly studied, exploring strategies based on single-molecule PSs (tetrapyrrolic compounds, dipyrrinate derivatives, metal complexes, etc.) or on conjunction with delivery systems. The present work describes some of the most relevant advances of the last 6 years, focusing on photosensitizers design, formulation, and potentiation, aiming for the disinfection of Gram-negative bacteria.Graphical abstract

Abstract:Photodynamic therapy (PDT) — which combines light, oxygen and photosensitizers (PS) to generate reactive oxygen species — has emerged as an effective approach for targeted ablation of pathogenic cells with reduced risk of inducing resistance. Some organic PS are now being applied for PDT in the clinic or undergoing evaluation in clinical trials. A limitation of the first-generation organic PS was their potential off-target toxicity. This shortcoming prompted the design of constructs that can be activated by the presence of specific biomolecules — from small biomolecules to large enzymes — in the target cells. Here, we review advances in the design and synthesis of activatable organic PS and their contribution to PDT in the past decade. Important areas of research include novel synthetic methodologies to engineer smart PS with tuneable singlet oxygen generation, their integration into larger constructs such as bioconjugates, and finally, representative examples of their translational potential as antimicrobial and anticancer therapies.

Abstract:Photodynamic therapy (PDT) is a long known, FDA approved, and highly promising therapeutic modality, which is still developing. Photosensitizers (PSs) are at the core of PDT action and a worldwide effort has been put to develop PSs having multiple functionalities. In the design of new generation PSs, an attractive approach is to target organelles to get an exceptional therapeutic outcome as damaging these vital subcellular compartments improve the photocytotoxic effect of the PSs. To this end, small molecule organic PSs are suitable candidates due to ease of chemical modification, which allows simple implementation of the organelle targeting units on the PS structure. Furthermore, they hold unique characteristics such as low dark toxicity, high reactive oxygen species (ROS) generation capacity, biocompatibility, and tunable photophysical and photochemical properties. Accordingly, a wide variety of organelle-targeted organic PSs have been developed and proved to be highly successful both in therapeutic action and bioimaging applications. In this review article, we have summarized the recent advances in organelle localizing organic-based PSs by discussing the design principles, organelle targeting strategies and related bio-applications in detail. PSs targeting seven different organelles have been included. We have also highlighted the current challenges and gave a comprehensive outlook for the future of organelle-targeted PSs.

Beyond Antibiotics: Photo/Sonodynamic Approaches for Bacterial Theranostics

Abstract:Abstract Rapid evolution and propagation of multidrug resistance among bacterial pathogens are outpacing the development of new antibiotics, but antimicrobial photodynamic therapy (aPDT) provides an excellent alternative. This treatment depends on the interaction between light and photoactivated sensitizer to generate reactive oxygen species (ROS), which are highly cytotoxic to induce apoptosis in virtually all microorganisms without resistance concern. When replacing light with low-frequency ultrasonic wave to activate sensitizer, a novel ultrasound-driven treatment emerges as antimicrobial sonodynamic therapy (aSDT). Recent advances in aPDT and aSDT reveal golden opportunities for the management of multidrug resistant bacterial infections, especially in the theranostic application where imaging diagnosis can be accomplished facilely with the inherent optical characteristics of sensitizers, and the generated ROS by aPDT/SDT cause broad-spectrum oxidative damage for sterilization. In this review, we systemically outline the mechanisms, targets, and current progress of aPDT/SDT for bacterial theranostic application. Furthermore, potential limitations and future perspectives are also highlighted.
