Abstract:OBJECTIVE:To investigate the association between genetic polymorphisms of inflammatory factors and susceptibility to coronary heart disease(CHD) in southern Chinese Han population.METHODS:Using matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry (MALDI-TOF-MS) method, the genotypes of five inflammatory factors (BRCA1-associated protein, a disintegrin and metalloproteinase 8, inter-alpha-trypsin inhibitor H3, interleukin-15, cyclooxygenase-2) were anaylzed in 283 CHD patients diagnosed by angiography and 176 controls.RESULTS:In these inflammatory factors, the 270T/C and 90A/G polymorphisms of the BRAP gene showed a significant association with CHD. The allele and genotype frequencies of BRAP gene were consistent with those predicted by Hardy-Weinberg equilibrium (chi-square=0.878, P> 0.05; chi-square=0.776, P> 0.05, respectively). The frequencecies of 270C and 90G alleles in CHD patients was significantly higher than those of the control group (29.51% vs. 21.31%, P=0.006; 30.04% vs. 21.31%, P=0.004, respectively). Compared with 270TT and 90AA, 270CC and 90GG genotypes had a significantly increased CHD risk by Logistic regression analysis (OR=4.51, 95%CI: 1.41-14.45, P=0.011; OR=5.09, 95%CI: 1.60-16.26, P=0.006, respectively). This association was still signifcant after adjustment for the sex, age, smoke, hypertension, diabetes, plasma total cholesterol and low density lipoprotein levels. No evidence of association was found for other single nucleotide polymorphisms.CONCLUSION:The 270T/C and 90A/G polymorphisms in the BRAP gene may contribute to an increased risk of CHD among southern Chinese Han population.

Abstract:Objective: We performed this study to investigate whether the EDNRA gene rs1878406 C > T polymorphism is associated with risk of large artery atherosclerosis (LAA) stroke in the Chinese Han population.Methods: Genotyping of rs1878406 was performed in 1,112 LAA stroke patients and 1,192 healthy controls. Multivariate logistic regression analyses were applied to assess the effect of the rs1878406 C > T polymorphism on susceptibility to LAA stroke.Results: A significant increase of LAA stroke risk was found in the recessive model (TT vs. CC/TC, OR = 1.74, 95% CI = 1.23-2.48, p = 0.002) and co-dominant model (TC vs. CC, OR = 1.06, 95% CI = 0.89-1.27, TT vs. CC, OR = 1.79, 95% CI = 1.25-2.55, p = 0.006). However, the interaction between age and genotypes of rs1878406 was not statistically significant, and no significant interactive effect was observed between the rs1878406 C > T polymorphism and sex (p > 0.05).Conclusion: The rs1878406 C > T polymorphism is associated with increased risk of LAA stroke in the Chinese Han population.
Correlation Between Rs198388 and Rs198389 Polymorphismsin Brainnatriuretic Peptide (NPPB) Gene and Susceptibility to Congenital Heart Diseases in a Chinese Population.

Abstract:OBJECTIVEWe discussed the correlation between SNP loci (rs198389 and rs198388) in brain natriuretic peptide gene (NPPB) and susceptibility to congenital heart diseases (CHD).METHODMultiplex SNaPshot technique was adopted for profiling of SNP genotypes at loci rs198389 and rs198388 in NPPB gene among 150 cases of CHDand 150 normal controls.RESULTSThe distribution frequency of 3 genotypes (AA, AG and GG) at locus rs198389 was 40.7%, 36.0% and 23.3% in CHD group, respectively, showing significant differences compared with the normal controls (P<0.001). Gallele was associated with higher risk of CHD (OR=2.48, 95% CI=1.77-3.48). The distribution frequency of CC, CTand TT genotypes at locus rs198388 was 60.7%, 17.3% and 22.0% in CHD group, respectively, also showing significant differences compared with the normal controls (P<0.001). C allele could increase the risk of CHD (OR=1.92, 95% CI=1.48-2.48).CONCLUSIONSNP loci rs198389 and rs198388 in NPPB gene were correlated with genetic susceptibility to CHD.
A Comprehensive Contribution of Genetic Variations of the Insulin-Like Growth Factor 1 Signalling Pathway to Stroke Susceptibility

Abstract:Background and aims: The insulin-like growth factor (IGF)-1 signalling pathway has been implicated in the pathogenesis of atherosclerosis; however, the mechanism underlying its role in stroke remains unexplained. Herein, we aimed to explore the effects of genetic polymorphisms in the IGF1 pathway on stroke in the Chinese Han population. Methods: Twenty-six single-nucleotide polymorphisms (SNPs) in IGF1 pathway genes were genotyped in a case-control study consisting of 2070 stroke cases and 2243 controls. Main genetic effects and gene-gene interactive effects of the IGF1 pathway were evaluated. Weighted genetic risk scores (wGRS) were computed, and the associations between wGRS and gene expression were analysed. Results: The variants at GHRH rs6032470 were significantly associated with high risk of hemorrhagic stroke (HS), and the adjusted OR (95%CI) was 1.368 (1.136-1.647). Significant additive interaction between rs6032470 and gender was detected for HS and ischemic stroke (IS). The association of rs6032470 and stroke was stronger in males than in females. Additionally, a significant gene-gene interaction of rs6032470-rs1874479 (IGFBP1) in relation to HS risk was identified (p < 0.05). IGF1 mRNA expression was significantly upregulated in IS, while it was linearly downregulated across rs6214 genotypes. In addition, IGFBP3 transcript variant 2 mRNA level was negatively correlated with wGRS (r = -0.285, p = 0.005). Conclusions: Our findings indicated that the IGF1 signalling pathway genes potentiated the risk of stroke through both main effects and gene-gene interactions. The genetic effect of GHRH rs6032470 on stroke was gender dependent. The wGRS of IGF1 pathway genes may be an independent predictor of stroke risk.

Abstract:Objective: To determine whether polymorphisms in the genes for coagulation factor. II, V, VII could predispose an individual to increase risk for coronary artery disease (CAD) and/or myocardial infarction (MI) in Chinese. Methods: We screened coagulation factor II(G20210A), V(G1691A), VII (R353Q and HVR4) genotype in 374 patients undergoing coronary angiography by polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) assay. Results: The R353Q and HVR4 gemotype of the factor VII distribution was in accordance with Hardy-Weinberg equilibrium. The frequencies of FVII genotype or allele did not show statistically significant differences between CAD group and controls or between male and female. The frequencies of the Q allele and (RQ+QQ) genotype were significantly higher among the CAD patients without myocardial infarction (MI) history than among those with MI history (P<0.05). However, HVR4 polymorphism was not significantly different within groups. We only find one normal control of factorII (G20210A) mutation. No coagulation factor V(G1691A) mutation was found in the CAD patients and controls. Conclusion: The factor II(G20210A), V(G1691A) mutation is absent and may not be a major genetic factor for CAD and/or MI; the Q allele of the R353 Q polymorphism of the factor VII gene may be a protective genetic factor against myocardial infarction in Chinese.

Abstract:Abstract Background Endometrial cancer is one of the most common female reproductive system tumors. Ninjurin2 ( NINJ2 ) is a new adhesion factor. As a vascular susceptibility gene, it is highly expressed in other cancers and promotes the growth of cancer cells. We conducted an association analysis between NINJ2 gene polymorphism and endometrial cancer risk. Methods Five SNPs rs118050317, rs75750647, rs7307242, rs10849390 and rs11610368 of NINJ2 gene were genotyped in 351 endometrial cancer patients and 344 healthy controls. The clinical index difference between cases and controls were tested by one-way analysis of variance. The allele and genotype frequency of cases and controls were been compared by Chi square test. The odds ratios (OR) with 95% confidence interval (95% CI) were examined by logistic regression analysis. Results The SNP rs118050317 mutant allele C and homozygote CC genotype were significant increased the endometrial cancer risk (OR 1.46, 95% CI 1.04–2.06, p = 0.028; OR 8.43, 95% CI 1.05–67.89, p = 0.045). In the clinical index analysis, there were significant higher quantities of CEA, CA125 and AFP in cases serum than controls. Conclusion The NINJ2 gene polymorphism loci rs118050317 mutant allele C was associated with an increased risk of endometrial cancer. CEA, CA125 and AFP quantities were significant higher in endometrial cancer patients.

Abstract:OBJECTIVE:To investigate the association between the NINJ2 gene rs11833579 polymorphism and stroke in Han Chinese population.METHODS:This study was a population-based cross-sectional case-control study. Polymerase chain reaction-restriction fragment length polymorphism (RFLP) and sequencing were used for the detection of NINJ2 genotypes in 790 patients with stroke (679 ischemic stroke) which were Han Chinese population from Fangshan First Hospital and 811 controls which were healthy Han Chinese population without family history of stroke in Fangshan district rural area.RESULTS:In rs11833579 locus of the NINJ2 gene, the frequencies of GG genotype and allele G were higher in ischemic stroke patients than that in controls (P<0.001). The frequency of allele G of the NINJ2 gene was higher in cerebral hemorrhage patients than that in controls (P=0.005). Genotype had little effect on the glucose, total cholesterol and triglyceride.CONCLUSION:There is significant association between rs11833579 site polymorphism of the NINJ2 gene and risk for stroke in Han Chinese population from Fangshan district.

Abstract:Recently, a genome-wide association study reported an association between two single nucleotide polymorphisms (SNPs) rs11833579 and rs12425791 near NINJ2 gene and ischemic stroke in Caucasians. Therefore, NINJ2 gene is an important candidate locus in the prevalence of ischemic stroke. We performed a hospital based genetic association study in Chinese Han subjects to investigate the relationship between NINJ2 gene and ischemic stroke. We genotyped 14 tagging single nucleotide polymorphisms (tSNP) in 749 ischemic stroke subjects and 924 control subjects and conducted the association between these tSNPs and ischemic stroke. We detected a tSNP rs10849373 in the first intron of the NINJ2 gene significantly associated with ischemic stroke (both genotype and allelic p=0.0001). The minor A allele increased the risk of ischemic stroke with a per-allele OR of 1.37 for the additive genetic model in univariate analysis (p=0.0001). The significance remained after adjustment for the covariates of age, gender, BMI, cigarette smoking, alcohol drinking, hypertension, and diabetes. Therefore, we report a new genetic variant, rs10849373, located in the first intron of the NINJ2 gene, conferring risk of ischemic stroke in Chinese Han subjects. Further genetic association and functional studies are required to search the causal functional variant in linkage disequilibrium with this polymorphism.

Abstract:In recent years, the genetic factor has become one of the important predisposing factors of nephropathy susceptibility. There is a high incidence of nephropathy in CCVd. The CYP2C19 enzyme metabolizes most the drugs, including proton pump inhibitors commonly used medicines to treat CCVd, CYP2C19 genetic polymorphisms is association with multi-pathogenesis factors of nephropathy. The purpose of the study is to reveal the association between CYP2C19 genotype and the susceptibility of nephropathy in the CCVd patients. The study is composed of 623 samples from CCVd treated by anticoagulation. The patients were studied, including CCVd with hyperuricemia, coronary heart disease, diabetes, and other complication. Biochemical tests and CYP2C19 variants measurements were performed by the gene chip method. The association among CYP2C19 variants, complications, and nephropathy was analyzed in the CCVd. There is no correlation between nephropathy and complications in CCVd. In hyperuricemia, coronary heart disease and diabetes groups, the differences of renal function tests were significant between CYP2C19 mutant (P<0.05). The nephropathy risk of wild genotype is 3.288 times higher than of mutation genotype in hyperuricemic group, 1.928 times higher than mutation genotype in coronary heart disease group, and 5.248 times higher than CYP2C19 mutation genotype in the diabetic group. There was significant correlation between the CYP2C19 wild type and the nephropathy susceptibility in CCVd patients. The CYP2C19 gene plays a potential maker to evaluate nephropathy in CCVd patients. We deduced that identification of CYP2C19 gene type may benefit for reducing and avoiding nephropathy caused by abnormal metabolism function in CCVd patients.
Genetic Polymorphisms of UTS2 Rs2890565 Ser89Asn in Coronary Heart Disease and Myocardial Infarction in Chinese Population

Abstract:Objective: Atherosclerosis plays a key role in the inducibility and persistence of coronary heart disease. Clinical evidence, in vitro and in vivo studies have implicated Urotensin II (U-II/UTS2) in the development of atherosclerosis and coronary artery disease, contributing to the (patho) physiological regulation of cardiovascular homeostasis in humans. Increased U-II plasma levels have been reported in patients with atherosclerosis and coronary heart disease. Considering these, our objective was to evaluate possible role of the UTS2 gene polymorphisms (Thr21Met and Ser89Asn) in the genetic susceptibility to coronary heart disease and myocardial infarction in a Chinese population. Methods: A case-control study was designed to compare the distribution of alleles and genotypes between case group (subjects with myocardial infarction, n = 409) and control group (subjects with coronary heart disease, n = 830). The detection of UTS2 gene polymorphisms was achieved with PCR-RFLP technique. Results: We did not identify statistically significant differences between the myocardial infarction and coronary heart disease groups, neither with regard to the frequency of genotype/variant at the Ser89Asn locus nor at the Thr21Met locus. When stratified by sex, differences in genotype distribution of polymorphism Ser89Asn were only seen in female subjects in both additive tested inheritance model (OR = 0.257, 95% CI: 0.074-0.896, P = 0.033) and recessive tested inheritance model (OR = 0.280, 95% CI: 0.082-0.955, P = 0.042). For subjects with myocardial infarction, we identified statistically significant differences between the ST-segment elevation myocardial infarction and non ST-segment elevation myocardial infarction groups. Differences in genotype distribution of polymorphism Ser89Asn not Thr21Met were seen in both additive tested inheritance model (OR = 0.202, 95% CI: 0.049-0.833, P = 0.027) and recessive tested inheritance model (OR = 0.208, 95% CI: 0.052-0.835, P = 0.027). When stratified by sex, differences in genotype distribution of polymorphism Ser89Asn were only seen in male subjects in both additive tested inheritance model (OR = 0.208, 95% CI: 0.049-0.890, P = 0.034) and recessive tested inheritance model (OR = 0.197, 95% CI: 0.047-0.824, P = 0.026). Conclusions: Ser89Asn (S89N) polymorphisms of the UTS2 gene were significantly associated with coronary heart disease and myocardial infarction in Chinese population. Additionally, we demonstrated that Genotype Asn89Asn may imply a potential benefit role for myocardial infarction.
A Novel Functional Polymorphism in the NINJ2 Promoter Predicts Risk of Large Artery Atherosclerotic Stroke

Abstract:A recent genome-wide association study (GWAS) identified two common polymorphisms (rs12425791 and rs11833579) on chromosome 12p13 that confer risk to stroke, particularly for large artery atherosclerotic (LAA) stroke. However, these two polymorphisms are located ∼11 kb upstream of the NINJ2 gene and their effects on NINJ2 expression have not been well characterized. Through linkage disequilibrium and fine-mapping analysis, we identified a novel functional polymorphism in the NINJ2 promoter (rs3809263 G > A) and examined its association with risk of LAA stroke in Chinese population. Rs3809263 was genotyped using the improved multiple ligase detection reaction in 414 patients with LAA stroke and 423 healthy controls. A significant decreased risk of LAA stroke was found for the rs3809263 GA (adjusted odd ratio [OR] = 0.63, 95 % confidence interval [CI] = 0.46–0.88) and AA (OR = 0.54, 95 % CI = 0.35–0.84) genotypes. Moreover, genotype-phenotype correlation analysis indicated that the AA genotype carriers had significantly increased NINJ2 mRNA expression levels in the Chinese population, suggesting that the rs3809263 G > A polymorphism is a functional SNP and a biomarker for risk of LAA stroke. Further validation of the functionality of the NINJ2 rs3809263 G > A polymorphism and its association with risk of LAA stroke in other ethnic populations is warranted.

Abstract:Genome-wide association studies have identified 2 single-nucleotide polymorphisms (SNPs) on chromosome arm 9p21, rs10757278, and rs2383207 that confer susceptibility to myocardial infarction. However, these data are mostly from Italian, American Caucasian, South Korean, and Japanese cohorts. This study is the first to investigate whether 6 SNPs (rs10757277, rs10757278, rs10757279, rs1333049, rs1333047, and rs10811656) are associated with acute coronary syndrome (ACS) in a Chinese Han population.We performed a case-control analysis in 359 patients with ACS diagnosed by coronary angiography and 398 non-ACS controls of Han background between April 2007 and January 2008 to determine whether these 6 SNPs were associated with ACS. Exon fragments were genotyped by polymerase chain reaction-restriction fragment length polymorphism.After we adjusted for clinical parameters, we found the rs10757278 GG genotype to be associated with a significantly elevated risk of ACS (odds ratio [OR], 1.91; 95% confidence interval [CI], 1.35-2.68; P = 0.00035), the rs10811656 T allele to be associated with a higher risk of ACS (OR, 1.67; 95% CI, 1.26-2.23; P = 0.0016) than the CC genotype, and the rs1333047 TT genotype also to be associated with a higher risk of ACS (OR, 1.57; 95% CI, 1.15-2.06; P = 0.0052) than the CC and CT genotypes. After 14.2 ± 4.5 months of follow-up, the end-point data were obtained: death (cardiac and noncardiac), nonfatal myocardial infarction, and recurrent angina leading to repeated coronary angiography. We found that the rs10757278 GG genotype was significantly associated with recurrent angina compared with the AA and AG genotypes (P = 0.013).Polymorphisms on 9p21 were associated with ACS in a Chinese Han population. The rs10757278 GG genotype was further associated with adverse cardiac outcomes after ACS.
Association Between CYP2D7 and TCF20 Polymorphisms and Coronary Heart Disease

Abstract:One of the causes of coronary heart disease (CHD) is genetic factors. In this study, we explored the relationship between CYP2D7 and TCF20 gene polymorphisms and the risk of CHD in the Han Chinese population. Three single nucleotide polymorphisms (CYP2D7 rs1800754, CYP2D7 rs2743461, and TCF20 rs760648) were selected and genotyped from 490 cases and 480 controls. The odds ratios (ORs) and 95% confidence intervals (CIs) were used to assess the association between CYP2D7 and TCF20 polymorphisms and the risk of CHD. The association between clinical indicators and polymorphisms was analyzed using one-way ANOVA and Tukey's HSD. The SNP-SNP interactions were obtained by performing multifactor dimensionality reduction (MDR). CYP2D7 rs1800754 and rs2743461 were closely associated with increased risk of CHD (alleles: p = 0.014, p = 0.031). Stratified analysis showed that CYP2D7 rs1800754 and rs2743461 were associated with an increased risk of CHD in men, age > 60 years, BMI ≥ 24, and smoking. Rs1800754 is also associated with an increased risk of CHD associated with alcohol consumption. In addition, TCF20 rs760648 was associated with a reduced risk of CHD in patients aged ≤ 60 years and with CALs. A significant association was found between CYP2D7 rs1800754 and rs2743461 genotypes and levels of UREA, Cr, and LDL-C; TCF20 rs760648 genotypes and levels of RBC. The MDR analysis showed that the three-locus interaction model was the best in the multi-locus model. In conclusion, CYP2D7 rs1800754 and rs2743461 polymorphisms were associated with CHD risk.
NINJ2 deficiency inhibits preadipocyte differentiation and promotes insulin resistance through regulating insulin signaling

Abstract:Objective Genetic variants in ninjurin‐2 (NINJ2; nerve injury‐induced protein 2) confer risk of ischemic strokes and coronary artery disease as well as endothelial activation and inflammation. However, little is known about NINJ2's in vivo functions and underlying mechanisms. Methods The phenotypes of NINJ2 knockout mice were analyzed, and mechanisms of NINJ2 that regulate body weight, insulin resistance, and glucose homeostasis and lipogenesis were investigated in vivo and in vitro. Results This study found that mice lacking NINJ2 showed impaired adipogenesis, increased insulin resistance, and abnormal glucose homeostasis, all of which are risk factors for strokes and coronary artery disease. Mechanistically, NINJ2 directly interacts with insulin receptor/insulin‐like growth factor 1 receptor (INSR/IGF1R), and NINJ2 knockdown can block insulin‐induced mitotic clonal expansion during preadipocyte differentiation by inhibiting protein kinase B/extracellular signal‐regulated kinase (AKT/ERK) signaling and by decreasing the expression of key adipocyte transcriptional regulators CCAAT/enhancer‐binding protein β (C/EBP‐β), C/EBP‐α, and peroxisome proliferator‐activated receptor γ (PPAR‐γ). Furthermore, the interaction between NINJ2 and INSR/IGF1R is needed for maintaining insulin sensitivity in adipocytes and muscle via AKT and glucose transporter type 4. Notably, adenovirus‐mediated NINJ2 overexpression can ameliorate diet‐induced insulin resistance in mice. Conclusions In conclusion, these findings reveal NINJ2 as an important new facilitator of insulin receptors, and the authors propose a unique regulatory mechanism between insulin signaling, adipogenesis, and insulin resistance.

Tissue Factor Pathway Inhibitor-2 Gene Polymorphisms Associate with Coronary Atherosclerosis in Chinese Population.

Abstract:Tissue factor pathway inhibitor-2 (TFPI-2) may play critical roles in the pathogenesis of atherosclerosis. In this study, we aimed to investigate the association between TFPI-2 gene polymorphisms and coronary atherosclerosis.Four hundred and seven patients with coronary atherosclerosis and 306 individuals with normal coronary artery were enrolled in the present study. Nine single-nucleotide polymorphisms (SNPs) (rs3763473, rs59805398, rs60215632, rs59999573, rs59740167, rs34489123, rs4517, rs4264, and rs4271) were detected with polymerase chain reaction-direct sequencing method. Severity of coronary atherosclerosis was assessed by Gensini score. After the baseline investigation, patients with coronary atherosclerosis were followed up for incidence of cardiovascular events (CVEs).Eight SNPs were in accordance with the Hardy-Weinberg equilibrium, and 8 haplotypes were constructed based on rs59999573, rs59740167, and rs34489123 after linkage disequilibrium and haplotype analysis. Two SNPs (rs59805398 and rs34489123) and 5 haplotypes correlated with coronary atherosclerosis even after adjustment by Gensini score. At follow-up (median 53 months, range 1-60 months), 85 patients experienced CVE. However, there was no strong association between the gene polymorphisms and the occurrence of CVE.Tissue factor pathway inhibitor-2 gene polymorphisms were associated with coronary atherosclerosis in the Chinese population, suggesting that the information about TFPI-2 gene polymorphisms was useful for assessing the risk of developing coronary atherosclerosis, but there was not enough evidence showing it could predict occurrence of CVE.

Abstract:Objective: To investigate the relation of tissue factor pathway inhibitor 2 (TFPI-2) gene polymorphism and acute coronary syndrome (ACS) in Chinese Han population. Methods: The polymorphism of TFPI-2 gene was detected by PCR-sequencing in all people recruited, and a case control study was carried out in three groups, the ACS group consisted of 140 ACS patients (including 68 unstable angina patients and 72 acute myocardial infarction patients), the SA group included 273 stable angina (SA) patients, and the control group consisted of 306 healthy people. Results: Eight single nucleotide polymorphism (SNP) sites were identified in TFPI-2 gene in all subjects, the allele and genotype frequencies were different on the two SNP sites: rs59805398 and rs34489123 among case and control group. Linkage disequilibrium among the SNPs of rs3763473, rs59999573, rs59740167, rs34489123 of TFPI-2 gene was found and C-C-G-G-G and T-C-G-G-G were predominant haplotypes in ACS patients. Linkage disequilibrium among the SNPs of rs59999573, rs59740167, rs34489123 of TFPI-2 gene was found and C-G-G-G and G-A-A-A were predominant haplotypes in SA patients. There was no significant difference between ACS group and SA group. Conclusions: C allele in rs59805398 and A allele in rs34489123 have correlation with coronary heart disrase (CHD), implying that they might be a higher risk factor of CHD among Chinese Han population. There is apparent linkage disequilibrium among SNPs of TFPI-2 gene in CHD patients.

Abstract:Objective To explore the pleiotropic effects of genotypes at rs10757274 locus in coronary heart disease(CHD) susceptibility gene CDKN2B-ASI on type 2 diabetes mellitus (T2DM) and hyperlipemia and to provide the new idea for genetic etiology study of cardiovascular and cerebrovascular diseases.Methods In present hospital-based case-control study,1 158 CHD patients were the subjects,which included 242 T2DM without hyperlipemia cases (20.9％),306 non-T2DM with hyperlipemia cases (26.4％).204 T2DM with hyperlipemia cases (17.6％) and 406 controls (non-T2DM without hyperlipemia cases,35.1％).All subjects were Han people.The present study was performed by questionnaire,physical examination and biochemical test for all subjects.The single base prime extension genotyping technology and GenomeLab SNP stream genotyping system were used to conduct genotyping.Multivariate unconditional logistic regression analysis were utilize to analyzethe correlation with adjustments for age,sex,BMI,smoking,alcohol drinking,hypertension and myocardial infarction.Results As compared with genotype GG,genotype GA/rs10757274 (OR=2.43.95％CI:1.58-3.70) and genotype AA/rs10757274 (OR=2.64.95％CI:1.62-4.30) could increase the risk ofT2DM.Genotype GA/rs10757274 (OR=2.25.95％CI:1.53-3.30) and genotype AA/rs10757274 (OR=1.89.95％CI:1.20-2.97) could increase the risk of hyperlipemia.Genotype GA/rs10757274 (OR=1.86.95％CI:1.15-3.01) and genotype AA/rs10757274 (OR=4.21.95％CI:2.55-6.92) could increase the risk of T2DM with hyperlipemia.Conclusion There are the pleiotropic effects of genotypes at rs10757274 locus of ge CDKN2B-AS1 on T2DM and hyperlioemia.

Abstract:Novel susceptibility genes related to ischemic stroke (IS) are proposed in recent literatures. Population-based replicate studies would cause false positive results due to population stratification. 229 recruit IS patients and their 229 non-IS siblings were used in this study to avoid population stratification. The family-based study was conducted in Beijing from June 2005 to June 2012. Association between SNPs and IS was found in the sibship discordant tests, and the conditional logistic regression was performed to identify effect size and explore gene–environment interactions. Significant allelic association was identified between NINJ2 gene rs11833579 (P = 0.008), protein kinase C η gene rs2230501 (P = 0.039) and IS. The AA genotype of rs11833579 increased 1.51-fold risk (95 % CI 1.04–3.46; P = 0.043) of IS, and it conferred susceptibility to IS only in a dominant model (OR 2.69; 95 % CI 1.06–6.78; P = 0.036]. Risk of IS was higher (HR 3.58; 95 % CI 1.54–8.31; P = 0.003) especially when the carriers of rs11833579 AA genotype were smokers. The present study suggests A allele of rs11833579 may play a role in mediating susceptibility to IS and it may increase the risk of IS together with smoking.
Genetic variation in cyclin dependent kinase inhibitor 2A/2B(CDKN2A/B) is associated with the risk of coronary artery disease in Chinese type 2 diabetes

Abstract:Objective To investigate the association of eyclin dependent kinase inhibitor 2A/2B (CDKN2A/B) polymorphisms with the risk of coronary artery disease in type 2 diabetics in China. Methods Of total 309 subjects with type 2 diabetes,220 individuals with coronary artery disease as cases (CAD) and 89 controls (Non-CAD)were studied. From the Hapmap CHB database, we selected 7 haplotype-tagging single nucleotide polymorphisms(SNPs) in CDKN2A/B genes: CDKN2A rs2811708 (G T), rs3088440(AG)and rs3731239(CT);CDKN2B rs3217986(AC),rs1063192(CT), rs2285327 (AG)and rs3217992(AG). All the subjects for the 7 SNPs were genotyped by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP)assay or by direct DNA-sequencing. Results In Chinese type 2 diabetic patients, allelic frequency of minor allele T at rs2811708 was higher in CAD group(26.4%)than in non-CAD group(17.4%)(OR=1.698, 95%CI 1.092～2.640, P=0.019). Genotypic frequency at rs2811708 was associated with the risk of CAD in type 2 diabetics(P=0.028). The carriers of genotype G/T had the increased risk of CAD compared to those of genotype G/G(OR= 2.074, 95%CI 1.194～3.601,P=0.010), and the association was consistent after adjustment for the other known risk factors of cardiovascular disease (adjusted OR′=2.503,95% CI 1.298～4.825, P′= 0.006). Conclusions Genetic variation in cyclin dependent kinase inhibitor 2A/2B(CDKN2A/B)genes is associated with cardiovascular risk in Chinese type 2 diabetics.
