Abstract:Objective:To explore the effect of propofol on the tPA release of hippocampal neurons in developing mice ,so as to investigate whether exogenous tPA could reverse the propofol‐induced neurotoxicity on developing neurons .Methods : The hippocampal neurons of developing mice cultured in vitro were divided into 4 groups(n= 20 each):control group(group C) , propofol group(group Pro) ,group tPA and propofol plus tPA group(group Pro+ tPA) .There was no specific treatment in group C .In group Pro ,propofol was added to the culture media with the final concentration of 5 ,10 ,30 μmol/L ,respectively , and the cells were then incubated for 1 h ,2 h ,3 h ,6 h ,respectively .In group tPA ,tPA was added to the culture media with the final concentration of 1 μmol/L ,and the cells were then incubated for 6 h .In group Pro+ tPA ,both propofol and tPA were added to the culture with the final concentration of 10μmol/L and 1μmol/L ,respectively ,and the cells were then incubated for 6 h .The neuron apoptosis rate and the expression level of actived‐caspase‐3 protein were detected in each group .Results:Compared with that in group C ,the apoptosis rate and the expression level of actived‐caspase‐3 protein in group Pro significantly increased (P< 0 .05) .The tPA level in group Pro was significantly higher than that in group C (P< 0 .05) . Compared with that in group Pro ,the apoptosis rate and the expression level of actived‐caspase‐3 protein in group Pro+ tPA significantly decreased (P< 0 .05) .Conclusions :Propofol induces neurotoxicity on hippocampal neurons of developing mice cultured in vitro by inhibiting neuronal tPA release .And exogenous tPA could partially reverse propofol‐induced neurotoxicity .

Abstract:Background: Tissue plasminogen activator (tPA) regulates fibrinolysis and is routinely used as ischemic stroke pharmacotherapy. We hypothesized that brain microvascular tPA expression and barrier properties of endothelial cells are substantially related. Methods: Human brain microvascular endothelial cells were incubated with two agents known to modify cAMP pathways: forskolin and rolipram. We analyzed development of endothelial barrier properties, i.e., trans-endothelial electrical resistance (TEER), permeability of endothelial cell monolayer, expression of influx transporter glut-1 and endothelial tight junction molecules occludin and claudin-5, tPA antigen release, and levels of endothelial tPA mRNA. Results: Forskolin plus rolipram-treated endothelial cells showed increased TEER compared to controls (174 +/- 20% of control at day six, p<0.01), while permeability to albumin and 70kDa dextran was reduced (21 +/- 6.8% of control and 3.8 +/- 0.3% of control, respectively, p<0.001). In addition, occludin and claudin-5 protein were up-regulated, occludin mRNA was increased to 206 +/- 60% of control (p<0.05), glut-1 mRNA was increased to 196 +/- 8% of control (p<0.05), levels of tPA protein were reduced to 35 +/- 7.0% of control (p<0.001) after six days, and tPA mRNA was reduced to 32 +/- 7.7% of control (p<0.01). TPA and occludin mRNA levels were inversely associated (r=-0.68, p<0.05). Conclusions: In this in vitro model, barrier properties were strongly linked (by inverse association) with tPA expression of brain microvascular endothelial cells. Copyright (C) 2011 S. Abstract:Aim: Inflammation cytokine tumor necrosis factor-alpha (TNF-alpha) induces apoptosis in neuronal cells. We hypothesized that propofol may attenuate TNF-alpha-induced apoptosis in mouse hippocampal HT22 cells and aimed to explore the underlying mechanisms. Methods: Mouse hippocampal HT22 cells were pretreated with propofol, and then stimulated with TNF alpha. Cell viability was measured by cell counting kit 8 (CCK8). Cell apoptosis was examined by flow cytometry analysis. The effect of propofol on TNF-alpha-modulated nitric oxide production was measured by a nitrate reductase assay kit, intracellular calcium release and mitochondrial membrane potential (MMP) depolarization were measured by flow cytometry analysis, and the expression of inducible nitric oxide synthase (iNOS), C/EBP homologous protein (CHOP), B-cell lymphoma 2 (Bcl2) family and caspases were detected by Western blot. Results: Compared with control, TNF-a concentration-and time-dependently increased HT22 cell apoptosis, which was attenuated by 25 mu mol/l propofol. TNF-alpha (40 ng/ml, 24 h) induced the overexpression of iNOS and the release of nitric oxide, caused the accumulation of intracellular Ca2+ and endoplasmic reticulum (ER) stress, and therefore leading to mitochondrial dysfunction. Importantly, these effects were alleviated by 25 mu mol/l propofol. Conclusions: We demonstrated that propofol could attenuate TNF-alpha-induced HT22 apoptosis. More importantly, we indicated that the underlying mechanism may involve iNOS/NO, Ca2+ and mitochondrial dysfunction. (C) 2017 Published by Elsevier Masson SAS.
Effect of propofol on temporal profile of apoptosis induced by focal cerebral ischemia-reperfusion in rats

Abstract:Objective To investigate the effect of propofol on apoptosis at different time points after focal cerebral ischemia-reperfusion (I/R) in a rat model of reversible middle cerebral artery occlusion (MCAO). Methods Sixty male SD rats weighing 240-260g were randomly divided into 2 groups ( n = 30 each): propofol group received intraperitoneal (i. p.) propofol 80 mg ·kg before MCAO and control group received equal volume of normal saline instead of propofol. The animals were anesthetized with 10% chloral hydrate 350mg·kg-1 i.p. . The right external, internal and common carotid artery were exposed. A 4-0 nylon thread with rounded tip was inserted via external carotid artery into internal carotid artery and threaded cranially until resistance was felt. MCAO was maintained for 90 min before reperfusion. The animals were killed and brains removed for detection of apoptotic neurons using TUNEL staining combined with electronic microscopic examination at 3 h, 6 h, 24 h, 3 days, 5 days and 7 days after reperfusion was started ( n = 5 at each time point in both groups) . Results In control group the ratio of apoptotic neurons peaked at 24 h of reperfusion and then gradually decreased; while the ratio of TUNEL positive neurons kept on increasing indicating a shift from apoptosis to necrosis after 24 h reperfusion. The ratio of TUNEL positive neurons and apoptosis were significantly lower in propofol group than in control between 6 h to 3 days after reperfusion was started. Conclusion Propofol pretreatment attenuates apoptosis induced by focal cerebral I/R and the maximum effect is reached and maintained between 6 h to 3 days of reperfusion.
Mechanisms of Propofol Attenuation of Ketamine-Induced Neonatal Brain Injury

Abstract:OBJECTIVE: We studied the mechanisms of protective effects of propofol on ketamine-induced damage to neonatal cognitive function.MATERIALS AND METHODS: We utilized a rat model of ketamine anaesthesia. Eighty neonatal rats (7 days after birth) were divided into four groups: normal saline group, ketamine group, and low-and high-dose propofol combined with ketamine groups. Six hours after anaesthesia, we obtained hippocampal tissue, and quantified apoptotic index and total protein concentration, and assessed global proteomics changes induced by two tested drugs. The latter changes were documented by two-dimensional electrophoresis and matrix-assisted laser desorption/ionization time of flight mass spectrometry. To evaluate cognitive functions, water maze test was applied after animals grew for 21 days. We further repeated proteomics studies at 21 days post-anaesthesia.RESULTS: Ketamine markedly up-regulated apoptotic index and decreased total protein concentration. Propofol dose-dependently reverted these adverse changes. Six hours post-anaesthesia, combined propofol and ketamine administration up-regulated the following proteins in the hippocampus: PD1A3, NDUFB10, HSPA8, ATP5JD, and PSMA1. Furthermore, the following proteins were down-regulated: PPIA, PKM2, GFAP, NSE, PPIA, PKM2, and GFAP. After 21 days, animals treated with ketamine showed marked disturbances in cognitive function as demonstrated by increased time of the water maze test, whereas propofol diminished these changes. In addition, expression of proteins largely normalized in propofol-treated animals, with only two up-regulated proteins (FUBP3 and PRDX5) and three down-regulated proteins (GAPDH, AKR1A1, and VCP).CONCLUSIONS: Adverse effects of ketamine on cognitive function are reverted by propofol, also through beneficial effects on protein expression in the hippocampus.

Abstract:The pathologic outcome of focal cerebral ischemia-reperfusion injury was displayed by co-existence of apoptosis and necrosis. A short duration of ischemia induced apoptosis. The therapeutic window for propofol initiated before the onset of ischemia and lasted until early stage of reperfusion. The neuroprotection of propofol might be attributed to the inhibition of apoptosis.
The Mechanism of TNF- α-Mediated Accumulation of Phosphorylated Tau Protein and Its Modulation by Propofol in Primary Mouse Hippocampal Neurons: Role of Mitophagy, NLRP3, and p62/Keap1/Nrf2 Pathway

Abstract:Background: Neuroinflammation-induced phosphorylated Tau (p-Tau) deposition in central nervous system contributes to neurodegenerative disorders. Propofol possesses neuroprotective properties. We investigated its impacts on tumor necrosis factor-α (TNF-α)-mediated p-Tau deposition in neurons. Methods: Mouse hippocampal neurons were exposed to propofol followed by TNF-α. Cell viability, p-Tau, mitophagy, reactive oxygen species (ROS), NOD-like receptor protein 3 (NLRP3), antioxidant enzymes, and p62/Keap1/Nrf2 pathway were investigated. Results: TNF-α promoted p-Tau accumulation in a concentration- and time-dependent manner. TNF-α (20 ng/mL, 4 h) inhibited mitophagy while increased ROS accumulation and NLRP3 activation. It also induced glycogen synthase kinase-3β (GSK3β) while inhibited protein phosphatase 2A (PP2A) phosphorylation. All these effects were attenuated by 25 μM propofol. In addition, TNF-α-induced p-Tau accumulation was attenuated by ROS scavenger, NLRP3 inhibitor, GSK3β inhibitor, or PP2A activator. Besides, compared with control neurons, 100 μM propofol decreased p-Tau accumulation. It also decreased ROS and NLRP3 activation, modulated GSK3β/PP2A phosphorylation, leaving mitophagy unchanged. Further, 100 μM propofol induced p62 expression, reduced Keap1 expression, triggered the nuclear translocation of Nrf2, and upregulated superoxide dismutase (SOD) and heme oxygenase-1 (HO-1) expression, which was abolished by p62 knockdown, Keap1 overexpression, or Nrf2 inhibitor. Consistently, the inhibitory effect of 100 μM propofol on ROS and p-Tau accumulation was mitigated by p62 knockdown, Keap1 overexpression, or Nrf2 inhibitor. Conclusions: In hippocampal neurons, TNF-α inhibited mitophagy, caused oxidative stress and NLRP3 activation, leading to GSK3β/PP2A-dependent Tau phosphorylation. Propofol may reduce p-Tau accumulation by reversing mitophagy and oxidative stress-related events. Besides, propofol may reduce p-Tau accumulation by modulating SOD and HO-1 expression through p62/Keap1/Nrf2 pathway.

Abstract:Primary objective: To determine whether propofol has a neuroprotective effect on hypoxic brain injury. Research design: A hippocampal slice, in artificial cerebrospinal fluid (ASCF) with glucose and oxygen deprivation (OGD), was used as an in vitro model for brain hypoxia. Methods and procedures: The orthodromic population spike (OPS) and hypoxic injury potentia1 (HIP) were recorded in the CA1 region when Schaffer collateral was stimulated in the CA3 region of the hippocampal slices during hypoxia. The concentrations of amino acid neurotransmitters in perfusion solution of hippocampal slices were directly measured using high performance liquid chromatography (HPLC). Morphological changes of neurons, astrocytes and mitochondria in CA1 region were observed using histology and electron microscopy. Neuronal apoptosis was evaluated with TUNEL assay. Main outcome and results: Propofol treatment delayed the elimination of OPS and improved the recovery of OPS; decreased frequency of HIP, postponed the onset of HIP and increased the duration of HIP. Propofol treatment also decreased the release of amino acid neurotransmitters such as aspartate, glutamate and glycine induced by hypoxia, but elevated the release of gamma-aminobutyric acid (GABA). Morphological studies showed that propofol treatment attenuated oedema of pyramid neurons in the CA1 region and reduced apoptosis. Conclusions: Propofol has a neuro-protective effect on hippocampal neuron injury induced by hypoxia.
Tissue Plasminogen Activator Causes Brain Microvascular Endothelial Cell Injury After Oxygen Glucose Deprivation by Inhibiting Sonic Hedgehog Signaling

Abstract:The thrombolytic activity of tissue plasminogen activator (tPA) has undisputed benefits. However, the documented neurotoxicity of tPA raises important issues. Currently, common treatments for stroke might not be optimum if exogenous tPA can pass through the blood–brain barrier and enter the brain, thus adding to the deleterious effects of tPA within the cerebral parenchyma. Here, we determined whether tPA could damage brain microvascular endothelial cells (BMECs) during cerebral ischemia. We showed that treatment of BMECs with tPA decreased trans-endothelial electrical resistance and cell proliferation, and blocked the cell cycle at the G0–G1 phase. In addition, the Sonic hedgehog (Shh) signaling pathway was involved in tPA-induced BMECs dysfunction. However, tPA-enhanced oxygen glucose deprivation-induced BMECs dysfunction was eliminated by Shh administration and the effects could be reversed by Shh inhibitors. Taken together, these results demonstrate that tPA administration might result in damage to the endothelial barrier owing to blocked Shh signaling pathway.

Abstract:Tissue-type plasminogen activator (tPA) a serine protease is constituted of five functional domains through which it interacts with different substrates, binding proteins, and receptors. In the last years, great interest has been given to the clinical relevance of targeting tPA in different diseases of the central nervous system, in particular stroke. Among its reported functions in the central nervous system, tPA displays both neurotrophic and neurotoxic effects. How can the protease mediate such opposite functions remain unclear but several hypotheses have been proposed. These include an influence of the degree of maturity and/or the type of neurons, of the level of tPA, of its origin (endogenous or exogenous) or of its form (single chain tPA versus two chain tPA). In this review, we will provide a synthetic snapshot of our current knowledge regarding the natural history of tPA and discuss how it sustains its pleiotropic functions with focus on excitotoxic/ischemic neuronal death and neuronal survival.

Effects of Different Concentrations of Propofol on Anoxia / Reoxygenation Injury to Primary Cultured Rat Hippocampal Neurons

Abstract:Objective To investigate the effects of different concentrations of propofol on anoxia / reoxygenation injury to primary cultured rat hippocampal neurons. Methods Newborn ( 24 h) Wistar rats were decapitated and their brains were removed under sterile condition. Hippocampus tissue was dissected and cut into small pieces ( 1 mm3 ) . Cell suspension was obtained by digestion with 0.25% trypsin, filtration and centrifugation. The cells were cultured for 10 days. Different concentrations of propofol was added to the primary cultured neurons. The final concentration of propofol was 1, 10, 50, 100 or 200 μmol·L-1(group 1-5) . In control group only the vehicle 2‰ DMSO was added. The neurons were then exposed to 3 h anoxia followed by 24 h reoxygenation. Neuron viability and lactate dehydrogenase (LDH) efflux were determined. Malondealdehyde (MDA) concentration and superoxide dismutase ( SOD) activity were measured. Results The viability of the neurons was significantly higher, while the LDH efflux rate was significantly lower in the 5 propofol groups than in the control group, but there was no significant difference among the 5 propofol groups. The MDA concentration in propofol group 1 and 2 (1, 10 μmol·L-1) was significantly lower than that in control group. But with increasing propofol concentration (5, 100, 200 μmol·L-1 ) , MDA concentration increased and was not significantly different from that in control group. The SOD activity was significantly higher in propofol group 1(1 μmol· L-1 ) than in control group. Further increase in propofol concentration the SOD activity gradually decreased and was even lower in propofol group 4 and 5 (100, 200 μmol·L-1 ) than in control group, but the difference was not statistically significant. Conclusion Propofol at the clinical relevant concentration can protect the primary cultured hippocampal neurons form anoxia / reoxygenation injury but further increase in propofol concentration does notprovide further neuroprotechon, on the contrary may lead to neurorial damage.
Effects of L-Tetrahydropalmatine on Electrophysiological Changs Induced by Ischemia in Hippocampal Slices

Abstract:OBJECTIVE To investigate the effect of L-tetrahydropalmatine on synaptic function after ischemia and reperfusion in hippocampal slices.METHODS Rat hippocampal slices placed in two compartment of a dual chamber were immersed in the superfusing artificial cerebral fluid(ACSF)with 95% O2+5%CO2.The population spike(PS) were recorded in CAl pyramidal cells by stimulating Schffer collateral pathways.The appearance time of attenuation and disappearance time of PS,the appearance of hypoxic injury potential and the recovery rate of PS after reoxygenation were recorded.RESULTS After the treatment with L-tetrahydropalmatine at the doses of 0.028,0.14,0.28 mmol·L-1,the appearance of attenuation and disappearance of population spike(PS) were significantly postponed comparing with the ischemia group.The appearance of hypoxic injury potential(HIP) was delayed significantly and the appearance probability of HIP was lower in L-tetrahydropalmatine group and the recovery amplitude of PS after reoxygenation was significantly higher than that of oxygen-glucose deprivation(OGD) group.CONCLUSION L-THP increased the ability against brain hypoxic injury and relieved the reversible and irreversible hypoxic injury in hippocampal slices.
HIF-α/PKM2 and PI3K-AKT Pathways Involved in the Protection by Dexmedetomidine Against Isoflurane or Bupivacaine-Induced Apoptosis in Hippocampal Neuronal HT22 Cells.

Abstract:The present study investigated the mechanism underlying the protective effect of dexmedetomidine (Dex) on hippocampal neuronal HT22 cell apoptosis induced by the anesthetics isoflurane and bupivacaine. The cellular morphology was observed using a phase contrast microscope. The effects of anesthetics on cell proliferation were assayed using a Cell Counting Kit-8 (CCK-8). The levels of apoptosis were examined by flow cytometry utilizing Annexin V-fluorescein isothiocyanate/propidium iodide double staining, and the protein expression levels of cleaved caspase-3, phosphorylated phosphoinositide 3-kinase (p-PI3K), p-protein kinase B (p-AKT), hypoxia inducible factor (HIF-), pyruvate kinase M2 (PKM2), B-cell lymphoma (Bcl-2)-associated X protein (Bax), Bcl-2 and cytochrome c were detected by western blot analysis. In vitro treatment with anesthetics was identified to decrease cell proliferation (P<0.01), the effect of which was then markedly inhibited by treatment with Dex (P<0.01) or a PI3K/AKT agonist. Exposure to anesthetics induced apoptosis in HT22 cells (75.4%), which was significantly attenuated by co-treatment with Dex (26.2%) or the PI3K/AKT agonist (28.1%). Analysis of the protein expression levels revealed that exposure to anesthetics resulted in the activation of cleaved caspase-3, Bax, cytochrome c, HIF- and PKM2 and decreased the expression levels of Bcl-2, p-PI3K and p-AKT. However, these changes were inhibited by treatment with Dex or the PI3K/AKT agonist. Dex protected hippocampal neuronal HT22 cells from anesthetic-induced apoptosis through the promotion of the PI3K/AKT pathway and inhibition of the HIF-/PKM2 axis.

Abstract:Recombinant tissue-type plasminogen activator (r-tPA/Actilyse) stands as the prevailing pharmacological solution for treating ischemic stroke patients, of whom because their endogenous circulating tPA alone is not sufficient to rescue reperfusion and to promote favorable outcome. Beyond the tPA contributed by circulating endothelial cells and hepatocytes, neurons also express tPA, sparking debates regarding its impact on neuronal fate ranging from pro-survival to neurotoxic properties. In order to investigate the role of neuronal tPA during brain injuries, we developed models leading to its conditional deletion in neurons, employing AAV9-pPlat-GFP and AAV9-pPlat-Cre-GFP along with tPA floxed mice. These models were subjected to N-methyl-D-aspartate (NMDA)-induced excitotoxicity or thromboembolic ischemic stroke in mice. Initially, we established that our AAV9 constructs selectively transduce neurons, bypassing other brain cell types. Subsequently, we demonstrated that tPA-expressing neurons exhibit greater resistance against NMDA-induced excitotoxicity compared to tPA negative neurons. The targeted removal of tPA in neurons heightened the susceptibility of these neurons to cell death and prevented a paracrine neurotoxic effect on tPA non-expressing neurons. Under ischemic conditions, the self-neuroprotective influence of tPA encompassed both excitatory (GFP + /Tbr1 + ) and inhibitory (GFP + /GABA + ) neurons. Our data indicate that endogenous neuronal tPA is a protective or deleterious factor against neuronal death in an excitotoxic/ischemic context, depending on whether it acts as an autocrine or a paracrine mediator.

Abstract:Propofol is one of the most used intravenous anesthetic agents, which is widely used in clinical anesthesia induction and maintenance of pediatric patients. Exposure of the developing brain to propofol has been reported to lead to adverse brain changes, which in turn can induce persistent behavioral abnormalities in adulthood. However, the mechanisms by which propofol exposure in the developing brain induces cognitive impairment remain unclear. Here we report that repeated propofol exposure during the second postnatal week impairs spatial learning and memory in young mice. The reduced excitatory synaptic function and synaptogenesis in hippocampal CA1 neurons underlie this cognitive impairment. Propofol exposure specifically activates Toll-like receptor 4 (TLR4)-myeloid differentiation primary response protein 88 (MyD88)-NF-κB signaling pathway. TLR4 deficiency recues propofol exposure-induced synaptic function and cognitive deficits in young mice. Thus, we provide evidence that the activation of the TLR4-mediated pathway by propofol exposure may serve as a crucial trigger for the cognitive impairment in young adulthood caused by repeated exposure to propofol in the developing brain.

Abstract:Background: To investigate the protective effects of propofol in brain ischemia/reperfusion injury and the role of heme oxygenase-1 (HO-1) in this process.Methods: Sprague-Dawley rats were randomly divided into 9 groups: sham-operated group; middle cerebral artery occlusion (MCAO) group; propofol 10, 20, and 50 mg/kg/h group (propofol 10, 20, 50 mg/kg/h infused at the onset of reperfusion for 30 min), ZnPPIX (a HO-1 activity inhibitor) group (ZnPPIX 20 mol/kg administered 24 h before MCAO), and the other 3 groups received ZnPPIX followed by 10, 20, and 50 mg/kg/h propofol respectively. At 24 hours of reperfusion, neurological examinations were performed, and after neurological test, brain infarct size and volume, neuronal apoptosis, the level of HO-1 protein expression and HO-1 activity were observed in each group.Results: Propofol significantly improved neurological deficits, reduced infarct volume, and attenuated neuron apoptosis compared with MCAO group alone. Increased HO-1 protein expression was observed in the ischemic penumbra and core of propofol group after transient MCAO. The selective HO-1 activity inhibitor, ZnPPIX, partially eliminated the neuroprotective effects of propofol.Conclusions: The neuroprotective effects of propofol postconditioning in brain ischemia/reperfusion injury may be partially through the induction of the HO-1 expression.
The protective effects of propofol against CoCl 2 -induced HT22 cell hypoxia injury via PP2A/CAMKIIα/nNOS pathway

Abstract:Background Perioperative cerebral ischemia/hypoxia could induce hippocampal injury and has been reported to induce cognitive impairment. In this study, we used cobalt chloride (CoCl 2 ) to build a hypoxia model in mouse hippocampal cell lines. Propofol, a widely used intravenous anesthetic agent, has been demonstrated to have neuroprotective effect. Here, we explored whether and how propofol attenuated CoCl 2 -induced mouse hippocampal HT22 cell injury. Methods Mouse hippocampal HT22 cells were pretreated with propofol, and then stimulated with CoCl 2 . Cell viability was measured by cell counting kit 8 (CCK8). The effect of propofol on CoCl 2 -modulated expressions of B-cell lymphoma 2 (Bcl-2), BAX, cleaved caspase 3, phosphatase A2 (PP2A), and the phosphorylation of Ca 2+ /Calmodulin (CaM)-dependent protein kinase II (pCAMKIIα), neuron nitric oxide synthase at Ser 1412 (pnNOS-Ser 1412 ), neuron nitric oxide synthase at Ser 847 (pnNOS-Ser 847 ) were detected by Western blot analysis. Results Compared with control, CoCl 2 treatment could significantly decrease cell viability, which could be reversed by propofol. Further, we found CoCl 2 treatment could up-regulate the expression of PP2A, BAX, cleaved caspase three and cause the phosphorylation of nNOS-Ser 1412 , but it down-regulated the expression of Bcl-2 and the phosphorylation of CAMKIIα and nNOS-Ser 847 . More importantly, these CoCl 2 -mediated effects were attentuated by propofol. In addition, we demonstrated that propofol could exert similar effect to that of the PP2A inhibitor (okadaic acid). Further, the PP2A activator (FTY720) and the CAMKIIα inhibitor (KN93) could reverse the neuroprotective effect of propofol. Conclusion Our data indicated that propofol could attenuate CoCl 2 -induced HT22 cells hypoxia injury via PP2A/CAMKIIα/nNOS pathway.
Effect of Intracarotid or Intravenous Injection of Propofol on the Neuronal Apoptosis of Hippocampus in Rats Undergoing Cardiopulmonary Bypass

Abstract:Aim To compare the different effect of intracarotid and intravenous injection of propofol on the neuronal apoptosis of hippocampus in rats undergoing cardiopulmonary bypass.Methods Fifty rats were randomly assigned to five groups. Shame group (Group Non-CPB,n=10); group cardiopulmonary bypass(group CPB,n=10);group intravenous injection of propofol 2 mg·kg -1·h -1 (group P_ 2V,n=10);group intravenous injection of propofol 10mg.kg -1·h -1 (group P_ 10V,n=10);group intracarotid injection of propofol 2 mg·kg -1·h -1 (group P_ 2a,n=10).Mean arterial pressure(MAP)and heart rate(HR) were monitored. Changes of the neuronal apoptosis of hippocampus were observed with TUNEL staining, Bax and bcl-2 mRNA and protein expressions were measured by RT-PCR and Western blot. In addition,neuronal ultrastructures were observed. Results Neuronal apoptosis and changes of neuronal ultrastructures of hippocampus were attenuated in group P_ 2a and P_ 10V,but the apoptosis of neurons was not attenuated in group P_ 2v. Bcl-2 and Bax mRNA expression increased undergoing CPB. Bax mRNA expression decreased after CPB in group P_ 2a and P_ 10V. Bcl-2 mRNA expression increased in group P_ 2a and P_ 10V. But Bcl-2 and Bax mRNA expression in group P_ 2V was not significantly different compared with that of group CPB. Bcl-2 and Bax protein expression measured by Western blot showed the same trend with mRNA expressions. Compared with baseline values, MAP and HR in group P_ 2V and P_ 2a remained unchanged. But the MAP and HR in group P_ 10V changed compared with group CPB and baseline value.Conclusions intracarotid injection of propofol attenuated the injury of brain at a fraction of the intravenous dose and with less adverse systemic and cerebrovascular side effects.
FUNDC1-dependent mitophagy induced by tPA protects neurons against cerebral ischemia-reperfusion injury

Abstract:<p>Autophagy of mitochondria, termed mitophagy, plays an important role in cerebral ischemia-reperfusion (IR) injury, but the mechanism is not yet clear. Tissue-type plasminogen activator (tPA) is the most important thrombolytic drug in the clinical treatment of ischemic stroke and has neuroprotective effects. Here, we explored the effects of tPA on neuronal apoptosis and mitophagy following IR. We found that knocking out the tPA gene significantly aggravated brain injury and increased neuronal apoptosis and mitochondrial damage. Exposure of neurons to tPA reduced injury severity and protected mitochondria. Further studies demonstrated that this protective effect of tPA was achieved via regulation of FUNDC1-mediated mitophagy. Furthermore, we found that tPA enhanced the expression level of FUNDC1 by activating the phosphorylation of AMPK. In summary, our results confirm that tPA exerts neuroprotective effects by increasing the phosphorylation of AMPK and the expression of FUNDC1, thereby inhibiting apoptosis and improving mitochondrial function.</p>

Propofol ameliorates ischemic brain injury by blocking TLR4 pathway in mice

Abstract:Abstract Ischemic brain injury is one of the most serious perioperative complications. However, effective preventative methods have not yet been established. This study aimed to investigate whether propofol has neuroprotective effects against ischemic brain injury, with a specific focus on Toll-like receptor 4 (TLR4). Focal brain ischemia was induced via a combination of left common carotid artery occlusion and distal left middle cerebral artery coagulation in mice. Either propofol (10 mg/kg) or vehicle was intravenously injected 10 min prior to the induction of brain ischemia in wild-type and TLR4 knockout mice. Infarct volume, pro-inflammatory cytokine expression, inflammatory cell infiltration, and neurobehavioral function were assessed. Propofol administration significantly reduced infarct volume in wild-type mice (26.9 ± 2.7 vs 15.7 ± 2.0 mm 3 at day 7), but not in TLR4 knockout mice. Compared with the control mice, the propofol-treated wild-type mice exhibited lower levels of IL-6 (0.57 ± 0.23 vs 1.00 ± 0.39 at 24 h), and smaller numbers of TLR4-expressing microglia in the penumbra (11.7 ± 3.1 vs 25.1 ± 4.7 cells/0.1 mm 2 ). In conclusion, propofol administration prior to ischemic brain insult attenuated brain injury by blocking the TLR4-dependent pathway and suppressing pro-inflammatory cytokine production.
