Abstract:Hair cosmetic agents are preparations intended for placing in contact with the hair and scalp, with the purpose of cleansing, promoting attractiveness, altering appearance, and/or protecting them in order to maintain them in good condition. Current shampoo formulations and styling products are tailored to the variations associated with age, gender, hair quality, hair care habit, and specific problems relating to the superficial condition of the scalp. The great amount of variables to be accounted for, some of them contradictory and hard to bring into accord, makes adequate product formulation a challenge and requires continuous research. Recent hair cosmetic developments include topical hair growth stimulants, photoprotectors, and anti-aging compounds. The advances in molecular technology have opened the avenue to the study of the molecular basis of hair growth and its deviations, as well as of the aging process. The discovery of potential pharmacological targets and the development of selective and effective delivery systems following topical application indicate further strategies for maintenance of healthy hair and scalp in the young and old.

Abstract:As early as can be traced, written documents testify endeavors shown by humanity to please by means of the hair. Hair care, color and style play an important role in people's physical appearance and self-perception. Dermatologists should be knowledgeable about the procedures people follow to look their best and should have the competence to provide patients with information on the benefits and hazards of hair cosmetics and pharmaceuticals. Shampooing is the most common form of cosmetic hair treatment. The diversity of qualities expected from a shampoo by today's consumer surpass the primary function of cleansing. Current shampoo formulations are adapted to the variations associated with hair quality, hair care habit and specific problems related to the superficial condition of the scalp. Hand in hand, test methods are developed to evaluate the efficacy of hair care products so that consumers are offered products that perform as claimed. Through the development of cosmetics with pharmaceutically active compounds, products are evolving that are becoming more similar to topical therapeutic agents (cosmeceuticals). The efficacy of cosmeceuticals that claim to act as hair growth stimulants should be measured by the standards set by the drugs minoxidil and, more recently, oral finasteride. Finally, health hazards associated with the use of hair care products, especially rinse-off products, have been overemphasized by the media and need careful correction by opinion leaders.

Abstract:Human hair is transformed into functional nano-dimensional material using a benign choline chloride–urea ionic liquid composite as a deep eutectic melt. The hair and isolated central fibres after treatment with the ionic liquid composite have the ability to immobilise microalgal cells for wastewater treatment and the cuticle cells are effective as templates in coating a sparingly soluble drug molecule which is slowly released at physiological pH. Thus, instead of human hair being a waste, it can be converted to end products with potentially high-end value, with reduced negative impact on the environmental.

Chemical bonds and hair behaviour-A review

Abstract:When undertaking any review of the structure of the hair and its mechanical properties it becomes apparent that the overall behaviour of keratin fibres is commonly attributed to the presence of hydrogen, disulfide and ionic bonds. The action of physico-chemical agents used during various cosmetic treatments is viewed as the result of an interaction with these bonds. Thus, the breaking of bonds by chemical agents, or via mechanical or thermal stresses, affects the relative balance of disulfide and hydrogen bonds and the contribution of hydrophobic interactions, which are all important factors that may alter hair behaviour. Generally, these chemical bonds are considered as responding homogeneously to the environmental and cosmetic factors. This unitary image is challenged, however, by evaluating the results of chemical, nanomechanical, tensile and thermal measurements, which suggest that disulfide bonds may be grouped into several types, according to their location within the fibre and the way they respond to various agents. A compensatory effect of newly formed hydrogen bonds for broken disulfide bonds may also be seen, and additionally involves different types of hydrogen bonds. As a result, the picture of chemical bonding in hair appears to be far from a homogeneous one. In addition, it is apparent that further investigation is required for clarifying the action of ionic bonds and hydrophobic interactions within the hair fibre. The present review aims, thus, at offering a deeper background for understanding how the hair behaves under various conditions.

Abstract:Keratin constitutes the major component of the feather, hair, hooves, horns, and wool represents a group of biological material having high cysteine content (7-13%) as compared to other structural proteins. Keratin -based biomaterials have been investigated extensively over the past few decades due to their intrinsic biological properties and excellent biocompatibility. Unlike other natural polymers such as starch, collagen, chitosan, the complex three-dimensional structure of keratin requires the use of harsh chemical conditions for their dissolution and extraction. The most commonly used methods for keratin extraction are oxidation, reduction, steam explosion, microbial method, microwave irradiation and use of ionic liquids. Keratin -based materials have been used extensively for various biomedical applications such as drug delivery, wound healing, tissue engineering. This review covers the structure, properties, history of keratin research, methods of extraction and some recent advancements related to the use of keratin derived biomaterials in the form of a 3-D scaffold, films, fibers, and hydrogels.

Abstract:Human hair fibre is subjected to various structural modifications due to the application of chemicals such as dyes, shampoos and bleaches and/or physical procedures such as heating, and often more than one procedure is performed on the same hair. The present work aims to analyze the changes incurred in hair samples of two ethnic groups, namely Caucasian and Afro, before and after different treatments such as thermal, bleaching and straightening. In addition to observing the damage caused by each treatment separately, the study of samples that received all three treatments was carried out. For molecular structural characterization, the Raman vibrational spectroscopic technique was used and scanning electron microscopy (SEM) was used for morphological analysis of the hair fibres. This investigation has shown, through vibrational spectroscopy, that several important bonds have been modified, such as the S-S, C-S, C-C and S-O bonds as well as the secondary structures of proteins that have indergone changes in their conformation as a result of the treatment. Hair from the two ethnic groups showed small differences in relation to each applied treatment. Excessive heat generated a higher rate of Raman spectral band intensity changes when compared to the other treatments and it was observed that the action of several treatments on the same hair fibres resulted in even more pronounced structural changes. Finally, scanning electron microscopy showed that each treatment caused a different morphological deformation pattern on the capillary surface of the human hair.

Abstract:Hair is a defining feature of mammals and has critical functions, including protection, production of sebum, apocrine sweat and pheromones, social and sexual interactions, thermoregulation, and provision of stem cells for skin homeostasis, regeneration, and repair. The hair follicle (HF) is considered a "mini-organ," consisting of intricate and well-organized structures which originate from HF stem and progenitor cells. Dermal papilla cells are the main components of the mesenchymal compartments in the hair bulb and are instrumental in generating signals to regulate the behavior of neighboring epithelial cells during the hair cycle. Mesenchymal-epithelial interactions within the dermal papilla niche drive HF embryonic development as well as the postnatal hair growth and regeneration cycle. This review summarizes the current understanding of HF development, repair, and regeneration, with special focus on cell signaling pathways governing these processes. In particular, we discuss emerging paradigms of molecular signaling governing the dermal papilla-epithelial cellular interactions during hair growth and maintenance and the recent progress made towards tissue engineering of human hair follicles.

Abstract:The hair is a filamentous biomaterial consisting of the cuticle , the cortex and the medulla , all held together by the cell membrane complex. The cortex mostly consists of helical keratin proteins that spiral together to form coiled-coil dimers, intermediate filaments, micro-fibrils and macro-fibrils. We used X-ray diffraction to study hair structure on the molecular level, at length scales between ∼3–90 Å, in hopes of developing a diagnostic method for diseases affecting hair structure allowing for fast and noninvasive screening. However, such an approach can only be successful if common hair treatments do not affect molecular hair structure. We found that a single use of shampoo and conditioner has no effect on packing of keratin molecules, structure of the intermediate filaments or internal lipid composition of the membrane complex. Permanent waving treatments are known to break and reform disulfide linkages in the hair. Single application of a perming product was found to deeply penetrate the hair and reduce the number of keratin coiled-coils and change the structure of the intermediate filaments. Signals related to the coiled-coil structure of the α -keratin molecules at 5 and 9.5 Å were found to be decreased while a signal associated with the organization of the intermediate filaments at 47 Å was significantly elevated in permed hair. Both these observations are related to breaking of the bonds between two coiled-coil keratin dimers.

Abstract:We present a coarse-grained molecular model of the surface of human hair, which consists of a supported lipid monolayer, in the MARTINI framework. Using coarse-grained molecular dynamics (MD) simulations, we identify a lipid grafting distance that yields a monolayer thickness consistent with both atomistic MD simulations and experimental measurements of the hair surface. Coarse-grained models for fully-functionalised, partially damaged, and fully damaged hair surfaces are created by randomly replacing neutral thioesters with anionic sulfonate groups. This mimics the progressive removal of fatty acids from the hair surface by bleaching and leads to chemically heterogeneous surfaces. Using molecular dynamics (MD) simulations, we study the island structures formed by the lipid monolayers at different degrees of damage in vacuum and in the presence of polar (water) and non-polar (n-hexadecane) solvents. We also use MD simulations to compare the wetting behaviour of water and n-hexadecane droplets on the model surfaces through contact angle measurements, which are compared to experiments using virgin and bleached hair. The model surfaces capture the experimentally-observed transition of the hair surface from hydrophobic (and oleophilic) to hydrophilic (and oleophobic) as the level of bleaching damage increases. By selecting surfaces with specific damage ratios, we obtain contact angles from the MD simulations that are in good agreement with experiments for both solvents on virgin and bleached human hairs. To negate the possible effects of microscale curvature and roughness of real hairs on wetting, we also conduct additional experiments using biomimetic surfaces that are co-functionalised with fatty acids and sulfonate groups. In both the MD simulations and experiments, the cosine of the water contact angle increases linearly with the sulfonate group surface coverage with a similar slope. We expect that the proposed systems will be useful for future molecular dynamics simulations of the adsorption and tribological behaviour of hair, as well as other chemically heterogeneous surfaces.
A consumer cross-over trial suggests that there are significant seasonal changes of the tensile properties (wet) of human hair

Abstract:Objectives: Through the cooperation with an industrial partner, we gained a set of data for the tensile properties (wet) of human hair. The hair samples originated from a cross-over study with two groups of individuals, using for a topical application sequentially two products (A and B). Each phase of the study lasted 6 months. The phases of the study by chance covered first largely the winter and then the summer period. Initially, tensile variables were chosen, which not only reflect the mechanical properties of hair (modulus, break strain, and break stress) but which are also considered to have a good connection to practice-relevant hair properties. The initial analysis of the data showed that changes were observed for the variables due to the treatment phases. However, these were either small or difficult to interpret. Methods: Against this background and using two-factor analysis of variance, we investigated the hypothesis that the tensile properties of hair (wet) may show significant seasonal changes. For this, we chose those two independent variables, which reflect the properties of the intermediate filaments (modulus) and the matrix (break strain) in the composite structure of the hair cortex. Results: The results support the 'seasonal' hypothesis and clearly show that the variables show significant changes from Winter to Summer (modulus: 10% increase; break strain: 3% decrease). The seasonal effect was thus a major reason, why the first stage of the analysis of the data was inconclusive. Conclusions: The tensile properties of the main morphological components of the cortex show distinct seasonal changes. Towards the summer, the hair becomes stiffer and more brittle. Furthermore, the results suggest that seasonal effects may need to be taken into account when conducting studies on lengths of hair grown during different seasons.

Abstract:The Workshop on the morphological approach to hair disorders brought together a group of clinicians involved in hair biology research. Six speakers spoke on a range of topics that can be grouped broadly into a central theme. It summarizes the evolution of medical research. The section by Tosti and coworkers describes a patient with a new unique syndrome. The section by Ferrando and colleagues provides a framework in which patients with rare hair disorders can be classified. The section by Whiting tries to define the normal anatomy of the hair follicle and both horizontal and vertical sections. It is only when normal anatomy has been absolutely defined that pathological deviations can be recognized. The section by Sinclair and coworkers attempts to estimate the reliability of histological diagnosis so that its true value of pathology can be recognized. The section by Zlotogorski and coworkers shows how accurate clinical and histological definition of disease acts as the cornerstone for gene discovery techniques. Once a causative mutation is found and a gene product identified, then the biological consequences of the altered protein product can be studied and the impact of the abnormal molecular function on hair biology can be understood. It is hoped that improved understanding of hair disease will then lead to useful therapeutic interventions. The final section by Leroy and Van Neste highlights the difficulties of evaluating therapeutic interventions in hair loss disease and proposes a new technique.

Abstract:Abstract: The hair is a distinguishing feature of animals that has several functions, such as providing warmth, providing protection, facilitating sweating and pheromone evaporation, providing sensory stimulation, and, in certain situations, acting as color-based camouflage. Hair grooming and styling have progressed significantly over the years, and modern hair treatments primarily focus on enhancing hair quality. However, standard active ingredients in formulations display suboptimal performance, and some may present toxicity issues, demanding a stronger formulation design that is appropriate for performance and safety. The need for natural ingredients, such as macromolecules and biomolecules sourced from plants, animals, and marine environments, is increasing among consumers. The favorable benefits induced by including lipid-rich components in Personal Care Products (PCP) formulations are regarded as valuable elements. A lipid-based preparation is specifically employed for innovative medicine-targeted distribution of bioactive substances among all medication administration strategies. The distribution of LBDDS transdermally and topically is exciting since the lipid bilayer might enter the uppermost layer of skin with ease and release medicine in a regulated way. It offers benefits in terms of stability, bioacceptability, and biodegradability, as well as receptor-mediated fast absorption by the particular site. Various lipid-based formulations for hair care therapy have been created, showing some improvement in hair care. This review focuses on several lipid-based formulations for hair care therapy, including their thorough mechanism of action, efficacy on hair care, patents, obstacles, and future prospects, along with the challenges and future prospects in the hair care industry

Abstract:Various cosmetic alterations done to the hair for aesthetic purposes have a profound damaging impact on its morphology. This study delves into the complex dynamics of hair structure by analysing the changes in the cuticle induced by chemical degradation. Advanced microscopic techniques like Microtopography and Scanning Electron Microscopy (SEM) were employed to investigate the grade of damage on treated hair as compared with virgin hair. On treatment with bleach, it could be observed that the cuticle structure in virgin hair exhibited deteriorating effects like partial degradation whereas the impact on treated hair samples strands that had previously undergone modifications exhibited much more intensifying consequences like complete deterioration exposing holes and gaps. The findings from the current exploration shed light on the nuanced relationships within the hair structure, providing valuable insights into the consequences of chemical degradation. Thus, laying a foundation for suggestive recommendations that include the impact of environment, lifestyle and genetics in the fields of trichology and cosmetology.
Extraction of keratin from keratinous wastes: current status and future directions

Abstract:This review aims to trace the developments of keratin extraction techniques over the last 70 years and possibilities for future research. Keratin is a fibrous structural protein naturally present in the appendages of animals such as hair, wool, feathers, hooves and hides. Currently millions of tons of these appendages are discarded as waste products by the meat, poultry, textile and leather industries. These keratin-rich wastes lack environmental-friendly disposal methods, and are often dumped in landfills or incinerated. Over the last few years, several studies have developed various methods to repurpose these wastes as a potential source for obtaining keratin. Keratin has recently been demonstrated to have applications within the biomedical field (as a scaffold material in tissue engineering, for drug delivery and wound healing), cosmetic products and environmental remediation. To meet the demand for keratin in the above-mentioned applications, research on extracting keratin from different natural sources including keratinous wastes and improving the extraction efficiency thereof is imperative. Hence, with a brief introduction to the structure and occurrence of keratin, the present review extensively focuses on current methods of its extraction from hair, feathers, wool, hides and hooves. Also, the review identifies gaps in current research and outlines future directions in this field.

Abstract:Synopsis: The mammalian hair-fibre, together with the “inner root-sheath” ( i.e. the axial layers of the follicle wall), grows upward by a proximal addition of cells. Changes in the inner root-sheath are responsible for the final shape and surface sculpture of the fibre. At its distal limit the inner root-sheath disintegrates owing to the effect of a de-keratinizing chemical agent. A bent, undulated, or “crimped” fibre is due to a differentiated progress of the changes leading to keratinization of the “fibre cortex”. The cells which give rise to a “medulla” are different chemically and less compressible than cortical cells.

Abstract:In contrast to the skin, aging of the hair has seemingly only recently found the attention of dermatological meetings, mainly promoted by the cosmetic industry for marketing purposes. In fact, basic scientists interested in the biology of hair growth and pigmentation have for some time already exposed the hair follicle as a highly accessible model with unique opportunities for the study of age-related effects. As a result, the science of hair aging focuses on two main streams of interest: the esthetic problem of aging hair and its management, in terms of age-related effects on hair color, quantity, and quality; and the biological problem of aging hair, in terms of microscopic, biochemical, and molecular changes underlying the aging process. Ultimately, the aim of hair anti-aging is to delay, lessen, or reverse the effects of aging on hair. According to the complex nature of the aging process, the treatment for lifetime scalp and hair health has to be holistic to include the multitude of contributing factors in a polyhedral and patient-specific manner. It comprises both medical treatments and hair cosmetics. Accordingly, the discovery of pharmacological targets and the development of safe and effective drugs for treatment of hair loss indicate strategies of the drug industry for maintenance of hair growth and quantity, while the hair care industry has become capable of delivering active compounds directed toward meeting the consumer demand for maintenance of hair cosmesis and quality. "Where there's life, there's hope" (Ecclesiastes 9:3-5).

Abstract:Publisher The hair or wool follicle is surrounded by a basement membrane around which lies the connective-tissue sheath. In hair follicles, the basement membrane is slightly periodic acid/schiff-positive; it contains highly sulfated glycosaminoglycans. This chapter discusses the events occurring during the formation of the wool or hair fiber. The complexity of the follicle is now well established, particularly with respect to the interrelationships between dermal and epithelial components and the different cell lines during differentiation. Cytological and cytochemical studies, although still in their infancy, have already indicated that many more levels of complexity exist. Moreover, several processes for which more mechanistic explanations have been used are being shown to have a cytological basis. This trend may be expected to continue. It is becoming increasingly evident that the full significance of many studies will not be revealed unless they can be related to known and specified cell components. Cytological techniques will play an important part in this area.

Abstract:Small-size models of keratin constituent units and products of its decomposition are proposed and computed to i) reproduce the interaction patterns that hold the macromolecule structure and its behavior under different reaction media and physico-chemical conditions; ii) estimate the energy of interactions that support the tridimensional structure of the protein, and consequently, the energy requirements to break it. To create these models and evaluate their performance, quantum-chemical, COSMO, and COSMO-RS calculations were performed. They seem to be capable to support process simulations during the conceptual design and optimization of operations and processes to transform the raw keratin into products of higher added value. Besides, proper analyses using these models demonstrated that the cleavage of the disulfide bonds is the primary condition to break the macromolecule but additionally, the intramolecular H-bond network should be dissociated enabling the intramolecular interaction with the chemicals used to dissolve and regenerate the keratin.

Abstract:Hair analysis plays an important role in forensic toxicology and biomonitoring tests. However, cosmetic treatments cause changes to the hair. Thus, a better understanding of the hair's structure and the factors that influence its composition is critical. It is known that oxidative treatments modify the hair chemical, structural and mechanical properties. These treatments also cause degradation of the melanin as well as of the structures present in the hair cuticle and cortex. Considering that the literature is unanimous regarding the increase in hydrophilicity and porosity promoted in human hair by bleaching, in this work we investigated how this oxidative damage is triggered. By combining several techniques, inelastic and quasi-elastic neutron scattering, differential scanning calorimetry, thermal gravimetric analysis and X-ray diffraction, we were able to connect the chemical and structural changes to a subtle dynamic modification of the proton mobility in the hair fibers. In addition, alterations in the thermal behavior evidenced a small denaturation of <i>α</i>-keratin intermediate filaments and a slight decrease in the amount of confined water in the hair fibers. Moreover, data obtained by neutron spectroscopy indicated that bleaching attacks the thioester groups of the proteins causing larger proton mobility of the hydrogenous components (water, protein and/or lipids).
