Abstract:A small series of fluorescent lysosome-targeting probes based on the BODIPY fluorophore and containing morpholine and nitro groups were rationally designed. These probes emitted light from green to NIR wavelengths, and provided specificity for imaging the lysosomes of hypoxic cells. The electron-withdrawing nitrophenyl group at the meso position was found to lead to highly efficient nonradiative decay of the S-1 state, and hence a recovery of fluorescence when reduction of the nitro group occurred under hypoxic conditions.

Abstract:An activatable hypoxia fluorescent probe was synthesized, named Azo‐TPE, by covalently linking tetraphenylethylene groups at both ends of the azobenzene group. In hypoxic cancer cells, nonfluorescent Azo‐TPE is reduced and cleaved to fluorescent amino derivative with aggregation‐induced emission characteristic by intracellular azoreductase, which turned on the fluorescence signal, to detect the degree of hypoxia by fluorescence imaging. Cellular hypoxia is a common pathological process in various diseases. Detecting cellular hypoxia is of great scientific significance for early diagnosis of tumors. The hypoxia fluorescence probe analysis method can efficiently and conveniently evaluate the hypoxia status in tumor cells. These probes are covalently linked by hypoxic recognition groups and organic fluorescent molecules. Currently, the fluorescent molecules used in these probes often exhibit the aggregation‐caused quenching effect, which is not conducive to fluorescence imaging in water. Herein, an activatable hypoxia fluorescence probe was constructed by covalently linking aggregation‐induced emission luminogens to the hypoxic recognition group azobenzene. It does not emit fluorescence in solution and in solid state under light excitation due to the presence of photosensitive azo bonds. It can be cleaved by intracellular azoreductase into fluorescent amino derivatives with aggregation‐induced emission characteristic. As the concentration of oxygen in cells decreases, its fluorescence intensity increases, making it suitable for fluorescence imaging to detect hypoxic environment in live cancer cells. This work broadens the molecular design approach for activatable hypoxia fluorescent probes.

Abstract:Hypoxia is a pathogenic characteristic of solid tumors owing to absent or abnormal vasculature in the tumor microenvironment and essential in tumor progression, angiogenesis, metastasis, invasion and resistance to immune system and therapy. In hypoxic environments, CYP450 enzymes are more efficient than in normoxia. Herein, based on the reductive capacity of CYP450 enzymes/NADPH system, we managed to cage aminoluciferin developing a reaction-based bioluminescent probe as well as an imaging method for the hypoxia detection. Exhibiting enhanced about 3-fold total flux in big (1.2 cm-diameter) tumors, Hypoxia BioLuminescent probe (HBL) can afford potential utility for in cellulo and in vivo hypoxia imaging in tumor model mice.

Abstract:Fluorescence imaging with tumor-specific fluorescent probe has emerged as a tool to aid surgeons in the identification and removal of tumor tissue. We report here a new lysosome-targeting fluorescent probe (NBOH) with BODIPY fluorephore to distinguish tumor tissue out of normal tissue based on different pH environment. The probe exhibited remarkable pH-dependent fluorescence behavior in a wide pH range from 3.0 to 11.0, especially a sensitive pH-dependent fluorescence change at pH range between 3.5 and 5.5, corresponding well to the acidic microenvironment of tumor cells, in aqueous solution. The response time of NBOH was extremely short and the photostability was proved to be good. Toxicity test and fluorescence cell imaging together with a sub-cellular localization study were carried out revealing its low biotoxicity and good cell membrane permeability. And NBOH was successfully applied to the imaging of tumor tissue in tumor-bearing mice suggesting potential application to surgery as a tumor-specific probe.

Abstract:The synthesis and characterization of a novel two-photon-absorbing fluorene derivative, LT1, selective for the lysosomes of HCT 116 cancer cells, is reported. Linear and nonlinear photophysical and photochemical properties of the probe were investigated to evaluate the potential of the probe for two-photon fluorescence microscopy (2PFM) lysosomal imaging. The cytotoxicity of the probe was investigated to evaluate the potential of using this probe for live two-photon fluorescence biological imaging applications. Colocalization studies of the probe with commercial Lysotracker Red in HCT 116 cells demonstrated the specific localization of the probe in the lysosomes with an extremely high colocalization coefficient (0.96). A figure of merit was introduced to allow comparison between probes. LT1 has a number of properties that far exceed those of commercial lysotracker probes, including higher two-photon absorption cross sections, good fluorescence quantum yield, and, importantly, high photostability, all resulting in a superior figure of merit. 2PFM was used to demonstrate lysosomal tracking with LT1.

Abstract:Hypoxia is a trait that widely exists in 90% of solid tumors. Under hypoxic conditions, biological reductase enzymes in tumors will be overexpressed (e.g., azoreductase). This feature is often used to design fluorescence probes to recognize hypoxic cells from normoxic cells. The presently reported fluorescence probes are mostly designed by covalent bonds, which requires tedious synthesis. In this study, a fluorescence nanoprobe was prepared by encapsulating azobenzene into assemblies of polymeric amphiphiles bearing fluorophores (denoted by PAF). The nanoprobes without azobenzene are strongly emitted in aqueous solution, and the emission was effectively quenched when azobenzene was included in the nanoprobes. The nanoprobes with azobenzene showed quite different emission intensities when cultured in hypoxic and normoxic cells, leading to fluorescence quantum yields (QYs) of 18.0% vs. 1.5%, respectively. Here, we demonstrated a facile method to prepare an azoreductase-responsive fluorescent nanoprobe that can selectively light up malignant cells from benign cells.

Abstract:Hydrogen sulfide (H2S) is an endothelial gasotransmitter which has been extensively studied recently in various physiological processes. H2S can induce lysosomal membrane destabilization leading to an autophagic event of precipitation apoptosis coupled with calpain activation, thus ensuring cellular demise. In this study, we developed a lysosome-targetable fluorescent probe for the recognition of H2S with considerable fluorescence enhancement. Through introducing a lysosome-targetable group 4-(2aminoethyl)-morpholine into the H2S probe N-imide termus of 4-azide-1,8-naphthalimide, the new compound Lyso-AFP can recognize H2S in lysosomes. This probe emerges as a more biocompatible analysis tool with low poison by-product than reported H2S fluorescent probes.

Abstract:The insufficient oxygen supply, known as hypoxia, is one of the most obvious features of tumors. In tumor regions, caused by the exaggerated growth of solid tumor, the depletion of oxygen in interior cells far exceeds their blood supply. Hypoxia will lead to a series of problems in tumor microenvironment , including increased tumor metastasis, drug resistance and pm-survival changes in gene expression. Thus, the construction of hypoxia-responsive fluorescent probe is vital to tumor diagnosis and prognosis. In this review, we discussed recent progress in fluorescent probe for hypoxia-triggered living cells imaging and cancer therapy. At first, we introduced three main sensing moieties which were sensitive to oxygen or oxygen-related enzymes, such as azoreductase, nitmreductase, quinone reductase and so on. Then, the latest progress in fluorescent probes for imaging under hypoxia conditions was summarized, including imaging for hypoxia-related parameter and hypoxia-related metabolic process. Third, based on different therapeutic strategies, including gene therapy, chemotherapy, photodynamic therapy and synergistic therapy, we discussed the research progress of hypoxiaresponsive fluorescent probe for cancer therapy. In the future, we look forwarded to develop probe with excellent ability of targeting and selecting for hypoxia-activated imaging and cancer therapy, which could further facilitate cancer diagnosis and treatment in clinical area.

Abstract:A lysosome-targeting fluorescent off-on probe has been developed by one-step synthesis for detecting lysosomal nitroreductase and hypoxia. The probe is constructed by incorporating morpholine (a lysosome-targeting unit) into 4-nitro-1,8-naphthalimide (as a fluorochrome and specific substrate for nitroreductase), and the detection mechanism is based on the nitroreductase-catalyzed reduction of the probe to 4-amino-1,8-naphthalimide, accompanied by a large fluorescence enhancement at a wavelength of 543 nm. The probe shows an accurate lysosome-targeting ability with high selectivity and sensitivity to nitroreductase (detection limit: 2.2 ng mL(-1)). Notably, the probe has been used to image the change of lysosomal nitroreductase in live cells during hypoxia, revealing that the increase of nitroreductase in lysosomes may be smaller than that in the cytoplasm. In addition, the probe is expected to be useful for studying the function of nitroreductase in the acidic organelle of lysosomes.

Abstract:Fluorescent probes capable of simultaneous visualization of cell death, lysosomes, and nucleus are significant molecular tools essential for the in-depth understanding on cell death. However, such probes have not been devoted yet. In this work, by modifying two alkalescence groups onto naphthimide and tuning the DNA affinity, we obtained a fluorescent probe enabling visualization of cell death via its subcellular immigration between lysosomes and nucleus. In living cells, the probe exclusive targeted lysosomes to give intense fluorescence. During cell death, the probe was released from lysosomes and immigrated into the nucleus. Since lysosomes localized in cytoplasm which exhibited a clear boundary from the nucleus, the cell death could be conveniently monitored via the subcellular localization of the probe molecules. In this manner, the lysosomes, nucleus, and cell death could be visualized concurrently with the probe. The cell death induced by H2O2 has been imaged, and the cell apoptosis caused by rotenone was also successfully visualized by means of the newly designed fluorescent probe. The probe is potential to deep our understanding on cell death in a new aspect, and is also a desirable tool to study the interaction between cell death, lysosomes, and nucleus.
A near-infrared multifunctional fluorescent probe for hypoxia monitoring and tumor-targeted therapy

Abstract:Hypoxia is one of the key characteristics of solid tumors. The over-expression of azoreductase resulting from hypoxia can be used as a target to visualize hypoxic levels and a trigger of the drug release system in tumor treatment. In this work, we developed a near-infrared fluorescent probe YLOD, composed of a near-infrared fluorophore, an azo bond, and an analogue of the anti-tumor drug melphalan. In the presence of azoreductase, YLOD displayed a red emission at 620 nm and released the anti-tumor drug concomitantly, thus achieving the integrated effects of hypoxic imaging and tumor treatment. Furthermore, YLOD successfully inhibited the growth of solid tumors during the tumor suppression experiments in nude mice. Considering all the results, YLOD emerges as a new fluorescence tool that can quickly determine the location and the edges of a tumor, showing concrete potential in clinical cancer treatment. Abstract:Hypoxia is a characteristic feature of most solid tumors, which promotes the proliferation, metastasis, and invasion of tumors and stimulates the resistance of cancer treatments, leading to the serious consequences of tumor recurrence. The exploration of hypoxia detection technology will aid tumor diagnosis and treatment. Fluorescence imaging technology is an accurate and efficient hypoxia detection technology. It has attracted significant research interest, but designing novel fluorescence probes, especially stimuli-responsive probes with high sensitivity and low toxicity is still challenging. In this work, we report a hypoxia-responsive molecular bioprobe lighted up by peptide self-assembly, which contains aggregationinduced emission (AIE) fluorescent molecule TPE, hypoxia-responsive azo group (-N═N-), the self-assembling peptide GFFY, and targeting ligand RGD. The resulting peptide derivative TPE-GFFY-N═N-EERGD forms supramolecular nanofibers but emit weak fluorescence because the azobenzene moiety can effectively quench the fluorescence of the TPE dye. However, the fluorescence-quenched nanofibers could be lighted up dramatically when the azo group is reduced. More importantly, this "turn-on" supramolecular fluorescence bioprobe enables effective detecting tumor hypoxia due to the overexpressed azoreductase in the tumor microenvironment. This work affords a paradigm of designing environmentsensitive fluorescent molecular probes for tumor hypoxia imaging.

Abstract:The organelle-specific pH is crucial for cell homeostasis. Aberrant pH of lysosomes has been manifested in myriad diseases. To probe lysosome responses to cell stress, we herein report the detection of lysosomal pH changes with a dual colored probe (CM-ROX), featuring a coumarin domain with "always-on" blue fluorescence and a rhodamine-lactam domain activatable to lysosomal acidity to give red fluorescence. With sensitive ratiometric signals upon subtle pH changes, CM-ROX enables discernment of lysosomal pH changes in cells undergoing autophagy, cell death, and viral infection.

Abstract:A new rhodamine B-based pH fluorescent probe has been synthesized and characterized. The probe responds to acidic pH with short response time, high selectivity and sensitivity, and exhibits a more than 20-fold increase in fluorescence intensity within the pH range of 7.5–4.1 with the pKa value of 5.72, which is valuable to study acidic organelles in living cells. Also, it has been successfully applied to HeLa cells, for its low cytotoxicity, brilliant photostability, good membrane permeability and no ‘alkalizing effect’ on lysosomes. The results demonstrate that this probe is a lysosome-specific probe, which can selectively stain lysosomes and monitor lysosomal pH changes in living cells.
Fluorescence Nanoprobe Imaging Tumor By Sensing The Acidic Microenvironment

Abstract:Visualizing tumors in their early stages is crucial to increase the survival rate of cancer patients. Optical imaging, as a developing imaging modality, proves to be promising in visualizing tumors non-invasively in vivo due to its high sensitivity, no radioactive irradiation and low running cost. The development of fluorescence probes with high tumor targeting specificity will accelerate the translation of optical imaging from the bench to bedside. Compared to the small molecular imaging probes, the tumor targeting nanoprobes demonstrate the advantages, such as the tunable circulation lifetime, the up-regulated tumor accumulation via the enhanced permeability and retention (EPR) effect, and improved targeting specificity/sensitivity by labeling multiple targeting/imaging domains on a single nanoparticle. Tumor microenvironment (TME) indicators such as acidosis, hypoxia and high interstitial pressure are universal phenomena of solid tumors. Therefore, fluorescent nanoprobes that respond to TME, especially the physiologically acidic pH environment, are promising in visualizing tumor and metastases regardless of the tumor types or even developmental stages. The signal of these pH "responsive" nanoprobes stays "silent" at the normal tissues, but it enhances significantly in the tumor acidic microenvironment. Therefore the signal ratio between the tumor and surrounding normal tissues can increase remarkably, which benefits the visualization of early stage/small volume tumors with high sensitivity. Here, we have summarized the recent advances in the development of fluorescence nanoprobes for tumor visualization by sensing the tumor acidic microenvironment.

Abstract:Non-invasive dynamic tracking of lysosomes and their interactions with other organelles is important for the study of lysosomal function and related diseases. However, many fluorescent dyes developed so far to target lysosomes cannot be used to monitor these processes due to the high concentrations required for imaging, long cell penetration times, and non-ideal photostability. In this regard, we synthesized three lysosomal targeting probes with large Stokes shifts, good stability, and high brightness. The Q-P-ARh dye, developed by us for the first time, can stain lysosomes at ultra-low concentrations (1.0 nM) without affecting the physiological functions of the lysosomes. More importantly, its excellent anti-interference ability and ultrafast lysosomal staining ability (within 1.0 min) clearly monitored the entire dynamic process of lipophagy. Ultimately, this method can greatly contribute to the study of autophagy pathways. This novel fluorescence platform shows great promise for the development of biological probes for application in pathological environments.
A Novel Off-on Fluorescent Probe for Imaging of Hypoxia in Tumor Cell

Abstract:A novel off-on fluorescent probe, developed for imaging the hypoxic status of tumor cells by means of detecting nitroreductase (NTR), consisted of a naphthalimide fluorophore and 4-Nitrobenzyl chloroformate as exogenous hypoxia markers. The detection mechanism is based on the fact that nicotinamide adenine dinucleotide (NADH) is utilized by nitroreductase as an electron source to reduce nitroaromatics to corresponding amino derivatives. The fluorescence of probe displays distinct change from very weak fluorescence to strong green fluorescence at 550 nm after reaction with NTR. The fluorescence intensity increase of probe shows a good linearity when the concentration of NTR is in the range of 0.1-0.3 mu g/mL. The probe shows significant specificity with NTR and good biocompatibility. The result of laser confocal fluorescence imaging confirmed that probe can successfully differentiate normoxic (non-fluorescent) from hypoxic conditions (strong green fluorescence) in U87 cell lines. All the results demonstrate that our probe is a promising optical imaging candidate for tumor hypoxia and can be potentially used for the detection of hypoxic-ischemic diseases.
