Abstract:An enantioselective one-pot Michael/Michael/Henry/hemiacetalization reaction between alpha,beta-unsaturated aldehydes, alpha-ketoamides, and nitroalkenes under mild conditions catalyzed by a diarylprolinol silyl ether has been developed. The sequential methodology provides a direct approach to a wide range of fully substituted chiral oxabicyclo[2.2.2]octanes with seven contiguous stereocenters in moderate to excellent yields (up to 99%), high to excellent diastereoselectivities (up to >25:1 dr), and high to excellent enantioselectivities (up to 99% ee).

Abstract:A facile synthesis of γ-butenolides and maleic anhydrides via annulation of α-keto acids and triazenyl alkynes is described. In this process, α-keto acids and triazenyl alkynes could undergo a self-catalyzed annulation at room temperature to deliver γ-butenolides efficiently, while the further addition of BF3-Et2O furnished maleic anhydrides. Overall, these processes have mild reaction conditions, broad scope, and high efficiency.

Abstract:A new and efficient protocol for the synthesis of 2-hydroxybenzophenone derivatives through [5C+1C] annulation of alpha-alkenoyl-alpha-aroyl ketene dithioacetals and nitroalkanes had been developed. In the presence of 1 equivalent of K2CO3 in DMF at 70 degrees C, a range of alpha-alkenoyl-alpha-aroyl ketene dithioacetals smoothly react with nitroalkanes to provide various 2-hydroxy benzophenone derivatives in excellent yields. Abstract:Comprehensive SummaryNiH‐catalyzed multicomponent reductive hydrofunctionalization of alkenes is an attractive but poorly explored approach to rapidly increasing molecular complexity. In this process, the selective generation of the desired product can be challenging. Herein, we report that a highly chemo‐, regio‐ and enantioselective reductive hydrocarbonylation of alkenes has been achieved using a chloroformate ester as a source of CO. A wide range of structurally diverse α‐hydroxy ketones, privileged structural elements in bioactive molecules and useful building blocks, were obtained from such reactions with high enantiomeric purity.

Abstract:A general and direct organocatalytic asymmetric vinylogous Michael reaction of gamma-butenolide with alpha,beta-unsaturated ketones was investigated with a multifunctional primary amine salt as catalyst. Abstract:γ-Butenolides are useful structural motifs in many pharmaceutically relevant compounds. In particular, halogenated γ-butenolides are attractive building blocks because the halogen handles can readily be manipulated to give various functional molecules. In this study, a catalytic synthesis of halogenated γ-butenolides from cyclopropene carboxylic acids was developed using zwitterionic catalysts and <i>N</i>-haloamides as the halogen sources. The catalytic protocol could also be applied to the synthesis of halogenated pyrrolones by using cyclopropene amides as the starting materials.

Abstract:An efficient nickel(0)-catalyzed highly regio- and stereoselective hydrocarboxylation of homopropargylic alcohols with ZnEt(2) in the presence of CO(2) (1 atm, balloon) to synthesize α-alkylidene-γ-butyrolactones is described. The catalyst is highly active and can be applied for the synthesis of (optically active) mono- or bicyclic α-alkylidene-γ-butyrolactones with excellent regio- and stereoselectivity and good functional group tolerance. The potential of the reaction has been demonstrated in the first synthesis of (±)-heteroplexisolide E.

Abstract:A highly efficient asymmetric [3+2] and [4+2]-annulation of cyclic N-sulfonyl ketimines with γ- or δ-hydroxy-α,β-unsaturated ketones has been developed. This innovative reaction employs an organocatalytic approach, utilizing a hydrogen-bonding bifunctional squaramide-based catalyst. The process enables precise synthesis of chiral polyheterotricyclic oxazolidines and 1,3-oxazinane derivatives, revealing intricate structures with incorporated chiral quaternary centers. Remarkably, this method delivers high yields and exceptional enantioselectivities and diastereoselectivities, achieving up to 99% yield, >20 : 1 dr, and >99% ee.

Abstract:The base-promoted [3 + 3]/[1 + 4] tandem reaction of tosyl-protectedo-amino alpha,beta-unsaturated ketones and crotonate-derived sulfur ylide for the efficiently diastereoselective synthesis of functionalized hydrocarbazoles is reported. The reaction proceeded smoothly and gave the desired products in high yields and excellent diastereoselectivities under mild conditions. The hydrocarbazoles could be readily transformed into functionalized carbazoles.

Abstract:Synthesis of α-heterosubstituted ketones was achieved through sulfur mediated difunctionalization of internal alkynes in one-pot. The reaction design involves: phenyl substituted internal alkyne attacking the triflic anhydride activated diphenyl sulfoxide to give a sulfonium vinyl triflate intermediate, hydrolysis to give the α-sulfonium ketone, and then substitution with various nucleophiles. This method provides a unified route to access α-amino ketones, α-acyloxy ketones, α-thio ketones, α-halo ketones, α-hydroxy ketones, and related heterocyclic structures, in a rapid fashion.

Abstract:The total synthesis of (–)-herbaric acid has been achieved through the stereoselective synthesis of 3-substituted isobenzofuranones with a new organocatalytic route. When combined with a catalytic amount of benzoic acid, quinidine thiocarbamate based bifunctional catalysts have demonstrated their efficiency for the diastereoselective halolactonization reaction of acrylate-type benzoic acids bearing a chiral alkoxycarbonyl group on the carbon–carbon double bond. High diastereomeric excesses were obtained thanks to a positive match effect between the (+)-menthyl ester group and the chiral organocatalyst.
