Abstract:Three new homoisoflavanones, 1-3, together with a known one, 4, were obtained from the AcOEt extract of the tuberous root of Ophiopogon japonicus (Liliaceae). They were identified as (3R)-2,3-(1,3-benzodioxol-5-methoxy-3-(4-methoxybenzyl)-6,8-dimethyl-4H-chromen-4-one (1), (3R)-3-(1,3-benzoldioxol-5-ylmethyl)-2,3-dihydro-7-hydroxy-5-methoxy-6-methyl-4H-chromen-4-one (2), (3R)-3-(1,3-benzodioxol-5-ylmethyl)-2,3-dihydro-7-hydroxy-6-methyl-4H-chromen-4-one (3), and ophiopogonanone A(4). Their structures were determined on the basis of extensive NMR-spectroscopic and mass-spcetrometric analyses. The three enw compounds are rare homoisoflavanones which contian a MeO group at C(5). Compounds 1 and 2 showed weak cyotoxicity against the HepG2 (human hepatoma G2), KB (human oral epidermoid carcinoma), and MCF-7 (human breast adenocarcinoma) cell lines in an MTT assay. Compound 3 exhibited weak cytotoxicity against HepG2 and MCF-7 and moderate cytotoxicity against KB cell lines. Compound 4 showed moderated cytotoxicity against HepG2, KB, and MCF-7 cell lines.

Abstract:A new homoisoflavanone, (3 R)-5,7-dihydroxy-8-methyl-3-(2′-hydroxy-4′-methoxybenzyl)-chroman-4-one (1), was isolated from the 95% ethanol extract of the rhizomes of Polygonatum cyrtonema. The structure of 1 was determined by spectroscopic methods, especially 2D NMR techniques. Quantum chemical TD DFT calculations of the theoretical ECD spectrum of 1 allowed assignment of the absolute configuration as 3R.

Abstract:A phytochemical investigation of the rhizomes of Polygonatum cyrtonema, a traditional Chinese medicine and healthy food named "Huang-jing" (Polygonati Rhizoma), led to the isolation of 22 phenolic derivatives including an undescribed (1) and nine (2-10) known C-methylated homoisoflavanones, two common homoisoflavanones (11, 12), four flavanones (13-16), one chalcone (17), and five octopamine derivatives (18-22). The structure of compound 1 was established mainly by spectroscopic methods, including 1D-/2D-NMR and electronic circular dichroism (ECD). The known compounds were identified by comparing the observed and reported physicochemical properties and spectroscopic data published in the literature. Polygonatone H (8) and 5,7,2 '-trihydroxy4 '-methoxy-6,8-dimethyl-homoisoflavanone (9) exhibited significant cytotoxic effects against four human cancer cell lines (HCT-116, AGS, U-87 MG, and PC-12) with IC50 values ranging from 2.2 to 9.8 mu M. Furthermore, the chemotaxonomic significance of the isolated C-methylated homoisoflavanones was briefly discussed.

Abstract:Twenty-two compounds, including five homoisoflavanones (1–3, 7 and 8), three isoflavones (9–11), flavone glycoside (19), one triterpenoid aglycon (5), two steroidal sapogenins (4, 6), five steroidal saponins (12, 13, 20–22), three lignanoids (14, 17 and 18) and two fatty acids (15 and 16) were isolated from the rhizomes of Polygonatum odoratum. The structures were elucidated by spectroscopic methods and by comparison of the reported spectral data. Additionally, the relationships between this plant and other species from Liliaceae family have also been discussed on the basis of the given compounds.

Abstract:A new steroidal glycoside (1), (25R)-14α, 17α-hydroxyspirost-5-en-3β-yl 3-O-α-L-rhamnpyranosyl-(1 → 2)-β-D-glucopyranosyl-(1 → 3)-β-D-glucopyranoside, together with three known steroidal glycosides, (25R)-3β-hydroxyspirost-5-en-1β-yl-3-O-α-L-rhamnopyranosyl-(1 → 2)-O-β-D-xylopyranosyl-(1 → 3)-α-L-arabinopyranoside (2), Cixi-ophiopogon B (3) and Cixi-ophiopogon A (4), were obtained from the tuberous roots of Ophiopogon japonicus (Liliaceae). Compound 2 was isolated from the Ophiopogon genus for the first time. Their structures were identified on the basis of extensive mass and nuclear magnetic resonance spectroscopic analysis.

Abstract:One new flavonostilbene glycoside, polygonflavanol C ( 1 ), two new dimeric stilbene glycosides, multiflorumiside M and multiflorumiside N ( 2 − 3 ), one new diphenyl ethanol glycoside, ( R )-2,3,5,4′-tetrahydroxy-diphenylethanol 2- O - β -D-glucopyranoside ( 4 ), and one new deoxybenzoin glycoside, 2,4,3′,5′-tetrahydroxy-6-methyl-deoxybenzoin 2- O - β -D-glucopyranoside ( 5 ), together with six known ones ( 6 − 11 ), were isolated from the roots of Polygonum multiflorum . Their structures were elucidated by the comprehensive spectroscopic analyses. In addition, compounds 1 and 7 showed significantly in vitro anti-inflammatory activity.

Abstract:A new homoisoflavonoid, (R)-3-(2,4-dihydroxybenzyl)-5,7-dihydroxy-8-methoxychroman-4-one ( 1 ), has been isolated from the rhizomes of Polygonatum alte-lobatum , together with seven known compounds ( 2–8 ). The structure of the new compound 1 was determined through spectroscopic and MS analyses. Among the isolated compounds, compounds 1 , 3 , 5 , and 6 showed potent inhibition with IC 50 values of 18.04 ± 2.14, 19.87 ± 2.70, 16.88 ± 1.35, and 18.50 ± 1.52 μM, respectively, against LPS-induced NO generation.

Abstract:Two new diastereoisomeric phenolic compounds ( 1 , 2 ) have been isolated from the methanol extract of Polygonatum cyrtonema Hua leaves. Their structures were elucidated as (1S,2R) and (1S,2S)-1-(4-hydroxy-3-methoxyphenyl)-1-ethoxypropan-2-ols by spectroscopic data ( 1 H NMR, 13 C NMR, HSQC, HMBC, HR-ESI-MS, and ORD). In addition, compounds 1 and 2 displayed potential aldose reductase inhibitory activities with IC 50 values of 29.05 and 22.38 μM, respectively.

Abstract:Two new phenolic compounds, (1R,2R)-1-(4-hydroxy-3-methoxyphenyl)-1-methoxypropan-2-ol ( 1 ) and (1S,2S)-1-(4-hydroxy-3,5-dimethoxyphenyl)-1-ethoxypropan-2-ol ( 2 ), and one known analog ( 3 ) have been isolated from the methanol extract of Polygonatum cyrtonema Hua flowers and identified by 1 H NMR, 13 C NMR, HSQC, HMBC, HR-ESI-MS, and optical rotations. In addition, compounds 1–3 displayed potential aldose reductase inhibitory activity with IC 50 values of 22.16, 17.63, and 28.74 μM, respectively.

Abstract:Phytochemical study on the root tubers of Ophiopogon japonicus resulted in the isolation and identification of 13 homoisoflavonoids, including three new compounds, 8-formyl-7-hydroxy-5,4-dimethoxy-6-methylhomoisoflavone (1), 6-formylisoophiopogonone B (2), and 8-formylophiopogonanone B (4), and the ten known homoisoflavonoids 3, and 5-13. The absolute configurations of 8-formylophiopogonanone B (4) and 8-formyl-7-hydroxy-5,4-dimethoxy-6-methylhomoisoflavanone (5) were confirmed by time-dependent density-functional-theory (TD DFT) calculations of their theoretical electronic circular dichroism (ECD) spectra. The structure of the formerly reported 6-aldehydoisoophiopogonone B' was revised to 8-formylophiopogonone B (3). All compounds were evaluated for their cytotoxic activities against the human-lung-tumor A549 cell line, and compounds 3, 9, 10, and 13 exhibited promising antiproliferative activities with IC50 values of 10.01, 6.40, 0.84, and 1.66M, respectively.

Abstract:A new cyclic diarylheptanoid, 1,5-epoxy-3-hydroxy-1-(3-methoxy-4,5-dihydroxyphenyl)-7-(4-hydroxyphenyl)-heptane (1), as well as a new monoterpene, 10-O-β-d-glucopyranosyl-hydroxy cineole (2) were isolated from the rhizomes of Zingiber officinale. The structures of compounds 1 and 2 were established based on their spectral data. In addition, the antioxidant activities of these compounds were also measured.

Abstract:Two new monoterpenoid glycosides, trans-1,8-cineole-3,6-dihydroxy-3-O-β-D-glucopyranoside (1), and 5,9-dihydroxy borneol 2-O-β-D-glucopyranoside (2), together with four known monoterpenoid glycosides (3–6), were isolated from the water-soluble constituents of the fresh rhizome of Tongling White Ginger (Zingiber officinale). Their structures were decisively elucidated by spectroscopic analysis. In vitro tests for antimicrobial activity showed that compounds 1 and 3 possess significant activity against two Gram-positive organisms, Staphylococcus aureus and Staphylococcus epidermidis.

Abstract:Polygonatum odoratum is widely used as a traditional food supplement and herbal medicine with strong antioxidant activity. However, systematic investigation of its antioxidants was limited. Ethanol extract of P. odoratum was fractioned on macroporous absorptive resin (D101) column. A bioassay-guided purification of flavonoid-rich fraction (IC50 value at 74.1 ± 11.9 μg/mL for DPPH scavenging) was realised via high-speed counter-current chromatography (HSCCC) using petroleum ether-ethyl acetate-methanol-water (2:3:3:2, v/v/v/v) as the solvent system combination with Sephadex LH-20 column chromatography (CC) eluting with MeCN-MeOH (1:1, v/v). Three novel homoisoflavonoids (1-3), along with eight homoisoflavonoids (4-11), were isolated. Their structures were elucidated by interpretating various spectroscopic data. All the isolated homoisoflavonoids showed potent antioxidant activities, while compounds 1, 4, and 6 with dihydroxylated B-rings exhibited stronger antioxidant activities (IC50 values at 3.8 ± 0.5, 4.9 ± 0.3 and 3.9 ± 0.4 μg/mL) than ascorbic acid (IC50 value at 5.3 ± 0.6 μg/mL).

Abstract:Three steroidal saponins were isolated and characterised from the rhizomes of Polygonatum odoratum. The structure of the compounds were determined as 3-O-beta-D-glucopyranosyl-(1 --> 2)-[beta-D-xylopyranosyl-(1 --> 3)]-beta-D-glucopyranosyl-(1 --> 4)-galactopyranosyl-25(S)-spirost-5(6)-en-3beta-ol (1), 3-O-beta-D-glucopyranosyl-(1 --> 2)-[beta-D-xylopyranosyl-(1 --> 3)]-beta-D-glucopyranosyl-(1 --> 4)-galactopyranosyl-25(S)-spirost-5(6),14(15)-dien-3beta-ol (2) and 3-O-beta-D-glucopyranosyl-(1 --> 2)-[beta-D-xylopyranosyl-(1 --> 3)]-beta-D-glucopyranosyl-(1 --> 4)-galactopyranosyl-25(S)-spirost-5 (6)-en-3beta, 14alpha-diol (3). Compound 2 was identified as a new compound. Compounds 1-3 were found to be strong against the growth of bacteria and plant pathogenic fungi.

Abstract:Zang, Jialin ; Li, Gang ; Zhao, Chaoyang ; Lv, Zhen ; We created a fast and effective thin-layer chromatography method to detect homoisoflavones in Polygonatum odoratum (Mill.) Druce (POD), demonstrating high sensitivity, specificity, and stability. Utilizing this method, we assessed the quality of 20 commercial batches of POD. Our results indicated that homoisoflavones (III, IV, and V) were present in 8 samples, absent in 7, and detected in 5. This method facilitates the effective evaluation of POD germplasm resources, laying a strong foundation for enhancing quality standards. This advancement aids in the standardization and quality control of POD, ensuring the consistency of its active ingredients and thereby supporting its therapeutic applications.

Abstract:Two new homoisoflavonoids, named ophiopogonone E (1) and ophiopogonanone H (2), together with thirteen known ones (3–15) were isolated from the tuberous roots of Ophiopogon japonicus. Their structures were elucidated by spectroscopic and chemical analyses. Compounds 7 and 15 were isolated from the genus for the first time. In addition, compounds 2–15 were evaluated for their effects on the inhibition of NO production induced by lipopolysaccharide in the murine microglial cell line BV-2. Compounds 2, 4, 6, 7, 10, 11 showed potent inhibitory effects on NO production with IC50 values of 20.1, 17.0, 7.8, 5.1, 19.2 and 14.4μM respectively.

Abstract:Two new iridoids, 3-(5-(methoxycarbonyl)-2-oxo-2H-pyran-3-yl)butanoic acid, named verbeofflin I (1), and 7-hydroxydehydrohastatoside (2), were isolated from the aerial part of Verbena officinalis L, along with three known iridoids, verbenalin (3), 3,4-dihydroverbenalin (4), hastatoside (5) by means of various column chromatography steps. The structures of these compounds were elucidated through analysis of their spectroscopic data obtained using 1D and 2D NMR and MS techniques. Verbeofflin I (1) is the new class of secoiridoid in the family Verbenaceae.
