Abstract:In the past thirty years, DNA molecule has received considerable attention as a promising building material due to its precise base-paring recognition, the programmable sequences, the predictable secondary structures, etc. Based on DNA self-assembly technique, DNA supramolecular hydrogels can be fabricated which are crosslinked networks swollen in an aqueous phase by the supramolecular interaction and also have widely applications in the biomedical field. In this paper, a new type of DNA-based supramolecular hydrogel was prepared by simply mixing two components: Polypeptide-DNA and G-quadruplex. We have successfully synthesized single-stranded DNA (ssDNA) grafted polypeptide via the copper-catalyzed click reaction. By introducing the intermolecular G-quadruplex structure, which is rich in guanine and is capable of forming a four-stranded structure in the presence of potassium, the hydrogel can be formed by DNA hybridization in a fast and in-situ manner and also obtain thermal and ionic responsive gel-sol transition, respectively. In this experiment, we firstly used polyacrylamide gel electrophoresis (PAGE), circular dichroism spectra (CD), UV-Vis spectroscopy to demonstrate the G-quadruplex structure formation, and the assembling process was efficient which are formed the parallel structure and the melting temperature (T-m) was about 43 degrees C. Then by the rheological tests, the shear-storage modulus (G') was obviously higher (up to 2.4 kPa) than the shear-loss modulus (G '') over the entire time range, and a minimum concentration of 0.5 wt% polypeptide-DNA was required to form a gel-like state, and the mechanical strength of the DNA hydrogels increased with increasing concentration of the DNA building blocks. What's more, the hydrogels can switch between the gel and the sol state when changing the temperature from 25 to 50 degrees C many times, indicating that the DNA hydrogels respond to thermal in a reversible way. At last, by using the properties of crown ether complexing potassium ion, the DNA hydrogel can obtain the ion responsiveness properties. Due to its designable responsiveness and good biocompatibility, this supramolecular hydrogel would have great potential applications in tissue engineering and 3D bioprinting field.

Abstract:In recent years, DNA supramolecular hydrogels have attracted much attention due to their injectability, biocompatibility, responsiveness and self-healing properties. In this work, we designed a linear DNA brick containing one duplex with two cytosine (C)-rich sequence on both ends. This brick can first assemble to form duplex under pH 8 condition. After adjusting the pH to 5, the C-rich sequence tends to form intermolecular i-motif structure, which joins the linear DNA molecules together to form interlocked cyclic structures and yield the DNA hydrogel. By adjusting the length and bending curvature of the duplex part of the molecule, one can change the basic unit of the hydrogel structure to tune the properties of the DNA hydrogel.

Abstract:A pH‐responsive cascaded DNA hydrogel that can be fine‐tuned by utilizing pH‐triggered DNA secondary structures, including A‐motif, i‐motif, and T·A‐T triplex, is reported following a stepwise pH 1.1 → 5.2 → 7.2 → 10.2. This study provides design principles for pH‐responsive DNA hydrogels and advances their single pH‐responsiveness to multiple cascaded pH values. Governed by pH‐configurable A‐motif, i‐motif and T·A‐T triplex configurations, a cascaded DNA hydrogel subjecting to diverse pH values is presented. Under highly acidic conditions (pH 1.1), N1 protonation of adenine (pKa 3.5) in A‐strands generates AH+‐H+A units, resulting in a parallel A‐motif duplex crosslinked hydrogel. Under mild acid conditions (pH 5.2), the dissociation of A‐motif duplex into single A‐strands occurs due to the deprotonation of adenine, while N3 protonation of cytosine (pKa 6.5) in C‐strands creates hemi‐protonated C:C+ units, resulting in i‐motif bridged hydrogel. At neutral pH (pH 7.2), deprotonation of cytosine separates i‐motif crosslinkers. Simultaneously, the addition of auxiliary T‐strands results in the N3 protonation of thymine (pKa 10), generating T·A‐T triplex stabilized hydrogel. Under mildly alkaline conditions (pH 10.2), T·A‐T triplex separates into single T‐stands and A‐T duplex, resulting in the disassembly of DNA hydrogel. Therefore, a stepwise pH‐cascaded DNA hydrogel dictates the formation of structures following the pH steps 1.1 → 5.2 → 7.2 → 10.2 where pH‐triggered DNA secondary structures, including A‐motif, i‐motif and T·A‐T triplex, stabilize the hydrogel. The study advances DNA hydrogels from single pH‐responsiveness to multiple cascaded pH values, which opens up new possibilities for the development of smart hydrogels capable of adapting to various environmental conditions.

Abstract:A supramolecular double network hydrogel is presented by physical interpenetration of DNA and cucurbit[8]uril networks. In addition to exhibiting an increase in strength and thermal stability, the double network hydrogel possesses excellent properties such as stretchability, ductility, shear-thinning, and thixotropy. Moreover, it is enzymatically responsive to both nuclease and cellulase, as well as small molecules, showing great potential as a new soft material scaffold.

Abstract:As a novel type of bio-functional material, DNA hydrogels have attracted more and more attention due to their successful applications in 3D cell culturing and tissue engineering for the designable and programmable responsiveness. Herein, we have developed a pH-triggered DNA hydrogel based on a clamped hybridization chain reaction(C-HCR). In this system, a DNA switch was designed, which can release the initiator strand in a controllable way via the formation of the C-G·C+ triplex under the pH stimuli. While the pre-gelation solution is stable in neutral environment, the C-HCR will trigger the sol-gel transition as the pH decreased to 5.0. This strategy has endowed the DNA hydrogel with good controllability for triggering, which also shows potential in intellectual responsiveness to certain stimuli.

Abstract:DNA hydrogel has aroused widespread attention because of its unique properties. In this work, the DNA-modified magnetic nanoparticles were integrated into the mainframe of DNA hydrogel, resulting in DNA-MNP hydrogel. Under the magnetic field, this hydrogel can be remotely deformed into various shapes, driven to-jump between two planes and even climb the hill. By applying various triggers, such as temperature, enzyme, and magnetic field, DNA-MNP hydrogel can specifically undergo sol gel transition. This work not only imparts DNA hydrogel with a new fold of property but also opens a unique platform of such smart materials for its-further applications.

Abstract:A guanine‐rich oligonucleotide based on a human telomeric sequence but with the first three‐nucleotide intervening stretch replaced by a putative 15‐nucleotide hairpin‐forming sequence shows a pH‐dependent folding into different quadruplex‐duplex hybrids in a potassium containing buffer. At slightly acidic pH, the quadruplex domain adopts a chair‐type conformation. Upon increasing the pH, a transition with a midpoint close to neutral pH to a major and minor (3+1) hybrid topology with either a coaxially stacked or orthogonally oriented duplex stem‐loop occurs. NMR‐derived high‐resolution structures reveal that an adenine protonation is prerequisite for the formation of a non‐canonical base quartet, capping the outer G‐tetrad at the quadruplex‐duplex interface and stabilizing the antiparallel chair conformation in an acidic environment. Being directly associated with interactions at the quadruplex‐duplex interface, this unique pH‐dependent topological transition is fully reversible. Coupled with a conformation‐sensitive optical readout demonstrated as a proof of concept using the fluorescent dye thiazole orange, the present quadruplex‐duplex hybrid architecture represents a potentially valuable pH‐sensing system responsive in a physiological pH range of 7±1.

Abstract:Deoxyribonucleic acid (DNA) is a natural polyester polymer with programmable sequences and precise molecular recognition. Therefore, DNA has been widely investigated in functional devices and materials recently. Taking advantage of the structural diversity of DNA, novel functional devices and materials have been endowed with a variety of physical, chemical and biological responsiveness, among which, optical regulation has attracted much attention due to its non-contact and precise temporal-spatial controllability. To bring optical responsiveness to DNA material, it is necessary to introduce rational designed unnatural bases into DNA, which has been a challenging frontier. In this work, a novel thymine phosphoramidite monomer protected by 1-(4,5-dimethoxy-2-nitrobenzyl) ethoxy group (DMONB) has been designed and synthesized. The monomer was synthesized through 6 step reactions to shield the hydrogen bond site of thymine, and the product can be achieved in gram scale with high purity. Then the monomer was applied to synthesize photo-responsive DNA with random sequence through commercial solid phase synthesis procedure, which was purified and detected by RP-HPLC with good stability. The coupling efficiency of the photo-responsive monomer is comparable to the commercially available phosphoramidite monomers, detected by monitoring the 4,4'-dimethoxytriphenylmethyl (DMT) groups. The DMONB group could effectively hinder the DNA hybridization, which was demonstrated by the 10% native polyacrylamide gel electrophoresis. Under UV light for 30 s, the DMONB group could be totally removed and result in the recover of the DNA hybridization, which illustrates that this approach is of high efficiency and fast-responsive. Taking advantage of this controllable hybridization strategy, a fast light-responsive pure DNA supramolecular hydrogel has been designed and prepared, which could realize photo-chemically control the formation of pure DNA supramolecular hydrogel. This photo-induced DNA hydrogel exhibits 730 Pa storage modulus and 100 Pa loss modulus, with reversible thermal responsiveness and shear-thining property, which is similar to previously reported DNA hydrogel by our group. Our research expands the diversity of artificial DNA monomers and provides an alternative way to construct light-responsive functional DNA based materials system.

Abstract:Stimuli-responsive DNA-based hydrogels are attracting growing interest because of their smart responsiveness, excellent biocompatibility, regulated biodegradability, and programmable design properties. Integration of reconfigurable DNA architectures and switchable supramolecular moieties (as cross-linkers) in hydrogels by responding to external stimuli provides an ideal approach for the reversible tuning structural and mechanical properties of the hydrogels, which can be exploited in the development of intelligent DNA-based materials. This review highlights recent advances in the design of responsive pure DNA hydrogels, DNA-polymer hybrid hydrogels, and autonomous DNA-based hydrogels with transient behaviors. A variety of chemically and physically triggered DNA-based stimuli-responsive hydrogels and their versatile applications in biosensing, biocatalysis, cell culture and separation, drug delivery, shape memory, self-healing, and robotic actuators are summarized. Finally, we address the key challenges that the field will face in the coming years, and future prospects are identified.

Abstract:DNA hydrogels play an increasingly important role in biomedicine and bioanalysis applications. Due to their high programmability, multifunctionality and biocompatibility, they are often used as effective carriers for packing drugs, cells, or other bioactive cargoes in vitro and in vivo. However, the stability of the DNA hydrogels prevents their in-demand rapid release of cargoes to achieve a full therapeutic effect in time. For bioanalysis, the generation of signals sometimes needs the DNA hydrogel to be rapidly degraded when sensing target molecules. To meet these requirements, stimulus-responsive DNA hydrogels are designed. By responding to different stimuli, self-degradable DNA hydrogels can switch from gel to solution for quantitative bioanalysis and precision cargo delivery. This review summarizes the recently developed innovative methods for designing stimuli-responsive self-degradable DNA hydrogels and showed their applications in the bioanalysis and biomedicines fields. Challenges, as well as prospects, are also discussed.

Abstract:The base sequence of nucleic acids encodes structural and functional information into the DNA biopolymer. External stimuli such as metal ions, pH, light, or added nucleic acid fuel strands provide triggers to reversibly switch nucleic acid structures such as metal-ion-bridged duplexes, i-motifs, triplex nucleic acids, G-quadruplexes, or programmed double-stranded hybrids of oligonucleotides (DNA). The signal-triggered oligonucleotide structures have been broadly applied to develop switchable DNA nanostructures and DNA machines, and these stimuli-responsive assemblies provide functional scaffolds for the rapidly developing area of DNA nanotechnology. Stimuli-responsive hydrogels undergoing signal-triggered hydrogel-to-solution transitions or signal-controlled stiffness changes attract substantial interest as functional matrices for controlled drug delivery, materials exhibiting switchable mechanical properties, acting as valves or actuators, and "smart" materials for sensing and information processing. The integration of stimuli-responsive oligonucleotides with hydrogel-forming polymers provides versatile means to exploit the functional information encoded in the nucleic acid sequences to yield stimuli-responsive hydrogels exhibiting switchable physical, structural, and chemical properties. Stimuli-responsive DNA-based nucleic acid structures are integrated in acrylamide polymer chains and reversible, switchable hydrogel-to-solution transitions of the systems are demonstrated by applying external triggers, such as metal ions, pH-responsive strands, G-quadruplex, and appropriate counter triggers that bridge and dissociate the polymer chains. By combining stimuli-responsive nucleic acid bridges with thermosensitive poly(N-isopropylacrylamide) (pNIPAM) chains, systems undergoing reversible solution ↔ hydrogel ↔ solid transitions are demonstrated. Specifically, by bridging acrylamide polymer chains by two nucleic acid functionalities, where one type of bridging unit provides a stimuli-responsive element and the second unit acts as internal "bridging memory", shape-memory hydrogels undergoing reversible and switchable transitions between shaped hydrogels and shapeless quasi-liquid states are demonstrated. By using stimuli-responsive hydrogel cross-linking units that can assemble the bridging units by two different input signals, the orthogonally-triggered functions of the shape-memory were shown. Furthermore, a versatile approach to assemble stimuli-responsive DNA-based acrylamide hydrogel films on surfaces is presented. The method involves the activation of the hybridization chain-reaction (HCR) by a surface-confined promoter strand, in the presence of acrylamide chains modified with two DNA hairpin structures and appropriate stimuli-responsive tethers. The resulting hydrogel-modified surfaces revealed switchable stiffness properties and signal-triggered catalytic functions. By applying the method to assemble the hydrogel microparticles, substrate-loaded, stimuli-responsive microcapsules are prepared. The signal-triggered DNA-based hydrogel microcapsules are applied as drug carriers for controlled release. The different potential applications and future perspectives of stimuli responsive hydrogels are discussed. Specifically, the use of these smart materials and assemblies as carriers for controlled drug release and as shape-memory matrices for information storage and inscription and the use of surface-confined stimuli-responsive hydrogels, exhibiting switchable stiffness properties, for catalysis and controlled growth of cells are discussed.

Abstract:DNA supramolecular hydrogels are hydrogels cross-linked through DNA hybridization. They have attracted wide attention due to their great molecular permeability, biocompatibility, degradability, thixotropy, and self-healing properties, which make them very useful in cell culture, tissue engineering, and 3D printing. The designability and responsiveness of DNA endows these hydrogels with specific stimuli responsiveness, thus enhancing their utility for more applications, e.g., detection devices and shape-memory materials. In this review, we outlined and discussed the development of responsive DNA supramolecular hydrogels, and their applications have also been summarized.

Abstract:Living organisms operate far from equilibrium in order to maintain their adaptive structures and functions by continuous energy consumption. The development of synthetic out-of-equilibrium systems using light as an input fuel will allow for remote and noninvasive control over a range of important chemical/biological processes. Here, we report on the rational design of light-fueled DNA hydrogels that exhibit life-like nonequilibrium and dissipative features. Such hydrogels are formed through a two-component mixing strategy, where merocyanine photoacid bearing a robust pH-adjustment capability delicately imparts photoinactive but pH-sensitive DNA components with an adaptive response to light. The transient hydrogel spontaneously dissipates to its sol state in the dark, enabling a highly reversible sol–gel transition that is fine-tuned by irradiation conditions. We construct reprogrammable DNA hydrogel shapes/patterns by alternating the photopatterning/dark-erasing processes using a single recyclable ink. Our work provides a general waste-free approach to developing other light-fueled out-of-equilibrium hydrogels for applications in smart materials and soft robotics.

Abstract:A unique type of novel supramolecular hybrid hydrogel based on the co-assembly of the anionic polyoxometalate and the cationic ABA triblock copolymer via electrostatic interaction was reported to show jellyfish-like switchable chromic luminescence.
