P46 Mechanism of excitatory effect of hydrogen sulfide on gastric motility

Abstract:Hydrogen sulfide (H2S) is regarded as the third gas transmitter and receiving increasing interest over the past decade or so. It is an endogenous, biologically active gas of both physiological and pathophysiological importance in the cardiovascular and nervous systems, and elsewhere. In gastrointestinal tissue, the level of H2S is much higher than in other tissues. So we tried to figure out the role of H2S in gastrointestinal motility. Application of H2S donor, 100 micromolar NaHS, significantly enhanced the basal tension of gastric antrum muscle strip. And kV blocker can inhibit the H2S effect on strip basal tension. In isolated antral smooth muscle cells, H2S inhibited the peak component of IKv rather than the later sustained component. So we analyzed the effect of H2S on KCND family which contributes to the IKv peak component in heterogenous expressed 293 cells. All three membranes of KCND family were tested in 293 cells separately and only Kv4.3 current was inhibited by H2S. H2S induced the membrane potential depolarization and knock down the Kv4.3 can inhibit the potential depolarization. Kv4.3 is sulfhydrated by H2S in muscle tissue and 293 cells. Blocking the free thiol or desulfhydrated by reducing agent inhibited the sulfhydration of Kv4.3 and inhibited the effect of H2S simultaneously. These results provided the evidence for predication that H2S inhibited IKv by sulfhydrating Kv4.3.
