ApoE and ABCG2 expression

Abstract:Deposition of beta-amyloid peptides incerebral vessels occurs in the majority of AD patients and results in cerebralamyloid angiopathy (CAA). We have found that ABCG2, a blood-brain barrier (BBB) transporter protein, is strongly up-regulated in the brains of AD/CAApatients as compared to age-matched non-demented controls (ND). We havedemonstrated that ABCG2 can act as a gatekeeper at the BBB to preventblood-borne beta-amyloid from entering the brain and that ABCG2 canprotect cells from ROS-induced toxicity and inflammation. Since ApoE plays arole in Alzheimer pathogenesis, we have investigated if ApoE isoforms affect ABCG2expression. Cultured human brain endothelial cells (HBEC) were treated with 3ÂμM ApoE3, ApoE4 recombinant protein or vehicle for 4,8, and 24h. Cells were harvested for RNA and protein isolation. RT/qPCR and Western blot were performed to examine ABCG2 expression. Thelevels of ApoE protein in AD, AD/CAA, and ND patients’ brains were examined byWestern blot. Brain samples from 8-week old ApoE3 tg, ApoE4 tg, andwild-type mice (6 mice/genotype) were examined by Western blot for Abcg2expression. RT/qPCR showed that both ApoE3 and ApoE4significantly up-regulated ABCG2 expression in HBEC at 4, 8 and 24h as comparedto controls. Increased ABCG2 mRNA expression wasconfirmed by Western blots. The level of ApoE protein in AD/CAAbrains was significantly higher than ND brains. The level of ApoE in ADbrain was higher than ND brains but was not statistically significant. The level of Abcg2 was comparable in the brains of ApoE3 tg, ApoE4 tg, andwild-type mice. In vitro study shows that ApoE3 and ApoE4are capable of up-regulating ABCG2 expression in HBEC. The level of ApoEprotein was significantly higher in AD/CAA brains than that in ND brains. These suggest that ApoE3 and ApoE4 may play a role in promoting ABCG2expression. However, the study with ApoE3 and ApoE4 transgenic mice didnot show increased Abcg2 expression as compared to wild-type miceof murine apoE genotype. Further studies will be conducted to comparethe levels of Abcg2 in ApoE2, ApoE3, and ApoE4 transgenic mice at differentstages and in different brain regions/substructures.
