GLYCATION OF TAU PROTEIN PROMOTES THE FORMATION OF FILAMENTS DURING TAU PHOSPHORYLATION

Abstract:Tauopathies are neurodegeneration disorders which accumulate intracellular filamentous aggregates composed of the microtubule-associated protein tau, including Alzheimer's disease (AD), progressive supranuclear palsy (PSP), corticobasal degeneration (CBD), frontotemporal dementia with Parkinsonism linked to chromosome 17(FTDP-17) and Pick's disease(PiD). Tau protein has several post translational modifications. The dynamic change of the process of tau phosphrylation and dephosphrylation maintain the microtubule stable, the form of tau posphrylation which is called hyper- phosphorylation tau were found abundant in brain of that disease. It related to GSK-3β phosphorylation pathway. However its mechanism is still unknown. In order to investigate the relationship of post translational modification of tau and tau aggregaton, we purified tau 4R2N protein by E.coli, and then phosphorylated the protein by GSK-3β or incubate with glucose to glycate the tau protein prior to protein phosphorlation. Thioflavine T (Th T) assay was used to measure the level of the aggregation, and mass spectrum detected the site of modification. Our data show that phosphorylation by GSK-3β accelerate the aggregation of tau protein. However tau glycation induced tau phosphorylation increasing, phosphorylation at S324, S404, and S400 found in tau protein which has both glycation and phosphorylation and not found in hosphoryalted tau. Results indicate that tau protein maybe first have glycation and then aggregation.
