Rnai-Mediated Silencing of Rap1B Gene Expression in Esophageal Squamous Cell Carcinoma Inhibits Tumor Growth and Invasion

Abstract:Background: Rap1b is known to play a role in the progression of angiogenesis and migration. Our previous study found that Rap1b was over-expression in esophageal squamous cell carcinoma (ESCC). Based on our previous results, we investigated the role of Nrf2 in cell proliferation and invasion in ESCC. Methods: Human ESCC cell lines Eca-109, Ec-9706, and TE-1 were selected for this study. We first detect the expression of Rap1b by western blot and Real-time PCR. Then, after inhibition of gene expression of Rap1b by specific small interfering RNA (Rap1b-siRNA), effects of Rap1b on cell growth, apoptosis, and invasion were determined by MTT, flow cytometry, and transwell assay respectively. Results: It was found that all of these cells had high expression levels of Rap1b in mRNA and protein levels. Eca-109, with highest expression of Rap1b, was chose for following experiment. By Rap1b-siRNA, the cell proliferation and invasion were both inhibited. Further, the cell apoptosis was significantly elevated by Rap1b-siRNA. Conclusion: These results indicate that Rap1b is essential for both tumor cell growth and invasion in ESCC. Therefore, Rap1b is supposed to be a potential molecular target for treatment of ESCC.
