Abstract:The specific interaction between sugar and lectin plays a key role in various human diseases and projects remarkable significance to the development of a lectin biosensor for diagnosis and drug discovery. An inorganic nano MoS2 field-effect transistor (FET)-based D-Mannose biosensor was developed in this paper using a simple assembly process. First, gold nanoparticles were functionalized on the surface of MoS2. Then, the beta-mercaptoethylamine was self-assembled on the gold surface via Au-S bonds to derivatize amino groups. Finally, the probe molecules D-Mannose for biological protein were immobilized on the FET via the Schiff base reaction. Results from scanning electron microscope and x-ray photoelectron spectroscopy analysis confirmed the entire assembly procedure and the feasible interaction of the FET sensor with Concanavalin A (ConA). The fabricated FET biosensors can reliably detect the target ConA with a detection limit below 105 nm. The current study shows the potential of MoS2 FET biosensors for ConA specific detection and clarifies the design of biosensors for the early diagnosis of glycomics and drug screening in medicine.

Abstract:In this paper, a novel method for detecting concanavalin A (Con A) was developed based on lectin-carbohydrate biospecific interactions. Multi-wall carbon nanotube-polyaniline (MWNT-PANI) nanocomposites, synthesized by in situ polymerization, were chosen to immobilize d-glucose through the Schiff-base reaction. The immobilized D-glucose showed high binding sensitivity and excellent selectivity to its target lectin, Con A. Cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS), transmission electron microscopy (TEM) and atomic force microscopy (AFM) were applied to characterize the assembly process of the modified electrode. Due to the high affinity of Con A for D-glucose and high stability of the propounded sensing platform, the fabricated biosensor achieved ultrasensitive detection of Con A with good sensitivity, acceptable reproducibility and stability. The changes of response current were proportional to the Con A concentrations from 3.3 pM to 9.3 nM, with a detection limit of 1.0 pM. Therefore, the combination of MWNT-PANI nanocomposites and the special binding force between lectin and carbohydrate provides an efficient and promising platform for the fabrication of bioelectrochemical devices.

Abstract:A sensitive sandwich-like electrochemiluminescent (ECL) biosensor was designed for the detection of concanavalin A (ConA) using a C60–reduced graphene oxide (C60–rGO) nanocomposite as a platform and glucose oxidase (GOX) decorated hollow gold nanospheres (HGNSs) as a label. First, C60–rGO with a large surface area was prepared for combining with phenoxy-derivatized dextran, which served as the recognition element for interacting with ConA by biospecific carbohydrate–protein (lectin) interactions. Then, GOX decorated HGNSs (GOX-HGNSs) were linked to the electrode surface through the biospecific interaction between the intrinsic carbohydrate residues of GOX and ConA. These localized GOX and HGNSs amplified the ECL signal of luminol intensely, which was achieved by the efficient catalysis of the GOX towards the oxidation of glucose to in situ generate an improved amount of hydrogen peroxide (H2O2) as a coreactant, and the excellent catalysis of HGNSs towards the ECL reaction of luminol–H2O2. The prepared biosensor exhibited a sensitive response for the determination of ConA, ranging from 0.10 to 100 ng mL−1 with a detection limit down to 30 pg mL−1 (signal to noise = 3). With excellent stability, sensitivity, selectivity and simplicity, the prepared biosensor showed great prospects in lectin sensing or carbohydrate sensing.

Abstract:An electrochemiluminescence (ECL) analytical platform was initially proposed based on the electrochemiluminescence resonance energy transfer (ECL-RET) mechanism for ultrasensitive detection of Concanavalin A (Con A). In this protocol, the glucose functionalized carboxylic g-C3N4 nanosheets (g-C3N4-COOH@Glu) and MnO2 nanoparticles covered carboxylic multi-wall carbon nanotubes (BSA@MnO2-MWCNTs-COOH@Glu) were synthesized and acted as ECL-RET electron donor and acceptor, respectively. Herein, glucose was served as the recognition element for binding Con A and MWCNTs was utilized as the carrier materials for loading MnO2. When the quenching probe BSA@MnO2-MWCNTs-COOH@Glu was incubated onto the modified electrodes via the specific carbohydrate-Con A interaction, the ECL signals of g-C3N4-COOH@Glu which used S2O82- as its coreactant have drastically declined. Under optimum conditions, this biosensor performed a sensitive detection of the Con A ranging from 1 x 10(-5) to 1 x 10(4) ng/mL with a detection limit of 2.2 fg/mL (S/N = 3). Moreover, favorable analytical outcomes for detecion Con A in actual serum samples were obtained, exhibiting huge applications in clinical diagnosis of this assay.

Abstract:In this work, we proposed a novel biosensor to homogeneously detect concanavalin A (ConA) using pyrene-conjugated maltose assembled graphene based on fluorescence resonance energy transfer (FRET). Maltose-grafted-aminopyrene (Mal-Apy) was synthesized and characterized by mass spectra, UV–vis and fluorescence spectra. The Mal-Apy was further employed for fluorescence switch and ConA recognition. When Mal-Apy was self-assembled on the surface of graphene by means of π-stacking interaction, its fluorescence was adequately quenched because the graphene acted as a “nanoquencher” of the pyrene rings due to FRET. As a result, in the presence of ConA, competitive binding of ConA with glucose destroyed the π-stacking interaction between the pyrene and graphene, thereby causing the fluorescence recovery. This method was demonstrated the selective sensing of ConA, and the linear range is 2.0×10−2 to 1.0μM with the linear equation y=1.029x+0.284 (R=0.996). The limit of detection for ConA was low to 0.8nM, and the detection of ConA could be performed in 5min, indicating that this method could be used for fast, sensitive, and selective sensing of ConA. Such data suggests that the graphene FRET platform is a great potential application for protein–carbohydrate studies, and would be widely applied in drug screening, bimolecular recognition and disease diagnosis.

Abstract:Carbohydrate–protein interactions mediate the important physiological and pathophysiological processes in living organism. Their study has attracted great attention due to its importance in understanding these biological processes and in fabricating biosensors for diagnostics and drug development. Here, by using concanavalin A (ConA) as a model protein, a novel surface plasmon resonance (SPR) sensor was developed for sensitive detection ConA. In this sensing platform, dextran (Dex) capped gold nanoparticles (Dex-Au NPs) were initially synthesized in one-pot and utilized as amplification reagent. After deposition of graphene oxide (GO) on the SPR gold film, phenoxy-derivatized dextran (DexP) was assembled onto the GO-modified gold chip surface through π–π interaction. The resultant GO/DexP sensing interface could specifically capture ConA which could further react with Dex-Au NPs through the specific interaction between ConA and Dex, forming a sandwich configuration. The morphologies and the electrochemistry of the formed sensing surface were investigated by using scanning electron microscopy and electrochemical techniques including electrochemical impedance spectroscopy and cyclic voltammogram. Owing to the high surface area of GO and the excellent amplification of Dex-Au NPs, the developed sandwich SPR sensor successfully fulfilled the sensitive detection of ConA in the range of 1.0–20.0μgmL−1 with a detection limit of 0.39μgmL−1. Compared to the direct assay format, the prepared sandwich SPR sensor led to an improvement of 28.7-fold in the sensitivity. The results demonstrated that the proposed method might provide a new direction in designing high-performance SPR biosensors for sensitive and selective detection of a wide spectrum of biomolecules.

Abstract:A solid-state electrochemiluminescence (ECL) biosensor for the detection of concanavalin A (Con A) was constructed using graphene quantum dots (GQDs) as luminophore, which were loaded on polyamidoamine (PAMAM) functionalized CeO2@Ag NPs by the interaction between the NH2 of PAMAM and the COOH of GQDs. The electrodepositing gold layers were used as matrix to immobilize glucose oxidase (GOx). GOx served as recognition elements to further capture Con A by the specific interaction between carbohydrate and Con A. With the increase of the concentration of Con A, the amount of GOx-CeO2@Ag-GQDs incubated onto the electrode was increased and a higher ECL signal was obtained. The constructed biosensor exhibited a sensitive response towards Con A and a low detection limit of 0.16 pg/mL was obtained. Such a construction strategy provides a new approach for applying GQDs to establish a solid-state ECL biosensor.

Abstract:A facile electrochemiluminescent (ECL) strategy for in situ label-free monitoring of carbohydrate expression on living cells was designed by integrating the specific recognition of lectin to carbohydrate with a carbohydrate-functionalized CdS nanocomposite. The mercaptopropionic acid-capped CdS quantum dots were firstly immobilized on carbon nanotubes modified electrode and then functionalized with carbohydrate using mannan as a model on the surface. The carbohydrate-functionalized CdS nanocomposite showed high ECL sensitivity and good stability, and could be used for competitive recognition to concanavalin A with the target cells in solution, which led to a change of ECL intensity due to the resistance of concanavalin A. The change depended on both the cell number and the expression level of cell surface carbohydrate. A wide linear response to cells ranging from 2×103 to 1×107cellsmL−1 with a detection limit of 1.2×103cellsmL−1 was obtained. The proposed biosensor could be used to in situ evaluate cell surface glycan, and the average number of mannose moieties on single living BGC cell was detected to be 8.7×107. This sensitive strategy was further used for facile monitoring of dynamic carbohydrate expression on living cells in response to drugs. The proposed method could be further expanded to high-throughput detection with the addition of more specific glycan–lectin pairs to the repertoire.

Abstract:A fullerene-promoted organic-inorganic hybrid electrochemiluminescence (ECL) biosensor was proposed for sensitive detecting concanavalin A (ConA). Here, platinum-modified inorganic TiO 2 (Pt-TiO 2 ) promotes charge transfer to enhance ECL intensity of organic luminescent probe—ammonolysis product of PTCDA (PTC). Fullerene interacts with K 2 S 2 O 8 co-reagent to produce more SO 4 •− free radicals, and its large specific-surface-area and electrophilic nature are beneficial to loading more luminescent PTC. The glucose oxidase (GOx) is chosen for specific recognition of ConA through a biospecific interaction. The constructed ECL biosensor shows a wide detection range of 1 × 10 −4 –1 × 10 3 ng mL −1 with a low detection limit (LOD) of 1.8 × 10 −5 ng mL −1 , as well as high stability, reproducibility, and detection capability of actual serum. This work provides a promising organic-inorganic hybrid biosensor platform for early biotherapy and clinical diagnosis.

Ternary Supramolecular Quantum-Dot Network Flocculation for Selective Lectin Detection

Abstract:We present a versatile, tuneable, and selective nanoparticle-based lectin biosensor, based on flocculation of ternary supramolecular nanoparticle networks (NPN), formed through the sequential binding of three building blocks. The three building blocks are β-cyclodextrin-capped CdTe quantum dots, tetraethylene glycol-tethered mannose-adamantane cross-linkers (ADTEGMan), and the tetravalent lectin Concanavalin A (ConA). The working principle of this selective sensor lies in the dual orthogonal molecular interactions of the linker, uniting adamantane-β-cyclodextrin and mannose-lectin interaction motifs, respectively. Only when the lectin is present, sequential binding takes place, leading to in situ self-organization of the sensor through the formation of ternary supramolecular networks. Monitoring the loss of fluorescence signal of the quantum dots in solution, caused by controlled network formation and consecutive flocculation and sedimentation, leads to selective, qualitative, and quantitative lectin detection. Fluorescent sedimented networks can be observed by the naked eye or under UV illumination for a lectin concentration of up to 10−8 M. Quantitative detection is possible at 100 min with a lower detection limit of approximately 5 × 10−8 M.
A Biorecognition System for Concanavalin a Using a Glassy Carbon Electrode Modified with Silver Nanoparticles, Dextran and Glucose Oxidase

Abstract:Concanavalin A (Con A) was chosen as a model lectin protein for an electrochemical study on the interaction between lectins and carbohydrates. A composite prepared from reduced graphene oxide and silver nanoparticles (rGO-AgNPs) acted as a redox probe, and phenoxy-derivatized dextran (DexP) and glucose oxidase (GOx) acted as recognition elements. The rGO-AgNPs, DexP and GOx were deposited on the surface of a glassy carbon electrode. The immobilized DexP and GOx showed high affinity for the target lectin Con A. The surface-modified biosensor was characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, UV–vis absorption spectra, cyclic voltammetry and electrochemical impedance spectroscopy. Due to the high affinity of Con A for both DexP and GOx, the biosensor displays a detection limit for Con A that is as low as 0.67 ng mL−1 at a working voltage of 48 mV. The response is linear in the 2.0–322 ng mL-1 concentration range, thus offering a sensitive means for the determination of Con A.

Abstract:A sandwich-configuration electrochemiluminescence (ECL) biosensor was constructed for detecting concanavalin A (ConA) based on peroxydisulfate/oxygen (S2O82−/O2) system. In this work, the gold nanoflower modified Zn-doped SnO2 was used as a substrate to adsorb recognition element horseradish peroxidase (HRP) for binding ConA. Then, Au nanoparticles-thiosemicarbazide functionalized PtNi nanocubes (AuNPs-TSC-PtNi NCs), as a novel ECL signal tracer, were incubated onto the electrode through a specific carbohydrate-ConA interaction, thus achieving a sandwiched structure. The integration of amplifying effect of both TSC and PtNi NCs on the ECL of S2O82−/O2 system endowed the biosensor a high sensitivity. The linear range for ConA detection was from 0.0010ng/mL to 10ng/mL with a detection limit of 0.0002ng/mL (S/N=3).
A Simple Electrochemical Lectin-Probe For In Situ Homogeneous Cytosensing And Facile Evaluation Of Cell Surface Glycan

Abstract:This work constructed a novel electrochemical team-probe, ferrocene-concanavalin A (Fc-ConA), for in situ monitoring of cell surface glycan by incorporating the specific recognition ability of lectin to glycan and favorable electrochemical property of ferrocenyl group. The covalent conjugation of ConA with ferrocenyl group was achieved by a carbodiimide coupling reaction and proved with UV-vis absorption spectroscopy and infrared spectroscopy. Cyclic voltammetric behavior of Fc-ConA at glassy carbon electrode demonstrated a reversible diffusion-controlled process A facile homogeneous cytosensing strategy was then developed using Fc-ConA probe for detection of K562 cells. The suspending cells specifically captured Fc-ConA via membrane mannosyl groups and decreased the concentration of free Fc-ConA, producing a response correlative with cell number and the content of cell surface glycan A wide linear response to cells ranging from 1 x 10(4) to 1 x 10(7) cells mL(-1) with a calculated detection limit of 3000 cells mL(-1) was obtained. The lectin-probe could be conveniently used to in situ evaluate cell surface glycan The average number of mannose moieties on single living K562 cell was detected to be 3.0 x 10(10), while this value increased by 81% on drug-treated cells. These results agreed with those from flow cyto-metric detection. Abstract:A localised surface plasmon resonance (LSPR) optical sensor with double-layer silver nanoparticle (Ag NP) arrays has been developed for the sensitive detection of concanavalin A (ConA). Based on the principle of carbohydrate-protein interactions, double-layer dextran capped Ag NP arrays were initially constructed. The increase of adsorption apparent mass and LSPR coupling between the two Ag NP arrays can effectively enhance the LSPR spectral shift response. Compared with the single-layer Ag NP arrays configuration, the double-layer Ag NP arrays configuration improves the limitation of detection of ConA concentration from 3.42 to similar to 1.14 mu g/ml.

Abstract:A CdS/AuNPs/NiO Z-scheme heterojunction was prepared on a fluorine-doped tin oxide (FTO) electrode by hydrothermal synthesis of NiO on FTO, electrodeposition of AuNPs on NiO/FTO electrode and then cast-coating of CdS quantum dots. The CdS/AuNPs/NiO/FTO electrode gave a notably increased photocurrent versus NiO/FTO, CdS/FTO, AuNPs/NiO/FTO, CdS/AuNPs/FTO and CdS/NiO/FTO electrodes. The CdS/AuNPs/NiO/FTO electrode was further cast-coated with chitosan to immobilize d-mannose by Schiff base reaction, and concanavalin A (ConA) and then horseradish peroxidase (HRP) were captured on the electrode surface by lectin-sugar binding. 4-Chloro-1-naphthol (4-CN) was oxidized to form an insoluble precipitate catalyzed by HRP in the presence of H2O2, and the presence of precipitate on the photoelectrode inhibited the photocurrent in the presence of holes scavenger ascorbic acid. The relevant electrodes were characterized by electrochemistry, quartz crystal microbalance (QCM), UV-vis spectrophotometry, scanning electron microscopy/energy dispersive spectroscopy, and transmission electron microscopy. The QCM revealed that the collection efficiency (η) of the 4-CN-electrooxidation precipitate on the electrode can be as high as 91.8%. Under the optimal conditions, the decline of photocurrent responded linearly to the common logarithm of ConA concentration from 50 pM to 500 nM, with a limit of detection of 17 pM (S/N = 3). Satisfactory results were obtained in the detection of real soybean samples.

Abstract:In this paper, we report a novel lectin-based electrochemical biosensor constructed by functionalized multiwalled carbon nanotubes (CNTs) for a competitive assay of glycan expression on living cells. The biosensor was fabricated by adsorbing poly(diallyldimethylammonium chloride) (PDDA)-functionalized CNTs (PDCNTs) onto a glassy carbon working electrode (GCE), followed by adsorbing glutathione (GSH)-protected gold nanoparticles (AuNPs). The resulting GCE/PDCNT/Au-GSH offers an effective platform for concanavalin A (Con A, a lectin) immobilization with high stability and bioactivity. A thiol-derivatized carbohydrate (thiomannosyl dimer) was synthesized to construct {CNT/thionine/Au-S-mannose} biocomposites, which exploit CNTs as carriers to load enormous amounts of thionine for generating an electrochemical signal and AuNPs for anchoring the thiomannosyl dimer. These two applications of the functionalized CNTs as a biosensing platform and biocomposites play important roles in signal enhancement for glycan detection. Using a competitive assay, the electrochemical biosensor shows good analytical performance with high sensitivity, selectivity and a rapid response for the analysis of mannose on human lung cancer cells as a model glycan. This proposed strategy possesses promising application for the assay of other glycans on living cancer cells with the selection of more lectins with the biosensor development.

Abstract:A novel signal-on electrochemiluminescence (ECL) biosensor for detecting concanavalin A (Con A) was fabricated with phenoxy dextran-graphite-like carbon nitride (DexP-g-C3N4) as signal probe. In this construction strategy, the nanocomposites of three-dimensional graphene and gold nanoparticles (3D-GR-AuNPs) were used as matrix for high loading of glucose oxidase (GOx), which served as recognition element for bounding Con A. Con A further interacted with DexP-g-C3N4 through a specific carbohydrate-Con A interaction to achieve a sandwiched scheme. With the increase of Con A incubated onto the electrode, the ECL signal resulted from DexP-g-C3N4 would enhance, thus achieving a signal-on ECL biosensor for Con A detection. Due to the integration of the virtues of 3D-GR-AuNPs and the excellent ECL performance of DexP-g-C3N4, the prepared biosensor exhibits a wide linear response range from 0.05 ng/mL to 100 ng/mL and a low detection limit of 17 pg/mL (S/N=3).

Abstract:In this paper, an improved method for detection of Concanavalin A (Con A) with label-free optical biosensors is reported. 1-Dodecanethiol (DDT) was self-assembled onto gold nanoparticles which were deposited on glass slides, and then glycolipid molecules were inserted into dodecanethiol by physical interactions only. The recognition between Con A and carbohydrate was observed by UV–vis spectrophotometry. The absorption spectrum shifted when Con A was bound to the sugar residues of glycolipids immobilized onto nanogold slides, while almost no spectrum change was observed when another nonspecific protein molecule met the nanogold slides. The self-assembled bilayer on nanogold substrates had very high sensitivity for Con A, the minimum detection concentration of Con A can be down to 0.1nM. In addition to the ultra sensitivity for investigating carbohydrate–lectin interaction, the self-assembled bilayer structure, is expected to replace many receptors which require time-consuming organic syntheses for the fixation to the transducer. The simplicity and sensitivity of this biosensor architecture once again show the prospect of nanogold application in biosensor.

Abstract:In this work, a novel “on-off” electrochemiluminescence (ECL) biosensor was successfully constructed for ultrasensitive detection of concanavalin A (ConA). The “signal-on” state with strong initial ECL signal was obtained by Ag-doped graphitic carbon nitride nanosheet (Ag-g-C3N4), which was modified onto the glassy carbon electrode (GCE) for immobilizing phenoxy dextran (DexP) through π-π interaction. Then, the target ConA was anchored onto the DexP/Ag-g-C3N4 modified film via the specific carbohydrate-ConA interaction. Successively, polyaniline-3,4,9,10-perylenetetracarboxylic acid-DexP conjugate (denoted as PANI-PTCA-DexP), as the ECL signal quenching probe, was incubated onto the electrode through the carbohydrate-ConA interaction to achieve the “signal-off” state. Here, PTCA was used as a matrix for the high loading of PANI and DexP, PANI served as the quencher towards Ag-g-C3N4 system, and DexP as the recognition element for bounding ConA. With the formation of the sandwiched structure of DexP, ConA and PANI-PTCA-DexP, a desirable quenching ECL signal was measured with S2O82− as the co-reactant of Ag-g-C3N4. The quenching effect of PANI towards Ag-g-C3N4 is positively correlated with the concentration of ConA. With such an “on-off” system, the detection of ConA was achieved with a wide linear range from 0.001ng/mL to 50ng/mL as well as a low detection limit down to 0.0003ng/mL.

Abstract:In this work, we report a novel label-free fluorescence "turn off-on" biosensor for lectin detection. The highly sensitive and selective sensing system is based on the integration of carboxymethyl chitosan (CM-CHIT), CuInS2 quantum dots (QDs) and Au nanoparticles (NPs). Firstly, CuInS2 QDs featuring carboxyl groups were directly synthesized via a hydrothermal synthesis method. Then, the carboxyl groups on the CuInS2 QDs surface were interacted with the amino groups (-NH2), carboxyl groups (-COOH) and hydroxyl groups (-OH) within CM-CHIT polymeric chains via electrostatic interactions and hydrogen bonding to form CM-CHIT-QDs assemblies. Introduction of Au NPs could quench the fluorescence of CM-CHIT- QDs through electron and energy transfer. In the presence of lectin, lectin could bind exclusively with CM-CHIT-QDs by means of specific multivalent carbohydrate-protein interaction. Thus, the electron and energy transfer process between CM-CHIT-QDs and Au NPs was inhibited, and as a result, the fluorescence of CM-CHIT-QDs was effectively "turned on". Under the optimum conditions, there was a good linear relationship between the fluorescence intensity ratio I/I-0 (I and I-0 were the fluorescence intensity of CM-CHIT-QDs-Au NPs in the presence and absence of lectin, respectively) and lectin concentration in the range of 0.2-192.5 nmol L-1, And the detection limit could be down to 0.08 nmol L-1. Furthermore, the proposed biosensor was employed for the determination of lectin in fetal bovine serum samples with satisfactory results. (C) 2016 Elsevier B.V. All rights reserved.
