Abstract:Tissue scaffolds must satisfy multiple design constraints, such as geometry, mechanical properties, and connectivity, to yield a functioning heterogeneous tissue. Onemethod that accounts for these multiple constraints is the unit-cellbased assembly approach. In this method, the volume that represents the natural tissue is filled with unit-cells that meet thedesign requirements of the volume. This approach requires data exchanges between several procedures including design, characterization, assembly, analysis, and fabrication procedures. In the paper, we present a data exchange system to store andretrieve the unit-cell information and customize data migrationamong applications. We also present our application of this dataexchange system to facilitate the management of data flow.

Abstract:Tissue engineering raised a high requirement to controlcell distributionin defined materials and structures. In "ink"-basedbioprintings, such as 3D printing and photolithography, cells wereassociated with inks for spatial orientation; the conditions suitablefor one ink are hard to apply on other inks, which increases the obstaclein their universalization. The Magneto-Archimedes effect based (Mag-Arch)strategy can modulate cell locomotion directly without impelling inks.In a paramagnetic medium, cells were repelled from high magnetic strengthzones due to their innate diamagnetism, which is independent of substrateproperties. However, Mag-Arch has not been developed into a powerfulbioprinting strategy as its precision, complexity, and throughputare limited by magnetic field distribution. By controlling the paramagneticreagent concentration in the medium and the gaps between magnets,which decide the cell repelling scope of magnets, we created simultaneouslymore than a hundred micrometer scale identical assemblies into designedpatterns (such as alphabets) with single/multiple cell types. Cellpatterning models for cell migration and immune cell adhesion studieswere conveniently created by Mag-Arch. As a proof of concept, we patterneda tumor/endothelial coculture model within a covered microfluidicchannel to mimic epithelial-mesenchymal transition (EMT) under shearstress in a cancer pathological environment, which gave a potentialsolution to pattern multiple cell types in a confined space withoutany premodification. Overall, our Mag-Arch patterning presents analternative strategy for the biofabrication and biohybrid assemblyof cells with biomaterials featured in controlled distribution andorganization, which can be broadly employed in tissue engineering,regenerative medicine, and cell biology research.
Transferring Optimized Unit Cell Based Tissue Scaffold Designs to a Freeform Fabrication System

Abstract:Designing for the freeform fabrication of heterogeneous tissue scaffold is always a challenge in Computer Aided Tissue Engineering. The difficulties stem from two major sources: 1) limitations in current CAD systems to assembly unit cells as building blocks to form complex tissue scaffolds, and 2) the inability to generate tool paths for freeform fabrication of unit cell assemblies. To overcome these difficulties, we have developed an abstract model based on skeletal representation and associated computational methods to assemble unit cells into an optimized structure. Additionally we have developed a process planning technique based on internal architecture pattern of unit cells to generate tool paths for freeform fabrication of tissue scaffold. By modifying our optimization process, we are able to transfer an optimized design to our fabrication system via our process planning technique.

Abstract:Three-dimensional (3D) porous tissue scaffolds are being engineered to promote cell attachment, cell proliferation and functioning heterogeneous tissue formation. A scaffold's structure and transport capabilities need to mimic cells' natural geometrical and architectural environment and will need to consistently provide a flux throughout the scaffold to deliver materials for growth and waste-removal. This paper presents our work on establishing a connectivity characterization of designed unit cell tissue structures for surface alignment between units to insure both geometric connectivity and mass and fluid transport within a heterogeneous tissue scaffold to meet both structural and biological requirements for cell growth and tissue growth. To capture this information, skeletal representations of the unit cell structures have been utilized for subsequent assembly processes.

Abstract:A novel targeted application of tissue engineering is the development of an in vitro 3D tissue model for drug screening and toxicology. This paper discusses the modeling, design, and freeform fabrication of 3D cell-embedded tissue constructs for creating a pharmacokinetic model. This is achieved using a combinatorial setup involving a CAD-driven automated syringe-based, layered direct cell writing process in conjunction with soft lithographic micro-patterning techniques. This enables the rational design of a microscale in vitro device housing a bioprinted 3D tissue construct (or micro-organ) that biomimics the cell’s physiological microenvironment for enhanced functionality. Abstract:The development and application of the tissue engineering technique has shown a significant potential in regenerative medicine. However, the limitations of conventional tissue engineering methods (cell suspensions, scaffolds and/or growth factors) restrict its application in certain fields. The novel cell sheet technique can overcome such disadvantages. Cultured cells can be harvested as intact sheets without the use of proteolytic enzymes, such as trypsin or dispase, which can result in cell damage and loss of differentiated phenotypes. The cell sheet is a complete layer, which contains extracellular matrix, ion channel, growth factor receptors, nexin and other important cell surface proteins. Mesenchymal stem cells (MSCs), which have the potential for multiple differentiation, are promising candidate seed cells for tissue engineering. The MSC sheet technique may have potential in the fields of regenerative medicine and tissue engineering in general. Additionally, induced pluripotent stem cell and embryonic stem cell-derived cell sheets have been proposed for tissue regeneration. Currently, the application of cell sheet for tissue reconstruction includes: Direct recipient sites implantation, superposition of cell sheets to construct three-dimensional structure for implantation, or cell sheet combined with scaffolds. The present review discusses the progress in cell sheet techniques, particularly stem cell sheet techniques, in tissue engineering.

Abstract:Organ printing offers a great potential for the freeform layer-by-layer fabrication of three-dimensional (3D) living organs using cellular spheroids or bioinks as building blocks. Vascularization is often identified as a main technological barrier for building 3D organs. As such, the fabrication of 3D biological vascular trees is of great importance for the overall feasibility of the envisioned organ printing approach. In this study, vascular-like cellular structures are fabricated using a liquid support-based inkjet printing approach, which utilizes a calcium chloride solution as both a cross-linking agent and support material. This solution enables the freeform printing of spanning and overhang features by providing a buoyant force. A heuristic approach is implemented to compensate for the axially-varying deformation of horizontal tubular structures to achieve a uniform diameter along their axial directions. Vascular-like structures with both horizontal and vertical bifurcations have been successfully printed from sodium alginate only as well as mouse fibroblast-based alginate bioinks. The post-printing fibroblast cell viability of printed cellular tubes was found to be above 90% even after a 24 h incubation, considering the control effect.

Abstract:Centimeter-scale tissue with angiogenesis has become more and more significant in organ regeneration and drug screening. However, traditional bioink has obvious limitations such as balance of nutrient supporting, printability, and vascularization. Here, with “secondary bioprinting” of printed microspheres, an innovative bioink system was proposed, in which the thermo-crosslinked sacrificial gelatin microspheres encapsulating human umbilical vein endothelial cells (HUVECs) printed by electrospraying serve as auxiliary component while gelatin methacryloyl precursor solution mixed with subject cells serve as subject component. Benefiting from the reversible thermo-crosslinking feature, gelatin microspheres would experience solid-liquid conversion during 37°C culturing and form controllable porous nutrient network for promoting the nutrient/oxygen delivery in large-scale tissue and accelerate the functionalization of the encapsulated cells. Meanwhile, the encapsulated HUVECs would be released and attach to the pore boundary, which would further form three-dimensional vessel network inside the tissue with suitable inducing conditions. As an example, vascularized breast tumor tissue over 1 cm was successfully built and the HUVECs showed obvious sprout inside, which indicate the great potential of this bioink system in various biomedical applications.

Abstract:Tissues and organs exhibit distinct shapes and functions nurtured by vascular connectivity. In order to mimic and examine these intricate structure-function relationships, it is necessary to develop efficient strategies for assembling tissue-like constructs. Many of the top-down fabrication techniques used to build microelectromechanical systems, including photolithography, are attractive due to the similar feature sizes, but are not suitable for delicate biological systems or aqueous environments. A layer-by layer approach has been proposed by us to pattern functional cell structures in three dimensions. Freeform cell structures are created by the inkjet method, in which cells are entrapped within hydrogels and crosslinked on demand. The cells are viable, functional and show potential for cell maturation as exemplified by the diversion of hematopoietic stem cells into multiple cell types. These results show promise for many tissue engineering applications.

Abstract:Engineered microstructures that mimic in vivo tissues have demonstrated great potential for applications in regenerative medicine, drug screening, and cell behavior exploration. However, current methods for engineering microstructures that mimic the multi-extracellular matrix and multicellular features of natural tissues to realize tissue-mimicking microstructures in vitro remain insufficient. Here, we propose a versatile method for constructing tissue-mimicking heterogeneous microstructures by orderly integration of macroscopic hydrogel exchange, microscopic cell manipulation, and encapsulation modulation. First, various cell-laden hydrogel droplets are manipulated at the millimeter scale using electrowetting on dielectric to achieve efficient hydrogel exchange. Second, the cells are manipulated at the micrometer scale using dielectrophoresis to adjust their density and arrangement within the hydrogel droplets. Third, the photopolymerization of these hydrogel droplets is triggered in designated regions by dynamically modulating the shape and position of the excitation ultraviolet beam. Thus, heterogeneous microstructures with different extracellular matrix geometries and components were constructed, including specific cell densities and patterns. The resulting heterogeneous microstructure supported long-term culture of hepatocytes and fibroblasts with high cell viability (over 90%). Moreover, the density and distribution of the 2 cell types had significant effects on the cell proliferation and urea secretion. We propose that our method can lead to the construction of additional biomimetic heterogeneous microstructures with unprecedented potential for use in future tissue engineering applications.

Abstract:AbstractComputer-aided tissue engineering (CATE) integrates advances of multi-disciplinary fields of Biology, Biomedical Engineering, Information Technology, and modern Design and Manufacturing. Application of CATE to the design and fabrication of tissue scaffolds can facilitate the exploration of many novel ideas of incorporating biomimetic and biological features into the scaffold design. This paper presents some of the salient applications of CATE, particularly in the modeling and design of scaffolds with controlled internal and external architecture; with vascular channels of different sizes; with modular and interconnecting subunits; with multi-layered heterogeneous dense and compact regions; and the scaffolds with designed artificial chambers for drug delivery, embedded growth factors and other sophisticated features.

Abstract:Designed microtissues that replicate highly ordered three-dimensional (3D) multicellular in vivo structures have shown huge potential in biomedical research and drug discovery. Through microencapsulation and microfluidic techniques, cell-laden microcapsules have been widely used as pathological or pharmacological models. However, most conventional microtissue construction strategies can only engineer simply predefined microcapsules with monotonous biological components in two dimensions. Here, we propose a flexible 3D microtissue construction method through self-bonding of real-time shape-programmable microcapsules. The microcapsules are prepared by photo-induced electrodeposition of cell-laden alginate hydrogel and flexibly tailored into tissue-specific shapes, sizes, and arbitrary biocomponents. With the local fluidics-guided assembly, the microcapsules are spatially organized into 3D perfectly aligned microtissues. To mimic in vivo intercellular connection, the aligned microcapsules are precoated with fibroblasts to self-bond the adjacent layers into a robust assemblage through fibroblast-extracellular matrix interactions, which highly reproduces the tissue morphogenesis in natural organisms. As a typical complex tissue model, the 3D hepatic lobule was engineered utilizing HepG2 cells seeded into microcapsules with a fibroblast coating, and its biofunction including albumin and urea secretion was improved by nearly two-fold compared with cells seeded without a fibroblast coating. We anticipate that our method will be capable of regenerating more complex multicellular constructs with unprecedented possibilities for future tissue engineering applications.

Abstract:The physical confinement of the extracellular matrix (ECM) has been proven to be a coconspirator in cancer metastasis. However, current experimental models are limited in accurately dissecting the effect of mechanical and topographical properties on cancer cell migration in a confined three-dimensional (3D) environment. In this study, we propose a facile strategy to produce precisely controlled channel-like hydrogel confinements on a microfluidic device via a “hydrogel-in-hydrogel” two-photon 3D printing approach. Within this model, it is demonstrated that breast cancer cells migrate faster in the stiffer confinements with an adapted migratory phenotype, and the transcriptomic profile of the migrating cells shows that the activation of YAP is involved in this process. This platform allows the fabrication of high-resolution hydrogel microstructures, real-time observation of the cell–environmental interplay, and convenient collection of migrating cell samples for sequencing analyses, which provides a powerful tool for investigating cell migration and mechanical interactions within a confined microenvironment.

Emerging Organ Models and Organ Printing for Regenerative Medicine

Abstract:Engineered materials that integrate advances in polymer chemistry, nanotechnology, and biological sciences have the potential to create powerful medical therapies. Our group aims to engineer tissue regenerative therapeutics using water‐containing polymer networks called hydrogels that can regulate cell behavior. Specifically, we have developed photo‐crosslinkable hybrid hydrogels that combine natural biomolecules with nanoparticles to regulate the chemical, biological, mechanical and electrical properties of gels. These functional scaffolds induce the differentiation of stem cells to desired cell types and direct the formation of vascularized heart or bone tissues. Since tissue function is highly dependent on architecture, we have also used microfabrication methods, such as microfluidics, photolithography, bioprinting, and molding, to regulate the architecture of these materials. We have employed these strategies to generate miniaturized tissues. To create tissue complexity, we have also developed directed assembly techniques to compile small tissue modules into larger constructs. It is anticipated that such approaches will lead to the development of next‐generation regenerative therapeutics and biomedical devices.

Abstract:Artificial organs are devices implanted into the living body as a substitute for damaged or diseased organs. Current efforts focus on the construction of fully functionalized artificial tissues/organs with vascular networks. Although engineering efforts have been made in creating artificial vessels with simple or complex configurations, building vascular networks with hierarchical architectures approximating native counterparts remains challenging. Herein, we give a perspective of cellular fluidics-based construction of vascular networks for tissue engineering, with inspirations drawn from a novel concept of 3D fluidic control platform based on unit-cell constructs. Through architected design of the unit cells, it enables programmed control over gas-liquid-solid interfaces and fluid flow processes in open-cell structures. This cellular-fluidics concept and the associated platform provide lots of inspirations for constructing artificial vascular networks. We believe that cellular fluidics opens a new avenue for fluid control and deterministic delivery, and would find vast opportunities in tissue engineering.

Abstract:Morphogenesis involves a complex series of cell signaling, migration and differentiation events that are coordinated as tissues self-assemble during embryonic development. Collective cell movements such as those that occur during morphogenesis have typically been studied in 2D with single layers of cultured cells adhering to rigid substrates such as glass or plastic. In vivo, the intricacies of the 3D microenvironment and complex 3D responses are pivotal in the formation of functional tissues. To study such processes as collective cell movements within 3D multilayered tissues, we developed a microfluidic technique capable of producing complex 3D laminar multicellular structures. We call this technique "3D tissue-etching" because it is analogous to techniques used in the microelectromechanics (MEMS) field where complex 3D structures are built by successively removing material from a monolithic solid through subtractive manufacturing. We use a custom-designed microfluidic control system to deliver a range of tissue etching reagents (detergents, chelators, proteases, etc.) to specific regions of multilayered tissues. These tissues were previously isolated by microsurgical excision from embryos of the African claw-toed frog, Xenopus laevis. The ability to shape the 3D form of multicellular tissues and to control 3D stimulation will have a high impact on tissue engineering and regeneration applications in bioengineering and medicine as well as provide significant improvements in the synthesis of highly complex 3D integrated multicellular biosystems.

Abstract:Adequate vascularization is a critical determinant for the successful construction and clinical implementation of complex organotypic tissue models. Currently, low cell and vessel density and insufficient vascular maturation make vascularized organotypic tissue construction difficult, greatly limiting its use in tissue engineering and regenerative medicine. To address these limitations, recent studies have adopted pre-vascularized microtissue assembly for the rapid generation of functional tissue analogs with dense vascular networks and high cell density. In this article, we summarize the development of module assembly-based vascularized organotypic tissue construction and its application in tissue repair and regeneration, organ-scale tissue biomanufacturing, as well as advanced tissue modeling.

Abstract:A novel modeling approach for designing and constructing CAD-based composite unit cells is presented. The modeling technique is developed based on a reasoning Boolean operation algorithm which consists of merging and extracting operation to construct composite unit cells at the heterogeneous material level. Owing to its CAD-based nature, the unit cell models can be readily implemented with advanced CAD/CAE/CAM systems for integrated design, simulation, and manufacturing of advanced composites. A seamless integration between the CAD model and finite element analysis, and results of stress and deformation in composite unit cells and in the fiber and matrix is demonstrated. The proposed modeling technique is able to capture the designed composite geometry, reinforced fiber architecture, and constituent material heterogeneity. Sample unit cells representing 2D woven fabric, 2D basket weave, 3D tri-axial braided, and 3D uni-directional composite are also presented.

Abstract:Three-dimensional (3D) micronano structures have attracted much attention in tissue engineering since they can better simulate the microenvironment in vivo. Two-photon polymerization (TPP) technique provides a powerful tool for printing arbitrary 3D structures with high precision. Here, the desired 3D biocompatible hydrogel microscaffolds (3D microscaffold) with structure design referring to fibroblasts L929 have been fabricated by TPP technology, particularly considering the relative size of cell seed (cell suspension), spread cell, strut and strut spacing of scaffold. Modulation of the cell behavior has been studied by adjusting the porosity from 69.7% to 89.3%. The cell culture experiment results reveal that the obvious modulation of F-actin can be achieved by using the 3D microscaffold. Moreover, cells on 3D microscaffolds exhibit more lamellipodia than those on 2D substrates, and thus resulting in a more complicated 3D shape of single cell and increased cell surface. 3D distribution can be also achieved by employing the designed 3D microscaffold, which would effectively improve the efficiency of information exchange and material transfer. The proposed protocol enables us to better understand the cell behavior in vivo, which would provide high prospects for the further application in tissue engineering.
