To date, little is known about the underlying cellular function and regulatory networks of EZH2 in PCa. This study aims to determine whether or not the signalling pathway of EZH2/microRNAs serves key functions in PCa development. METHODS Whole-genome RNA sequencing data were downloaded from the TCGA database. Bioinformatics and integrative analytical approaches were employed to identify the relationships of EZH2 to specific cancer-related gene sets. Transfection efficiency of sh-EZH2 was determined by Western blot analysis. The effects of sh-EZH2 on prostate cancer cells were evaluated by microarray, quantitative RT-PCR (qRT-PCR), Cell Counting Kit-8(CCK-8) assay, glucose assay, lactate assay, as well as Western blot analysis. Dual-luciferase reporter assay was used to identify the relationship between EZH2, miR-181b, and IGF-1R. RESULTS The enrichment of gene sets about glucose metabolism progression was associated with EZH2 expression. The depletion of EZH2 in cell experiments inhibited PCa cell growth and blocked glucose metabolism accompanying the downregulation of IGF-1R expression. Furthermore, miR-181b served as an important mediator between EZH2 and IGF-1R. Compared with scrambled control cells, sh-EZH2 cells showed lower H3K27me3 expression and higher miR-181b expression. Western blot and luciferase reporter assays showed that miR-181b inversely modulated IGF-1R by directly targeting the binding site within 3'UTR. Moreover, decreased miR-181b expression largely abrogated the effect of sh-EZH2 on IGF-1R/AKT/PKM2 expression and glucose metabolism progression. Immunohistochemistry and in situ hybridization revealed a significant correlation among EZH2, miR-181b, and IGF-1R expression in human PCa tissues. CONCLUSIONS These findings revealed the novel role and mechanism of EZH2 in regulating cellular growth and glucose metabolism via miR-181b-mediated IGF-1R/AKT/PKM2 pathway in prostate cancer cells. © 2015 by American Urological Association Education and Research, Inc.FiguresReferencesRelatedDetails Volume 193Issue 4SApril 2015Page: e678 Advertisement Copyright & Permissions© 2015 by American Urological Association Education and Research, Inc.MetricsAuthor Information Tao Tao More articles by this author Chunhui Liu More articles by this author Yeqing Huang More articles by this author Han Guan More articles by this author Ming Chen More articles by this author Expand All Advertisement Advertisement PDF downloadLoading ...
