AIB1, also known as Steroid Receptor Coactivator-3, is overexpressed and/or amplified in various cancer types, derived from steroid targeted tissues, such as prostate and breast. Recently, it has been implicated in UCB, as a new independent prognostic marker. However, its underlying mechanism is ill defined in non-steroid targeted bladder cancer cells. METHODS Western Blot (WB) tests AIB1 protein expression in clinic specimen and bladder cell lines. When AIB1 was knocked down in bladder cancer cell lines (T24 and 5637) by RNA interference and specific degrade by chemical-gossypol, FCM, MTT, soft agar and xenograft were utilized to detect the effects of downregulation of AIB1. The glycolytic phenotypes of bladder cancer, such as glucose consumption, medium pH value, lactate and ATP production were also examined under normoxia and hypoxia. The interaction of AIB1 and HIF-1α was investigated by Co-IP. qRT-PCR, WB, Luciferase assay and ChIP test whether AIB1 is necessary for glycolytic genes transcription. The independent statistics analysis of microarray was employed to identify the correlation between AIB1 and glycolytic genes. RESULTS Our results demonstrated knockdown of AIB1 in bladder cancer cells induced G1 arrest and Our results demonstrated knockdown of AIB1 in bladder cancer cells induced G1 arrest and apoptosis, inhibited colony formation and tumorigenesis in vivo. Cell cycle inhibitor, p27, was up-regulated and survival protein BCL2 was reduced in AIB1 knockdown cells independence of cell type. Depletion of AIB1 by RNA interference and pharmacologic inhibition reduced glucose consumption, lactate and ATP production in T24 cancer cells under hypoxia. By interacting with HIF1α, AIB1 was required for HIF1α transcriptional activity and its glycolytic target gene expression, through direct recruitment onto promoters of HIF1α target genes. AIB1expression level showed positive correlation with glycolytic genes in clinic UCB specimen. CONCLUSIONS Together, oveexpression of AIB1 is required for glycolysis in bladder cancer cells through HIF1α, eventually to facilitate tumorigenesis.
