Here, we investigated the potential role of resveratrol and its interactive effects with TRAIL on apoptosis in renal cell carcinoma (RCC) cells and the possible mechanisms. METHODS 786-O and ACHN cells were treated with resveratrol or/and TRAIL and MTT was used to detect cell growth rate. Apoptosis and mitochondrial transmembrane potential (δψm) were determined by flow cytometry after treatment of resveratrol or/and TRAIL. Death receptor (DR) 4, DR 5, and apoptosis associated proteins were detected using Western blot. Effects of resveratrol alone or combination with TRAIL on growth of xenografts in nude mice were observed. RESULTS Resveratrol inhibited cell viability in a dose- and time-dependent manner. TRAIL failed to inhibit cell viability in 786-O and ACHN cells, whereas cell viability was repressed in the presence of resveratrol. 20 µmol/L resveratrol pre-treatment for 24 h was able to induce apoptosis in RCC cells, whereas TRAIL was failed to induce apoptosis. Notably, combination of resveratrol and TRAIL resulted in significant apoptosis in RCC cells. Furthermore, a significant loss of δψm was obtained in resveratrol alone and combination of resveratrol and TRAIL treated cells compared to control cells, and TRAIL alone failed to trigger δψm collapse. Resveratrol alone or combination with TRAIL inhibited growth of 786-O xenografts in nude mice, and the combination of resveratrol and TRAIL was more effective in inhibiting tumor growth, however, TRAIL alone failed to repress tumor growth. Mechanistically, resveratrol upregulated the expression DR4 and DR5, and downregulated the expression of Bcl-2, Bcl-XL, and survivin. CONCLUSIONS Our data suggests that resveratrol inhibits RCC 786-O and ACHN cells growth in a time- and dose-dependent manner and induces cellular apoptosis. In addition, resveratrol could sensitize RCC cells to TRAIL-induced apoptosis.
