Abstract:Abstract Predatory natural enemies play key functional roles in biological control. Abundant predatory arthropod species have been recorded in tea plantation ecosystems. However, few studies have comprehensively evaluated the control effect of predatory arthropods on tea pests in the field. We performed a 1‐year field investigation and collected predatory arthropods and pests in the tea canopy. A total of 7931 predatory arthropod individuals were collected, and Coleosoma blandum (Araneae, Theridiidae) was the most abundant species in the studied tea plantation. The population dynamics between C. blandum and four main tea pest species (Aleurocanthus spiniferus, Empoasca onukii, Ectropis grisescens, and Scopula subpunctaria) were established using the individual number of predators and pests in each month. The results showed that C. blandum appeared to co‐occur in the tea canopy with A. spiniferus, Em. onukii, and Ec. grisescens in a longer period. The prey spectrum of C. blandum was further analyzed using DNA metabarcoding. Among prey species, A. spiniferus, Em. onukii, and Ec. grisescens were included, and the relative abundance and positive rates of target DNA fragments of A. spiniferus were greater than that of other two pests. Combined with the high dominance index of C. blandum, co‐occurrence between C. blandum and A. spiniferus in time and space and high positive rate and relative abundance of target DNA fragments of A. spiniferus, C. blandum was identified to prey on A. spiniferus, and C. blandum may be an important predator of A. spiniferus. Thus, C. blandum has potential as a biological control agent of A. spiniferus in an integrated pest management strategy.
(+)-Catechin, epicatechin and epigallocatechin are important inducible defensive compounds against Ectropis grisescens in tea plants

Abstract:The tea plant, Camellia sinensis (L.) O. Kuntze, is an economically important, perennial woody plant rich in catechins. Although catechins have been reported to play an important role in plant defences against microbes, their roles in the defence of tea plants against herbivores remain unknown. In this study, we allowed the larvae of Ectropis grisescens, a leaf-feeding pest, to feed on the plants, and alternatively, we wounded the plants and then treated them with E. grisescens oral secretions (WOS). Both approaches triggered jasmonic acid-, ethylene- and auxin-mediated signalling pathways; as a result, plants accumulated three catechin compounds: (+)-catechin, epicatechin and epigallocatechin. Not only was the mass of E. grisescens larvae fed on plants previously infested with E. grisescens or treated with WOS significantly lower than that of larvae fed on controls, but also artificial diet supplemented with epicatechin, (+)-catechin or epigallocatechin gallate reduced larval growth rates. In addition, the exogenous application of jasmonic acid, ethylene or auxin induced the biosynthesis of the three catechins, which, in turn, enhanced the resistance of tea plants to E. grisescens, leading to the coordination of the three signalling pathways. Our results suggest that the three catechins play an important role in the defences of tea plants against E. grisescens.

Abstract:Tea, Camellia sinensis (L) O. Kuntze, is the most popular non‐alcohol beverage and has over a thousand years of cultivation history. Sustainable tea cultivation is hampered by insect and mite pests and heavily relies on conventional pesticides. However, such management strategies can cause environmental pollution and insect resistance. Well‐planted tea ecosystem naturally provides a suitable micro‐climatic condition for the prosperity of natural enemies, especially entomopathogenic fungi (EPF) that are susceptible to moisture, temperature and sunlight. Hence, EPF have been widely used in tea plantations for pest management. Here, we summarized the approaches of research pertaining to EPF diversity in ecosystems. We also describe the general infection process and discuss the environmental factors that impact EPF efficiency. Finally, we listed the successful cases of EPF in tea pest management. Overall, this review has provided the application status and biological potential of EPF, which will contribute to agricultural productivity, and ecological, economic and social sustainability in the tea ecosystem.

Correlation of tea green leafhopper occurrence with leaf structure and biochemical components in different tea cultivars

Abstract:Being one of the most disastrous pierce-sucking pests of tea plants [Camellia sinensis (L.) O. Kuntze], tea green leafhopper (Empoasca onukii Matsuda) greatly affects tea yield and quality. In this paper, correlations among leaf structure, biochemical components and tea green leafhopper occurrence in tea cultivars with larger planting area were deeply analyzed, aiming to find the key factor impacting the host selectivity of tea green leafhopper. Tea cultivar 'Longjing 43', together with 'Shuchazao', showed the most susceptibility to this pest with high tea polyphenols content. And the best resistant tea plant cultivar, 'Caoxi No.1', largely relied on the thicker leaf, epidermis (both upper and lower), cuticle and spongy tissue as revealed by principal component analysis. Here, we suggested that tea polyphenols were considered as a biochemical marker for determining the resistance ability to tea green leafhopper of different tea cultivars, along with leaf upper and lower structures as mechanical barriers, which can potentially be used to promote breeding of insect-resistant tea plants.

Abstract:Background: Six-spotted spider mite (Eotetranychus sexmaculatus) is one of the most damaging pests of tea (Camellia sinensis). E. sexmaculatus causes great economic loss and affects tea quality adversely. In response to pests, such as spider mites, tea plants have evolved resistance mechanisms, such as expression of defense-related genes and defense-related metabolites. Results: To evaluate the biochemical and molecular mechanisms of resistance in C. sinensis against spider mites, "Tianfu-5" (resistant to E. sexmaculatus) and "Fuding Dabai" (susceptible to E. sexmaculatus) were inoculated with spider mites. Transcriptomics and metabolomics based on RNA-Seq and liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) technology were used to analyze changes in gene expression and metabolite content, respectively. RNA-Seq data analysis revealed that 246 to 3,986 differentially expressed genes (DEGs) were identified in multiple compared groups, and these DEGs were significantly enriched in various pathways, such as phenylpropanoid and flavonoid biosynthesis, plant-pathogen interactions, MAPK signaling, and plant hormone signaling. Additionally, the metabolome data detected 2,220 metabolites, with 194 to 260 differentially abundant metabolites (DAMs) identified in multiple compared groups, including phenylalanine, lignin, salicylic acid, and jasmonic acid. The combined analysis of RNA-Seq and metabolomic data indicated that phenylpropanoid and flavonoid biosynthesis, MAPK signaling, and Ca2+-mediated PR-1 signaling pathways may contribute to spider mite resistance. Conclusions: Our findings provide insights for identifying insect-induced genes and metabolites and form a basis for studies on mechanisms of host defense against spider mites in C. sinensis. The candidate genes and metabolites identified will be a valuable resource for tea breeding in response to biotic stress.

Abstract:Agriophara rhombata is a tea leaf moth that is considered one of the most destructive pests of Camellia sinensis (tea plant). Several recent studies have shown that many insects acquire part of the microbiome from their host and soil, but the pattern and diversity of their microbiome have not been clearly demonstrated. The present study aimed to investigate the bacterial and fungal communities present in the rhizospheric soil and leaf of tea plant compared to the gut of tea moth at different developmental stages (larvae, pupae, adult female and male) using Illumina MiSeq technology. Alpha diversity (Shannon index) showed higher (p 0.05) between larvae, pupae, female, and male guts. Beta diversity also revealed more distinct bacterial and fungal communities in soil and leaf samples compared with tea moth gut samples, which had a more similar microbiome. Furthermore, Proteobacteria, Firmicutes, and Tenericutes were detected as the dominant bacterial phyla, while Ascomycota, Basidiomycota, and Mortierellomycota were the most abundant fungal phyla among all groups, but their relative abundance was comparatively higher (p < 0.05) in soil and leaf samples compared to tea moth gut samples. Similarly, Klebsiella, Streptophyta, and Enterococcus were the top three bacterial genera, while Candida, Aureobasidium, and Strelitziana were the top three fungal genera, and their relative abundance varied significantly (p < 0.05) among all groups. The KEGG analysis also revealed significantly higher (p < 0.5) enrichment of the functional pathways of bacterial communities in soil and leaf samples than in tea moth gut samples. Our study concluded that the bacterial and fungal communities of soil and tea leaves were more diverse and were significantly different from the tea moth gut microbiome at different developmental stages. Our findings contribute to our understanding of the gut microbiota of the tea moth and its potential application in the development of pest management techniques.

Abstract:Introduction Tea is one of the most widely consumed beverages around the world. Larvae of the moth, Ectropis obliqua Prout (Geometridae, Lepidoptera), are one of the most destructive insect pests of tea in China. E. obliqua is a polyphagus insect that is of increasing concern due to the development of populations resistant to certain chemical insecticides. Microbial biological control agents offer an environmentally friendly and effective means for insect control that can be compatible with “green” and organic farming practices. Methods To identify novel E. obliqua biological control agents, soil and inset cadaver samples were collected from tea growing regions in the Fujian province, China. Isolates were analyzed morphologically and via molecular characterization to identity them at the species level. Laboratory and greenhouse insect bioassays were used to determine the effectiveness of the isolates for E. obliqua control. Results Eleven isolates corresponding to ten different species of Metarhizium were identified according to morphological and molecular analyses from soil and/or insect cadavers found on tea plants and/or in the surrounding soil sampled from eight different regions within the Fujian province, China. Four species of Metarhizium including M. clavatum, M. indigoticum, M. pemphigi , and M. phasmatodeae were documented for the first time in China, and the other species were identified as M. anisopliae, M. brunneum, M. lepidiotae, M. majus, M. pinghaense , and M. robertsii . Insect bioassays of the eleven isolates of Metarhizium revealed significant variation in the efficacy of each isolate to infect and kill E. obliqua . Metarhizium pingshaense (MaFZ-13) showed the highest virulence reaching a host target mortality rate of 93% in laboratory bioassays. The median lethal concentration (LC 50 ) and median lethal time (LT 50 ) values of M. pingshaense MaFZ-13 were 9.6 × 10 4 conidia/mL and 4.8 days, respectively. Greenhouse experiments and a time-dose-mortality (TDM) models were used to further evaluate and confirm the fungal pathogenic potential of M. pingshaense MaFZ-13 against E. obliqua larvae. Discussion Isolation of indigenous microbial biological control agents targeting specific pests is an effective approach for collecting resources that can be exploited for pest control with lowered obstacles to approval and commercialization. Our data show the presence of four different previously unreported Metarhizium species in China. Bioassays of the eleven different Metarhizium strains isolated revealed that each could infect and kill E. obliqua to different degrees with the newly isolated M. pingshaense MaFZ-13 strain representing a particularly highly virulent isolate potentially applicable for the control of E. obliqua larvae.

Abstract:Tea is the second most consumed beverage after water; thus, tea plants are economically important crops in many countries. The frequent application of chemical pesticides over large plantations of tea monoculture has led to pest outbreaks. In recent years, high amounts of highly water-soluble pesticides have been applied because of the proliferation of piercing-sucking insects; however, this method poses health hazards for humans and has negative environmental effects. This review outlines the effects of pesticide applications on the succession of tea pest populations, the risks posed by the use of highly water-soluble pesticides, and the principles of tea pest management. Various pest control techniques, including physical, biological, chemical-ecological, chemical pesticide, and cultural control methods, have been used in the last few decades. We discuss future prospects and challenges for the integrated pest management of tea plantations.

Abstract:Background: The tea plant Camellia sinensis (L.) O. Kuntze is a perennial crop, invaded by diversity of insect pest species, and pink tea mite is one of the most devastating pests for sustainable tea production. However, molecular mechanism of defense responses against pink tea mites in tea is still unknown. In this study, metabolomics and transcriptome profiles of susceptible and resistant tea varieties were compared before and after pink tea mite infestation. Results: Metabolomics analysis revealed that abundance levels of polyphenol-related compounds changed significantly before and after infestation. At the transcript level, nearly 8 GB of clean reads were obtained from each sequenced library, and a comparison of infested plants of resistant and susceptible tea varieties revealed 9402 genes with significant differential expression. An array of genes enriched in plant pathogen interaction and biosynthetic pathways of phenylpropanoids showed significant differential regulation in response to pink tea mite invasion. In particular, the functional network linkage of disease resistant proteins, phenylalanine ammonia lyase, flavanone -3-hydroxylase, hydroxycinnamoyl-CoA shikimate transferase, brassinosteroid-6-oxidase 1, and gibberellin 2 beta-dioxygenase induced dynamic defense signals to suppress prolonged pink tea mite attacks. Further integrated analyses identified a complex network of transcripts and metabolites interlinked with precursors of various flavonoids that are likely modulate resistance against to pink tea mite. Conclusions: Our results characterized the profiles of insect induced metabolic and transcript reprogramming and identified a defense regulatory network that can potentially be used to fend off pink tea mites damage.

Abstract:The tea lace bug, Stephanitis chinensis Drake, 1948, a notorious pest, inflicts significant damage on commercial tea cultivation and ornamental gardens across China. In this study, we investigated the genetic diversity, population structure, and demographic history of S. chinensis based on the genetic and phylogenetic analysis of mitochondrial cytochrome c oxidase I (cox1) sequences. The dataset exhibited 453 polymorphic sites, 315 parsimony informative sites, and 138 singleton variable sites, attesting to the species' genetic complexity. Haplotype diversity varied across populations, with the highest in Ziyang County of Shaanxi Province (SXZY) and lowest in Yongchuan District of Chongqing City (CQYC). Genetic differentiation was primarily within populations, while some pairs showed significant divergence, indicative of geographic barriers and local adaptation. Inconsistent signals from neutrality tests and mismatch distribution analysis suggested historical stability interspersed with growth or bottleneck events. Phylogenetic analysis depicted distinct haplotype clusters, with Mabian County of Sichuan province of Hunan Province (HNGW). These findings illuminate S. chinensis' ecological adaptability, historical dynamics, and potential for adaptive responses to agricultural practices. In addition, the results indicated species' evolutionary trajectory.

Abstract:Habitat suitability analysis using species distribution models (SDMs) presents a valuable approach for determining appropriate land utilization strategies and crop planting decisions, which are vital to achieve optimal agricultural productivity and ensure environmental sustainability in the context of rapid global change. Aside abiotic conditions, pests emerge as the most important biological factor affecting crop growth. It can be an effective avenue to comprehensively assess land use suitability by building and integrating SDMs for both crops and their pests. Using the MaxEnt model and a bivariate map, we predicted and integrated the habitat suitability of tea ( Camellia sinensis ) and its primary insect herbivore ( Empoasca onukii ) in China. Our study explicitly revealed spatial heterogeneity in suitability between the two species, despite finding that they shared common environmental predictors (i.e., temperature, elevation, and soil pH). Results support South China and Southwest China as the most favorable areas for tea cultivation, while highlighting the adverse climatic challenges on tea growth in the middle and lower reaches of the Yangtze River and in the northern regions of the Yangtze River. Meanwhile, analyses emphasize the necessity of sustainable leafhopper management strategies in the middle and lower reaches of the Yangtze River and in regions along the southeast coast. These findings demonstrate the significance of comparative analysis on the habitat suitability of both crops and insect herbivores, providing valuable insights for agricultural production planning and sustainable pest management. Furthermore, the methods employed in this study hold potential for the application in diverse context, ranging from the control of invasive species to the conservation of endangered biodiversity.

Abstract:Tea green leafhopper [Empoasca (Matsumurasca) onukii Matsuda] is one of the most devastating pests of tea plants (Camellia sinensis), greatly impacting tea yield and quality. A thorough understanding of the interactions between the tea green leafhopper and the tea plant would facilitate a better pest management. To gain more insights into the molecular and biochemical mechanisms behind their interactions, a combined analysis of the global transcriptome and metabolome reconfiguration of the tea plant challenged with tea green leafhoppers was performed for the first time, complemented with phytohormone analysis. Non-targeted metabolomics analysis by ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry (UPLC-QTOF MS), together with quantifications by ultra-performance liquid chromatography triple quadrupole mass spectrometry (UPLC-QqQ MS), revealed a marked accumulation of various flavonoid compounds and glycosidically bound volatiles but a great reduction in the level of amino acids and glutathione upon leaf herbivory. RNA-Seq data analysis showed a clear modulation of processes related to plant defense. Genes pertaining to the biosynthesis of phenylpropanoids and flavonoids, plant-pathogen interactions, and the biosynthesis of cuticle wax were significantly up-regulated. In particular, the transcript level for a CER1 homolog involved in cuticular wax alkane formation was most drastically elevated and an increase in C29 alkane levels in tea leaf waxes was observed. The tea green leafhopper attack triggered a significant increase in salicylic acid (SA) and a minor increase in jasmonic acid (JA) in infested tea leaves. Moreover, transcription factors (TFs) constitute a large portion of differentially expressed genes, with several TFs families likely involved in SA and JA signaling being significantly induced by tea green leafhopper feeding. This study presents a valuable resource for uncovering insect-induced genes and metabolites, which can potentially be used to enhance insect resistance in tea plants.

Abstract:Insect and mite pests are damaging stressors that are threatening the cultivation of tea plants, which result in enormous crop loss. Over the years, the effectiveness of synthetic pesticides has allowed for its prominent application as a control strategy. However, the adverse effects of synthetic pesticides in terms of pesticide residue, environmental contamination and insect pest resistance have necessitated the need for alternative strategies. Meanwhile, microbial pesticides have been applied to tackle the damaging activities of the insect and mite pests of tea plants, and their performances were scientifically adjudged appreciable and environmental friendly. Herein, entomopathogenic microbes that were effective against tea geometrid (Ectropis obliqua Prout), tea green leafhopper (Empoasca onukii Matsuda), paraguay tea ampul (Gyropsylla spegazziniana), tea mosquito bug (Helopeltis theivora Waterhouse) and red spider mite (Oligonychus coffea Nietner) have been reviewed. The current findings revealed that microbial pesticides were effective and showed promising performances against these pests. Overall, this review has provided the basic and integrative information on the integrated pest management (IPM) tool(s) that can be utilized towards successful control of the aforementioned insect and mite pests.

Abstract:The achievements in the 20th century and the future development trend of tea breeding and tea cultivation in China were described in the present paper. China, with 1,076,220 ha tea field, is now one of the major tea producing and tea exporting countries in the world and its tea field area ranked the first in the world. Chinese tea industry has been greatly developed in the 20th century because of the development of tea breeding and cultivation techniques.The development of tea breeding in China was benefited from the techniques of vegetable propagation, cross breeding, chemically and physically induced mutation as well as bio-technique and molecular biological technique. There were five stages in the development of tea breeding in China, i.e. the non-organized stage, clonal tea breeding stage, yield breeding stage, quality breeding stage and resistance breeding stage.The tea production in China has been fast developed in the 20th century, especially in the last 50 years. It was mainly benefited from the increase of planting density, the popularization of improved tea cultivars, the manuring technique, the efficient and low-cost cultivation technique.The development trend in Chinese tea breeding will be pest-resistance breeding, high quality breeding and breeding of some special use tea cultivars such as those with high content of catechins, low content of caffeine and low activity of polyphenol oxidase. Intensive management, low-cost and sustainable production will be the important research topics in Chinese tea cultivation in the early decades of the 21st century.
A checklist of spiders in tea plantations of China

Abstract:Background Spiders are the most dominant predatory natural enemies of insect pests in the tea plantation ecosystem. There has been a large amount of literature published about the investigation of spider species in Chinese tea plantations from 1982 to 2020. Here, the spider species in Chinese tea plantations has been summarised and the dominant spider species in each regional tea plantation recorded. To date, there were 535 spider species from 40 families reported in Chinese tea plantations. New information There are 245 spider species from 13 families now being added to the checklist. A total of 89 spider species from 19 families were the dominant species, amongst them, Agelena labyrinthica, Allagelena difficilis, Neoscona theisi, Clubiona deletrix, Clubiona japonicola, Hylyphantes graminicola, Pardosa laura, Oxyopes sertatus, Evarcha albaria, Plexippus paykulli, Coleosoma octomaculatum, Ebrechtella tricuspidata and Xysticus ephippiatus were recorded in many tea plantations. The checklist will provide important data for the biodiversity and distribution of spiders in tea plantations of China.

Abstract:Scopula subpunctaria (Herrich-Schaeffer) (Lepidoptera: Geometridae) is a leaf-eating pest in tea plantations that often causes serious economic losses. Arma chinensis (Fallou) (Hemiptera: Pentatomidae) as a polyphagous insect has become one of the main biological control agents for tea plantation pests due to its wide feeding habit, predatory ability, and adaptability. However, studies related to the predation using A. chinensis on the third instar S. subpunctaria have not been reported. In this study, we used the age-stage, two-sex life table method to analyze the developmental duration and fecundity of S. subpunctaria fed on tea, and A. chinensis fed on third instar S. subpunctaria larvae, under a 25 °C regime. The growth, development, survival, fecundity, and predation rates of the insect populations were investigated. The results showed that the predator and the prey can complete their respective life histories, but the developmental durations at each stage were different, and the developmental stages overlapped significantly. In addition, we used the computer program TIMING-MSChart to project the stage structure and the total population size of A. chinensis and S. subpunctaria. We also simulated the population changes of S. subpunctaria using an A. chinensis intervention. These results showed that S. subpunctaria can support A. chinensis to finish its life history and A. chinensis has great potential to control S. subpunctaria. This study contributes to the understanding of the biological characteristics of S. subpunctaria and provides a theoretical basis for releasing A. chinensis in the field to suppress S. subpunctaria.

Abstract:Insect tea, listed in the “Compendium of Materia Medica” by Li Shizhen, is not only a traditional drink for the ethnic minority in southwest China, but also one of China's traditional export commodities. Insect tea is made of the feces of insects that feed on plants, and characterized by minimal dose, enjoyable tea flavor, few tea-residues, and superb transparency. Insect tea has been used to clear summer heat, protect the spleen and stomach, and facilitate digestion. Modern research has suggested that insect tea is safe and nutritional, and it has blood lipid lowering, antihypertensive and hypoglycemic effects. At present, due to the household production of insect tea, there are a variety of species of tea-producing insects and feeding plants. In the present review, we summarized the types, civilian applications, nutritional value, pharmacological activity and safety of insect tea, in an effort to provide scientific knowledge for future study.

Soil Carbon Sequestration, Plant Nutrients and Biological Activities Affected by Organic Farming System in Tea (camellia Sinensis(l.) O. Kuntze) Fields

Abstract:There is growing interest in investigations into soil carbon (C) sequestration, plant nutrients and biological activities in organic farming since it is regarded as a farming system that could contribute to climate mitigation and sustainable agriculture. However, most comparative studies have focused on annual crops or farming systems with crop rotations, and only a few on perennial crops without rotations, e.g. tea (Camellia sinensis (L.) O. Kuntze). In this study, we selected five pairs of tea fields under organic and conventional farming systems in eastern China to study the effect of organic farming on soil C sequestration, plant nutrients and biological activities in tea fields. Soil organic C, total nitrogen (N), phosphorus (P), potassium (K) and magnesium (Mg), available nutrients, microbial biomass, N mineralization and nitrification were compared. Soil pH, organic C and total N contents were higher in organic tea fields. Soil microbial biomass C, N and P, and their ratios in organic C, total N and P, respectively, net N mineralization and nitrification rates were significantly higher in organic fields in most of the comparative pairs of fields. Concentrations of soil organic C and microbial biomass C were higher in the soils with longer periods under organic management. However, inorganic N, available P and K concentrations were generally lower in the organic fields. No significant differences were found in available calcium (Ca), Mg, sodium (Na), iron (Fe), manganese (Mn), copper (Cu) and zinc (Zn) concentrations between the two farming systems. These findings suggest that organic farming could promote soil C sequestration and microbial biomass size and activities in tea fields, but more N-rich organic fertilizers, and natural P and K fertilizers, will be required for sustainable organic tea production in the long term.
