814 Prevention of Recurrent Ulcer Bleeding in Arthritis Patients with High Cardiovascular and High Gastrointestinal Risks: A 18-Month, Double-Blind, Randomized Trial

Abstract:lysozyme (Paneth) and phyloxine/tartrizine/alcian blue (PTAB; intermediate).H&E stained sections were used for quantifying granule size.Crypts from wild type, Mist1 KO , and Atoh1 KO (negative control) mice were isolated, plated in matrigel, and cultured in Wnt-deficient medium for 7 days to evaluate Paneth cell function in vitro.Results: IF showed Mist1 protein expression confined to Paneth cells within the WT intestinal epithelium, and that Mist1 protein expression was absent in Mist1 KO intestine.No differences in number of crypt or villus enteroendocrine, villus goblet, or crypt lysozyme + cells between WT and Mist1 KO mice were observed.PTAB staining demonstrated a significant decrease in mature Paneth cells and a significant increase in intermediate cells in Mist1 KO mice compared to WT mice.This was confirmed by lysozyme and muc2 co-IF.Granule size was significantly decreased in Mist1 KO mice compared to WT mice.Culture of WT, Mist1 KO , and Atoh1 KO mice in Wnt-deficient medium demonstrated that WT crypts were able to form enterospheres and transition to enteroids, while Atoh1 KO crypts were unable to progress past the entersphere stage, as expected.Mist1 KO crypts were able to form enterospheres and transition to enteroids as WT crypts did, but had an increased budding capacity compared to WT crypts.Conclusions: Mist1 expression is not necessary for allocation of secretory progenitor cells to the Paneth cell lineage.However, Mist1 is necessary for maturation of Paneth cells, and its absence leads to accumulation of less mature intermediate cells in place of Paneth cells.Paneth cell-expressed Mist1 plays a role in controlling budding of intestinal crypts in culture.
