Abstract:A new Schiff-base fluorescence probe CS containing benzimidazole and coumarin fluorophores was synthesized and characterized by NMR, IR and ESI-MS. Fluorescence and absorption spectra show the unique changes in the presence of Hg2+ and Cu2+. Hg2+ induces a remarkable fluorescence enhancement and results in mercury-promoted hydrolysis of a mercury-CS complex to convert the imine group to the aldehyde group. Cu2+ leads to a colorimetric change from colorless to pale yellow and causes formation of a copper-CS complex. Thus, CS can act as a fluorescent chemodosimeter for Hg2+ and a naked-eye chemosensor Cu2+. The cell imaging experiments for Hg2+ show a slow enhancement of a bright blue fluorescence with increasing incubation time, while Cu2+ instantly quenches the fluorescence and then the fluorescence intensity has no further change by extending the incubation time. Therefore, the probe CS shows its potential application for simultaneous differential detection of Hg2+ and Cu2+ in living cells based on the fluorescence response time. (C) 2016 Elsevier B.V. Abstract:Mercury (Hg) is a heavy metal with high toxicity and easy migration; it can be enriched through the food chain, and cause serious threats to the natural environment and human health. So, the development of a method that can be used to detect mercury ions (Hg2+) in the environment, in cells, and in organisms is very important. Here, a new 7-hydroxycoumarin-derived carbonothioate-based probe (CC-Hg) was designed and synthesized for detection of Hg2+. After addition of Hg2+, a large fluorescence enhancement was observed due to the formation of 7-hydroxyl, which reinforced the intramolecular charge transfer process. The CC-Hg probe had good water solubility and selectivity. Moreover, the probe was able to detect Hg2+ quantitatively over the concentration range 0-2 mu M and with a detection limit of 7.9 nM. Importantly, we successfully applied the probe to detect Hg2+ in water samples, in living cells, and in zebrafish. The experimental results demonstrated its potential value in practical applications.

Abstract:Herein, a novel near-infrared fluorescent probe (Cy-PT) for the detection of Hg2+ was designed and synthesized, which contained a hemicyanine skeleton as the near-infrared (NIR) fluorophore and carbonothioate moiety as the recognition unit. The very weak fluorescence of Cy-PT is greatly enhanced due to the destruction of thiocarbonate moiety by Hg2+, generating a hydroxyl group. The study shows that Cy-PT exhibits many advantages including NIR emission (708 nm), colorimetric change, off-on fluorescence, remarkably large Stokes shift (120 nm), rapid response, excellent sensitivity and high selectivity. In addition, Cy-PT can be used as a direct visualization detector for Hg2+ detection, allowing further application using test paper. Subsequent fluorescence imaging shows that Cy-PT probe has high ability to enter living cells and bodies, generating strong NIR fluorescence response in the presence of Hg2+. This study indicates that Cy-PT probe is a good candidate for Hg2+ detection in colorimetric and near-infrared fluorescence, which can be developed into an efficient method for Hg2+ detection in complex living samples.

Abstract:A water-soluble fluorescent probe for Hg(2+) based on a rhodamine B derivative was designed and synthesized. The new probe showed reversible colorimetric and fluorescent response to Hg(2+) in a fully aqueous solution. The probe exhibited real-time detection of Hg(2+) with high selectivity in media containing less than 1% organic cosolvent. Furthermore, bioimaging studies indicated that the new probe was cell permeable and suitable for the real-time imaging of Hg(2+) in living cells by confocal microscopy.

Abstract:A highly sensitive and selective Hg2+ probe 4 based on rhodamine-b was developed and characterized. In consideration of environmental and biological application, we connected a water soluble receptor group (sulfonated β-naphtol) and rhodamine-b together through hydrazine hydrate in high yield. The result turns out that this compound not only exhibits excellent sensitivity and selectivity toward Hg2+, but also shows well cell permeability and compatibility in vitro.

Abstract:A highly selectivity and senstivity fluorescent probe for Hg2+. A simple naphthalimide‐based fluorescent probe was designed and synthesized for the determination of mercury ion (Hg2+). The probe showed a noticeable fluorescence quenching response for Hg2+. When added with Hg2+, the fluorescence intensity of the probe at 560 nm was remarkably decreased with the color changed from yellow to colorless under ultraviolet (UV) light. The probe had a notable selectivity and sensitivity for Hg2+ and displayed an excellent sensing performance when detecting Hg2+ at low concentration (19.5 nM). The binding phenomenon between the probe and Hg2+ was identified by Job's method and high‐resolution mass spectrometry (HRMS). Moreover, the probe was not only utilized to identify Hg2+ in real samples with satisfactory results (92.00%–110.00%) but also was successfully used for bioimaging in cells and zebrafish. The recognition mechanism has been verified by transmission electron microscopy (TEM) for the first time. All the results showed that the probe could be used as a potent useful tool for detection of Hg2+.

Abstract:A new benzothiazole-based fluorescent probe 2-(benzo[d]thiazol-2-yl)-4-(1,3- dithian-2-yl)phenol (BT) with two different reaction sites, a thioacetal group (site 1 for Hg2+), and O and N atoms of the benzothiazole dye (site 2 for Cu2+), was designed and synthesized. The probe BT showed ratiometric fluorescent response to Hg2+ and fluorescence quenching behavior to Cu2+, which induces naked-eye fluorescent color changes from green to blue and colorless, respectively. Moreover, it displayed highly sensitivity and selectivity toward Hg2+ and Cu2+ without interference from other metal ions. The sensing mechanisms were also confirmed by 1H NMR titration, mass spectrum and Job's plot analyses. Finally, probe BT was successfully used for fluorescent imaging of Hg2+ and Cu2+ in living cells, demonstrating its potential applications in biological science.

Abstract:A novel hemicyanine-based near-infrared (NIR) fluorescence probe CY1OH2S for Hg2+ ions detection was synthesized, which can selectively and sensitively responds to Hg2+ ions based on the Hg2+-promoted deprotection of thioacetal. The probe exhibits good response and anti-interference performance for Hg2+ ions based on the fluorescence enhancement in the NIR region under physiological conditions. The detection limit of probe CY1OH2S for Hg2+ ions was as low as 0.32 mu M. Probe-coated test paper was fabricated as a "naked-eye" indicator for Hg2+ ions in water samples. In addition, probe CY1OH2S exhibited good cell permeability and was successfully applied in living cells by NIR fluorescence imaging.

Abstract:Mercury ion (Hg2+) is considered a harmful neurotoxin, and real-time monitoring of Hg2+ concentrations in environmental and biological samples is critical. Fluorescent probes are a rapidly emerging visualization tool owing to their simple design and good selectivity. Herein, a novel fluorescence (FL) probe 2-(4-((6-((quinolin-8-yloxy)methyl)pyridin-2-yl)methyl)piperazin-1-yl)anthracene-9,10-dione (QPPA) is designed using piperazine as a linker between the anthraquinone group, which serves as a fluorophore, and N4O as the Hg2+ ligand. The probe exhibits FL "turn-on" sensing of Hg2+ because the complex inhibits the photo-induced electron transfer (PET) process. Moreover, QPPA can overcome the invasion by other possible cations, resulting in a clear color change from orange to colorless with the addition Hg2+. The chelation of QPPA with Hg2+ in a 1:1 ratio. Subsequently, the theoretically determined binding sites of the ligand to Hg2+ are validated through 1H NMR titration. The in situQPPA-Hg2+ complex can be subjected to Hg2+ extraction following the introduction of S2- owing to its robust binding capacity. The exceptional limit of detection values for Hg2+ and S2- are obtained as 63.0 and 79.1 nM (S/N = 3), respectively. Moreover, QPPA can display bright red FL in the presence of Hg2+ in different biological specimens such as HeLa cells, zebrafish, onion root tip tissues, and water flea Daphnia carinata, further providing an effective strategy for environmental monitoring and bioimaging of Hg2+ in living organisms.

Abstract:It is an effective and precise strategy to design the fluorescence probe for Hg2+ based on a new specific reaction. A novel turn-on fluorescent probe, 4-{244-(2-Mercapto-benzoyloxy)-phenyl]-vinyl)-1-methyl-pyridinium[e]iodide (MBPVP) for Hg2+ was developed. The mechanism was based on Hg2+-mediated ester hydrolysis to release the fluorophore, 442-(4-Hydroxy-phenyl)-vinyl]-1-methyl-pyridinium[e]iodide, which showed outstanding intramolecular charge transfer (ICT) and the yellow-green fluorescence were observed. The turn-on probe showed rapid response time, outstanding selectivity and sensitivity for Hg2+ over other metal ions in 100% PBS buffer (pH = 7.4). The linear range was 2-16 mu M and the detection limit was 6.5 nM. Furthermore, MBPVP was used to determine Hg2+ in real samples and in the solid state.
A Novel Ratiometric Fluorescent Probe for Hg2+ Imaging in Biological System

Abstract:A novel ratiometric fluorescent Hg2+ probe, CDTT, was synthesized based on the changing of intramolecular charge transfer (ICT) process caused by the specific Hg2+ triggered aldehyde recovery. A large excitation shift from 410 nm in the absence of Hg2+ to 485 nm in the presence of Hg2+ was observed. The excitation intensity ratio of F 485/ F 410 increased from 0.06 to 5.02. CDTT exhibited high selectivity and fast response to Hg2+, and its detection limit was determined to be 0.6 ppb, lower than the US EPA standard for drinking water. The ratiometric imaging ability in living cells and 5-day-old zebra fished was confirmed using laser scanning confocal fluorescent microscopy, the preliminary study of the distribution and the toxicity of Hg2+ in zebra fish larvae were also conducted.

Abstract:A fluorescence turn-on chemosensor based on rhodamine B derivative (FD10) has been developed as a highly sensitive chemosensor for Hg 2+ . A prominent fluorescence enhancement was measured in the presence of Hg 2+ , which was in agreement with the changes in the absorption spectrum. Furthermore, by means of laser scanning fluorescence microscopy experiments, it was demonstrated that FD10 was cell-permeable and could be used as a fluorescent probe for monitoring Hg 2+ in living cells.

Abstract:This paper described the optimized design, synthesis and application of a novel rhodamine thiospirolactam derivative as an 'off-on' fluorescent probe for the detection of Hg2+ in aqueous samples. The 'off-on' fluorescence and color signal change of the probe is based on an Hg2+-triggered domino reaction which brings on the opened-ring form of the rhodamine spirolactam to regain the conjugated system of the rhodamine skeleton. In the well designed probe, the thiospirolactam serves as both Hg2+ binding unit and electron-defect carbon centre, a phenolic hydroxyl with very strong nucleophilicity after deprotonation is chosen as the attacking unit, and a benzene ring is introduced on the linker to afford steric effects, which benefits an efficient nucleophilic reaction, with a high sensitivity towards Hg2+. It exhibits a stable response for Hg2+ from 1.0 x 10(-8) to 1.0 x 10(-6) M, with a detection limit of 3.0 x 10(-9) M. The response of the probe to Hg2+ is highly selective and pH-insensitive, with a fast response time. All these unique features make it particularly favorable for cellular Hg2+ imaging applications. It has been preliminarily used for highly sensitive monitoring of Hg2+ levels in living cells with satisfying resolution.

Abstract:Mercury ions (Hg 2+ ) are a class of heavy metal ions that can cause serious harm to the human body due to their toxicity, fluidity, and bioaccumulation. Direct contact with Hg 2+ can cause adverse effects on the human body. In addition, long-term exposure to Hg 2+ can lead to extreme toxicity of enzymes and proteins in the body, disrupting the cell cycle and thus endangering human life. It is becoming increasingly important to develop a method that is reliable, stable, and sensitive for detecting Hg 2+ . In this study, a near-infrared "turn-on" fluorescent probe based on dicyanomethylene-4 H -pyran, DCM-Hg, was designed and synthesized to determine Hg 2+ over other metal ions. Probe DCM-Hg had a detection limit (LOD) of 3.35 μM when used to detect Hg 2+ . The Hg 2+ concentrations and the fluorescence intensities at 695 nm showed a good linear correlation. Additionally, Hg 2+ in HepG2 cells was successfully visualized using the current probe.

Abstract:An easily synthesized reaction-based probe for the fluorescence detection of Hg 2+ ions using was reported. The designed probe exhibited "turn-on" fluorescence response towards Hg 2+ ions via deprotection of the thiocarbonate-protecting group to alcohol in the HEPES/EtOH=8:2 (v/v, 5 mM, pH 7.4). The investigation results of probe Bzp-1 properties for Hg 2+ detection indicated that probe Bzp-1 has satisfactory high selectivity and sensitivity. The detection limit of Bzp-1 for Hg 2+ was found to be 4.2 nM. The recognition mechanism of Bzp-1 for Hg 2+ was confirmed by ESI-MS. Moreover, the probe Bzp-1 has been successfully used to rapidly detect trace amounts of hazardous Hg 2+ ions in real samples such as tap water, seafood and soil with good recoveries and less the relative standard deviations. Moreover, the Bzp-1 can also be used for fluorescence imagining of Hg 2+ in living cells.

Abstract:Mercury is highly toxic. In this work, a fluorescent probe BTD‐Hg‐Lyso was designed and developed for the specific detection of Hg2+. BTD‐Hg‐Lyso revealed a large Stokes shift (210 nm). It was successfully applied to detect Hg2+ in lysosomes. In addition, BTD‐Hg‐Lyso showed good fluorescence enhancement on Hg2+ in HeLa cells, zebrafish and tobacco seedlings (rhizomes and stems). Mercury is highly toxic, and appropriate amounts of the mercury‐selenium complex can protect plasma. However, when excessive mercury (II) ions (Hg2+) are exposed to human skin or ingested directly, it can lead to irreversible accumulation in the body. Therefore, detecting the presence of Hg2+ in cells is important. A novel fluorescent probe BTD‐Hg‐Lyso was designed and constructed based on the intramolecular charge transfer mechanism. Thiotaldehyde could specifically recognize Hg2+ so that the probe could produce a fluorescence enhancement effect. In addition, the fluorescent probe BTD‐Hg‐Lyso exhibited the advantages of large Stokes shift (210 nm) and good selectivity. More importantly, the probe BTD‐Hg‐Lyso could be used for the determination of Hg2+ in cellular lysosomes. BTD‐Hg‐Lyso was able to image Hg2+ in HeLa cells, zebrafish, and tobacco seedlings (rhizome and stem) successfully.
