Abstract:In this work, water-soluble highly fluorescent gold (Au) nanoclusters capped with eptifibatide (eptifibatide@Au NCs) were synthesized with sodium borohydride (NaBH4) as the reducing agent, in which sodium bromide (NaBr) was used as an excellent fluorescence sensitizer in the neutral media. The corresponding fluorescent intensity can enhance 3.37 times in comparison with that in the absence of NaBr, with the fluorescence quantum yield of 1.8%. Furthermore, the as-prepared fluorescent probe exhibits bright green fluorescence, high optical stability, excitation wavelength dependent behavior, good biocompatibility and low toxicity. Furthermore, the cell imaging experiments demonstrate that eptifibatide@Au NCs can stain the cell membrane and cytoplasm of HeLa and Chromaffin cells. Abstract:A simple, convenient and fast preparation for successful fabrication of water-soluble DL-homocysteine capped Au nanoclusters (hcy-AuNCs) was demonstrated. This preparation allowed the generation of water-soluble Au NCs within a short time of 15 min. The resulting hcy-AuNCs were characterized by photoluminescence, UV-Vis absorption, X-ray photoelectron spectroscopy, and transmission electron microscopy (TEM). The mean diameter of hcy-AuNCs was found to be 1.6 +/- 0.2 nm. The Au NCs exhibited an orange-yellow fluorescence emission at 560 nm with a large Stokes shift of 120 nm, a quantum yield of 3.01% and a good stability over the physiologically relevant pH range and ionic strength. Furthermore, cytotoxicity studies showed that the Au NCs exhibited negligible effects in altering cell proliferation or triggering apoptosis. Cancer cell imaging of HeLa cell lines indicated that the obtained Au NCs could serve as a promising fluorescent bioprobe for bioimaging. This strategy, based on the one-step preparation using DL-homocysteine as a reducing/stabilizing reagent, may offer a novel approach to fabricate other water-soluble metal nanoclusters for application in biolabelling and bioimaging.

Abstract:We report a facile strategy to synthesize water-soluble, fluorescent gold nanoclusters (AuNCs) in one step by using a mild reductant, tetrakis(hydroxymethyl)phosphonium chloride (THPC). A zwitterionic functional ligand, D-penicillamine (DPA), as a capping agent endowed the AuNCs with excellent stability in aqueous solvent over the physiologically relevant pH range. The DPA-capped AuNCs displayed excitation and emission bands at 400 and 610 nm, respectively; the fluorescence quantum yield was 1.3%. The effect of borohydride reduction on the optical spectra and X-ray photoelectron spectroscopy (XPS) results indicated that the AuNC luminescence is closely related to the presence of Au(I) on their surfaces. In a first optical imaging application, we studied internalization of the AuNCs by live HeLa cells using confocal microscopy with two-photon excitation. A cell viability assay revealed good biocompatibility of these AuNCs. Our studies demonstrate a great potential of DPA-stabilized AuNCs as fluorescent nanoprobes in bioimaging and related applications.

Abstract:Here, a simple, green, and economic method was developed for the synthesis of fluorescent carbon nanoparticles (CPs) by one-step hydrothermal treatment of grape juice, without any additives (e.g. salts, acids, bases, and organic solvents). The as-prepared CPs showed bright blue fluorescence under the irradiation of UV light, with a high quantum yield of 13.5%, good water solubility, excellent stability, and low toxicity, which can be used as an excellent fluorescent probe for cellular imaging.

Abstract:We describe a method for the preparation of water-soluble gold nanoclusters (Au-NCs) from chloroauric acid using denatured-casein as both a reducing and stabilizing agent. The resulting Au-NCs were characterized by photoluminescence, UV–vis absorption, and X-ray photoelectron spectroscopies, and by transmission electron microscopy. The Au-NCs have an average diameter of 1.7 ± 0.2 nm and exhibit orange-red fluorescence emission peaking at 600 nm (with a Stokes’ shift as large as 237 nm), a quantum yield of 4.3 %, and good stability over the physiologically relevant range of pH values and ionic strength. Cytotoxicity studies showed the Au-NCs to display negligible effects in terms of altering cell proliferation or triggering apoptosis. Fluorescence imaging of HeLa cancer cells was accomplished by loading such cells with the Au-NCs. The fluorescence of the Au-NCs is found to be strongly quenched by Hg(II) ions, and thus the Au-NCs can be used for detecting and, possibly, imaging of Hg(II). An assay was worked out for the determination of Hg(II), and its limit of detection is 1.83 nM, which is 5.5 times lower than the maximum allowed concentration of Hg(II) in drinking water as defined by the US EPA.
Chitosan-Stabilized Self-Assembled Fluorescent Gold Nanoclusters for Cell Imaging and Biodistribution in Vivo.

Abstract:Biocompatible, near-infrared luminescent gold nanoclusters were synthesized in situ using as-prepared chitosan grafted with N-acetyl-l-cysteine (NAC-CS). The fluorescent gold nanoclusters coated with chitosan-N-acetyl-l-cysteine (AuNCs@NAC-CS) were aggregated by multiple ultrasmall gold nanoclusters closing with each other, with strong fluorescence emission at 680 nm upon excitation at 360 nm. AuNCs@NAC-CS did not display any appreciable cytotoxicity on cells even at a concentration of 1.0 mg mL-1. AuNCs@NAC-CS were more insensitive to H2O2 and trypsin compared with fluorescent gold nanoclusters coated with Albumin Bovine V (AuNCs@BSA), which make them have long time imaging in HeLa cells. Furthermore, the obvious fluorescence signal of AuNCs@NAC-CS appeared in the liver and kidney of the normal mice after 6 h injection. And the fluorescence intensity decreased after that because of the highly efficient clearance characteristics of ultrasmall nanoparticles. These findings demonstrated that AuNCs@NAC-CS possessed good fluorescence, low cytotoxicity, and low sensitivity to some content of cells, allowing imaging of the living cells.

Abstract:A simple, green, and solvent-free method was developed for large-scale preparation of fluorescent nitrogen-sulfur-codoped carbon nanoparticles (NSCPs) by direct thermal treatment of gentamycin sulfate at 200 degrees C. The as-prepared NSCPs displayed high water-solubility, long lifetime (14.01 ns), high quantumyield (27.2%), excellent stability, and low cytotoxicity, and can be used as a probe for cellular imaging.

Abstract:We propose an ingenious method for synthesizing cross-linked hollow fluorescent carbon nanoparticles (HFCNs) with green emission by simply mixing acetic acid, water, and diphosphorus pentoxide. This is an automatic method without external heat treatment to rapidly produce large quantities of HFCNs, in contrast to other syntheses of fluorescent carbon nanoparticles that required high temperature, complicated operations, or long reaction times. Characterizations of HFCNs through high-resolution transmission electron microscopy, infrared/Raman spectroscopy, and X-ray diffraction indicate that abundant small oxygenous graphite domains existed and endowed the HFCNs with fluorescent properties. After simple post-treatments, the cross-linked HFCNs can be used for cell-imaging applications. Compared with traditional dyes and CdTe quantum dots, HFCNs are the superior fluorescent bioimaging agent according to their low toxicity, stability, and resistance to photobleaching. The HFCNs were also applied to watermark ink and fluorescent powder, showing their promising potentials for further wide usage.

Abstract:Fluorescent metal nanoclusters (NCs) have many attractive features, including ultrasmall size, good biocompatibility and large Stokes shift, thus making them promising alternatives as fluorescent markers for biological applications. We have developed a facile method of synthesizing water-soluble, near-infrared (NIR) luminescent AuNCs. By virtue of the long fluorescence lifetime of these AuNCs, we explored their application in cellular fluorescence lifetime imaging (FLIM). Furthermore, the interaction of AuNCs with biomolecules such as proteins has been investigated. Upon protein association, the fluorescence of AuNCs was found to be enhanced and their luminescence lifetime was prolonged. Our studies underline the importance of studying the behavior of AuNCs within the complex biological environment for their application as novel cellular imaging probes.

Abstract:A facile strategy to synthesize water-soluble fluorescent gold nanoclusters (Au NCs) stabilized with the bidentate ligand dihydrolipoic acid (DHLA) is reported. The DHLA-capped Au NCs are characterized by UV-vis absorption spectroscopy, fluorescence spectroscopy, transmission electron microscopy, and X-ray photoelectron spectroscopy. The Au NCs possess many attractive features including ultrasmall size, bright near-infrared luminescence, high colloidal stability, and good biocompatibility, making them promising imaging agents for biomedical and cellular imaging applications. Moreover, their long fluorescence lifetime (>100 ns) makes them attractive as labels in fluorescence lifetime imaging (FLIM) applications. As an example, the internalization of Au NCs by live HeLa cells is visualized using the FLIM technique.

Abstract:Background Tumor-target fluorescence bioimaging is an important means of early diagnosis, metal nanoclusters have been used as an excellent fluorescent probe for marking tumor cells due to their targeted absorption. We have developed a new strategy for facile synthesis of Au/Ce nanoclusters (NCs) by doping trivalent cerium ion into seed crystal growth process of gold. Au/Ce NCs have bright fluorescence which could be used as fluorescent probe for bioimaging. Results In this study, we synthesized fluorescent Au/Ce NCs through two-step hydrothermal reaction. The concentration range of 25–350 μM, Au/Ce NCs have no obvious cell cytotoxicity effect on HeLa, HepG2 and L02 cells. Furthermore, normal cells (L02) have no obvious absorption of Au/Ce NCs. Characterization of synthesized Au/Ce NCs was done by using TEM, EDS and XPS. Then these prepared Au/Ce NCs were applied for in vitro/in vivo tumor-target bioimaging due to its prolonged fluorescence lifetime and bright luminescence properties. Conclusions The glutathione stabilized Au/Ce NCs synthesized through hydrothermal reaction possess stable and bright fluorescence that can be readily utilized for high sensitive fluorescence probe. Our results suggest that Au/Ce NCs are useful candidate for in vitro/in vivo tumor bioimaging in potential clinical application.

Abstract:A novel fluorescent Au@SiO2 nanocomposite, with average size of ca. 30 nm in the diameter, was prepared via a simple microemulsion method. Additionally, transmission electron microscopy (TEM), UV-Vis absorption spectra, Fourier transform infrared (FT-IR) spectra and fluorescence spectra were used to characterize this nanocomposite. This newly synthesized, silica-wrapped, gold nanocluster has the following advantages: good water solubility, exceptional biocompatibility, favorable surface properties and excellent fluorescence properties. Because of these advantages, a Au@SiO2 nanocomposite is exceptionally suitable for biological applications. In this study, cellular imaging, as a form of biological application, has been fully investigated, and it was discovered, after covalent conjugation of folic acid (FA), that the nanocomposite effectively recognized over expressed folic acid receptors (FARs) on the HeLa cell's surface. Facile Construction of Highly Luminescent and Biocompatible Gold Nanoclusters by Shell Rigidification for Two-photon pH-Edited Cytoplasmic and In Vivo Imaging

Abstract:Gold nanoclusters (AuNCs), as a novel fluorescent material, have been extensively explored and developed for bioimaging because of their attractive advantages such as ultrasmall size, low toxicity and exceptional two-photon excitation properties. However, it still remains challenging in producing water-soluble, biocompatible and ultrabright AuNCs. Herein, we report on a novel one-pot synthesis of highly luminescent and biocompatible AuNCs by using polyvinyl pyrrolidone (PVP), a water-soluble polymer, to rigidify the primary stabilizing layer (shell) that is composed of 6-aza-2-thiothymine (ATT) ligands bound to the particle. Such shell-rigidification resulted in significant enhancement of the fluorescence efficiency, reaching a quantum yield of 39% under the best condition, about 35-fold increase from the intrinsically weak fluorescence of the AuNCs stabilized by only ATT. The fluorescence enhancement mechanism was systematically characterized, and the results indicate that PVP coating rigidifies the ATT ligand shell through steric hindrance and reduces the nonradiative relaxation of excited states. The biocompatible PVP-AuNCs were further examined for two-photon cellular and sentinel lymph node (SLN) bioimaging, and we observed pH dependent cytoplasmic images and intense green fluorescence in SLN and lymphatic vessels.

Abstract:Water-soluble silver nanoclusters (AgNCs) as a new type of fluorescent material have attracted much attention for their remarkable optical properties and excellent cytocompatibility. However, it is still challenging to synthesize water-soluble AgNCs with good cytocompatibility and excellent fluorescence. Herein, the dialdehyde nanofibrillated cellulose (DANFC)- reduced water-soluble AgNCs capped by glutathione (GSH) with tunable fluorescence emissions were first reported. The DANFC provides a mild reduction environment and crystal growth system for the coordination between silver ions and GSH compared to conventional methods using strong reducing agents. The AgNCs with intense red fluorescence (R-AgNCs@GSH, size ~2.24 nm) and green fluorescence (G-AgNCs@GSH, size ~1.93 nm) were produced by varying the ratios of silver sources and ligands, and could maintain stable fluorescence intensity over 6 months. Moreover, the CCK-8 study demonstrated that the R-AgNCs@GSH and G-AgNCs@GSH reduced by DANFC of excellent cytocompatibility (cell viability > 90%) and enable precise multicolor intracellular imaging of Hela cells in 1 h. This work proposes a novel method to synthesize water-soluble AgNCs with tunable fluorescence emission at room temperature based on the classical silver- mirror reaction (SMR) using DANFC as reducing agent, and the synthesized fluorescent AgNCs have great potential as novel luminescent nanomaterials in biological research.

Abstract:Gold nanoclusters (Au NCs) as ultrasmall fluorescent nanomaterials possess discrete electronic energy and unique physicochemical properties, but suffer from relatively low quantum yield (QY) which severely affects their application in displays and imaging. To solve this conundrum and obtain highly-efficient fluorescent emission, 2D exfoliated layered double hydroxide (ELDH) nanosheets were employed to localize Au NCs with a density as high as 5.44 x 10(13) cm(-2,) by virtue of the surface confinement effect of ELDH. Both experimental studies and computational simulations testify that the excited electrons of Au NCs are strongly confined by MgAl-ELDH nanosheets, which results in a largely promoted QY as well as prolonged fluorescence lifetime (both similar to 7 times enhancement). In addition, the as-fabricated Au NC/ELDH hybrid material exhibits excellent imaging properties with good stability and biocompatibility in the intracellular environment. Therefore, this work provides a facile strategy to achieve highly luminescent Au NCs via surface-confined emission enhancement imposed by ultrathin inorganic nanosheets, which can be potentially used in bio-imaging and cell labelling.

Abstract:In this study, we have developed a facile, rapid one-pot strategy for synthesis of orange fluorescent gold nanoclusters (MBISA-AuNCs) at room temperature by using 2-Mercapto-5-benzimidazolesulfonic acid (MBISA) as both reducing and protecting reagent. The prepared AuNCs exhibited prominent advantages including high fluorescent quantum yield, good water solubility, excellent biocompatibility and low toxicity. Based on these superior properties, the AuNCs have been multi-talented applied in cell imaging, hydrogen sulfide detection and pH sensing. Confocal fluorescence imaging showed that the MBISA-AuNCs could enter living cells and distribute in the cytoplasm. Additionally, the obtained AuNCs could visualize H2S fluctuations in living cells without interference of autofluorescence based on their high sensitivity and selectivity detection for H2S with a linear range of 2.7 x 10(-11)-8.5 x 10(-4) M and a detection limit of 0.024 nM. Most strikingly, the AuNCs exhibited strong pH dependent behavior and responded linearly, rapidly and reversibly to minor pH fluctuations within the range of 2.01-12.00. Therefore, the AuNCs could also image pH fluctuations in live cells with negligible autofluorescence and provided a new method of analysis to visualize monitoring wide pH range change in live cells.

Abstract:Copper nanoclusters (Cu NCs) are generally formed by several to dozens of atoms. Because of wide range of raw materials and cheap prices, Cu NCs have attracted scientists’ special attention. However, Cu NCs tend to undergo oxidation easily. Thus, there is a dire need to develop a synthetic protocol for preparing fluorescent Cu NCs with high QY and better stability. Herein, we report a one-step method for preparing stable blue-green fluorescent copper nanoclusters using glutathione (GSH) as both a reducing agent and a stabilizing agent. High-resolution transmission electron microscopy (HRTEM), X-ray photoelectron spectroscopy (XPS) and electrospray ionization mass spectrometer (ESI-MS) were used to characterize the resulting Cu NCs. The as-prepared Cu NCs@GSH possess an ultrasmall size (2.3 ± 0.4 nm), blue-green fluorescence with decent quantum yield (6.2%) and good stability. MTT results clearly suggest that the Cu NCs@GSH are biocompatible. After incubated with EB-labeled HEK293T cells, the Cu NCs mainly accumulated in nuclei of the cells, suggesting that the as-prepared Cu NCs could potentially be used as the fluorescent probe for applications in cellular imaging.

Abstract:Synthesis of ultrasmall water-soluble fluorescent gold nanoclusters is reported. The clusters have a decent quantum yield, high colloidal stability, and can be readily conjugated with biological molecules. Specific staining of cells and nonspecific uptake by living cells is demonstrated.
