Abstract:Polydiacetylenes (PDAs) have received increasing attention as smart materials owing to their unique optical and electronic properties. PDAs are most commonly synthesized as self-assembled bilayer vesicles via 1,4 addition polymerization. Vesicles are often perceived as closely mimicking the cell membrane, thus functioning as convenient biomimetic platforms. PDAs undergo colorimetric and fluorescent transition in response to external stimuli. Therefore, PDA vesicles can be used in the field of medical detection including bacteria, viruses, proteins, enzymes, etc. In this review, the preparation of PDA vesicle sensing systems and the mechanism of solid phase polymerization are summarized. The applications of PDA vesicle sensing systems in the field of medical detection are introduced here in detail. Besides, the shortcomings and issues to be solved, and future directions are also discussed and prospected.

Abstract:We report a type of polymer microgel that can undergo a rapid and highly sensitive volume change upon adding H2O2. Such a H2O2-sensitive microgel is made of dextrantyramine and horseradish peroxidase (HRP), which are interpenetrated in chemically cross-linked gel networks of poly(oligo(ethylene glycol) methacrylates). Unlike the H2O2-sensitive microgels reported in previous arts that typically involve degradation processes related to H2O2-induced cleavability of specific bonds, the proposed microgels can shrink upon adding H2O2 owing to the HRP-catalyzed coupling reaction of tyramine residues via decomposition of H2O2. While a fast (<10 s) and stable shrinkage of the microgels can be reached upon adding H2O2 over a concentration range 50.0 mu M1.0 mM, the response time can be modulated by the dispersion temperature in a nonmonotonous way over 1038 degrees C. With the microgels as probes, the H2O2 detection limit was approximately 6.8 ?M. In a combined use of the microgels with glucose oxidase for glucose detection, the glucose detection limit was approximately 83.1 mu M.

Abstract:It was found that Fe3O4 nanoparticles (Fe3O4 NPs) possess intrinsic enzyme mimetic activity similar to that found in natural peroxidase. Here, we applied Fe3O4 NPs to the construction of efficient electrochemical sensor to detect the concentration of hydrogen peroxide. The sensor was fabricated with layer-by-layer assembly of Fe3O4 NPs and poly(diallyldimethylammonium chloride) (PDDA) through the electrostatic interaction, and the multilayer film was characterized with UV–vis absorption spectra, atomic force microscopy, and cyclic voltammetry. Moreover, the sensor showed prominent electrocatalytic activity toward the reduction of H2O2, and the interferences of ascorbic acid (AA) and uric acid (UA) were completely avoided. Unlike the inherent instability of enzyme, Fe3O4 NPs-based sensor demonstrated outstanding stability.

Abstract:Under UV irradiation, ordered diacetylene molecules can be polymerized and form polydiacetylene (PDA) vesicle. Generally speaking, optical absorption in PDA occurs via a pi-to-pi* absorption within the linear pi-conjugated polymer backbone. When decorated with specific probe( receptors) PDA vesicles will undergo dramatic color changes from blue to red in the direct responding to the ligands on the surface of bio-molecules (eg. DNA, antibody-antigen and the bacteria) to be recognized by ligand-receptor binding at the interface and act as a "reporter" of molecular recognition. In some circumstances the red phase transition accompanies strong fluorescence. The unique chromatic properties of molecular recognition made PDA vesicles a promising biosensor and many biological species, such as virus, bacteria, lipolytic enzyme, antimicrobial peptide, mammalian peptide, ions, antibody-antigen and oligonucleotide etc. have been detected. PDA vesicles can also combine with nanogold for fabricating hollow spheres, which can be used as carrier of nanogold having a better performance for DNA immobilization, recognition and separation. With the improvement of detection conditions and influence factors, PDA vesicles of high sensitivity and high selectivity of recognition could be obtained, showing a great prospect to be applied as biosensors.
The Dual Colorimetric Discrimination of Dicyanovinyl−Substituted Polydiacetylene Vesicle Towards Both Dichloromethane and Hydrazine with High Selectivity

Abstract:Functionalized poly(diacetylenes) (PDAs), which can exhibit distinct colorimetric and fluorescent transition in response to certain external stimuli, have received increasing interest as smart materials. In this contribution, a dicyanovinyl substituted PDA derivative was polymerized in the form of vesicle and its thermal stability was studied at first. Then some commonly used organic solvents were added into the vesicle aqueous solution, among which only dichloromethane induced a distinct, immediate, blue to red color change of the PDA vesicle, while the other solvents could not induce any detectable chromatic changes of PDA even after a prolonged period of time. More interestingly, upon the addition of various kinds of anions, cations, and chemical compounds, the PDA based vesicle solution only showed a clear blue−to−purple change in the presence of hydrazine, but it remained blue in the presence of other analytes. The specific mechanism for such high selectivity of this PDA derivative was studied depending on detailed experimental characterizations as well as theoretical calculation. To the best our knowledge, this is the first work reporting the dual colorimetric discrimination behavior of PDA material against two different kinds of external stimuli, which would broaden the application prospect of functional PDAs in the area of biochemical sensing and intelligent recognition.
Preparation of Ag nanoparticle-decorated poly(m-phenylenediamine) microparticles and their application for hydrogen peroxide detection.

Abstract:The chemical oxidation polymerization of m-phenylenediamine (MPD) by ammonium persulfate (APS) at room temperature results in the formation of poly(m-phenylenediamine) (PMPD) microparticles. The subsequent treatment of such microparticles with an aqueous AgNO3 solution produces Ag nanoparticle (AgNP)-decorated PMPD microparticles. It was found that as-formed AgNPs exhibited remarkable catalytic performance toward the reduction of hydrogen peroxide (H2O2). The enzymeless H2O2 sensor constructed with such composites showed a fast amperometric response time of less than 5 s, and the corresponding linear range and detection limit were estimated to be from 0.1 to 30 mM and 4.7 mu M, respectively, at a signal-to-noise ratio of 3.

Abstract:Hydrogen peroxide (aqueous solution of H2O2) is one of the most used reagents i n medical sterilization, environmental disinfection, food storage, and other fields. However, hydrogen peroxide has the potential to cause serious harm to biological health and environmental safety. There are many methods (especially electrochemistry) for H2O2 detection in liquid phase systems, but a lack of methods for vapor detection. This is due to its colorless and tasteless nature, as well as the oxidative activity of the molecule and its coexistence with humidity. In this study, poly(3,4-ethylenedioxythiophene):polystyrene sulfonate (PEDOT:PSS), one of the most commercially successful and widely used conductive polymers, was employed to fabricate an all-organic chemiresistive sensor for simple, real-time, and on-site sensing of hydrogen peroxide vapor (HPV) at room temperature. In comparison with pristine PEDOT:PSS film, the PEDOT:PSS/PEDOT film was prepared by in situ electrochemical polymerization. Upon exposure to different concentrations of HPV, it was found that the hydrophobic and porous PEDOT layer could weaken the interference of humidity in HPV sensing, resulting in a more sensitive and accurate response. At 1.0 ppm HPV concentration, the resistance signal response was increased by nearly 89% compared with the pristine PEDOT:PSS film. This PEDOT-film-based chemiresistive sensor showcases the possibility for further development of nonenzymatic HPV monitoring technology.

Abstract:Polydiacetylenes are prepared from amphiphilic diacetylenes first through self-assembly and then polymerization. Different from common supramolecular assemblies, polydiacetylenes have stable structure and very special optical properties such as absorption, fluorescence, and Raman. The hydrophilic head of PDAs is easy to be chemically modified with functional groups for detection and imaging applications. PDAs will undergo a specific color change from blue to red, fluorescence enhancement and Raman spectrum changes in the presence of receptor ligands. These properties allow PDA-based sensors to have high sensitivity and specificity during analysis. Therefore, the PDAs have been widely used for detection of viruses, bacteria, proteins, antibiotics, hormones, sialic acid, metal ions and as probes for bioimaging in recent years. In this review, the preparation, polymerization, and detection mechanisms of PDAs are discussed, and some representative research advances in the field of bio-detection and bioimaging are highlighted.

Abstract:The detection of hydrogen in air is essential because of the flammability of hydrogen gas (H-2). Optical hydrogen sensor is safer than electrical hydrogen sensor because it can eliminate the potential risk of sparks by reading out remotely. Palladium-based optical hydrogen sensors have attracted lots of attentions. However, impurity poisoning of the sensing materials is a severe problem for the utilization of hydrogen sensors. Here, we report that a palladium (Pd) -PMMA bilayer on a polydimethylsiloxane (PDMS) substrate can eliminate the adverse effect of the impurity (such as oxygen) in air. With a 16-nm PMMA layer coating on the Pd surface, the hydrogenation time of a 25-nm Pd film on PDMS can decrease to 5 seconds exposed to 4% H-2 mixed with dry air. Meanwhile, the Pd-PMMA bilayer on PDMS shows a high reflectance change over 22.7% at the whole visible band with the reflectance contrast more than 370% due to the light scattering on the wrinkling surface induced by the volume expansion of the Pd film on PDMS during hydrogenation. By setting the alarm threshold of the reflectance change of 4% at the 600-nm wavelength, the response time of the sensor can be reduced to 1 second. This optical hydrogen sensor is low-cost and easy-operation with high optical contrast and fast response rate, showing great potential for practical applications.

Abstract:A novel hemin/phytic acid doped polyaniline (PA-PANI) hydrogel composite was prepared through a simple chemical and self-assembly method, which was modified onto electrode for electrochemical detection of H2O2 released from living cells. It showed good analytical performance with high sensitivity, selectivity and a rapid response for the analysis of H2O2 in the range of 2 to 102 mu M, with the detection limit of 1.2 mu M. The favorable results mainly originated from both the high conductivity of PA-PANI hydrogel and its network structure preventing hemin from self-dimerization to provide active catalytic species. Furthermore, PA-PANI with good biocompatibility allowed living cells to adhere and resulted in a short diffusion distance between H2O2 released from cells and electrode.

Abstract:Trace vapor detection of hydrogen peroxide (H2O2) represents a practical approach to nondestructive detection of peroxide-based explosives, including liquid mixtures of H2O2 and fuels and energetic peroxide derivatives, such as triacetone triperoxide (TATP), diacetone diperoxide (DADP), and hexamethylene triperoxide diamine (HMTD). Development of a simple chemical sensor system that responds to H2O2 vapor with high reliability and sufficient sensitivity (reactivity) remains a challenge. We report a fluorescence ratiometric sensor molecule, diethyl 2,5-bis((((4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)benzyl)oxy)carbonyl)amino)terephthalate (DAT-B), for H2O2 that can be fabricated into an expedient, reliable, and sensitive sensor system suitable for trace vapor detection of H2O2. DAT-B is fluorescent in the blue region, with an emission maximum at 500 nm in the solid state. Upon reaction with H2O2, DAT-B is converted to an electronic "push-pull" structure, diethyl 2,5-diaminoterephthalate (DAT-N), which has an emission peak at a longer wavelength centered at 574 nm. Such H2O2-mediated oxidation of aryl boronates can be accelerated through the addition of an organic base such as tetrabutylammonium hydroxide (TBAH), resulting in a response time of less than 0.5 s under 1 ppm of H2O2 vapor. The strong overlap between the absorption band of DAT-N and the emission band of DAT-B enables efficient Förster resonance energy transfer (FRET), thus allowing further enhancement of the sensing efficiency of H2O2 vapor. The detection limit of a drop-cast DAT-B/TBAH film was projected to be 7.7 ppb. By combining high sensitivity and selectivity, the reported sensor system may find broad application in vapor detection of peroxide-based explosives and relevant chemical reagents through its fabrication into easy-to-use, cost-effective kits.
A Self-Assembled Ratiometric Polymeric Nanoprobe for Highly Selective Fluorescence Detection of Hydrogen Peroxide

Abstract:In this study, a dual-emission fluorescence resonance energy transfer (FRET) polymeric nanoprobe by single wavelength excitation was developed for sensitive and selective hydrogen peroxide (H2O2) detection. Polymeric nanoprobe was prepared by simple self-assembly Of functional lipopolymers, which were 4-carboxy-3-fluorophenylboronic acid (FPBA)-modified DSPE-PEG (DSPE-PEG-FPBA) and 7-hydroxycoumarin :(HC)-conjugated DSPE-PEG (DSPE-PEG-HC). Subsequent binding of alizarin red S (ARS) to FPBA endowed the nanoprobe with a new fluorescence emission peak at around 600 nm. Because of the perfect match of the fluorescence emission spectra of HC with the absorbance spectra of ARS-FPBA, FRET was achieved between them. The sensing strategy for H2O2 was based on H2O2-induced deboronadon reaction and boronic acid-mediated ARS fluorescence. Interaction between phenylboronic acid and ARS was revisited herein and it was found that electron-donating or-withdrawing group on phenylboronic acid (PBA) has significant influence.on the fluorescence property of ARS, which enabled sensitive and selective H2O2 sensing. The nanoprobe displayed two well-separated emission bands (150 nm), providing high specificity and sensitivity for ratiometric detection of H2O2. Further application was exploited for the determination of glucose and the results demonstrated that the proposed strategy showed ratiometric response capability for glucose detection. The current method does not involve complicated organic synthesis and opens a new avenue for the construction of multifunctional polymeric fluorescent nanoprobe.

Abstract:Polydiacetylene vesicles were used to detect oligonucleotides in the present paper. For this system, probe-oligonucleotide-functionalized polydiacetylene vesicles were used as indicators of color transition and amplification tags at the same time in the detection of oligonucleotides. The method described is simple, rapid, and suitable for detection of oligonucleotides by naked eye. The influences of dimyristoylphosphatidylcholine (DMPC) in the color changeable polydiacetylene vesicles on the detection of oligonucleotides were also studied. The experimental results demonstrated that DMPC can effectively adjust the mechanical stress of polydiacetylenic structure and the content of DMPC in polydiacetylene structure is a crucial factor for a more sensitive polydiacetylene-based sensor. The maximum colorimetric response for the detection of oligonucleotides was obtained for the polydiacetylene vesicle composed of TCDA/DMPC/probe-DNA with molar ratio of 0.60:0.39:0.01. Both cases for too much or for too less DMPC in polydiacetylene vesicles are not benefit to the detection for this system. The TEM images further confirmed the results obtained.
Visual Hydrogenation of Palladium Membranes on an Elastic Substrate and Their Applications in Hydrogen Sensing

Abstract:AbstractThe study of metal‐hydrogen interaction is essential for hydrogen‐related applications. Due to the small permittivity variation of palladium (Pd), it is difficult to visualize its hydrogenation processes. Soft substrates can release the expansion stress of Pd membrane and form wrinkling during hydrogenation, which can be utilized to track the hydrogen diffusion. Here, we investigate the hydrogenation processes of Pd membranes with various shapes and sizes on polydimethylsiloxane (PDMS) substrates. It was found that the Pd membranes priorly wrinkles from the edges during hydrogenation and the saturation time of hydrogen absorption reduces with the decreasing size of Pd membranes. With these measurements, we fabricate a high‐performance optical hydrogen sensor consisted of a Pd nanostripe array on PDMS. Due to the transition from specular reflection to scattering during the surface wrinkling of the Pd nanostripe, when exposed to 4% hydrogen gas mixed with nitrogen gas, the sensor shows a high‐reflectance contrast of more than 800% with the absolute reflectance change exceeding 32% over the visible band, and the response time t90 is about 1.5 seconds. These results suggest that the Pd membranes on an elastic substrate is a promising platform for visualization of hydrogen diffusion and hydrogen detection.
Design and development of a fluorescent probe for monitoring hydrogen peroxide using photoinduced electron transfer

Abstract:A novel fluorescent probe, 7-hydroxy-2-oxo-N-(2-(diphenylphosphino)ethyl)-2H-chromene-3-carboxamide (DPPEA-HC) was developed for use in monitoring hydrogen peroxide (H2O2) production. DPPEA-HC, which consists of a diphenylphosphine moiety and a 7-hydroxycoumarin moiety, reacts with H2O2 to form DPPEA-HC oxide, which is analogous to the reaction of triphenylphosphine with hydroperoxides such as H2O2 to form triphenylphosphine oxide. Photoinduced electron transfer (PET) was applied in the design of DPPEA-HC. Since the diphenylphosphine moiety and the 7-hydroxycoumarin moiety would act as the PET donor and the acceptor, respectively, it would be expected that DPPEA-HC would rationally cancel the PET process via the formation of DPPEA-HC oxide, based on the calculated energy levels of the donor and the acceptor moieties using the B3LYP/6-31G*//AM1 method. The fluorescence intensity of DPPEA-HC increased on the addition of a H2O2 solution in 100 mM sodium phosphate buffer (pH7.4), as predicted from the energy level calculation and a good correlation between increase in the fluorescence of DPPEA-HC and the concentration of H2O2 was observed. DPPEA-HC was also fluoresced by H2O2, which was enzymatically produced in xanthine/xanthine oxidase/superoxide dismutase (XA/XOD/SOD) system. The increase in the fluorescence of DPPEA-HC in the presence of H2O2 immediately ceased on the addition of catalase (CAT), which catalyzes the disproportionation of H2O2. In addition, DPPEA-HC was found to have a much higher selectivity for H2O2 and a greater resistance to autoxidation than 2',7'-dichlorodihydrofluoresein (DCFH). Time-resolved fluorescence measurements of DPPEA-HC and DPPEA-HC oxide confirmed that the fluorescence off/on switching mechanism of DPPEA-HC is based on the PET on/off control.

Abstract:We report the straightforward synthesis of two types of H2O2-cleavable poly(ester-amide)s (P1 and P2) via the Passerini multicomponent polymerization (P-MCP) of 4-formylbenzeneboronic acid pinacol ester with 1,6-diisocyanohexane and 1,6-hexanedioic acid or a polyethylene glycol (PEG) dicarboxylic acid. The H2O2-cleavable phenylboronic acid ester was integrated into the polymer backbone by the in situ formed benzyl ester bond. GPC and 1H NMR confirmed the complete H2O2-triggered degradation of these polymers in aqueous medium by a mechanism of sequential oxidation of phenylboronic acid ester and self-immolative elimination. Compared with the hydrophobic polymer P1, the PEG-based water-soluble polymer P2 degraded much faster even at a lower H2O2 concentration. Cytocompatible nanoparticles of polymer P1 loaded with fluorescent Nile red were fabricated, and controlled release of Nile red in response to H2O2 was achieved, thus, demonstrating the utility of these polymers as potential H2O2-responsive delivery vehicles.

Abstract:The composites (LDH-CMC) composed of carboxymethyl chitosan (CMC) and 2D ZnAl layered double hydroxide (LDH) were successfully prepared using the one-step urea method; these composites were characterized by XRD, FT-IR, UV-vis DRS, SEM, BJH/BET, TG-DTG and pHzpc analyses, cyclic voltammetry, and electrochemical impedance spectroscopy. The use of CMC could impact the textural and surface chemical properties of the LDH-CMC composites, where the composites still maintained the 2D layered structure. Incorporating a moderate amount of CMC could increase both the surface area and the permanent charge density of the composites, leading to improved electrochemical performances. The LDH-CMC composite was used as a support matrix for the immobilization of horseradish peroxidase (HRP) on the glass carbon (GC) electrode to construct a biosensor that provides a biocompatible microenvironment for HRP and a pathway for H2O2 diffusion via the high surface area. The HRP biosensor displayed a satisfactory sensitivity and fast response (<3 s) toward H2O2 over a wide linear range of 0.02-6.0 mmol·L-1 with a low detection limit of 12.4 μmol·L-1, good anti-interference ability and long-term storage stability. The proposed HRP biosensor was found to be a sensitive, rapid, and disposable sensor with low cost, easy preparation and high selectivity; thus, the proposed biosensor can be used for the real-time detection of trace H2O2 in the biological, clinical and environmental fields.
A Novel Fluorescent Glycopolymer for Endogenous Hydrogen Peroxide Imaging in Living Cells in a Fully Aqueous Environment

Abstract:The development of H2O2-responsive fluorescent glycopolymers with high specificity, sensitivity, and water solubility is a promising strategy for developing probes to detect the production and delivery of endogenous H2O2. Herein, two series of fluorescent glycopolymers (boronic acid derivatives, PG-PFE, and borate derivatives, PG-PFW) were synthesized through esterification and the Williamson reaction, respectively. To increase the sensitivity of the fluorescent glycopolymers, the effect of the reaction conditions on the grafting ratio was investigated in detail. Compared with the PG-PFE samples, the PG-PFW samples had a higher grafting ratio due to the highly efficient Williamson reaction. The structure and properties of the fluorescent glycopolymers were characterized by using nuclear magnetic resonance spectroscopy, gel permeation chromatography, ultraviolet spectroscopy, and photoluminescent spectroscopy. The obtained results showed that all of these fluorescent glycopolymers had good water solubility, low cytotoxicity, and selective responsiveness toward H2O2. HPG-PFW-5, with the highest grafting ratio, displayed a higher sensitivity toward H2O2 and higher stability during hydrolysis than the other materials. In confocal laser scanning microscope images, all of the fluorescent glycopolymers were observed only in cellular mitochondria, and HPG-PFW-5 exhibited a high degree of overlap with Mitotracker Red because of its high sensitivity.

Abstract:The hydrophobic polymer HDDA-EDT-SADA with aggregation-induced emission (AIE) property was synthesized via the thiol-ene click copolymerization of salicylaldehyde azine (SA) derivative diacrylate monomer SADA and commercially available 1,2-ethanedithiol (EDT) and 1,6-hexanediol diacrylate (HDDA). HDDA-EDT-SADA was found to display unexpected intense fluorescence in acetone and DMSO compared to that in other good solvents, which dramatically decreased by adding small amount of water. We demonstrate that HDDA-EDT-SADA can be used as a fluorescent sensor for water detection in acetone and DMSO. To improve the water solubility of the AIE-active polymer, SADA was copolymerized with dithiothreitol (DTT) and poly(ethylene glycol) diacrylate (PEGDA) to give an amphiphilic AIE polymer PEGDA-DTT-SADA. Similar fluorescence phenomenon was also found in DMF, suggesting that PEGDA-DTT-SADA could serve as an indicator for water detection in DMF. Thanks to the PEG segments and DTT units, the PEGDA-DTT-SADA was able to well-disperse in water via the formation of stable micelles. The PEGDA-DTT-SADA selectively complexed with Cu2+ and thus could be used as a fluorescent sensor for Cu2(+) detection in aqueous media. Moreover, the PEGDA-DTT-SADA could be utilized as a fluorescent sensor for hemolysis ratio detection of red blood cells (RBCs) due to the inner filter effect.
