Abstract:The shake flask aerobic biodegradation experiment was used to study the effect on biodegradation of 17α-ethinylestradiol (EE2) in the sediment/water system affected by mono-rhamnolipid (RL-F1) and di-rhamnolipid (RL-F2). The results showed that RL-F1 and RL-F2 had no inhibitory effect on EE2 biodegradation. The effect of RL-F1 on enhanced biodegradation of EE2 was larger than that of RL-F2. In 6 mM and 10 mM of the rhamnolipidic functioned systems, EE2 biotransformation rates affected by RL-F1 were respectively 7.6 and 8.2 times of those of RL-F2 corresponding systems. The accumulation and change rule of the intermediate metabolites M.1 with the polarity greater than EE2 and M.3 with the polarity weaker then EE2 in the biodegradation process were analyzed. In the RL-F2 systems, the time for the accumulative amounts of M.1 and M.3 up to the peak was correspondingly lagged and their output was relatively lower. The testing result of rhamnolipids in the sample showed that RL-F1 was more biodegradable than RL-F2. The bioavailability of di-rhamnolipidic micelle solubilized with EE2 was inferior to that of mono-rhamnolipid, making the organic uptake and metabolism process of microorganisms slow down.
Sorption and Desorption of 17Α-Ethinylestradiol Onto Sediments Affected by Rhamnolipidic Biosurfactants.

Abstract:Many studies have addressed the desorption and mobilization performances of sorbed contaminants affected by different rhamnolipidic biosurfactants. Study results have been mixed and complicated. Rhamnolipids are always microbial produced with variable homologues. In this study, two representative rhamnolipidic fractions (i.e., RL-F1 and RL-F2, which are mono- and di-rhamnolipids, respectively) were investigated and compared to determine their influence on 17α-ethynylestradiol (EE2) distribution within sediment-water sorption and desorption systems. In general, the coexistence of RL-F1 and EE2 enhanced EE2 sorption in a wider monorhamnolipidic dosage range when freshly treated sorbate was used. The sorbed EE2 concentration decreased as the RL-F1 dosage increased in the aged sorbate desorption systems. However, RL-F2 facilitated EE2 mobilization in both sorption and desorption processes. Experimental data were estimated using a conceptual model that considered the sorbed rhamnolipids and aqueous micelles for organic partitioning. The model results indicated that the rhamnolipid type is an important factor influencing organic distribution, in addition to sorbate aging process and sediment characteristics. The use of a rhamnolipidic mixture containing both mono- and di-rhamnosyl components may not achieve the desired effect when the biosurfactant-enhanced mobilization or immobilization approach is selected. These results are significant for selecting and applying rhamnolipids to remediate contaminants.

Abstract:Environmental context The steroid hormone 17α-ethinylestradiol (EE2) has been established as a highly estrogenic substance, which potentially causes serious harm to environmental health. Rhamnolipids are a widely researched glycolipid used for the degradation of organic pollutants. Therefore, this study focused on the change of biodegradation of EE2 affected by rhamnolipids in sediment/water systems, showing increased and more rapid degradation. Abstract Rhamnolipids can enhance the remediation of hydrophobic organic pollutants in the environment. However, the rhamnolipid-associated biodegradation of hormones has rarely been investigated. In this study, aerobic biodegradation shake-flask experiments were conducted to investigate biosurfactant-associated biodegradation of 17α-ethinylestradiol (EE2) in sediment/water systems and to assess how the biodegradation rate is influenced by rhamnolipids produced by Pseudomonas aeruginosa MIG-N146. Results showed that EE2 biodegradability is significantly increased with increasing rhamnolipid concentration. An improved pseudo-first-order kinetic equation was established to simulate enhanced EE2 biodegradation at varying rhamnolipid concentrations. The biodegradation rate (k) initially increased marginally, and then increased rapidly with rhamnolipid concentrations exceeding the effective critical micelle concentration. The degree of enhancement of organic biodegradation was mainly affected by organic mass transfer, owing to rhamnolipidic micellar solubilisation, and by rhamnolipids acting as a primary substrate to stimulate the microbial consortium. Analysis results through various techniques indicated the formation of three main types of metabolic intermediates, with diverse polarity and biodegradability characteristics, in the process of EE2 biodegradation. Thus, it was concluded that the presence of rhamnolipids did not negatively affect the processes of EE2 biotransformation by indigenous microorganisms in the original sediment/water systems. This study presents an effective potential application of rhamnolipidic surfactants for enhancement of EE2 biodegradation in sediment/water systems.

Abstract:A cationic hydrophobically modified ethylene oxide polymer (HM-EO) was used to form aqueous two-phase system with only one phase-forming composition. It was found that the phase separation could be achieved under room temperature. The phase behavior and charge properties of HM-EO aqueous two-phase systems were studied systematically. The amphiphilic property of HM-EO molecule resulted in the micellization and aggregation of micelles in water. The micellar aggregates formed in the bottom phase could be charged, and the net-charge could be adjusted by the pH and conductivity of bulk solution and adding sodium dodecyl sulfate (SDS) The increase of pH would improve the phase separation of HM-EO aqueous two-phase system. The effect of salts on the phase behavior was observed. The influence of cations varied as K+ > Na+ and that of anions as SO42- > F- > Cl- > Br- > I-. In addition,the interaction between HM-EO polymer and anion surfactant SDS was studied.The results indicated that the addition of SDS caused the formation of mixed micellar aggregates of SDS and HM-EO which changed significantly the charge property of HM-EO aqueous two-phase systems.
Enzymatic Resolution of (R, S)-2-Octanol by Ultrastable-Y Molecular Sieve Immobilized-Lipase in Microaqueous Media

Abstract:The resolution of (R, S)-2-octanol was performed in microaqueous media by P. expansum PED-03 lipase (PEL) immobilized onto Ultrastable-Y modified molecular sieve. It was found that conversion(c), enantiomeric excess (e. e. ), and enantioselectivity (E) of the reaction catalyzed by PEL immobilized onto Ultrastable-Y modified molecular sieve were much higher, compared with free PEL and PEL immobilized by other carriers. Media type and water content played an important role in the resolution of (R,S)-2-octanol catalyzed by Ultrastable-Y modified molecular sieve immobilized-PEL, and the reaction eonversion(c) reached 97.68% of the theoretical value and the enantiomeric excess(e. e. ) reached 98.75% in n-hexane with 0.8% water at '' memorial '' pH=9.5 and 50 degrees C for 24 h. The modified Ultrastable-Y molecular sieve immobilized-PEL was a good biocatalyst with a fine enantio selectivity and stability, presenting a good promise in the enzymatic resolution of(R,S)-2-octanol.
Partitioning of Proteins Using A Hydrophobically Modified Ethylene Oxide/Sds Aqueous Two-Phase System

Abstract:A novel aqueous two-phase system containing hydrophobically modified ethylene oxide (HM-EO) and sodium dodecyl sulphate (SDS) was developed to enhance the selectivity of protein partitioning in two phases. Phase diagrams of HM-EO/H2O and HM-EO/SDS/H2O were measured, and the mechanism of interaction between HM-EO polymer and the anionic surfactant sodium dodecyl sulphate (SDS) was also discussed. It was found that the improvement of selectivity of protein partitioning was related to the increase of electrostatic potential difference between the two phases because of the charged network formed by mixed micelles of HM-EO and SDS in the bottom phase. With bovine serum albumin (BSA) and lysozyme as model proteins, some factors, such as pH, SDS concentration, conductivity and temperature of the system, were investigated for the influences of protein partition in HM-EO/SDS/H2O systems. The results showed that the addition of SDS not only changed the phase behaviour, but also played an important role in protein partitioning.

Abstract:The resolution of (R,S)-2-octanol was performed in microaqueous media by the lipase from Penicillium expansum PED-03 lipase (PEL) immobilized on modified ultrastable-Y (USY) molecular sieve. It was found that the conversion, enantiomeric excess (ee), and enantioselectivity (E) of the resolution catalyzed by PEL immobilized on modified USY molecular sieve were much higher than that of the reaction catalyzed by free PEL and PEL immobilized on other supports. Media type, “memorial” pH value and water content were of great importance in the resolution of (R,S)-2-octanol by immobilized PEL, and the conversion of the reaction catalyzed by PEL immobilized on modified USY molecular sieve reached 48.84% with excellent enantioselectivity (average E of eight batches>460) in n-hexane with 0.8% (v/v) water at “memorial” pH 9.5, 50°C for 24h, which showed a good application potential in the production of optically pure (R,S)-2-octanol.
Effects of rhamnolipids on the bioavailability of 17α-ethinylestradiol in sediment

Abstract:Liquid chromatography/mass spectrometry（HPLC-ESI-MS） was used to analyze the crude extracts of fermentation liquor produced by Pseudomonas aeruginosa mutant strain MIG-N146.Fifteen types of rhamnolipid homologues were identified,and the critical micelle concentration（CMC） of rhamnolipids（RL） was 0.125 mmol · L-1 with average minimal surface tension of water being 29.2 mN · m-1,indicating a good surface activity of RL.The effects of different concentrations of rhamnolipids on the bioavailability of 17α-ethinylestradiol（EE2） in sediment were tested by shaking flask experiment.The results indicated that the biodegradation rate constant k of EE2 increased with increasing dosages of rhamnolipids,and its bioavailability was thus enhanced.EE2 biodegradation was slightly improved at RL dosages below 2 mmol · L-1,but the biodegradation rate constant k was higher than 1.346 d-1 at RL dosages above 6 mmol · L-1,which was more than 2.9 times of that without RL.Almost 90% of EE2 were removed in 2 d by adding 10 mmol · L-1 RL.The results of HPLC-PDA displayed that RL was easily biodegraded by microorganism,and thus would not residue in the environment for a long time.Two intermediates of EE2 were also detected during its biodegradation,the dosages of which were affected by the presence of RL,but the metabolic pathway of EE2 was not affected by RL.

Extraction Separation of Rhamnolipids by N-Hexane Via Forming Reverse Micelles

Abstract:Rhamnolipids (RL) have been regarded to be insoluble in n-hexane. Unexpectedly, we have noticed that RL could be extracted together with vegetable oil by n-hexane at analyzing oil content of fermentation broth. This paradoxic phenomenon was assumed to be due to the formation of reverse micelles. As found in this paper, the micelle size as well as conductivity increased due to water solubilization, illustrating the formation of reverse micelles of RL in n-hexane. In this reverse micellar system, the maximal water solubility was detected to be 6.26 mol H2O/mol RL while the reverse critical micelle concentration of RL was around 3% (w/w). Hence, it seems that the presence of water could increase the dissolution of RL in n-hexane via forming the reverse micelles of RL/n-hexane/water. Lastly, the formation of reverse micelles was further applied for extraction of RL by n-hexane. Using this method, over 99.0% of RL was extracted under the appropriate pH of 4.5. This extraction separation using n-hexane instead of chloroform proposed a much cleaner and safer strategy in RL manufacture. Moreover, the capability of RL in forming reverse micelle system could benefit the future application on improving enzymatic reactions, stabilizing nanoparticles, environmental treatment, and protein folding, etc.
Mechanistic Investigation of Drug Supersaturation in the Presence of Polysorbates as Solubilizing Additives by Solution Nuclear Magnetic Resonance Spectroscopy

Abstract:The introduction of solubilizing additives has historically been an attractive approach to address the ever-growing proportion of poorly water-soluble drug (PWSD) compounds within the modern drug discovery pipeline. Lipid-formulations, and more specifically micelle formulations, have garnered particular interest because of their simplicity, size, scalability, and avoidance of solid-state limitations. Although micelle formulations have been widely utilized, the molecular mechanism of drug solubilization in surfactant micelles is still poorly understood. In this study, a series of modern nuclear magnetic resonance (NMR) methods are utilized to gain a molecular-level understanding of intermolecular interactions and kinetics in a model system. This approach enabled the understanding of how a PWSD, 17β-Estradiol (E2), solubilizes within a nonionic micelle system composed of polysorbate 80 (PS80). Based on one-dimensional (1D) 1H chemical shift differences of E2 in PS80 solutions, as well as intermolecular correlations established from 1D selective nuclear Overhauser effect (NOE) and two-dimensional NOE spectroscopy experiments, E2 was found to accumulate within the palisade layer of PS80 micelles. A potential hydrogen-bonding interaction between a hydroxyl group of E2 and a carbonyl group of PS80 alkane chains may allow for stabilizing E2-PS80 mixed micelles. Diffusion and relaxation NMR analysis and particle size measurements using dynamic light scattering indicate a slight increase in the micellar size with increasing degrees of supersaturation, resulting in slower mobility of the drug molecule. Based on these structural findings, a theoretical orientation model of E2 molecules with PS80 molecules was developed and validated by computational docking simulations.

Abstract:Molecular behavior of rhamnolipid mixed with a biobased zwitterionic surfactant at an n-hexadecane/water interface has been studied, and the effects of a rhamnose moiety and composition are evaluated. Results showed that rhamnolipid abundantly interacts with biobased surfactant EAB by means of hydrophobic interactions between aliphatic tails and electrostatic interactions between headgroups, including the attractive interaction between COO- of rhamnolipids and N+ of biobased surfactants and the repulsive interaction between COO- of both surfactants. Dirhamnolipid has a larger number of bound Na+ and a more stable bound structure of COO- ∼ Na+, which screens the repulsive interaction between two kinds of surfactants and shows a more homogeneous distribution with biobased surfactants. The interfacial tension between n-hexadecane and water has been synergistically reduced by dirhamnolipids mixed with biobased surfactants at a higher molar ratio of biobased surfactants. Monorhamnolipids show a strengthened interaction with N+ of biobased surfactants and a more stable hydrogen bond with water relative to that of dirhamnolipids, and there is no synergistic effect in lowering the interfacial tension for the mixture of monorhamnolipids and biobased surfactants. The present work provides details of the molecular behavior of biosurfactant rhamnolipids mixed with biobased surfactants and obtains the key factor in affecting the interfacial properties of the binary system.

Biorelevant dissolution media: aggregation of amphiphiles and solubility of estradiol

Abstract:Biorelevant dissolution media containing bile salt and lecithin at concentrations appropriate for fed and fasted state are useful when testing oral solid formulations of poorly water-soluble drugs. Dilution of amphiphile solutions affects the aggregation state of the amphiphiles because bile salt is partitioned between the aqueous phase and the aggregates. The aim of the investigation was to study the effect of dilution on the size distribution of aggregates and its effect on the solubilization capacity. Clear buffered solutions of four intestinal amphiphiles (sodium glycocholate, lecithin, monoolein, and oleic acid) and a combination of these were prepared at high bile salt concentration. Micelles in the glycocholate solutions decreased in size when diluted. The addition of insoluble amphiphiles led to bigger micelles with no clear correlation between size of the micelles and amphiphile concentration. Dilution of the two- and four component media caused enlargement of the mixed micelles and formation of vesicles. The solubility of estradiol in the buffer solution was increased with addition of the amphiphiles. A good correlation (R(2) = 0.987) was found between estradiol solubility and mass concentration of the amphiphiles. The results demonstrate that, in the case of estradiol, the concentration of amphiphiles rather than the aggregation state determines the solubilization capacity of the medium.
Sub-CMC solubilization of dodecane by rhamnolipid in saturated porous media

Abstract:Experiments were conducted with a two-dimensional flow cell to examine the effect of monorhamnolipid surfactant at sub-CMC concentrations on solubilization of dodecane in porous media under dynamic flow conditions. Quartz sand was used as the porous medium and artificial groundwater was used as the background solution. The effectiveness of the monorhamnolipid was compared to that of SDBS, Triton X-100, and ethanol. The results demonstrated the enhancement of dodecane solubility by monorhamnolipid surfactant at concentrations lower than CMC. The concentrations (50-210 μM) are sufficiently low that they do not cause mobilization of the dodecane. Retention of rhamnolipid in the porous medium and detection of nano-size aggregates in the effluent show that the solubilization is based on a sub-CMC aggregate-formation mechanism, which is significantly stronger than the solubilization caused by the co-solvent effect. The rhamnolipid biosurfactant is more efficient for the solubilization compared to the synthetic surfactants. These results indicate a strategy of employing low concentrations of rhamnolipid for surfactant-enhanced aquifer remediation (SEAR), which may overcome the drawbacks of using surfactants at hyper-CMC concentrations.
Insight into the aggregation and phase behavior for aqueous solution of ethylene glycol monobutyl ether with self-assembly induced optical and electrical properties

Abstract:Ethylene glycol monobutyl ether (EGMBE) is a polar small molecule with amphiphilic characteristics, which can form various aggregates in aqueous solution. The solubility of EGMBE decreases as the temperature increasing with a lower critical solution temperature (LCST). The influence of temperature and salinity on aggregation behavior and solubility of EGMBE were thoroughly investigated in this paper. The phase transition temperatures of EGMBE aqueous solutions with different concentrations were determined and compared with literature. The completed phase diagram of the ternary system (water + NaCl + EGMBE) was constructed. The measured liquid-liquid equilibrium (LLE) data can be well fitted to the commonly used empirical equations Othmer–Tobias, Bancroft and Setschenow. The dynamic light scattering (DLS) measurement shows that the particle size of EGMBE aggregates increases with increasing concentration, reaching its maximum at EGMBE mass fraction of about 30 %. Increasing temperature and inorganic salt concentration can both promote the increase of particle size, especially when approaching the phase separation point, the particle size increases sharply, and the corresponding viscosity of EGMBE solution abnormally increases with increasing temperature. The Tyndall effect of EGMBE solution was also observed to be most significant at mass fraction around 30 %. It is interesting to note that the conductivity of EGMBE solution increases with increasing concentration, reaching its maximum value when the EGMBE mass fraction is about 30 %, though EGMBE is not conductive. The investigation of pyrene fluorescence probes and surface/interfacial tension also demonstrated the aggregation and surface adsorption of EGMBE.

Unraveling the Differential Aggregation of Anionic and Nonionic Monorhamnolipids at Air-Water and Oil-Water Interfaces: A Classical Molecular Dynamics Simulation Study

Abstract:The molecular structure of a surfactant molecule is known to have a great effect on the interfacial properties. We employ molecular dynamics simulations for a detailed atomistic study of monolayers of the nonionic and anionic form of the most common congener of monorhamnolipids, α-rhamnopyranosyl-β-hydroxydecanoyl-β-hydroxydecanoate (( R, R)-Rha-C10-C10), at the air-water and oil-water interfaces. An atomistic-level understanding of monolayer aggregation is necessary to explain a recent experimental observation indicating that nonionic and anionic Rha-C10-C10 show surprisingly different surface area per molecule at the critical micelle concentration. Surface-pressure analysis, interface formation energy calculations, and mass density profiles of the monolayers at the air-water interface show similar properties between nonionic and anionic Rha-C10-C10 aggregation. It is found that there is a significant difference in the headgroup conformations of Rha-C10-C10 in the nonionic and anionic monolayers. Hydrogen bonding interactions between the Rha-C10-C10 molecules in the monolayers is also significantly different between nonionic and anionic forms. Representative snapshots of the simulated system at different surface concentrations show the segregation of molecular aggregates from the interface into the bulk water in the anionic Rha-C10-C10 monolayer at higher concentrations, whereas in the nonionic Rha-C10-C10 monolayer, the molecules are still located at the interface. The present work provides insight into the different aggregation properties of nonionic and anionic Rha-C10-C10 at the air-water interface. Further analyses were carried out to understand the aggregation behavior of nonionic and anionic Rha-C10-C10 at the oil-water interface. It is observed that the presence of oil molecules does not significantly influence the aggregation properties of Rha-C10-C10 as compared to those of the air-water interface.

Abstract:Mono-rhamnolipid（RL-F1） and di-rhamnolipid（RL-F2） were separated and purified through column chromatography of rhamnolipid crude extracts（RL-CE） produced by Pseudomonas aeruginosa mutant MIG-N146.The surface and micellization properties of three rhamnolipidic fractions were tested and their solubilizations of polar hydrophobic triclosan were investigated by solution equilibrium experiments.Results showed that the critical micelle concentration（CMC） of RL-CE,RL-F1 and RL-F2 were 60.31 mg · L^-1,43.82 mg · L^-1 and 115.00 mg · L^-1,while the surface tension（γCMC） were 29.1 mN · m-1,31.6 mN · m-1 and 28.8 mN · m-1,respectively.RL-F2,with the structure of two hydrophilic groups,had the lowest saturated surface excess and the largest mean surface area per molecule.The solution of RL-F2 included several complexes of molecular polymers,micelles,vesicles and multi-vesicles,with larger aggregates showing higher abundance.Among the three rhamnolipidic fractions,RL-F1 had the smallest average micelle grainsize and lower abundance.Grainsize of the complexes in RL-CE displayed a bi-peak dispersion trend,which mainly contributed to large micelles.When the concentration of rhamnolipid was above the CMC,aqueous apparent solubility of triclosan was significantly enhanced by micelle solubilization.The molar solubilization ratio（MSR） of RL-CE,RL-F1 and RL-F2 to triclosan could be determined as 0.2858,0.2548 and 0.3505,respectively,indicating that the solubility of triclosan was effectively enhanced.The larger interior volume of micelles,the stronger solubilization of triclosan by rhamnolipid displayed.The order of solubilization of triclosan was RL-F2 RL-CE RL-F1.

Abstract:The present experimental study was to characterize the adsorption behavior of 17 α-ethyl estradiol (EE2) onto marine sediment in both the single and binary solute systems. Stepwise spiking was innovatively performed to better understand the competition effects. Adsorption of EE2 on the marine sediment can be well fitted by the Freundlich model with an affinity coefficient (KF) varying from 15.8 to 39.8L/kg. It was significantly influenced by SOM and the particle properties. Co-presence of BPA brought about a significant competition effect on the adsorption of EE2, leading to a reduced EE2 adsorption. The competitive effect imposed by EE2 to BPA, however, was even more serious owing probably to the large molecular structure and high hydrophobicity of EE2. The sediment sample with the highest SOM and SSA presented a mild competition effect, while the sediment with the lowest SOM and largest particle size exhibited the most serious competition effect.
Solubilization of a novel antitumor drug ribociclib in water and ten different organic solvents at different temperatures

Abstract:Objective: This study reports new solubility and physicochemical data for ribociclib (RCB) in water and ten organic solvents including "methanol (MeOH), ethanol (EtOH), isopropyl alcohol (IPA), n-butanol (n-BuOH), propylene glycol (PG), polyethylene glycol-400 (PEG-400), acetone, ethyl acetate (EA), Transcutol-HP (THP), and dimethyl sulfoxide (DMSO)" at 293.2-313.2 K and 101.1 kPa. Significance: The obtained data are useful for the industrial applications of RCB. Methods: The solubility of RCB was measured and regressed using "van't Hoff, Buchowski-Ksiazczak λh, the modified Apelblat, and Jouyban models." Results: The overall deviations of <4.0% were recorded for all four models. The maximum mole fraction solubility of RCB was 2.66 × 10-2 in PEG-400 at 313.2 K, however, the lowest one was in the water. The RCB solubility increased with temperature and the order followed in the water and ten different organic solvents was PEG-400 (2.66 × 10-2) > THP (1.00 × 10-2) > PG (5.39 × 10-3) > DMSO (5.00 × 10-3) > n-BuOH (3.23 × 10-3) > acetone (3.11 × 10-3) > IPA (1.58 × 10-3) > EA (1.41 × 10-3) > EtOH (1.37 × 10-3) > MeOH (8.10 × 10-4) > water (2.38 × 10-5) at 313.2 K. The maximum solute-solvent interactions were found in RCB-PEG-400 in comparison with other combination of RCB and solvents. "Apparent thermodynamic analysis" indicated an "endothermic and entropy-driven dissolution" of RCB in water and ten organic solvents. Conclusions: Based on all these data and observations, PEG-400 can be used as the best co-solvent for RCB solubilization.
Polymer Effects on the Equimolar Mixed Cationic–anionic Surfactants

Abstract:Interactions between poly(ethylene oxide) (PEO), and equimolar mixed sodium decylsulfonate and decyltriethylammonium bromide were studied using surface tension, fluorescence, dynamic light scattering and freeze-fracture transmission electron microscopy techniques. Below the cmc of the equimolar mixed cationic–anionic surfactants, no obvious interaction was observed under the experimental conditions. However, PEO had dramatic effects on the morphology of the aggregates of the equimolar mixed cationic–anionic surfactants. Spontaneous, stable and narrow distributed vesicles could be prepared by mixing PEO and equimolar mixed cationic–anionic surfactants. Furthermore, the size of the vesicles could be controlled by turning the total surfactant concentration, PEO concentration and PEO molecular weight. We found the effect of PEO on the vesicles could be explained by the Asakura–Oosawa model.
