Abstract:Visible light-driven nano/micromotors are promising candidates for biomedical and environmental applications. This study demonstrates blue light-driven Ag/AgCl-based spherical Janus micromotors, which couple plasmonic light absorption with the photochemical decomposition of AgCl. These micromotors reveal high motility in pure water, i.e., mean squared displacements (MSD) reaching 800 µm2 within 8 s, which is 100× higher compared to previous visible light-driven Janus micromotors and 7× higher than reported ultraviolet (UV) light-driven AgCl micromotors. In addition to providing design rules to realize efficient Janus micromotors, the complex dynamics revealed by individual and assemblies of Janus motors is investigated experimentally and in simulations. The effect of suppressed rotational diffusion is focused on, compared to UV light-driven AgCl micromotors, as a reason for this remarkable increase of the MSD. Moreover, this study demonstrates the potential of using visible light-driven plasmonic Ag/AgCl-based Janus micromotors in human saliva, phosphate-buffered saline solution, the most common isotonic buffer that mimics the environment of human body fluids, and Rhodamine B solution, which is a typical polluted dye for demonstrations of photocatalytic environmental remediation. This new knowledge is useful for designing visible light driven nano/micromotors based on the surface plasmon resonance effect and their applications in assays relevant for biomedical and ecological sciences.

Abstract:Possessing both unique asymmetric structures and remote-controlled active movement, light-responsive Janus micro-/nanomotors offer the possibility of breaking through the limitations of traditional biomedicine, and have fascinated and inspired researchers. Despite many obstacles toward the clinical application, impressive progress of light-responsive Janus micro-/nanomotors for bioanalytical applications has been made over the past decades. In this review, we first briefly introduced several main light-driven Janus micro-/nanomotors, then focused on their typical bioanalytical applications such as biosensing, bioimaging, and theranostic. In the end, we summarized the remaining challenges of light-responsive Janus micro-/nanomotors in the practical application and also proposed potential solutions in the future.Graphical abstract

Abstract:A near-infrared light-driven Janus nanomotor is constructed by collagenase-coated gold nanorods and chitosan-functionalized mesoporous organosilica nanoparticles with Mn 2+ as the bridging ion. The nanomotors with excellent motility and collagenase activity can potently penetrate into tumors to sufficiently activate innate immune responses, significantly enhancing anti-tumor immune efficacy in vivo .

Abstract:A versatile and high‐yield fabrication method is developed for Janus micromotors featuring a spherical tip configuration, achieved through the localized photocuring of SU‐8 on a TiO2 microsphere array. These motors exhibit various functionalities by loading dyes onto the spheres or modifying the composition of tips. Moreover, the efficacy of this method in the production of multifunctional double‐tipped motors is demonstrated. Synthetic Janus micro/nanomotors can efficiently convert ambient energy into asymmetrical self‐propulsive force, overcoming random thermal fluctuations and enabling autonomous migration. Further modifications to the motors can equip them with different functional modules to meet different needs. However, developing a versatile and high‐yield fabrication method for multifunctional Janus micromotors remains challenging. In this study, a modular fabrication approach for micromotors with a particle‐tip structure based on the self‐focusing lithography induced by an array of TiO2 microspheres is presented. By adjusting the tip composition or loading, precise programming of motor functionality is achieved, allowing for various capabilities such as photoredox reaction‐induced propulsion, fluorescent imaging, electric and magnetic navigation. Furthermore, the flexibility of this fabrication method by selectively loading materials onto two tips is demonstrated to achieve multifunctionality within a micromotor unit. This study proposes a straightforward and versatile approach for modular functional micromotors.

Controlled Molecular Assembly Toward Self-propelled Micro-/Nanomotors

Abstract:Micro-/nanomotors (MNMs) are able to propel themselves in fluids through converting different energies from environment into kinetic energy. Recently, layer-by-layer (LbL) assembly, a versatile assembly approach, has been employed to access MNMs with advantages such as regulated motion, stimuli-response properties, and multifunctionality. In this chapter, we review the recent progress on controlled fabrication, motion control, and biomedical applications of MNMs based on controlled molecular assembly. Through integrating diverse functional building blocks such as nanoparticles, enzymes, and metal shells, MNMs with various structures (e.g., hollow capsules and nanotubes) have been prepared, and the control over the on/off state of the MNM motion has been realized. In addition, we also discuss a special type of MNMs which is derived from the combination of as-assembled biological aggregates and artificial nanostructures. These MNMs can be driven by bubble recoil, irradiation by near-infrared light, and ultrasonic fields. We have also demonstrated the potential applications of these assembled MNMs in biomedical fields such as targeted drug delivery, photothermal therapy, and detoxification.
Self-propelled Janus mesoporous silica nanomotors with sub-100 nm diameters for drug encapsulation and delivery

Abstract:The synthesis of an innovative self-propelled Janus nanomotor with a diameter of about 75 nm that can be used as a drug carrier is described. The Janus nanomotor is based on mesoporous silica nanoparticles (MSNs) with chromium/platinum metallic caps and propelled by decomposing hydrogen peroxide to generate oxygen as a driving force with speeds up to 20.2 μm s(-1) (about 267 body lengths per second). The diffusion coefficient (D) of nanomotors with different H2 O2 concentrations is calculated by tracking the movement of individual particles recorded by means of a self-assembled fluorescence microscope and is significantly larger than free Brownian motion. The traction of a single Janus MSN nanomotor is estimated to be about 13.47×10(-15) N. Finally, intracellular localization and drug release in vitro shows that the amount of Janus MSN nanomotors entering the cells is more than MSNs with same culture time and particle concentrations, meanwhile anticancer drug doxorubicin hydrochloride loaded in Janus MSNs can be slowly released by biodegradation of lipid bilayers in cells.

Abstract:Biological motors are marvels of nature that inspire creation of their synthetic counterparts with comparable nanoscale dimensions, high efficiency and diverse functions. Molecular motors have been synthesized, but obtaining nanomotors through self-assembly remains challenging. Here we describe a self-assembled colloidal motor with a repetitive light-driven rotation of transparent micro-particles immersed in a liquid crystal and powered by a continuous exposure to unstructured ~1 nW light. A monolayer of azobenzene molecules defines how the liquid crystal's optical axis mechanically couples to the particle's surface, as well as how they jointly rotate as the light's polarization changes. The rotating particle twists the liquid crystal, which changes polarization of traversing light. The resulting feedback mechanism yields a continuous opto-mechanical cycle and drives the unidirectional particle spinning, with handedness and frequency robustly controlled by polarization and intensity of light. Our findings may lead to opto-mechanical devices and colloidal machines compatible with liquid crystal display technology.

Abstract:Janus nano/micromotors have been developed into various sizes, shapes, and functions for a blaze of applications especially in biomedical and environmental fields. Here, a fabrication method of Janus micromotors is reported by capping hydrogel microspheres with functional nanoparticles (NPs). Microspheres are prepared in droplet microfluidics relying on hydrogel polymerization to obtain spheres with diameters from 20 to 500 mu m. By solidifying a hydrogel layer onto microspheres, functional NPs of MnO2 (catalyst of H2O2), TiO2 (photocatalyst), and Fe3O4 (magnetic guidance) are adhered onto microspheres resulting in Janus micromotors revealing different functionalities. Dynamics of Janus micromotors (diameter around 250 mu m) are explored by analyzing their trajectories in terms of mean squared displacement when immersed in H2O2 solutions of different concentrations, illuminated by light and guided in an external magnetic field. TiO2 Janus micromotors perform well for water purification tasks as is exemplarily demonstrated with a degradation of Methylene Blue dye in water. The proposed fabrication method is versatile and enables to achieve adjustable coverage of a microsphere with NPs as well as to realize multifunctional Janus micromotors by adhering different NPs (e.g., MnO2 and Fe3O4) on a sphere. This method provides an attractive way to fabricate multifunctional Janus micromotors in a cost-effective manner for environmental applications.
Remotely Controlling Drug Release by Light-Responsive Cholesteric Liquid Crystal Microcapsules Triggered by Molecular Motors

Abstract:Stimuli-responsive smart nanocarriers are an emerging class of materials applicable in fields including drug delivery and tissue engineering. Instead of constructing responsive polymer shells to control the release and delivery of drugs, in this work, we put forward a novel strategy to endow the internal drugs with light responsivity. The microcapsule consisted of molecular motor (MM)-doped cholesteric liquid crystals (CLCs) and drugs. The drug in gelatin-gum arabic microcapsules can protect the carried drugs for a long time with a low release speed totally resulting from drug diffusion. Under UV light, the MM isomerizes and the chirality changes, inducing the alteration of the superstructure of the CLCs. In this process, the cooperative molecular disturbance accelerates the diffusion of the drugs from the microcapsule core to the outside. As a result, thanks to the cooperative effect of liquid crystalline mesogens, molecular-scale geometric changes of motors could be amplified to the microscale disturbance of the self-organized superstructure of the CLCs, resulting in the acceleration of the drug release. This method is hoped to provide opportunities in the design and fabrication of novel functional drug delivery systems.

Abstract:Phycocyanin, a macromolecular protein known for its robust fluorescence, proves to be highly suitable for verifying the successful deposition of imprinted layers. In this study, an acid-propelled magnetic micromotor was successfully fabricated by utilizing surface imprinting and self-propelled nanomotor technology to achieve selective loading and capture of targets such as phycocyanin for future applications in environmental monitoring and precision drug delivery in vivo. This micromotor features a distinct recognition layer achieved through a template electrodeposition method. The outermost imprint layer of the micromotor was meticulously crafted using poly(3,4-ethylenedioxythiophene)/poly(sodium-4-styrenesulfonate) in the presence of a template, while the Pt layer serves as the supportive foundation, the Ni layer acts as the magnetic guidance component, and the innermost layer consists of metal Zn. In acidic environments, the Zn reacts to generate bubbles, which propels the micromotor's motion. The micromotor was comprehensively characterized using techniques such as scanning electron microscopy. Findings highlight the exceptional self-propulsion of the Zn-based micromotor, which is a fusion of molecular imprinting and micromotor technologies. This innovative design achieves an impressive maximum velocity of approximately 100 μm/s, as well as commendable magnetic steering performance. Furthermore, the micromotor demonstrates the ability to imprint target protein through the imprint layer, enabling selective recognition and capture for transport of specific phycocyanin. In vitro cytotoxicity tests have also demonstrated that the micromotors are non-toxic to cells. This breakthrough concept offers a novel avenue for realizing targeted capture and transport of specific nutrients within the human gastric environment.

Abstract:Metal-phenolic particles (MPPs) have received increasing attention due to their excellent physicochemical properties (e.g., high porosity, high stability, versatile coordination chemistry) for molecular adsorption. Herein, we constructed NIR light-propelled Janus () micromotors by depositing Au nanoparticles (NPs) onto the half surface of MPPs, to achieve the active molecular adsorption. Due to excellent photothermal conversion efficiency of Au half nanoshells and its asymmetrical Janus distribution, micromotors can make directional movement under the NIR-light irradiation. The translational diffusion coefficient of is 5.0 μm 2 /s and their velocity can reach 8.5 μm/s under NIR laser irradiation (808 nm, 3 W/cm 2 ). Noteworthily, micromotors show the enhanced removal efficiency of 10.2 % for methylene blue (MB) dye under NIR light irradiation.

Abstract:Micromotors are the backbone of material research as they are small-sized, self-propelled, intelligent systems capable of performing multiple tasks ranging from biomedicine to environmental monitoring. One of the primary obstacles the field faces is the live detection and differentiation of individual units through a complex environment. In this study, we demonstrate a facile approach for designing light-activated dye-tagged micromotors on a large scale. Both rod and sphere shaped micromotors made of titanium dioxide (TiO2)/copper oxide (Cu2O)-silica are designed to self-propel under the illumination of low-intensity UV/Visible light in aqueous peroxide medium. The micromotors were modified with site-specific coatings of different dyes, such as Alq3, Alizarin, Zinc Phthalocyanine, etc. The fabrication of micromotors and coating with dyes was performed using a simple and versatile physical vapor deposition-based glancing angle deposition (GLAD) technique. The fluorescent dyes help to detect the motion and position of micromotors independently. Moreover, they also help to identify the swimming direction as well as differentiate the micromotors in a complex medium consisting of similar configurations of other particles (bacteria and passive fluorescent particles). Light provides full control over the dynamics as well as the fluorescent nature of micromotors. To present the versatility of our design scheme, micromotors of different shapes, materials, and dye coatings are designed and explored for fluorescence-based observations. The simplistic design approach with easy-to-load multiple fluorescent dyes at specific locations is an interesting feature that makes the micromotors suitable candidates for various microfluidic and lab-on-a-chip studies, including biological or fluorescent samples.

X-ray-Powered Micromotors

Abstract:Light-powered wireless manipulation of small objects in fluids has been of interest for biomedical and environmental applications. Although many techniques employing UV-vis-NIR light sources have been devised, new methods that hold greater penetrating power deep into medium are still in demand. Here, we develop a method to exploit X-rays to propel half-metal-coated Janus microparticles in aqueous solution. The Janus particles are simultaneously propelled and visualized in real-time by using a full-field transmission X-ray microscope. Our real-time observation discovers that the propulsive motion follows the bubble growth enhanced by water radiolysis near the particle surface under X-ray irradiation. We also show that the propulsion speed is remotely controlled by varying the radiation dose. We expect this work to open opportunities to employ light-powered micro/nanomotors in opaque environments, potentially by combining with medical imaging or nondestructive testing.

Abstract:Emerging light‐driven micro/nanorobots (LMNRs) showcase profound potential for sophisticated manipulation and various applications. However, the realization of a versatile and straightforward fabrication technique remains a challenging pursuit. This study introduces an innovative bulk heterojunction organic semiconductor solar cell (OSC)‐based spin‐coating approach, aiming to facilitate the arbitrary construction of LMNRs. Leveraging the distinctive properties of a near‐infrared (NIR)‐responsive organic semiconductor heterojunction solution, this technique enables uniform coating across various dimensional structures (0D, 1D, 2D, 3D) to be LMNRs, denoted as "motorization". The film, with a slender profile measuring approximately 140 nm in thickness, effectively preserves the original morphology of objects while imparting actuation capabilities exceeding hundreds of times their own weight. The propelled motion of these microrobots is realized through NIR‐driven photoelectrochemical reaction‐induced self‐diffusiophoresis, showcasing a versatile array of controllable motion profiles. The strategic customization of arbitrary microrobot construction addresses specific applications, ranging from 0D microrobots inducing living crystal formation to intricate, multidimensional structures designed for tasks such as microplastic extraction, cargo delivery, and phototactic precise maneuvers. This study advances user‐friendly and versatile LMNR technologies, unlocking new possibilities for various applications, signaling a transformative era in multifunctional micro/nanorobot technologies. Abstract:Light, as an external stimulus, is capable of driving the motion of micro/nanomotors (MNMs) with the advantages of reversible, wireless and remote manoeuvre on demand with excellent spatial and temporal resolution. This review focuses on the state-of-the-art light-driven MNMs, which are able to move in liquids or on a substrate surface by converting light energy into mechanical work. The general design strategies for constructing asymmetric fields around light-driven MNMs to propel themselves are introduced as well as the photoactive materials for light-driven MNMs, including photocatalytic materials, photothermal materials and photochromic materials. Then, the propulsion mechanisms and motion behaviors of the so far developed light-driven MNMs are illustrated in detail involving light-induced phoretic propulsion, bubble recoil and interfacial tension gradient, followed by recent progress in the light-driven movement of liquid crystalline elastomers based on light-induced deformation. An outlook is further presented on the future development of light-driven MNMs towards overcoming key challenges after summarizing the potential applications in biomedical, environmental and micro/nanoengineering fields.

Abstract:Light-driven micro/nanorobots (LMNRs) are tiny, untethered machines with great potential in fields like precision medicine, nano manufacturing, and various other domains. However, their practicality hinges on developing light-manipulation strategies that combine versatile functionalities, flexible design options, and precise controllability. Our study introduces an innovative approach to construct micro/nanorobots (MNRs) by utilizing micro/nanomotors as fundamental building blocks. Inspired by silicon Metal-Insulator-Semiconductor solar cell principles, we design a new type of optomagnetic hybrid micromotors (OHMs). These OHMs have been skillfully optimized with integrated magnetic constituent, resulting in efficient light propulsion, precise magnetic navigation, and the potential for controlled assembly. One of the key features of the OHMs is their ability to exhibit diverse motion modes influenced by fracture surfaces and interactions with the environment, streamlining cargo conveyance along "micro expressway" - the predesigned microchannels. Further enhancing their versatility, a template-guided assembly strategy facilitates the assembly of these micromotors into functional microrobots, encompassing various configurations such as "V-shaped", "N-shaped", and 3D structured microrobots. The heightened capabilities of these microrobots, underscore the innovative potential inherent in hybrid micromotor design and assembly, which provides a foundational platform for the realization of multi-component microrobots. Our work moves a step toward forthcoming microrobotic entities boasting advanced functionalities.

Abstract:We describe the creation of polymeric microcapsules that can exhibit autonomous motion along defined trajectories. The capsules are made by cross-linking aqueous microdroplets of the biopolymer chitosan using glutaraldehyde. A coflow microfluidic tubing device is used to generate chitosan droplets containing nanoparticles (NPs) with an iron (Fe) core and a platinum (Pt) shell. The droplets are then incubated in a Petri dish with the cross-linking solution, and an external magnet is placed below the Petri dish to pull the NPs together as a collective "patch" on one end of each droplet. This results in cross-linked capsules (∼150 μm in diameter) with an anisotropic (patchy) structure. When these capsules are placed in a solution of H2O2, the Pt shell of the NPs catalyzes the decomposition of H2O2 into O2 gas, which is ejected from the patchy end in the form of bubbles. As a result, the capsules (which are termed micromotors) move in a direction opposite to the bubbles. Furthermore, the micromotors can be steered along specific paths by an external magnet (the magnetic response arises due to the Fe in the core of the NPs). A given micromotor can thus be directed to meet with and adhere to an inert capsule, i.e., a model cargo. Adhesion occurs due to the soft nature of the two structures. Once the cargo is picked up, the micromotor-cargo pair can be moved along a specific path to a destination, whereupon the cargo can be released from the micromotor. We believe these soft micromotors offer significant benefits over their existing hard counterparts because of their biocompatibility, biodegradability, and ability to encapsulate a variety of payloads.
Twin-Engine Janus Supramolecular Nanomotors with Counterbalanced Motion

Abstract:Supramolecular nanomotors were created with two types of propelling forces that were able to counterbalance each other. The particles were based on bowl-shaped polymer vesicles, or stomatocytes, assembled from the amphiphilic block copolymer poly(ethylene glycol)-block-polystyrene. The first method of propulsion was installed by loading the nanocavity of the stomatocytes with the enzyme catalase, which enabled the decomposition of hydrogen peroxide into water and oxygen, leading to a chemically induced motion. The second method of propulsion was attained by applying a hemispherical gold coating on the stomatocytes, on the opposite side of the opening, making the particles susceptible to near-infrared laser light. By exposing these Janus-type twin engine nanomotors to both hydrogen peroxide (H2O2) and near-infrared light, two competing driving forces were synchronously generated, resulting in a counterbalanced, "seesaw effect" motion. By precisely manipulating the incident laser power and concentration of H2O2, the supramolecular nanomotors could be halted in a standby mode. Furthermore, the fact that these Janus stomatocytes were equipped with opposing motile forces also provided a proof of the direction of motion of the enzyme-activated stomatocytes. Finally, the modulation of the "seesaw effect", by tuning the net outcome of the two coexisting driving forces, was used to attain switchable control of the motile behavior of the twin-engine nanomotors. Supramolecular nanomotors that can be steered by two orthogonal propulsion mechanisms hold considerable potential for being used in complex tasks, including active transportation and environmental remediation.
