Isolation and characterization of 12 microsatellite loci in the Chinese Cornsnake, Elaphe bimaculata (Colubridae)

Abstract:The Chinese Cornsnake ( Elaphe bimaculata ) is especially adapted to cool temperatures and higher humidity environments in central China. During the past few decades, E. bimaculata has dramatically declined due to the effect of human activities and over hunting for traditional Chinese medicine. In this study, we isolated and characterized 12 microsatellite loci for this species and evaluated polymorphism using 186 individuals from a population in Zhejiang province. The mean number of alleles per locus ranged from 13 to 20 (mean of 15.25), with expected ( H E ) and observed ( H O ) heterozygosity ranging from 0.761 to 0.880 and from 0.397 to 0.865, respectively. None departures from Hardy–Weinberg equilibrium were observed at all loci. There was no evidence of linkage disequilibrium among pairs of loci. These microsatellite markers will be valuable tools for the long term management and population-level studies (the population structure, genetic variation and evolutionary history) of the species.
