Abstract:Discovering novel Xylaria species. The genus Xylaria comprises a diverse group of fungi with a global distribution and significant ecological importance, known for being a source of bioactive secondary metabolites with antibacterial, antioxidative, anticarcinogenic, and additional properties. In this study, we present a comprehensive taxonomic revision of the species of Xylaria found in some parts of southern China, characterized by an extensive multilocus phylogeny analysis based on internal transcribed spacer (ITS), TUB2 (β‐tubulin), and DNA‐directed RNA polymerase II subunit 2 (rpb2) gene regions. Morphological examination and detailed comparative analyses of the collected specimens were conducted to determine the distinctiveness of each species. The multilocus phylogeny approach allowed us to infer evolutionary relationships and assess species boundaries accurately, leading to the identification of 40 novel Xylaria species hitherto unknown to science. The newly described species are: X. baoshanensis, X. bawanglingensis, X. botryoidalis, X. dadugangensis, X. doupengshanensis, X. fanglanii, X. glaucae, X. guizhouensis, X. japonica, X. jinghongensis, X. jinshanensis, X. kuankuoshuiensis, X. liboensis, X. negundinis, X. orbiculati, X. ovata, X. pseudoanisopleura, X. pseudocubensis, X. pseudobambusicola, X. pseudoglobosa, X. pseudohemisphaerica, X. pseudohypoxylon, X. puerensis, X. qianensis, X. qiongzhouensis, X. rhombostroma, X. serratifoliae, X. shishangensis, X. shuqunii, X. shuangjiangensis, X. sinensis, X. tongrenensis, X. umbellata, X. xishuiensis, X. yaorenshanensis, X. yinggelingensis, X. yumingii, X. yunnanensis, X. zangmui, and X. zonghuangii. The study's findings shed light on the distinctiveness of the newly described species, supported by both morphological distinctions and phylogenetic relationships with their close relatives. This taxonomic revision significantly contributes to our understanding the diversity of Xylaria in China and enriches the knowledge of fungal biodiversity worldwide.

Abstract:This study presents the identification of four novel Xylaria species, discovered in the karst region of China. The discovery was facilitated by a rigorous analysis that encompassed both morpho-anatomical features and multi-locus phylogenetics utilizing sequences from the ITS, rpb2, and TUB2 loci. The newly identified species are designated as Xylariajichuanii sp. nov., X.nanningensis sp. nov., X.orientalis sp. nov., and X.taiyangheensis sp. nov. The distinction of these species from their known counterparts was verified through comparison of morphological features and phylogenetic analysis. The study further provides detailed morphological descriptions, illustrative representations, and a phylogenetic tree, all of which contribute to the taxonomic positioning of these novel species.

Abstract:Xylaria is a widely distributed genus in the Ascomycota phylum that can decompose wood. It is an essential decomposer in ecosystems and a source of bioactive secondary metabolites. Based on morphological characteristics and molecular evidence, this article thoroughly describes two new species discovered on the fallen leaves in Hainan Tropical Rainforest National Park, along with illustrations and comparisons with similar species. Xylaria diaoluoshanensis is characterized by filamentous stromata with long infertile apexes, ascospores sometimes with non-cellular appendages. Xylaria fulvotomentosa differentiates itself from other Xylaria species that grow on fallen leaves by its stroma surface, being yellow tomentose. These two new species of the genus Xylaria were found by phylogenetic analysis using the ITS-β-tubulin-RPB2 sequence dataset. Furthermore, a species first discovered in China, X. petchii, is described. Finally, a search table for 44 species related to fallen leaves and petioles in the world is established.

Abstract:Based on inferences from morphological, chemical and phylogenetic analysis using ITS sequences, a new species of Alectoria is described from the Hengduan Mountains in southwestern China, A. spinosa Li S. Wang & Xin Y. Wang sp. nov, it has dull yellowish fruticose thallus with abundant pseudocyphellae, which is the main character of Alectoria, and differs from all the other Alectoria species by having isidia-like spinules on the soralia. Two species of Alectoria are confirmed in China, their morphological and chemical traits, together with their geographical distributions in China, are discussed, and a key to the species is provided.

Abstract:Morphological and phylogenetic analyses on samples of Xylaria species associated with fallen fruits from China were carried out, and two new species were described, namely X. aleuriticola and X. microcarpa . Xylaria aleuriticola is found on fallen fruits of Aleurites moluccana , and characterized by stromata dichotomously branched several times with long acute sterile apices, fertile parts roughened with perithecia and tomentose, and ellipsoid to fusiform ascospores. Xylaria microcarpa differs in its very small stromata with dark brown tomentum, light brown ascospores with an inconspicuous straight germ slit, and grows on leguminous pods. The differences between the new species and morphologically similar species are discussed. Phylogenetic analyses on ITS - RPB2 - TUB sequences confirmed that the two species are clearly separated from other species of the genus Xylaria . Xylaria liquidambaris is reported as a new record from China. A key to the Xylaria species associated with fallen fruits and seeds reported from China is provided to facilitate future studies of the genus.

Abstract:Until not so long ago, proteases were considered to be mere scissors, with the ECM as their main target, either for degradative or remodeling purposes. However, it is becoming obvious that protease actions are not limited to ECM degradation. Indeed, cells themselves have been shown to be targets of proteases, and PARs have proven to be an important family of cell membrane receptors through which proteolytic enzymes may exert some of their activities [1]. In the mid-1960s, the seminal work of the Laurell and Eriksson group [2] indirectly identified proteases as likely culprits in the pathogenesis of the genetic form of emphysema (e.g., "ZZ emphysema") and suggested that antiproteinases, such as A1Pi (and others), could be of therapeutic use in that pathology. Rodent models of protease instillation indeed subsequently confirmed that when compared with lung homeostasis (Fig. 1A), proteases can induce emphysemateous lesions (Fig. 1B) [3]. These important breakthroughs led to the theory of "protease-antiprotease imbalance," now widely applied to other diseases, including other chronic lung diseases (see below). One of the most important proteases involved in that process was identified as NE, a protease later identified as an antimicrobial mediator at homeostasis [4] and whose function was redefined more recently at the molecular level [5]. Irrespectively, in the late 1980s/early 1990s, pathologists started recognizing that the emphysemateous lung was by no means homogenous and that within the same lung, areas of remodeling/fibrosis could coexist with air space-enlarged areas, one of the cardinal features of emphysema [6, 7]. This suggested that the definition of this condition, put forward by the National Heart, Lung, and Blood Institute in 1984 as "a condition of the lung characterized by abnormal, permanent enlargement of airspaces distal to the terminal bronchiole, accompanied by the destruction of their walls, and without obvious fibrosis," may have been misleading. The paper, by Gregory et al. [8], published in this issue of the Journal of Leukocyte Biology, adds credence to the possibility that NE, traditionally known as an "emphysema/chronic obstructive pulmonary disease inducer" (causing alveolar-wall damage in the peripheral lung and increasing mucus production in the airways), may, under certain conditions, also yield a fibrotic phenotype and may potentially participate in the generation of full-blown IPF. There is certainly no shortage of neutrophils, the main (or only) source of NE in IPF, and previous studies have shown that SLPI, an important mucosal inhibitor of NE, was protective in a hamster model of bleomycin-induced fibrosis [9]. With the use of a similar bleomycin model, Chua et al. [10] showed that NE mice were protected when compared with WT mice, and they found reduced tissue levels of TGF-b, an important repair/fibrogenic cytokine. Gregory et al. [8] convincingly show in the present manuscript that NE can, at nanomolar concentrations, induce fibroblast proliferation, promote wound closure of a fibroblast cell layer in vitro, and induce myofibroblast differentiation (Fig. 1C). In vivo, with the use of a different model than the one mentioned above (i.e., asbestos-mediated instead of bleomycin-mediated fibrosis), these authors also demonstrate lesser fibrotic lesions in NE mice when compared with WT mice. Mechanistically, the authors demonstrate that NE acts partly through cleaving IRS-1, activating the PI3K/AKT pathway, events leading to fibroblast proliferation (Fig. 1D). By contrast, the mechanisms underlying NE-induced myofibroblast differentiation were not fully deciphered in the present study and were shown to be mothers against decapentaplegic homolog 3 dependent (SMAD-3) but TGF-b independent. Abstract:A Gram-stain-negative, coccoid, yellow, non-motile, aerobic bacterium, designated strain S36T, was isolated from soil of the Xixi wetland in Zhejiang province, PR China. Phylogenetic analysis, based on 16S rRNA gene sequences, revealed that strain S36T could represent a novel species of genus Altererythrobacter showing highest similarity to Altererythrobacter atlanticus 26DY36T (96.31 % 16S rRNA gene sequence similarity). The temperature, pH and NaCl concentration ranges for growth were 10-37 °C (optimum 32 °C), pH 5.0-10.0 (optimum pH 7.0) and 0.5-3 % (optimum 1 %, w/v), respectively. The predominant respiratory quinone of strain S36T was Q-10. The major fatty acids were C16 : 0, C17 : 1ω6c, C18 : 1ω7c and summed feature 3 (C16 : 1ω7c and/or iso-C15 : 0 2-OH). The G+C content of the genomic DNA was 62.7 mol%. These data all support the affiliation of strain S36T to the genus Altererythrobacter. The polar lipids profile of strain S36T comprised diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylglycerol, two unidentified phospholipids and two unidentified glycolipids. The results of physiological and biochemical tests allowed differentiation of strain S36T from other members of the genus Altererythrobacter. Therefore, strain S36T represents a novel species of the genus Altererythrobacter, for which the name Altererythrobacter xixiisoli sp. nov. is proposed; the type strain is S36T (=CGMCC 1.12804T=NBRC 110413T).

Abstract:Aster yaoshanensis sp. nov., a new species of the family Asteraceae is here described and illustrated. The species is presently known only from rock crevices of mountain valleys in Dayaoshan National Nature Reserve, Guangxi autonomous region, China. Phylogenetic analyses based on ITS sequences and complete plastome data have shown that this new species is a member of genus Aster with high support. Morphologically, it mostly resembles A. jishouensis, but it can be easily distinguished from the latter by bract indumentum (glabrous except margin ciliate vs. villous especially on veins abaxially, glabrous adaxially) and color (green vs. purple), shorter corolla (3.2-3.5 mm vs. 4.5-5.3 mm), bract stalk (obvious, ca.1.2 mm vs. sessile), and different distribution (Guangxi vs. Hunan). The detailed description, distribution map, and photos are provided. This study further elucidates the species identification, phylogeny and characteristic evolution of Aster.
Morpho-Molecular Characterization Reveals a New Genus, Three Novel Species and Two New Host Records in Xylariomycetidae

Abstract:Xylariomycetidae comprises extremely diverse taxa that are widespread on decaying wood worldwide. An investigation of the diversity of microfungi on oil tree plantations in Sichuan Province was conducted during 2020–2021. Twelve saprobic taxa representing five species were identified as members of Amphisphaeriales and Xylariales through morphological comparisons. Phylogenetic analyses of combined ITS, LSU, rpb2, tub2 and tef1 sequence data indicated a distinct clade formed by three strains within Xylariomycetidae, unrelated to any currently recognized families. Thus, a novel anthostomella-like genus, Bicellulospora, is proposed and treated as Xylariales genera incertae sedis. Bicellulospora is characterized by dark brown to black, immersed, subglobose ascomata with a clypeus, cylindrical asci, and hyaline to yellowish brown, inequilaterally ellipsoidal ascospores with a large upper cell and a dwarf lower cell. Two new species of Amphisphaeria, namely A. oleae and A. verniciae, are introduced based on multi-gene phylogenetic analyses (ITS, LSU, rpb2 and tub2) coupled with morphological characteristics. Amphisphaeria micheliae and Endocalyx ptychospermatis are reported as new host records.

Abstract:A novel aerobic Gram-negative strain, designated as YB25 T , was isolated from an activated sludge sample collected from a seafood processing plant in Zhoushan, Zhejiang Province, China, and characterized by using a polyphasic taxonomic approach in this study. Strain YB25 T was motile by gliding, and short-rod-shaped. The isolate grew at 4–37 °C (optimum 28 °C), pH 6.0–9.0 (optimum pH 7.0) and 0.0–10.0% NaCl (optimum 2.0%, w/v). Phylogenetic analysis based on 16S rRNA gene indicated that strain YB25 T belonged to the genus Gramella, and showed the highest sequence similarity of 97.59% to Gramella lutea YJ019 T . The DNA G + C content was 39.5%. In silico DNA-DNA hybridization (DDH) and average nucleotide identity (ANI) values between strain YB25 T with most closely strains were below the threshold, which is considered to the phylogenetic definition of a novel species. Chemotaxonomic analysis indicated that the only respiratory quinone was menaquinone-6 and the major fatty acids were iso-C 15:0 , anteiso-C 15:0 , iso-C 17:0 3-OH, and summed feature 9 (iso-C 17:1 ω9c and C 16:0 10 methyl). The polar lipid profile was composed of phosphatidylethanolamine, an unidentified phospholipid, two unidentified amino lipids, three unidentified glycolipids, and four unidentified lipids. Compared with the reference strains, strain YB25 T contained higher abundance of genes for carbohydrates metabolism,nitrogen metabolism, sulfur metabolism and respiration based on its genomic metabolic pathways and had been found a certain potential in the degradation of pectin. On the basis of the taxonomic evidence, strain YB25 T represents a novel species of the genus Gramella , for which the name Gramella crocea sp. nov. is proposed. The type strain is YB25 T (= KCTC 82680 T = MCCC 1K05761 T ).

Abstract:Lyophyllum lixivium sp. nov., collected from southwestern China, is described, illustrated, and discussed based on morphological characteristics and molecular data (ITS and nrLSU). Lyophyllum lixivium has umbonate to slightly depressed pileus, a cylindrical stipe that slightly enlarges towards the base, fuzz at the base, basidiospores mostly broadly ellipsoid, rarely fusiform lacrymoid or amygdaliform with an acute apex in side view, abundant clamp connections at the pileus surface and hymenophoral trama. Morphologically and phylogenetically similar species were compared with L. lixivium in this study.

Abstract:Patients with X-linked Ig deficiency with normal or elevated IgM (HIGMX-1) fail to switch from IgM/IgD to other Ig isotypes. Interaction between the B cell antigen CD40 and the CD40 ligand expressed on activated T cells is critical for T cell driven isotype switching. We have reported that T lymphocytes from three unrelated male patients with HIGMX-1 failed to express CD40 ligand on their surface, but the mRNA for CD40 ligand was of an apparently normal size and level. Analysis of CD40 ligand cDNA from two of the patients revealed deletions that alter the reading frame. Patient 1 displayed two mutations: a C-->A transversion at nucleotide 590 and the deletion of an adjacent C nucleotide. The second patient had a 58 bp deletion from nucleotides 289-346. Furthermore, neither patient expressed a protein product detectable by the CD40L mAb, 5c8.

Abstract:A new corticioid fungus from Guizhou Province in southwest China, Asterostroma rhizomorpharum, is described and illustrated based on morphological and molecular evidence. It is characterized by producing annual, resupinate basidiomata with long rhizomorphs, a dimitic hyphal structure with simple septa on generative hyphae, asterosetae frequently present in subiculum, asterohyphidia present in hymenium, ellipsoid to subglobose, and echinulate and amyloid basidiospores (5.5–6.8 × 4.6–5.9 μm). The phylogenetic analysis based on ITS+nLSU rDNA sequences shows that the new species belongs to the section Asterostroma.

Abstract:Molecular phylogeny and morphological characteristics of collections of diatrypaceous fungi from Guizhou Province, China, lead to the establishment of a new genus, Stromatolinea , and the identification of four new species and two new combinations. The taxa were found growing on the dead culms of Phyllostachys bamboo. The new genus is distinguished by its well-developed, discrete linear stromata with yellow interior tissue and allantoid subhyaline ascospores. The newly described species are Stromatolinea grisea , S. guizhouensis , S. hydei , and S. xishuiensis . Additionally, two new combinations, Stromatolinea linearis and S. phaselina , are proposed based on comparative analysis and morphology. Phylogenetic analyses were conducted using ITS and TUB2 sequences. The study includes comprehensive morphological descriptions, illustrations, and a phylogenetic tree depicting the placement of the new taxa.

Abstract:Xylariomycetidae (Ascomycota) is a highly diversified group with variable stromatic characters. Our research focused on inconspicuous stromatic xylarialean taxa from China, Italy, Russia, Thailand and the United Kingdom. Detailed morphological descriptions, illustrations and combined ITS-LSU-rpb2-tub2-tef1 phylogenies revealed 39 taxa from our collections belonging to Amphisphaeriales and Xylariales. A new family (Appendicosporaceae), five new genera (Magnostiolata, Melanostictus, Neoamphisphaeria, Nigropunctata and Paravamsapriya), 27 new species (Acrocordiella photiniicola, Allocryptovalsa sichuanensis, Amphisphaeria parvispora, Anthostomella lamiacearum, Apiospora guiyangensis, A. sichuanensis, Biscogniauxia magna, Eutypa camelliae, Helicogermslita clypeata, Hypocopra zeae, Magnostiolata mucida, Melanostictus longiostiolatus, M. thailandicus, Nemania longipedicellata, N. delonicis, N. paraphysata, N. thailandensis, Neoamphisphaeria hyalinospora, Neoanthostomella bambusicola, Nigropunctata bambusicola, N. nigrocircularis, N. thailandica, Occultitheca rosae, Paravamsapriya ostiolata, Peroneutypa leucaenae, Seiridium italicum and Vamsapriya mucosa) and seven new host/geographical records are introduced and reported. Divergence time estimates indicate that Delonicicolales diverged from Amphisphaeriales + Xylariales at 161 (123–197) MYA. Amphisphaeriales and Xylariales diverged 154 (117–190) MYA with a crown age of 127 (92–165) MYA and 147 (111–184) MYA, respectively. Appendicosporaceae (Amphisphaeriales) has a stem age of 89 (65–117) MYA. Ancestral character state reconstruction indicates that astromatic, clypeate ascomata with aseptate, hyaline ascospores that lack germ slits may probably be ancestral Xylariomycetidae having plant-fungal endophytic associations. The Amphisphaeriales remained mostly astromatic with common septate, hyaline ascospores. Stromatic variations may have developed mostly during the Cretaceous period. Brown ascospores are common in Xylariales, but they first appeared in Amphisphaeriaceae, Melogrammataceae and Sporocadaceae during the early Cretaceous. The ascospore germ slits appeared only in Xylariales during the Cretaceous after the divergence of Lopadostomataceae. Hyaline, filiform and apiospores may have appeared as separate lineages, providing the basis for Xylariaceae, which may have diverged independently. The future classification of polyphyletic xylarialean taxa will not be based on stromatic variations, but the type of ring, the colour of the ascospores, and the presence or absence or the type of germ slit.

Abstract:Asterostroma roseoalbum sp. nov. was found in Yunnan Province, China, is described here as a new species based on its morphology and phylogeny. Asterostroma roseoalbum is characterised by its felted-membranous to pellicular, fimbriate basidiomata with a smooth hymenophore and a dimitic hyphal system bearing generative hyphae with simple-septate and echinulate, subglobose to globose basidiospores measuring as 8–9.5 × 7–9 μm. Phylogenetic analyses of the new species were based on the internal transcribed spacer (ITS) and large subunit (nrLSU) of ribosomal DNA (rDNA) sequences. The phylogenetic analysis indicated that the new species belongs to the genus Asterostroma , and is closely related to two species A. macrosporum and A. muscicola . A full description, illustrations and phylogenetic analysis results of the new species are provided. Furthermore, a key to the genus Asterostroma in China is provided.

Novel discoveries of Xylariomycetidae (Ascomycota) taxa from peat swamp forests and other terrestrial habitats in Thailand

Abstract:In a comprehensive survey of fungi conducted in the northern (Chiang Rai Province) and southern (Narathiwat Province) regions of Thailand, several xylariales-like specimens were discovered. Through the integration of molecular phylogeny and morphological analyses, one previously undocumented taxon, Oxydothisnarathiwatensis sp. nov., was identified, along with Xylariabawanglingensis and Hypoxylonhypomiltum as new host and geographical records from Afzeliaxylocarpa, and Dalbergiacana, respectively. In addition, Annulohypoxylonthailandicum was identified as a new host record from Swieteniamacrophylla in Thailand. The morphological characters, including ascomata, asci, and ascospores, were compared with known Oxydothis, Xylaria, Hypoxylon, and Annulohypoxylon species. Multi-locus phylogenetic analyses based on ITS, LSU, and SSU (for Oxydothidaceae), ITS, rpb2, tub2, and act (for Xylariaceae), and ITS, LSU, rpb2, and tub2 (for Hypoxylaceae) gene regions were carried out to refine the taxonomic classifications of these specimens further. This research contributes to understanding fungal diversity in these ecologically significant regions, highlighting insights into the relationships among xylariales-like species.

Abstract:Cryptothecia and Myriostigma are important elements of crustose lichen communities in tropical to subtropical forests, but little research has been done on these two genera in China. Morphological and molecular phylogenetic approaches to investigate species diversity of Cryptothecia and Myriostigma from Southern China were carried out in this study. We find five species of Cryptothecia and Myriostigma in our study, including three new species (M. flavescens, M. hainana and M. laxipunctata) and two new records (C. bartlettii and C. inexspectata). In addition, a phylogenetic tree based on mtSSU, RPB2 and nLSU illustrates the placement of the five species and supports the delimitation of the three new taxa. Detailed descriptions of morphological, ecological and chemical characteristics and illustrations are provided for every species. A key to all known Chinese Cryptothecia and Myriostigma species is also provided.

Abstract:A new species Xya leshanensis sp. nov. of the genus Xya Latreille, 1809 from Sichuan, China is described in this paper. It is similar to Xya nitobei (Shiraki, 1913), but differs from latter by apical part of antennae joint almost as same as basal part in width; width of pronotum 1.25 times its length; hind wing long, extending over the end of abdomen distinctly; hind femur with four small yellowish-white spots on upper side; epiproct with a pair of large spines laterally directed on posterior angle, apical half narrow distinctly. Type specimens are deposited in the College of Life Science, Leshan Normal University, Leshan 614004, China.
