Abstract:In this paper, a simple and highly efficient ruthenium-catalyzed direct C3 alkylation of indoles with various α,β-unsaturated ketones without chelation assistance has been developed. This novel C-H activation methodology exhibits a broad substrate scope such as different substituted indoles, pyrroles, and other azoles. Further synthetic applications of the alkylation products can lead to more attractive 3,4-fused tricyclic indoles.

Abstract:Ru(II)-catalyzed redox-neutral [3+2] annulation reactions of N-ethoxycarbamoyl indoles and internal alkynes via C-H bond activation are reported. This method features a broad internal alkyne scope, including various aryl/alkyl-, alkyl/alkyl-, and diaryl-substituted alkynes, good to excellent regioselectivity, diverse functional group tolerance, and mild reaction conditions. The N-ethoxycarbamoyl directing group, temperature, CsOAc, and ruthenium catalyst proved to be crucial for conversion and high regioselectivity. Additionally, preliminary mechanistic experiments were conducted, and a possible mechanism was proposed.

Abstract:The synthesis of substituted indoles has received great attention in the field of organic synthesis methodology. C–H activation makes it possible to obtain a variety of designed indole derivatives in mild conditions. Ruthenium catalyst, as one of the most significant transition-metal catalysts, has been contributing in the synthesis of indole scaffolds through C–H activation and C–H activation on indoles. Herein, we attempt to present an overview about the construction strategies of indole scaffold and site-specific modifications for indole scaffold via ruthenium-catalyzed C–H activations in recent years.

Abstract:We report herein a new strategy of the Rh(III)-catalyzed C-H activation/cyclization of indoles and pyrroles, for the divergent synthesis of privileged heterocycles. A simple derivation of indoles and pyrroles to N-carboxamides with oxidative bidentate directing group could enable rhodacycle formation and late-stage redox-neutral cyclization with alkynes, alkenes and diazo compounds, for access to five- and six-membered fused heterocycles, such as pyrimido[1,6-a]indol-1(2H)-one, 3,4-dihydropyrimido[1,6-a]indol-1(2H)-one, and 1H-imidazo[1,5-a]indol-3(2H)-ones. Kinetic isotope effect study was conducted, and a plausible mechanism was proposed. Furthermore, this protocol was applied to concise synthesis of 5-HT3 receptor antagonist in gram-scale.

Abstract:A versatile rhodium-catalyzed/copper-mediated C-H/C-C activation and cascade annulation reaction was described to form 2-arylindole derivatives. Highly selective C-C bond cleavage of γ-substituted tert-propargyl alcohols occurred, together with pyridine-directed ortho C(sp2)-H bond activation, affording a series of 2-arylindoles with yields up to 90%. Subsequent derivations were smoothly conducted to access polyfunctionalized 2-arylindoles, illustrating the potential applications of this method.

Abstract:A new ruthenium(II)-catalyzed tandem C–H activation/cyclization/hydrolysis cascade process of 2H-imidazoles and alkynes for facile and regioselective access to α-ketone-isoquinolines has been successfully developed. 2H-Imidazole as the novel traceless directing group has been well applied in this paper. The protocol features mild reaction conditions and easily accessible starting materials, and α-ketone-isoquinolines could be applied to further construct more complex heterocyclic compounds.

Abstract:The Rh(III)-catalyzed regioselective C2-H bond carbenoid insertion/cyclization of N-amidoindoles with α-acyl diazo compounds has been developed. This method provides a novel approach to 2H-pyrimido[1,6-a]indol-1-ones with a broad range of functional group tolerance. Abstract:An efficient method for the synthesis of indole derivatives from readily available pyrimidyl-substituted anilines and diazo compounds via rhodium(III)-catalyzed CH bond activation has been developed. This cyclization reaction displays excellent functional group compatibility and regioselectivity, which overcomes some drawbacks of the classical indole synthetic methods and provides a facile approach for the construction of multi-substituted indole derivatives. The redox-neutral intermolecular annulation procedure comprises tandem CH bond activation, cyclization, and condensation steps, releasing water and nitrogen as by-products.

Abstract:Reported herein is the rhodium(III)-catalyzed C-H activation of anilines bearing an N-isoquinolyl directing group for oxidative [3+2] annulation with four classes of internal alkynes, leading to atroposelective indole synthesis via dynamic kinetic annulation with C-N reductive elimination constituting the stereo-determining step. This reaction proceeds under mild conditions with high regio- and enantioselectivity and functional group compatibility.
