Abstract:OBJECTIVE:To investigate the association between genetic polymorphisms of inflammatory factors and susceptibility to coronary heart disease(CHD) in southern Chinese Han population.METHODS:Using matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry (MALDI-TOF-MS) method, the genotypes of five inflammatory factors (BRCA1-associated protein, a disintegrin and metalloproteinase 8, inter-alpha-trypsin inhibitor H3, interleukin-15, cyclooxygenase-2) were anaylzed in 283 CHD patients diagnosed by angiography and 176 controls.RESULTS:In these inflammatory factors, the 270T/C and 90A/G polymorphisms of the BRAP gene showed a significant association with CHD. The allele and genotype frequencies of BRAP gene were consistent with those predicted by Hardy-Weinberg equilibrium (chi-square=0.878, P> 0.05; chi-square=0.776, P> 0.05, respectively). The frequencecies of 270C and 90G alleles in CHD patients was significantly higher than those of the control group (29.51% vs. 21.31%, P=0.006; 30.04% vs. 21.31%, P=0.004, respectively). Compared with 270TT and 90AA, 270CC and 90GG genotypes had a significantly increased CHD risk by Logistic regression analysis (OR=4.51, 95%CI: 1.41-14.45, P=0.011; OR=5.09, 95%CI: 1.60-16.26, P=0.006, respectively). This association was still signifcant after adjustment for the sex, age, smoke, hypertension, diabetes, plasma total cholesterol and low density lipoprotein levels. No evidence of association was found for other single nucleotide polymorphisms.CONCLUSION:The 270T/C and 90A/G polymorphisms in the BRAP gene may contribute to an increased risk of CHD among southern Chinese Han population.

Abstract:Recent studies have shown that interleukin (IL)-16 is an immunomodulatory cytokine, which plays an important role in some inflammatory and autoimmune diseases. We aimed to investigate the association between the IL-16 gene polymorphisms and presence of coronary artery disease (CAD) where inflammatory processes are involved.This case-control study enrolled 651 CAD patients confirmed by coronary angiography and 428 controls. Four tag single nucleotide polymorphisms (rs8034928-rs3848180-rs4577037-rs1131445) within the IL-16 gene and the related haplotypes were genotyped by using polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP). Plasma IL-16 concentration was measured by enzyme-linked immunosorbent assay.In patients with CAD, the plasma concentration of IL-16 was significantly higher than in controls (97.6±10.7, 66.5±9.6, respectively P<0.001). By using multivariate logistic regression analysis, the allele and genotype frequencies of rs8034928 were different between CAD and control groups (P<0.001). However, the associations of the polymorphisms rs3848180, rs4577037, and rs1131445 with CAD were not observed. The haplotypes TTTT and TGGT significantly increased risk to CAD (OR, 95% CI: 1.43, 1.26-1.63; 1.47, 1.16-1.85; respectively), whereas the haplotypes CTTT and TTGT referred to protection of CAD (OR, 95% CI: 0.45, 0.33-0.62; 0.50, 0.33-0.76; respectively).The study indicated that the IL-16 rs8034928 T/C polymorphism and haplotypes were associated with the presence of CAD in Chinese Han population. The IL-16 gene polymorphisms may be a useful predictor to the susceptibility of CAD.

Abstract:Our aim was to investigate the association between IL-16 gene polymorphisms (rs4778889 C/T and rs11556218 G/T) and coronary artery disease (CAD).The initial cohort consisted of 300 CAD patients and 397 controls from the Chinese Han population. Genotyping was performed by using polymerase chain reaction (PCR)-restriction fragment length polymorphism (RFLP). The positive association between polymorphism and CAD was replicated in another independent cohort, which included 424 CAD cases and 332 controls.In the initial study, the allele and genotype frequencies of rs4778889 were not different between in CAD and controls (P>0.05). However, The G allele frequency of rs11556218 was significantly higher in the CAD cases than in the controls (CAD, 46.8% vs. controls, 22.8%, P<0.001). The risk of CAD was significantly higher in the G allele carriers than in the non-carriers (P<0.001, adjusted odds ratio = 7.27; 95% confidence interval, 4.13-12.8). In the replication cohort, G carriers of rs11556218 also had a higher risk of CAD (P = 0.005, adjusted OR = 2.33; 95% confidence interval, 1.45-3.74).Our study suggested that IL-16 rs11556218 G/T polymorphism is significantly associated with the risk of CAD in the Chinese Han population.

Abstract:Growing evidence has shown that inflammation plays crucial roles in the development of coronary artery disease (CAD). Interleukin-16 (IL-16), a multifunctional cytokine, is involved in a series of inflammatory disorders. The aim of this study was to investigate the association between IL-16 polymorphisms and risk of CAD. We analyzed two polymorphisms of IL-16 (rs4778889 T/C and rs11556218 T/G) in 157 patients with CAD and 202 healthy controls by using polymerase chain reaction-restriction fragment length polymorphism and DNA sequencing methods. The TG/GG genotypes of rs11556218 T/G were associated with a significantly increased risk of CAD as compared with the TT genotype (odds ratio = 1.77; 95% confidence intervals, 1.16-2.71). This finding indicates that IL-16 may be used as a genetic marker for CAD susceptibility.
Interleukin-16 Gene Polymorphism Is Associated With Acute Coronary Syndrome In The Chinese Han Population

Abstract:Acute coronary syndrome (ACS) is clinical syndromes caused by coronary atherosclerosis plaque rupture or attack, which can lead to complete or incomplete pathological basis of occlusive thrombosis. Evidences have indicated the importance of gene polymorphism in the pathological progression of coronary heart disease risk. The purpose of this study was to investigate the associations between polymorphisms in interleukin-16 (IL-16) single nucleotide polymorphisms (SNP) and the risk of ACS in Chinese Han population. A total of 238 patients and 178 healthy individuals were recruited in this retrospective cohort. Genotyping of IL-16 rs8034928, rs3848180, rs1131445, rs4778889 and rs11556218 SNPs was performed by PCR-restriction fragment length polymorphisms. Results showed that the G allele in rs3848180 and C allele in rs1131445 were higher frequencies in ACS group than healthy group. We found that G/G genotype in rs1131445 showed a significant higher risk of CAS (OR=1.28, 95% CI=0.94-2.55). Variant of rs3848180 presented a significant increased risk of CAS in dominant (OR=1.44, 95% CI=1.04 -2.68). The allele C demonstrated a significant risk of CAS (OR=1.26, 95% CI=1.12-2.08) and T/T in rs3848180 were found to be associated with increasing risk of CAS (OR=1.66, 95% CI=1.07-4.25). In conclusion, these results showed that rs1131445 and rs3848180, SNPs of IL-16 are associated with CAS risk in Chinese Han population, which provide the possibility of statistical power calculation to predict the susceptibility of ACS.

Abstract:OBJECTIVE: To investigate the proposed associations between interleukin-18 (IL-18) polymorphisms and coronary heart disease (CHD) susceptibility among the human population. STUDY DESIGN: A comprehensive search was conducted to retrieve published studies relevant to IL-18 polymorphisms and CHD susceptibility. Odds ratios (ORs) of CHD with 95% confidence interval (95% CI) were used to compare effects of IL-18 mutations on CHD susceptibility in 5 different inheritance models. Statistical software was utilized for all data analyses. RESULTS: A sum of 9 published studies containing 5,394 CHD patients and 6,008 healthy controls in total were enrolled in our study. Both rs187238 G>C and rs549908 A>C polymorphisms were correlated with an elevated risk of CHD in the allele model (rs187238 G>C: OR=1.23, 95% CI 1.10 similar to 1.37, p<0.001; rs549908 A>C: OR=1.31, 95% CI 1.04 similar to 1.64, p=0.022), and the rs1946519 C>T polymorphism was associated with an increased risk of CHD in both the allele model and dominant model (both p<0.05). Ethnicity subgroup analysis clarified that rs187238 G>C, rs1946519 C>T, and rs549908 A>C were associated with CHD risk in Caucasians (all p<0.05), while only rs549908 A>C was associated with CHD risk in Asians (OR=1.62, 95% CI 1.07 similar to 2.46, p=0.024). CONCLUSION: Results of our metaanalysis suggested that 3 IL-18 polymorphisms, rs1946519 C>T, rs187238 G>C, and rs549908 A>C, were related to increased risk of CHD, and these polymorphisms may contribute to our understanding of mechanisms underlying CHD.

Abstract:Objective:To observe the frequencies of the polymorphism of IL-1αC-889T in Chinese and the relationship between the polymorphism of IL-1αC-889T and coronary artery disease(CHD). Method:IL-1αC-889T polymorphism was performed in 282 patients who were performed selective coronary angography by using PCR-RFLP methods. The patients were classfied as 126 CHD patients group and 156 controls group. The relationship between the polymorphism of IL-1αC-889T and the plasma levels of CRP were also investigated. Result:The frequencies of genotype in Chinese was CC ~78.7 %,CT ~20.6 %, TT ~0.7 %. The frequencies of allele was C 502 (~89.0 %),T 62 (~11.0 %).The genotype distribution was in accordance with Hardy- Weinberg equilibrilum. A significant increase was found for the IL-1a T allele in CHD patients compared with controls(~28.6 % vs~15.4 % , χ2=~7.24 ,P~0.01 ). There was no relationship between the polymorphism of IL-1αC-889T and myocardial infarction、the number of stenotic vessels. There was no relationship between the polymorphism and the plasma levels of CRP. Hypertension, hypercholesterolemia and type 2 diabetes were not the confounding factors. Conclusion: The polymorphism of IL-1αC-889T is present in Chinese and the T allele is a major risk factor for CHD patients.

Abstract:Interleukin (IL)-16, a multifunctional cytokine, plays a fundamental role in inflammatory diseases, as well as in the development and progression of tumors. Genetic variation in the DNA sequence of the IL-16 gene may lead to altered cytokine production and/or activity, and this variation may modulate an individual's susceptibility to both colorectal cancer (CRC) and gastric cancer (GC). To test this hypothesis, we investigated the association of IL-16 gene polymorphisms with serum levels of IL-16 and the risk of CRC and GC in a Chinese population. We analyzed single-nucleotide polymorphisms of the IL-16 gene in 596 cancer patients (376 patients with CRC and 220 patients with GC), and also in 480 age- and sex-matched controls using polymerase chain reaction-restriction fragment length polymorphism and DNA sequencing methods. Serum IL-16 levels were measured by enzyme-linked immunosorbent assay. The rs11556218 T/G polymorphism of the IL-16 gene was significantly associated with the susceptibility to CRC and GC patients. Both male and female patients carrying the G allele had a significantly higher risk for developing CRC and GC compared with individuals carrying the T allele. Alternatively, women carrying the T allele (rs4072111 C/T) showed a decreased risk for CRC and GC compared with individuals carrying the C allele. In patients with CRC or GC, IL-16 serum levels were significantly higher than those in the healthy controls, although no significant association between IL-16 polymorphisms and serum levels of IL-16 was observed. Our data indicate that IL-16 polymorphisms may contribute to CRC and GC susceptibility.

Abstract:Background: Acute coronary syndrome (ACS) is one of the most common forms of heart disease. Recent studies have shown that interleukin (IL)-8 plays a key role in the development of atherosclerotic plaques, but the relationship between the common genetic variants of IL-8 and ACS has not been extensively studied.Methods: This case-control study in the Chinese Han population included 675 patients with ACS and 636 age- and sex-matched controls. We investigated IL-8 polymorphisms and their association with susceptibility to ACS. The investigation was replicated in the second study comprising 360 cases and 360 control subjects. The plasma concentration of IL-8 was measured by enzyme-linked immunosorbent assay.Results: IL-8 -251 A/T polymorphism was associated with increased susceptibility to ACS (P = 0.004; odds ratio = 1.30; 95% confidence interval: 1.12-1.53). The second study yielded similar results. An increased IL-8 level was found in the plasma of acute myocardial infarction patients, suggesting that IL-8 -251 A/T may affect the expression of IL-8.Conclusion: IL-8 -251 A/T polymorphism is associated with ACS risk in the Chinese Han population and the A allele of IL-8 -251 A/T may be an independent predictive factor for ACS. Abstract:Objective: To explore the association between the polymorphlsm of IL-23R gene and Crhon's disease(CD).Methods: Total 98 CD patients,and 90 healthy individuals were studied.The alleles and gene type of rs11465779 were analyzed by restriction frag-ment length polymorphism(PCR-RFLP).Results: The frequency of alleles and gene type were similar between CD and normal group(P0.05).Further analysis also did not display any relationship between the polymorphism and the onsetage or sex of CD(P0.05).Conclusion: The rs11465779 of IL-23R gene is not associated with susceptibility to Crhon's disease in Han population of South China.

Abstract:Objective: To investigate the relationship between the Interleukin-6(IL-6) gene promoter region-572C / G single nu-cleotide polymorphism and coronary artery disease in Han population of North China.Methods: Polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) method was used to detect the IL-6 genotype in 434 cases of coronary artery disease(CAD) patients and 417 non-coronary artery disease(control group) and the relationship between-572C/G single nucleotide polymorphism and coronary heart disease was discussed.Results: IL-6 gene-572C / G polymorphism of the CC,CG and GG genotypes in the CAD group was 59.68%,37.09%,3.23%,while C and G allele frequencies were 78.23%,21.77%,respectively.Genotype in healthy adults who were 67.87%,30.22%,1.92%,C and G allele frequencies were 82.97%,17.03%.The distribution of IL-6 gene-572C/G polymorphism in the two groups was significant difference(p0.05);After adjustment of sex,age,body mass index,hypertension,diabetes,high cholesterol,smoking and other conventional risk factors with Logsitic regression analysis,IL-6 gene-572C / G single nucleotide polymorphism was an independent coronary artery disease risk factors(P 0.05);With the relative risk of allele frequency analysis,the risk of cornoary artery disease was markedly higher in the G allel carriers than that in the C allele carriers(OR=1.356,95%CI=1.0648～1.7279).Conclusion: IL-6 gene-572C/G single nucleotide polymorphism is correlated with the incidence of cornoary artery disease in Han population of North China and G allele is an important genetic markers with coronary heart disease.

Abstract:Objectives: Some polymorphisms of the interleukin-16 (IL-16) gene are associated with various cancers. To resolve inconsistencies in published data, we performed a meta-analysis of studies of IL-16 polymorphisms and cancer risk. Materials and Methods: Seven eligible studies pooling 1678 cases and 1937 controls were quantitatively analyzed to evaluate three IL-16 polymorphisms (rs4778889, rs4072111, rs11556218) and cancer risk. Hardy-Weinberg equilibrium (HWE) for controls was evaluated by goodness-of-fit chi-squared tests. Odds ratios (ORs) and their 95% confidence intervals (CIs) were calculated for each genetic model and allelic comparison. Data were pooled using fixed- or random-effects models depending on heterogeneity across studies. Results: Our meta-analysis demonstrated that the IL-16 polymorphism rs11556218 was significantly associated with increased susceptibility to cancer in several models, including allelic contrast (OR=1.307; 95% CI, 1.108-1.541), heterozygote contrast (OR=1.650; 95% CI, 1.424-1.911), and dominant model (OR=1.605; 95% CI, 1.391-1.845). The result remained consistent after adjustment for age and gender. No significant association was found between IL-16 polymorphisms rs4778889 rs4072111 and cancer risk. Conclusions: The rs11556218 T/G polymorphism of the IL-16 gene was significantly associated with elevated cancer risk in Asian populations. Our results warrant larger, better-designed studies, including a greater ethnic variety.

Abstract:Interleukin-17A/F (IL-17A/F) might play a role in the pathophysiology of osteoarthritis (OA), but several studies exploring the association between IL-17A/F single nucleotide polymorphisms and OA in different populations present inconsistent results. Thus, this case-control study, involving 410 OA cases and 507 controls, was aimed to investigate such association in a Chinese population. Genotyping was performed using standard polymerase chain reaction and restriction fragment length polymorphism. It was found that AA genotype or A allele carriers of IL-17A gene rs2275913 polymorphism were associated with OA susceptibility. Stratified analyses showed IL-17A rs2275913 polymorphism was evidently associated with a significantly increased risk for OA among males, <60 years old patients, smokers, and drinkers. No significant association was observed between IL-17F gene rs763780 polymorphism and OA risk. In conclusion, IL-17A rs2275913 polymorphism is involved in the development of OA in Chinese Han population. This finding should be confirmed by a larger study from diverse ethnic populations.

We determined the association between SNPs of IL-16 gene (i.e. rs11556218 G/T, rs4778889 C/T, and rs4072111 C/T) and systemic lupus erythematosus (SLE).One hundred thirty-eight SLE patients and 199 controls were genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and DNA sequencing assay.In the SLE group, the individuals carrying the G allele of rs11556218, the C allele of rs4778889 and the T allele of rs4072111 are at a significantly higher risk of SLE as compared with those carrying the T allele of rs11556218, the T allele of rs4778889, and the C allele of rs4072111 respectively (for rs11556218, OR=2.264, 95% CI, 1.64-3.127; for rs4778889, OR=1.927, 95%CI, 1.359-2.731; and for rs4072111, OR=2.417, 95% CI, 1.691-3.455).The IL-16 gene polymorphisms may be associated with the risk of SLE.

Abstract:Objective To investigate the association between -174 G/C -634C/G of interleukin-6 gene polymorphism and coronary heart disease in Chinese population.Methods The genotype and allele frequency of interleukin-6 gene polymorphisms was assayed by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) in 199 patients with evident of CHD and 189 healthy individuals. The relation of gene polymorphisms of interleukin-6 and levels of serum lipids, lipoproteins, and apolipoproteins were also studied.Results The IL-6 -174 G allele was detected in 0.99 of normal controls and 0.99 of patients, while IL-6 -174 C allele was detected in 0.01of normal controls and 0.01 of patients. The IL-6( -634C) allele was detected in 0.82 of normal controls and 0.76 of patients, while IL-6 -634G allele was detected in 0.18 of normal controls and 0.24 of patients. The frequencies of GG genotype and IL-6 -634G allele were higher in coronary heart disease than in controls ( P 0.05 ).Conclusion These results suggest that the interleukin-6 gene -174G/C polymorphism was not related to CHD. But -634C/G polymorphism was associated with CHD in Chinese population. The -634G allele might be a risk factor for CHD.

This fact and plethora of gene polymorphism data prompted us to investigate IL1 gene polymorphisms in breast cancer (BC) patients. Totally, 530 patients with BC and 628 healthy control women were studied. The genetic polymorphisms for IL1 were analyzed by Massarray Sequencing method. Three single nucleotide polymorphisms (SNPs) identified in IL1B, IL1R1 gene are thought to influence breast cancer risk. The results of the association between IL-1B, IL1R1 polymorphisms and breast cancer risk have significant. We found that the variant TT genotype of rs10490571 was associated with a significantly increased breast cancer risk (TT vs. CC: OR = 2.82, 95% CI = 1.12-7.08, P = 0.047 for the codominant model). For rs16944 (AG vs. GG: OR = 0.60, 95% CI = 0.41-0.90, P = 0.034 for the codominant model) and rs1143623 (CG vs. CC: OR = 0.65, 95% CI = 0.45-0.94, P = 0.023 for the codominant model) have significant associations were found in genetic models. In conclusion, the present analysis suggests a correlation of polymorphic markers within the IL-1 gene locus with the risk in developing breast cancer. Taken together with our finding that IL1B, IL1R1 gene three SNP are also associated with the risk for the disease, we suggest that inflammation via innate and adaptive immunity contributes to multifactorial hereditary predisposition to pathogenesis of the breast cancer.
Genetic polymorphisms of interleukin-6 gene and susceptibility to coronary artery disease in Chinese population: Evidence based on 4582 subjects

Abstract:The aim of this study was to explore whether interleukin-6 (IL-6) gene (-174 G/C and -572 C/G) polymorphisms are associated with susceptibility to coronary artery disease (CAD) risk in Chinese population. All the statistical tests were performed using Stata version 11.0. Twelve articles involving 16 studies were included in this meta-analysis, covering a total of 2309 CAD cases and 2273 controls. For IL-6 gene -572 C/G polymorphism, the results showed evidence for significant association between IL-6 gene -572 C/G polymorphism and CAD risk (for G allele vs. C allele: OR=1.48, 95% CI=1.26-1.74, p<0.001; for G/G vs. C/C: OR=2.60, 95% CI=1.54-4.39, p<0.001; for G/G vs. G/C+C/C: OR=2.15, 95% CI=1.35-3.42, p=0.001; for G/G+G/C vs. C/C: OR=1.55, 95% CI=1.29-1.85, p<0.001). However, for IL-6 gene -174 G/C polymorphism, no significant association was found between this variation and CAD risk. In summary, our meta-analysis showed evidence that IL-6 gene -572 C/G polymorphism may be a risk factor for CAD susceptibility. For IL-6 gene -174 G/C polymorphism, no significant association was found between this variation and CAD risk.

Abstract:Objective:To investigate the association of interleukin-18 gene 105 genetic polymorphism in the coding region with susceptibility to coronary heart disease (CHD).Methods:IL-18 gene 105 genotypes in 162 CHD patients and 134 controls were detected using PCR-restriction fragment length polymorphism(RFLP) assay,and the association of genetic polymorphism with susceptibility to CHD was analyzed. Results:The frequencies of A/A,A/C,C/C types of interleukin-18 gene 105 polymorphism in CHD patients and controls were 70.4%,29.0% and 0.6%vs 88.1%,11.9% and 0%. Compared with AA genotype,the relative risk for CHD in people with AC genotype was 3.041(95% confidence interval [CI] 1.631-5.669). The relative risk for CHD in people with C allele was 2.806 higher than people with A allele(95%CI 1.556-5.061)(P0.01). Conclusions:IL-18 gene 105 genetic polymorphism is associated with CHD in Han nationality of Fujian province,people with AC or CC genotypes take higher risk of suffering from CHD.

Abstract:Background: Interleukin-18 (IL-18) has been suggested to play an important role in coronary arterial disease and its sequelae. The aim of the present study was to investigate the association between IL-18 promoter functional polymorphisms (-607C/A and - 137G/C) and acute myocardial infarction (AMI) in northern Chinese Han population.Methods: We performed a case-control study including 234 patients with AMI and 216 age-and sex-matched controls. Genotyping was performed using sequence specific primer-polymerase chain reaction (PCR-SSP).Results: There were significant differences in the genotype and allele distribution of -607C/A polymorphism of the IL-18 gene between cases and controls. Logistic regression analysis with adjustments for other well-established risk factors revealed that the -607C allele carriers had a significantly increased risk of AMI compared with the non-carriers (OR=1.890, 95% CI 1.196-2.985, p=0.006). No relationship between -137G/C polymorphism and AMI was found.Conclusions: This study shows for the first time that the IL-18 gene promoter -607C/A polymorphism may be considered a genetic risk factor for AMI in northern Chinese Han population. Abstract:The association of the Interleukin 6 (IL-6) -572G>C polymorphism and risk of coronary heart disease (CHD) have been implicated in a large number of investigations, but the results remain debatable. This meta-analysis was performed to provide more compelling evidence for the connection between the IL-6 -572G>C polymorphism and CHD risk. Studies eligible for this meta-analysis were identified through electronic search of PubMed, EMBASE, and CNKI. The fixed effects model was performed to summarize an odds ratio (OR) with 95 % confidence interval (CI). The meta-analysis of 3,985 patients and 7,153 controls from 17 studies showed that the CC genotype carriers had 0.84-fold lower risk of developing CHD when compared with the carriers with the GC+GG genotypes (OR(CC vs. GC+GG) = 0.84; 95% CI = 0.75-0.95; P = 0.414; I(2) = 3.5%). The decreased risk of CHD was also found in Asians (OR = 0.87; 95% CI = 0.77-0.98; P = 0.227; I(2) = 22.7%) and Caucasians (OR = 0.60; 95% CI = 0.40-0.92; P = 0.958; I(2) = 0) under the same genetic comparison. The results of our meta-analysis revealed that the IL-6 -572G>C polymorphism may be linked with risk of CHD in a protective model.
