Abstract:Protein kinase C has been shown to play a central role in the cardioprotection of ischemic preconditioning. However, the mechanism underlying PKC-mediated cardioprotection is not completely understood. Given that caveolae are critical for PKC signaling, we sought to determine whether hypoxic preconditioning promotes translocation and association of PKC isoforms with caveolin-3. A cellular model of hypoxic preconditioning from adult rat cardiac myocytes (ARCM) or H9c2 cells was employed to examine PKC isoforms by molecular, biochemical and cellular imaging analysis. Hypoxia was induced by incubating the cells in an airtight chamber in which O2 was replaced by N2 with glucose-free Tyrode's solution. Cells were subjected to hypoxic preconditioning with 10 minutes of hypoxia followed by 30 minutes of reoxygenation. Western blot data indicated that the band intensity for PKCε, PKCδ or PKCα, but not PKCβ and PKCζ was enhanced significantly by hypoxic preconditioning from the caveolin-enriched plasma membrane interactions. Immunoprecipitation experiments from the caveolin-enriched membrane fractions of ARCM showed that the level of PKCε, PKCδ and PKCα in the anti-caveolin-3 immunoprecipitates was also increased by hypoxic preconditioning. Further, our FRET analysis in H9c2 cells suggested that there is a minimum FRET signal for caveolin-3 and PKCε along cell peripherals, but hypoxic preconditioning enhanced the FRET signal, indicating a potential interaction between caveolin-3 and PKCε. And also treatment of the cells with hypoxic preconditioning led to a smaller amount of translocation of PKCε to the mitochondria than that to the membrane. We demonstrate that hypoxic preconditioning promotes rapid association of PKCε, PKCδ and PKCα with the caveolin-enriched plasma membrane microdomain of cardiac myocytes, and PKCε via direct molecular interaction with caveolin-3. This regulatory mechanism may play an important role in cardioprotection.
Involvement of the Mitochondrial Calcium Uniporter in Cardioprotection by Ischemic Preconditioning

Abstract:The objective of the present study was to determine whether the mitochondrial calcium uniporter plays a role in the cardioprotection induced by ischemic preconditioning (IPC). Isolated rat hearts were subjected to 30 min of regional ischemia by ligation of the left anterior descending artery followed by 120 min of reperfusion. IPC was achieved by two 5-min periods of global ischemia separated by 5 min of reperfusion. IPC reduced the infarct size and lactate dehydrogenase release in coronary effluent, which was associated with improved recovery of left ventricular contractility. Treatment with ruthenium red (RR, 5 microM), an inhibitor of the uniporter, or with Ru360 (10 microM), a highly specific uniporter inhibitor, provided cardioprotective effects like those of IPC. The cardioprotection induced by IPC was abolished by spermine (20 microM), an activator of the uniporter. Cyclosporin A (CsA, 0.2 microM), an inhibitor of the mitochondrial permeability transition pore, reversed the effects caused by spermine. In mitochondria isolated from untreated hearts, both Ru360 (10 microM) and RR (1 microM) decreased pore opening, while spermine (20 microM) increased pore opening which was blocked by CsA (0.2 microM). In mitochondria from preconditioned hearts, the opening of the pore was inhibited, but this inhibition did not occur in the mitochondria from hearts treated with IPC plus spermine. These results indicate that the mitochondrial calcium uniporter is involved in the cardioprotection conferred by ischemic preconditioning.

Abstract:Objective To investigate the possible mechanisms of caveolin-3(Cav-3)in protein kinase C(PKC)isoforms mediated cardioprotection of ischemic preconditioning. Methods Hypoxia of adult rat cardiac myocytes(ARCM)were induced in a closed container to establish a cell model of hypoxic preconditioning. The changes of five PKC isoforms,PKCα,PKCβ1,PKCe,PKCδ and PKCζ,before and after ischemic preconditioning were determined by Western blot and fluorescence resonance energy transfer(FRET),based on which the relationship of Cav-3 to the expression,translocation and activation of PKC was evaluated. Results Western blot indicated the selective translocations of PKCe,PKCδ and PKCα,but not PKCβ1 or PKCζ,onto the Cav-riched plasma membrane. The ischemic preconditioning enhanced the FRET signal,promoted the translocation of PKCe to the Cav-rich plasma membrane,and increased the coupling level of Cav-3 to PKCe. Conclusion Ischemic preconditioning might promote the associations of PKCe,PKCδ and PKCα with the Cav-rich plasma membrane microdomain of cardiac myocytes possibly via direct molecular interaction with Cav-3,which plays an important role in cardioprotection.
[Mechanisms of Cardioprotection Induced by Preconditioning after Activation of MITOK(ATP) Channel].

Abstract:AIMTo determine mechanisms of cardioprotection induced by combination angiotensin-converting enzyme inhibitors (ACEI) with subthreshold preconditioning after activation of mitochondrial ATP-sensitive potassium (mitoK(ATP)) channel.METHODSThe Langendorff model of isolated rat heart was used. The time of the onset of uncoupling, the activities of sarcolemmal Na+/K+ -ATPase and Ca2+/Mg2+ -ATPase were measured.RESULTSThe subthreshold preconditioning (2 min of ischemia and 10 min reperfusion) or captopril (an ACEI) alone did not protect hearts against injury of sustained ischemia. However combination captopril with subthreshold preconditioning increased LVDP. Pretreatment hearts with mitoK(ATP) channel inhibitor 5-HD abolished the protection effect. Combination captopril with subthreshold preconditioning delayed the onset of uncoupling, and enhanced the activities of sarcolemmal Na+/K+ ATPase and Ca2+/Mg2+ -ATPase in ischemia/reperfusion hearts. But 5-HD cancelled these cardioprotection effects.CONCLUSIONCombination ACEI with subthreshold preconditioning delays the onset of cellular uncoupling induced by acute ischemia, and promotes the stability of sarcolemmal ion channels, in which activation of the mitoK(ATP) channels may be involved.
Cardioprotection by Mitochondrial KATP Channel in Both Early and Late Preconditioning

Abstract:Activation of the mitochondrial K-ATP channel (mitoK(ATP)) has been shown to confer an early and late phase of cardioprotection against ischemic injury via protein kinase C (PKC) signaling pathways. However, the exact relationship between PKC or its isoforms and mitoK(ATP) channels has not yet been clarified in both early and late preconditioning (PC). The hypothesis of this study is that the translocation of PKC to mitochondria is important for early cardiac protection elicited by mitoK(ATP) channels. PKC was down-regulated by treatment of PMA (4mug/kg) for 24-hours prior to subsequent experiments. Langendorff-perfused rat hearts were subjected to 40-min of ischemia followed by 30-min reperfusion. In late PC, nitric oxide was hypothesized to be trigger for the opening of mitoK(ATP) channels to enable the heart to protect itself against ischemic damage. This was tested in NOS knockout mouse hearts. Effects of PKC down-regulation or blockade of PKC activity by chelerythrine on the activation of mitoK(ATP) channel and other interventions on hemodynamic, biochemical and pathological changes were assessed. Subcellular distribution of PKC isoforms by immunocytochemistry demonstrates that immunostaining of PKC-delta was observed in the mitochondria after diazoxide (DE, 80mumol/L) pretreatment, PKC-epsilon was translocated to intercalated disc and PKC-beta1 was translocated to the nucleus. In DE treated hearts, a significant improvement in cardiac function and attenuation of cell injury was observed. In the PKC down-regulated hearts or hearts pretreated with chelerythrine, the protection was abolished because mitoK(ATP) channels could not be activated by DE. These data suggest that PKC activation is required for the opening of mitoK(ATP) channels in the protection against ischemia and this effect is linked with isoform specific translocation of 6 to mitochondria. NO is an important trigger for the opening of mitoK(ATP) channels after 24 hours of initial preconditioning stimulus. DE was found to be totally ineffective in iNOS knockout mice. DE activated NFkB via PKC signaling pathway and that lead to NOS up-regulation for further activation of the mitoK(ATP) after 24 hours upon ischemic stimulus.

Abstract:The aims of this study were to determine whether preconditioning blocks cardiocyte apoptosis and to determine the role of mitochondrial ATP-sensitive K+ (K-ATP) channels and the protein kinase C epsilon-isoform (PKC-epsilon) in this effect. Ventricular myocytes from 10-day-old chick embryos were used. In the control series, 10 h of simulated ischemia followed by 12 h of reoxygenation resulted in 42 +/- 3% apoptosis (n = 8). These results were consistent with DNA laddering and TdT-mediated dUTP nick-end labeling (TUNEL) assay. Preconditioning, elicited with three cycles of 1 min of ischemia separated by 5 min of reoxygenation before subjection to prolonged simulated ischemia, markedly attenuated the apoptotic process (28 +/- 4%, n = 8). The selective mitochondrial K-ATP channel opener diazoxide (400 mumol/l), given before ischemia, mimicked preconditioning effects to prevent apoptosis (22 +/- 4%, n = 6). Pretreatment with 5-hydroxydecanoate (100 mumol/l), a selective mitochondrial K-ATP channel blocker, abolished preconditioning (42 +/- 2%, n = 6). In addition, the effects of preconditioning and diazoxide were blocked with the specific PKC inhibitors Go-6976 (0.1 mumol/l) or chelerythrine (4 mumol/l), given at simulated ischemia and reoxygenation. Furthermore, preconditioning and diazoxide selectively activated PKC-epsilon in the particulate fraction before simulated ischemia without effect on the total fraction, cytosolic fraction, and PKC delta-isoform. The specific PKC activator phorbol 12-myristate 13-acetate (0.2 mumol/l), added during simulated ischemia and reoxygenation, mimicked preconditioning to block apoptosis. Opening mitochondrial K-ATP channels blocks cardiocyte apoptosis via activating PKC-epsilon in cultured ventricular myocytes. Through this signal transduction, preconditioning blocks apoptosis and preserves cardiac function in ischemia-reperfusion.

PKCε modulates NF-κB and AP-1 via mitogen-activated protein kinases in adult rabbit cardiomyocytes

Abstract:We have previously shown that protein kinase C (PKC)-ε, nuclear factor (NF)-κB, and mitogen-activated protein kinases (MAPKs) are essential signaling elements in ischemic preconditioning. In the present study, we examined whether activation of PKCε affects the activation of NF-κB in cardiac myocytes and whether MAPKs are mediators of this signaling event. Activation of PKCε (+108% above control) in adult rabbit cardiomyocytes to a degree that has been previously shown to protect myocytes against hypoxic injury increased the DNA-binding activity of NF-κB (+164%) and activator protein (AP)-1 (+127%) but not that of Elk-1. Activation of PKCη did not have an effect on these transcription factors. Activation of PKCε also enhanced the phosphorylation activities of the p44/p42 MAPKs and the p54/p46 c-Jun NH2-terminal kinases (JNKs). PKCε-induced activation of NF-κB and AP-1 was completely abolished by inhibition of the p44/p42 MAPK pathway with PD98059 and by inhibition of the p54/p46 JNK pathway with a dominant negative mutant of MAPK kinase-4, indicating that both signaling pathways are necessary. Taken together, these data identify NF-κB and AP-1 as downstream targets of PKCε, thereby establishing a molecular link between activation of PKCε and activation of NF-κB and AP-1 in cardiomyocytes. The results further demonstrate that both the p44/p42 MAPK and the p54/p46 JNK signaling pathways are essential mediators of this event.

Abstract:Activation of protein kinase C (PKC) via adenosine receptors is known to be involved in the cardioprotection of ischemic preconditioning (IPC). Specifically, activation of PKCε is critical for cardioprotection. There is ample evidence that PKCε resides in cardiac mitochondria. However, the signals that promote translocation of PKCε are largely unknown. The present study was designed to determine whether and how adenosine receptor activation induces translocation of PKCε to mitochondria. Freshly isolated adult rat cardiac myocytes and rat heart-derived H9c2 were used in the study. Immunofluorescence imaging of isolated mitochondria showed that PKCε but not PKCδ was localized in mitochondria and this mitochondrial localization of PKCε was significantly increased by adenosine treatment. The adenosine-induced increase in PKCε-positive mitochondria was largely prevented not only by PKC inhibitor chelerythrine, but also by the HSP90 inhibitor geldanamycin and by siRNA targeting HSP90. Immunoblot analysis from percoll-purified mitochondria further demonstrated that adenosine mediated a significant increase in mitochondrial PKCε but not PKCδ. This effect was blocked by inhibiting PKC activity with chelerythrine and bisindolylmaleimide. Furthermore, co-immunoprecipitation data showed that PKCε but not PKCδ was associated with TOM70 and HSP90, and this association was enhanced by adenosine treatment. Moreover, adenosine-induced association of PKCε with TOM70 was reduced by suppressing HSP90 expression with siRNA. In conclusion, we demonstrate that adenosine induces HSP90-dependent translocation of PKCε to mitochondria, possibly through mitochondrial import machinery TOM70. These results point out a novel mechanism in regulating PKC in mitochondria and suggest an important implication in ischemic preconditioning or postconditioning.
Post-conditioning Protects Rat Cardiomyocytes Via PKCε-mediated Calcium-Sensing Receptors

Abstract:Protein kinase C (PKC) plays a role in cardioprotection through reduction of intracellular Ca2+ concentration [Ca2+]i during ischemic preconditioning (IPC). Cardioprotection against ischemic post-conditioning (PC) could be associated with reduced [Ca2+]i through PKC. The calcium-sensing receptor (CaR), G protein-coupled receptor, causes accumulation of inositol phosphate (IP) to increase the release of intracellular Ca2+. However, this phenomenon can be negatively regulated by PKC through phosphorylation of Thr-888 of the CaR. This study tested the hypothesis that the prevention of cardiomyocyte damage by PC is associated with [Ca2+]i reduction through an interaction of PKC with the CaR. Isolated rat hearts were subjected to 40min of ischemia followed by 90min of reperfusion. The hearts were post-conditioned after the 40min of ischemia by three cycles of 30s of reperfusion and 30s of re-ischemia applied before the 90min of reperfusion. Immunolocalization of PKCε in the cell membrane was observed with IPC and PC, and in hearts exposed to GdCl3 during PC. CaR was expressed in cardiac cell membrane and interacted with PKC in IPC, PC, and exposure to GdCl3 during PC groups. On laser confocal microscopy, intracellular Ca2+ was significantly decreased with IPC, PC, and exposure to GdCl3 during PC compared with the I/R and PKC inhibitor groups, and cell structure was better preserved and promoted the recovery of cardiac function after reperfusion in the same groups. These results suggested that PKC is involved in cardioprotection against PC through negative feedback of a CaR-mediated reduction in [Ca2+]i.
PKCepsilon modulates NF-kappaB and AP-1 via mitogen-activated protein kinases in adult rabbit cardiomyocytes.

The Role of Protein Kinase C in Immature Myocardial Ischemic Preconditioning

Abstract:Objective To investigate the mechanism of protein kinase C(PKC) in immature myocardial ischemic preconditioning in order to further its clinical applicability. Methods Langendorff perfusion heart models of 24 rabbits were set up and they were randomly divided into 4 groups: ischemic reperfusion group(I/R group),myocardial ischemic preconditioning group(MIP group),chelerythrine group(CLT group) and protein kinase C group(PKC group).The hemodynamics,biochemistry and myocardial ultrastructure were observed.Results The heart function recovery and myocardial water content in the MIP and the PKC groups were better than those of the I/R and the CLT groups(P<0.01).The adenosine triphosphate(ATP) content,superoxide dismutase activity,mitochondrial Ca2+-ATPase activity and synthesizing ATP activity of mitochondria in the MIP and the PKC groups were significantly higher than those of the I/R and the CLT groups(P<0.01).The dehydrogenase and creatine kinase leakage,malondialdehyde content,myocardial cell Ca2+ content and mitochondrial Ca2+ content in the MIP and the PKC groups were significantly lower than those of the I/R and the CLT groups(P<0.01).The myocardial ultrastructure injuries in the MIP and the PKC groups were less than that of the I/R and the CLT groups.Conclusion Myocardial ischemic preconditioning plays an important role in protecting immature myocardium,which is probably realized by the activation of PKC.
Ischemic Preconditioning Exaggerates Cardiac Damage In Pkc-Delta Null Mice

Abstract:Ischemic preconditioning confers cardiac protection during subsequent ischemia-reperfusion, in which protein kinase C (PKC) is believed to play an essential role, but controversial data exist concerning the PKC-delta isoform. In an accompanying study (26), we described metabolic changes in PKC-delta knockout mice. We now wanted to explore their effect on early preconditioning. Both PKC-delta(-/-) and PKC-delta(+/+) mice underwent three cycles of 5-min left descending artery occlusion/5-min reperfusion, followed by 30-min occlusion and 2-h reperfusion. Unexpectedly, preconditioning exaggerated ischemia-reperfusion injury in PKC-delta(-/-) mice. Whereas ischemic preconditioning increased superoxide anion production in PKC-delta(+/+) hearts, no increase in reactive oxygen species was observed in PKC-delta(-/-) hearts. Proteomic analysis of preconditioned PKC-delta(+/+) hearts revealed profound changes in enzymes related to energy metabolism, e.g., NADH dehydrogenase and ATP synthase, with partial fragmentation of these mitochondrial enzymes and of the E-2 component of the pyruvate dehydrogenase complex. Interestingly, fragmentation of mitochondrial enzymes was not observed in PKC-delta(-/-) hearts. High-resolution NMR analysis of cardiac metabolites demonstrated a similar rise of phosphocreatine in PKC-delta(+/+) and PKC-delta(-/-) hearts, but the preconditioning-induced increase in phosphocholine, alanine, carnitine, and glycine was restricted to PKC-delta(+/+) hearts, whereas lactate concentrations were higher in PKC-delta(-/-) hearts. Taken together, our results suggest that reactive oxygen species generated during ischemic preconditioning might alter mitochondrial metabolism by oxidizing key mitochondrial enzymes and that metabolic adaptation to preconditioning is impaired in PKC-delta(-/-) hearts.
PKC epsilon modulates NF-kappa B and AP-1 via mitogen-activated protein kinases in adult rabbit cardiomyocytes

Abstract:We have previously shown that protein kinase C (PKC)-epsilon, nuclear factor (NF)-kappa B, and mitogen-activated protein kinases (MAPKs) are essential signaling elements in ischemic preconditioning. In the present study, we examined whether activation of PKC epsilon affects the activation of NF-kappa B in cardiac myocytes and whether MAPKs are mediators of this signaling event. Activation of PKC epsilon (+108% above control) in adult rabbit cardiomyocytes to a degree that has been previously shown to protect myocytes against hypoxic injury increased the DNA-binding activity of NF-kappa B (+164%) and activator protein (AP)-1 (+127%) but not that of Elk-1. Activation of PKC eta did not have an effect on these transcription factors. Activation of PKC epsilon also enhanced the phosphorylation activities of the p44/p42 MAPKs and the p54/p46 c-Jun NH2-terminal kinases (JNKs). PKC epsilon-induced activation of NF-kappa B and AP-1 was completely abolished by inhibition of the p44/p42 MAPK pathway with PD98059 and by inhibition of the p54/p46 JNK pathway with a dominant negative mutant of MAPK kinase-4, indicating that both signaling pathways are necessary. Taken together, these data identify NF-kappa B and AP-1 as downstream targets of PKC epsilon, thereby establishing a molecular link between activation of PKC epsilon and activation of NF-kappa B and AP-1 in cardiomyocytes. The results further demonstrate that both the p44/p42 MAPK and the p54/p46 JNK signaling pathways are essential mediators of this event.

Abstract:Several studies have suggested that protein kinase C (PKC) plays a key role in the mechanism of cerebral ischemic/hypoxic preconditioning (I/HPC). However, detailed information regarding PKC isoforms in response to brain ischemia/hypoxia and their potential role in neuroprotection is unclear. Previous studies in our laboratory have demonstrated that the levels in membrane translocation of conventional PKC (cPKC) betaII, gamma, and novel PKCepsilon (nPKC), but not cPKCalpha, betaI, nPKCdelta, eta, mu, theta, and atypical PKC (aPKC) zeta and iota/lambda, were increased significantly in the hippocampus and cortex of intact mice with hypoxic preconditioning. To further detect cPKC and nPKC isoforms activation following prolonged hypoxia in vitro, we tested the membrane translocation (an indicator of PKC activation) of cPKCalpha, betaI, betaII, and gamma, and nPKCdelta, epsilon, eta, mu, and theta in a human neuroblastoma SH-SY5Y cell line following sustained hypoxic exposure (1% O(2)/5% CO(2)/94% N(2)). Using Western blot and immunocytochemistry methods, we found that the levels of cPKCalpha, betaI, betaII, and nPKCepsilon, but not nPKCdelta, eta, mu, and theta, membrane translocation were increased significantly (P < 0.05, n = 8) in a time-dependent manner (from 0.5 to 24 h) following sustained hypoxic exposure. Similarly, the immunostaining experiment also showed a noticeable translocation of cPKCalpha, betaI, betaII, and nPKCepsilon from the cytosol to the perinuclear or membrane-related areas after 6 h posthypoxic exposure. In addition, no cPKCgamma was detected in this cell line under either a normoxic or hypoxic condition. These results suggested that prolonged hypoxia may induce the activation of cPKCalpha, betaI, betaII, and nPKCepsilon by triggering their membrane translocation in SH-SY5Y cells.

Abstract:Recently, accumulated studies have suggested that protein kinases C (PKC) play a central role in the development of ischemic-hypoxic preconditioning (1/HPC) in the brain. However, which types of PKC isoforms might be responsible for neuroprotection is still not clear, especially when the systematic investigation of PKC isoform-specific changes in brain regions was rare in animals with ischemic-hypoxic preconditioning. By using Western blot, we have demonstrated that the levels of cPKC beta II and gamma membrane translocation were increased in the early phase of cerebral hypoxic preconditioning. In this study, we combined the Western blot and immunostaining methods to investigate the effects of repetitive hypoxic exposure (H1 - H4, n = 6 for each group) on membrane translocation and protein expression of several types of PKC isoforms, both in the cortex and hippocampus of mice. We found that the increased membrane translocation of nPKC epsilon (P < 0.05, versus normoxic H0) but not its protein expression levels in both the cortex and hippocampus during development of cerebral HPC in mice. However, there were no significant changes in both membrane translocation and protein expression levels of nPKC delta, theta, eta, mu, and aPKC iota/lambda, zeta in these brain areas after hypoxic preconditioning. Similarly, an extensive subcellular redistribution of cPKC beta II, gamma, and nPKC epsilon was observed by immunostaining in the cortex after three series of hypoxic exposures (H3). These results indicate that activation of cPKC beta II, gamma, and nPKC epsilon; might be involved in the development of cerebral hypoxic preconditioning of mice. (c) 2005 Elsevier B.V. Role of Protein Kinase A in Ischemia Preconditioning for Inhibiting Binding Activation of Nuclear Factor-κB-DNA in Cardiomyocyte

Abstract:Objective:To investigate the role of protein kinase A(PKA) in ischemia preconditioning for inhibiting binding activation of nuclear factor-κB-DNA(NF-κB-DNA) in cardiomyocyte. Methods:Forty SD rats were divided into four groups and each group contained 10 rats.Ischemia/reperfusion(I/R) group, Ischemia preconditioning(IPC) group,Pretreatment with PKA inhibitor H89(H89) group,and NF-κB inhibitor PDTC(PDTC) group.The isolated hearts were Langendorff-perfused by K-H solution for 10 minute as stabilization period in each group.Then, additional treatment was followed in I/R group:perfusion 30 min,rest 1 hour,and re-perfusion 30 min;in IPC group:rest 5 min, reperfusion 5 min,repeated this procedure for 3 times,then,rest 1 hour and reperfusion 30min;in H89 group and PDTC group: rest 5 min,then reperfused by 10μM H89 and 100μM PDTC for 5 min respectively,and repeated this procedure for 3 times. Heart rate(HR) and left ventricular developed pressure(LVDP),difference between left ventricular end systolic pressure and end diastolic pressure were recorded at baseline,post-reperfusion at 10 min,20 min and 30 min.At baseline and 30 min of reperfusion, coronary flow(CF) was measured,and contents of lactate dehydrogenase(LDH) and creatine kinase(CK) in the coronary effluent were analyzed.The heart tissues were sampled at 30 min of reperfusion and then rapidly stored in liquid nitrogen.The levels of phosphorylated cAMP response element binding protein on Ser133[p-CREB(Ser133)]were measured using Western blot analysis,NF-κB-DNA activation was analyzed using electrophoretic mobility shift assay(EMSA). Results:Compared with I/R group,the levels of p-CREB(Ser133) in IPC and PDTC groups were increased by 2.31 folds and 1.65 folds,respectively(P0.01),compared with IPC group,the levels of p-CREB(Ser133) in H89 group was significantly lower(P0.01).Compared with I/R group,the binding activity of NF-κB-DNA was significantly lower in IPC group(P0.01), compared with IPC group,the binding activity of NF-κB-DNA in H89 and PDTC groups were significantly higher(P0.05). Compared with IPC group,in I/R,H89 and PDTC groups,at 30min of reperfusion,HR,LVDP and CF were improved significantly (P0.05) and the contents of LDH and CK were decreased significantly in IPC group(P0.05 or 0.01). Conclusion:The results demonstrated that the activated PKA inhibited the binding activity of NF-κB-DNA and attenuated I/R injury in the cardioprotection of IPC.

Abstract:We applied a combined proteomic and metabolomic approach to obtain novel mechanistic insights in PKC epsilon-mediated cardioprotection. Mitochondrial and cytosolic proteins from control and transgenic hearts with constitutively active or dominant negative PKC epsilon were analyzed using difference in-gel electrophoresis (DICE). Among the differentially expressed proteins were creatine kinase, pyruvate kinase, lactate clehydrogenase, and the cytosolic isoforms of aspartate amino transferase and malate dehydrogenase, the two enzymatic components of the malate aspartate shuttle, which are required for the import of reducing equivalents from glycolysis across the inner mitochondrial membrane. These enzymatic changes appeared to be dependent on PKC epsilon activity, as they were not observed in mice expressing inactive PKC epsilon. High-resolution proton nuclear magnetic resonance (H-1-NMR) spectroscopy confirmed a pronounced effect of PKC epsilon activity on cardiac glucose and energy metabolism: normoxic hearts with constitutively active PKC epsilon had significantly lower concentrations of glucose, lactate, glutamine and creatine, but higher levels of choline, glutamate and total adenosine nucleotides. Moreover, the depletion of cardiac energy metabolites was slower during ischemia/reperfusion injury and glucose metabolism recovered faster upon reperfusion in transgenic hearts with active PKC epsilon. Notably, inhibition of PKC epsilon resulted in compensatory phosphorylation and mitochondrial translocation of PKC delta. Taken together, our findings are the first evidence that PKC epsilon activity modulates cardiac glucose metabolism and provide a possible explanation for the synergistic effect of PKC delta and PKC epsilon in cardioprotection.
