Abstract:Hypochlorous acid (HOCl) plays a crucial role in daily life and mediates a variety of physiological processes, however, abnormal levels of HOCl have been associated with numerous human diseases. It is therefore of significant interest to establish a simple, selective, rapid and sensitive fluorogenic method for the detection of HOCl in environmental and biological samples. A hydrazide-containing fluorescent probe based on a rhodamine scaffold was facilely developed that could selectively detect HOCl over other biologically relevant reactive oxygen species, reactive nitrogen species and most common metal ions in vitro. Via an irreversible oxidation-hydrolysis mechanism, and upon HOCl-triggered opening of the intramolecular spirocyclic ring during detection, the rhodamine hydrazide-based probe exhibited large fluorescence enhancement in the emission spectra with a fast response, low detection limit and comparatively wide pH detection range in aqueous media. The probe was further successfully applied to monitoring trace HOCl in tap water and imaging both exogenous and endogenous HOCl within living cells. It is anticipated that this simple and useful probe might be an efficient tool with which to facilitate more HOCl-related chemical and biological research. Copyright © 2015 John Wiley & Sons, Ltd.

Abstract:A rhodamine 6G hydrazide fluorescent chemosensor was prepared for the rapid HOCl detection in aqueous media. The system makes good use of the irreversible HOCl-mediated selective oxidation reaction to generate fluorescent response proportional to the amount of HOCl in neutral buffer. This probe exhibits great photostability, high sensitivity, and good selectivity for HOCl over other reactive species and most of the common metal ions. Furthermore, the probe is cell membrane permeable, and its applicability has been successfully demonstrated for fluorescence imaging of both exogenous and endogenous HOCl within living cells. Cytotoxicity assays prove that this probe is almost nontoxic to the cultured cell lines under the experimental conditions.

Abstract:A BODIPY-based ratiometric fluorescent probe for HOCl has been designed based on the transduction of thioether to sulfoxide function. This probe features a marked absorption and emission blue-shift upon the HOCl-promoted rapid transduction, enabling the highly selective and ratiometric detection. In addition, the probe works excellently within a wide pH range of 4–10, addressing the existing pH dependency issue. Living cells studies demonstrate that the probe is cell membrane permeable and can be employed successfully to image endogenous HOCl generation in macrophage cells.

Abstract:In this paper, we designed and synthesized a novel phenylazo-based fluorescent probe (RHN) for the sensing and imaging of hypochlorous acid (HClO) in mitochondria in living cells. In this process, HClO promoted the oxidation of the phenylazo group to generate a free Rhodol fluorophore moiety, which in turn restored strong fluorescence and realized the detection of HClO. As expected, RHN exhibited high selectivity, high sensitivity and rapid response, with detection limits as low as 22 nM (1.155 ng/mL). Importantly, the results of the cell imaging experiments indicated that RHN has the ability to image and sense HClO in mitochondria, which is of great significance for exploration of the specific role of HClO in both the immune system and diseases.

Abstract:Hypochlorous acid (HOCl) as a biologically important reactive oxygen species (ROS) plays crucial roles in the physiological and pathological processes. It is thus meaningful to explore an effective method for detecting basal HOCl in living systems. We herein report a large Stokes shift (about 115 nm) fluorescent probe LSS-HA for highly specific and ultrasensitive detection of HOCl with excellent water-solubility. Probe LSS-HA exhibited a remarkable fluorescence enhancement ((similar to) 227-fold) due to the adoption of high-quantum-yield fluorophore in aqueous solution. Besides, probe LSS-HA showed ultrasensitivity (detection limit is 3.30 nM) and fast response ( < 5 s) for HOCl. Furthermore, probe LSS-HA was successfully applied for imaging basal HOCl in living cells and zebrafish.

Abstract:A benzothiazole-based turn-on fluorescent probe with a large Stokes shift (190nm) has been developed for hypochlorous acid detection. The probe displays prompt fluorescence response for HClO with excellent selectivity over other reactive oxygen species as well as a low detection limit of 0.08μM. The sensing mechanism involves the HClO-induced specific oxidation of oxime moiety of the probe to nitrile oxide, which was confirmed by HPLC-MS technique. Furthermore, imaging studies demonstrated that the probe is cell permeable and can be applied to detect HClO in living cells.

Abstract:As one of the reactive oxygen species (ROS), hypochlorous acid (HOCl) plays a key role in the pathogenesis of a range of diseases. The detection of HOCl has been a hot research topic. Herein, this paper proposes a new conjugate of 4-hydroxy-1,8-naphthalimide and p-methylaminophenylether as fluorescent probe HNPE with simple synthesis and excellent performances to detect HOCl. The probe has a good linear relationship with HOCl (0-1 mu M), and its detection limit is 6.56 nM. Moreover, this probe has good selectivity, which can distinguish HOCl from other common analytes. Importantly, the probe HNPE has some achievements in HOCl imaging of living cells and zebrafish. Hence, the probe HNPE is competent as a tool to visualize HOCl in biosystems.

Abstract:A highly sensitive and selective fluorescein-based probe 1 for hypochlorite anion was synthesized. The probe 1 has favorable characteristics for biological imaging, including high water solubility, high fluorescence yield, pH-independent fluorescence, and biocompatibility. Results show that it has a detection limit of 40 nM to hypochlorite anion. In addition, confocal fluorescence microscopy imaging using RAW264.7 cells showed that the new probe 1 could be used as an effective fluorescent probe for detecting HOCl in living cells.

Abstract:The development of simple techniques for monitoring hypochlorous acid (HOCl) in living cells is urgently needed to disclose the important roles of HOCl in biological systems. Although a number of fluorescent probes have been exploited to detect HOCl, most of them are easily influenced by other competing reactive oxygen species (ROS) and reactive nitrogen species (RNS), and disturbed by many factors including variabilities in excitation, probe distribution and environmental conditions due to the recognition of HOCl based on the fluorescence changes in single wavelength. Herein, in this work, we present a simple fluorescent probe AETU-HOCl containing a new recognition group (2-amino-ethyl)thiourea (AETU) for the ratiometric detection of HOCl in living cells. Probe AETU-HOCl exhibits outstanding selectivity for HOCl over other ROS/RNS at higher concentrations (0.5 mM, 100 equiv.). More importantly, the rapid response (1 min) and high sensitivity (a detection limit of 3.9 nM) allow it to successfully visualize the intracellular HOCl levels by the ratiometric imaging. The present study would offer a simple, ratiometric, rapid, and ultrasensitive assay for the accurate determination of HOCl in the biological systems.

Abstract:As a significant reactive oxygen species (ROS), hypochlorous acid (HClO) plays a key role in the human immune system, ensuring the normal function of the organism. However, abnormal HClO levels have been linked to immune system disorders, neurodegenerative damage, and cancer. Therefore, rapid, and sensitive monitoring of HClO is important for the diagnosis of certain diseases. Here, we designed a probe XL for the detection of HClO in living cells, which is a mixture of phenothiazine and semi-cyanine. The probe XL is highly sensitive, selective and has an ultra-fast (<10 seconds) response to HClO in solution. The detection limit of probe XL for HClO was as low as 28 nM, and we also successfully used the test paper prepared with probe XL for the colorimetric fluorescence detection of HClO in real water samples. In addition, probe XL showed good response to both endogenous and exogenous HClO in cells. Finally, we successfully used probe XL for imaging HClO in zebrafish.

Abstract:Hypochlorous acid (HOCl), one of the reactive oxygen species (ROS), is a key microbicidal agent which is used for natural defense. However, it is also linked to varieties of human diseases owing to the overproduction of HOCl. Much effort has been made to exploit selective fluorescent sensors for the detection of HOCl, but most of them have some disadvantages such as short excitation wavelength, low selectivity, and slow response and so on. These restrict the biological application of the probes. In this work, BR-O was designed and synthesized on the base of phenoxazine for the detection of HOCl. BR-O exhibited a violent fluorescence enhancement in the presence of HOCl, showing excellent selectivity and high sensitivity. More importantly, the probe BR-O was capable of detecting exogenous and endogenous HOCl in living cells.

Abstract:As a vital reactive oxygen species (ROS), endogenous hypochlorous acid (HOCl) deeply engaged in masses of pathophysiological processes owing to its high reactivity, thus the detection of HOCl is awfully valuable for elucidating HOCl-related pathophysiology mechanism. Despite the extensive development of outstanding HOCl fluorescent probes so far, very few of them could be applied for monitoring HOCl in the pure water system. Herein, we present a novel rhodol-based fluorescent probe 6 for the unambiguous detection of HOCl in pure water, which was effectively designed and synthesized by using the spirocyclic hydrazide group as the HOCl sensitive site and introducing two hydroxyethyl groups for obtaining high hydrophilicity into the rhodol skeleton, a unique rhodamine structure. Compared to the control compound 5 without hydrophilic groups only capable to be used in the aqueous system containing organic solvent, 6 achieves the detection of HOCl not only in the above regular system but also in pure water, indicating that the introduction of hydrophilic hydroxyethyl group is an effective and reasonable approach to achieve the broader-spectrum detection of HOCl. More importantly, different with 5, highly water-soluble 6 regardless of dissolving in the regular system or pure water system can achieve the visual monitoring of HOCl with prominent sensitivity and high spatiotemporal resolution in living HepG2 cells and zebrafish, confirming the robust imaging ability of our probe 6 to HOCl in complex biosystem.

Abstract:We have developed a ratiometric fluorescent probe BRT based on boron dipyrromethene (BODIPY) and rhodamine-thiohydrazide Förster resonance energy transfer (FRET) platform for sensing hypochlorous acid (HOCl) with high selectivity and sensitivity. The probe can detect HOCl in 15 s with the detection limit of 38 nM. Upon mixing with HOCl the fluorescence colour of probe BRT changed from green to orange. Moreover, probe BRT was applied to successfully monitor HOCl in living RAW 264.7 cells.

Abstract:Hypochlorous acid (HOCl), one of the physiologically important reactive oxygen species (ROS), is engaged in a diverse variety of biological and pathophysiological functions. Excessive HOCl buildup can cause a variety of illnesses, therefore, the identification of events in real-time of abnormal levels of exogenous and endogenous HOCl in biological settings is extremely important. In the present paper, we reported a new near-infrared emissive fluorescent probe D-HBT-HOCl for the detection and bioimaging of HOCl in live cells. With regard to the construction of D-HBT-HOCl, we combined the dicyanoisophorone motif with a 2-aminobenzenethiol unit, resulting in the near-infrared emission wavelength of 700 nm and an ultra-large Stokes shift of 180 nm, which potentially endowed D-HBT-HOCl with superiority in biological imaging. In addition, the N, N-dimethylthiocarbamate served as an efficient recognition group for HOCl as well as a fluorescence quenching group in the probe. The results of the experiments showed that the newly designed probe exhibited excellent sensing capacity, including outstanding selectivity, a high signal-to-noise ratio, and a rapid response time for the detection of HOCl. What's more, this probe was further effectively leveraged for the imaging of both extrinsic and intrinsic HOCl in live cells, demonstrating its versatility, and possessed the potential to be used to further investigate the physiological and pathological activity of HOCl in biological systems.

Abstract:A novel fluorescent probe CR-ClO for detection of HOCl based on a carbazole fused rhodamine was designed and synthesized. The probe utilized a HOCl-promoted oxidation reaction, which lead to the ring opening of the compound with a strong enhancement of the fluorescent emission at about 587 nm.The new probe CR-ClO has excellent selectivity and high sensitivity to ClO−,whose detection limit to ClO− is 1.16 × 10−6 M. Furthermore, the new probe has been successfully applied in living cells for detection of ClO−.

Abstract:Hypochlorous acid (HOCl), one of the most reactive and deleterious reactive oxygen species (ROS), plays a vital role in many pathological and physiological processes. However, as a result of the highly reactive and diffusible nature of HOCl, its uncontrolled production may lead to an adverse effect on host physiology. Because of its biological importance, many efforts have been focused on developing selective fluorescent probes to image HOCl. However, it is still challenging to design a fluorescent probe with exclusive selectivity towards HOCl. In this study, a novel fluorescent probe for HOCl, Probe 1 was rationally designed based on 1,8-naphthalimide. As the concentration of HOCl increased, the fluorescence intensity of the probe gradually decreased, and the solution color changed from yellow-green to colorless, indicating this is a "naked-eye sensor". Probe 1 has a large Stokes shift (120 nm), which can effectively avoid fluorescence self-absorption. In addition, Probe 1 shows excellent selectivity to HOCl among different ions including common ROS, high sensitivity, fast response (<2 min), high fluorescence quantum yield (phi = 0.93) and low detection limit (0.237 mu M). Finally, the imaging results in HeLa cells showed that the probe could be used for the detection of exogenous and endogenous HOCl, and proved the potential of the probe as a biosensor for the detection of HOCl.

Abstract:We present a lysosome-targetable and ratiometric fluorescent probe, TJ2, for visually monitoring exogenous and endogenous HOCl in living cells by using a built-in S atom within the a-extended rhodamine fluorophore as a pH-independent recognition site. Based on the oxidation-promoted intramolecular charge transfer processes, TJ2 demonstrates fast ratiometric fluorescence response, excellent selectivity, and high sensitivity (2.3 nM) toward HOCl. Furthermore, TJ2 is highly lysosome-targetable, which could be used for ratiometric monitoring of the levels of HOCl in lysosomes.
