Abstract:A new Schiff-base fluorescence probe CS containing benzimidazole and coumarin fluorophores was synthesized and characterized by NMR, IR and ESI-MS. Fluorescence and absorption spectra show the unique changes in the presence of Hg2+ and Cu2+. Hg2+ induces a remarkable fluorescence enhancement and results in mercury-promoted hydrolysis of a mercury-CS complex to convert the imine group to the aldehyde group. Cu2+ leads to a colorimetric change from colorless to pale yellow and causes formation of a copper-CS complex. Thus, CS can act as a fluorescent chemodosimeter for Hg2+ and a naked-eye chemosensor Cu2+. The cell imaging experiments for Hg2+ show a slow enhancement of a bright blue fluorescence with increasing incubation time, while Cu2+ instantly quenches the fluorescence and then the fluorescence intensity has no further change by extending the incubation time. Therefore, the probe CS shows its potential application for simultaneous differential detection of Hg2+ and Cu2+ in living cells based on the fluorescence response time. (C) 2016 Elsevier B.V. Abstract:Lead ions (Pb 2+ ) are a widely distributed and highly toxic heavy metal pollutant, which seriously threatens the environment, economy and human safety. Here, a label-free ratiometric fluorescent biosensor was constructed for Pb 2+ detection using DNAzyme-driven target cycling and exonuclease III (Exo III)-mediated DNA cycling as a dual signal amplification strategy. The SYBR Green I (SGI) and N -methyl mesoporphyrin IX (NMM) used in this study are characterized by low cost, storage resistance, and short preparation time compared with conventional signaling probes labeled with fluorescent groups. Unlike the single-emission fluorescence strategy, monitoring the fluorescence intensity ratio of SGI and NMM can effectively reduce external interference to achieve accurate detection of Pb 2+ . DNAzyme structures on the surface of magnetic beads (MBs) can recognize Pb 2+ and activate the target circulatory system to cleave single-stranded DNA (ssDNA). The ssDNA further initiated the Exo III-assisted DNA circulatory system to digest double-stranded DNA (dsDNA) and release guanine-rich G1. Finally, the fluorescence signals of SGI and NMM were weakened and enhanced, respectively. The sensing strategy achieved a wide linear range from 0.5 to 500 nM and a low limit of detection (LOD) of 26.4 pM. Furthermore, its anti-interference ability and potential applicability for Pb 2+ detection in actual samples were verified. This work ingeniously combines the dual signal amplification strategy with the ratiometric sensing strategy constructed by structure-specific fluorescent dyes, which provides a promising method for constructing sensitive and accurate fluorescent biosensors.

Label-free Detection of Sub-Nanomolar Lead(ii) Ions in Aqueous Solution Using a Metal-Based Luminescent Switch-on Probe.

Abstract:A label-free oligonucleotide-based luminescent switch-on assay has been developed for the selective detection of sub-nanomolar Pb2+ ions in aqueous solution and real water samples. An iridium(III) complex was employed as a G-quadruplex specific luminescent probe and a guanine rich DNA (PS2.M, 5′-GTG3TAG3CG3T2G2-3′) was employed as recognition unit for Pb2+ ions. The PS2.M exists in a single-stranded conformation in the absence of Pb2+ ions, and the weak binding of the iridium(III) probe to ssDNA results in a weak luminescence signal. Upon binding to Pb2+ ions, the single-stranded DNA sequence (PS2.M) is induced into a G-quadruplex conformation, which greatly enhances the luminescence emission of the iridium(III) probe. The assay can detect Pb2+ ions in aqueous media with a limit of detection of 600pM. It also exhibits good selectivity for Pb2+ ions over other heavy metal ions. Furthermore, the application of the assay for the detection of Pb2+ ions in spiked river water samples has been demonstrated.
A Fluorescence Sensor for Pb 2+ Detection Based on Liquid Crystals and Aggregation-Induced Emission Luminogens

Abstract:Heavy metals, such as lead ions, are regarded as the main environmental contaminants and have a negative impact on human bodies, making detection technologies of lead ions critical. However, most existing detection methods suffer from time consumption, complicated sample pretreatment, and expensive equipment, which hinder their broad use in real-time detection. Herein, we show a new fluorescence sensor for detecting lead ions derived from liquid crystals doped with an aggregation-induced emission luminogen. The mechanism is based on the variation of fluorescence intensity caused by the disturbance of an ordered liquid crystal configuration in the presence of Pb2+, induced by DNAzyme and its catalytic cleavage. The proposed fluorescence sensor exhibits a low detection limit of 0.65 nM, which is 2 orders of magnitude lower than that previously reported in an optical sensor based on liquid crystals. The detection range of the Pb2+ fluorescence sensor is broad, from 20 nM to 100 μM, and it also selects lead ions from numerous metal ions exactly, resulting in a highly sensitive, highly selective, simple, and low-cost detection strategy of Pb2+ with potential applications in chemical and biological fields. This approach to designing a liquid crystal fluorescence sensor offers an inspiring stage for detecting biomacromolecules or other heavy metal ions by varying decorated molecules.This article has not yet been cited by other publications.

Abstract:To meet the severe water pollution status, multi-target biosensor becomes one of research focus. We herein report a novel duplex functional fluorescent biosensor for the label-free and separate detection of Pb2+ and Hg2+ with high sensitivity and selectivity. A K+-induced fluorescent G-quadruplex probe was assembled by a guanine-rich sequence (AGRO100) and N-methyl mesoporphyrin IX(NMM). It changed into a more stably non-fluorescent G-quadruplex structure and a hairpin-like structure upon binding Pb2+ and Hg2+ ions, respectively. As a result, the fluorescence decreased with relation to the addition of targets, allowing the separate detection of Pb2+ and Hg2+ ions at concentrations as low as 5 nM and 18.6 nM. The linear correlation existed between the fluorescence intensity and the concentration of Pb2+ and Hg2+ over the range of 10-200 nM (R2 = 0.98) and 20-1000 nM (R2 = 0.99), respectively. All real lake samples, even containing Pb2+ or Hg2+ at 50 nM, could be determined by the duplex functional fluorescent probe with recovery rates 96% and 104%, respectively. Considering that this sensor is label-free, simple, time-saving, cost-effective and easy-to-handle, it is possible to pave its way for Pb2+ and Hg2+ detection in various application fields.

Abstract:Due to the threat of lead pollution to health, environmental and food safety, developing simple and fast detection methods is highly required. Whereas, traditional single-mode probe suffers from limited application scenario. In this study, a colorimetric and fluorometric dual-mode probe for Pb 2+ determination was constructed by using bifunctional G-quadruplex-hemin complex. In this dual-mode probe, enzyme strand and substrate strand of 8–17 DNAzyme are labeled with G-quadruplex-hemin complex and fluorophore, respectively. In the absence of Pb 2+ , the self-assembly of enzyme strand and substrate strand inhibits intrinsic mimic peroxidase of G-quadruplex-hemin complex by base-pairing, which also quench the fluorescence via in proximity effect. When the DNAzyme is activated by Pb 2+ , the quenched fluorescence is restored as well as the inherent peroxidase mimetic activity, leading to dual signal output. Under optimal conditions, this dual-mode probe exhibit a good linear relationship between logarithm of Pb 2+ concentration and signal difference within the range from 1.5 nM to 20 nM and 0.5 nM to 10 nM for colorimetric and fluorescence mode, respectively. The detection limits for the corresponding mode were estimated to be 1.29 nM and 0.16 nM, respectively. This dual-mode probe also successfully applied for the spiked river water assay with satisfactory recovery in the range of 93.2 %–115.3 %. This work paves a new way for DNAzyme based dual-mode probe construction.

Abstract:A new compound, 2-boronobenzaldehyde-(2'-hydroxyl-4'-sulfonic acid) naphthalene hydrazone (1), was synthesized and its structure was characterized by proton nuclear magnetic resonance, mass and element analyses. The presence of Pb2+ led 1 to undergo colorimetric and fluorescent changes, which were detectable with the naked eye. Thus, a dual spectral response for Pb2+ detection was introduced. In KH2PO4-NaOH buffer aqueous solution (pH 6.0), 1 exhibited fluorescence enhancement at 568 nm and hyperchromicity at 595 nm upon the addition of Pb2+. The fluorescent intensity change was proportionate to the concentration of Pb2+ with a dynamic working range of 5.0 x 10(-7) mol L-1 to 1.0 x 10(-4) mol L-1 and a detection limit of 3.7 x 10(-8) mol L-1. The fluorometric method was successfully applied for the detection of Pb2+ water of Qianhu Lake and soil in Nanchang university campus. The recoveries were 111-116% for water and 97.6% for soil respectively, determined via the standard addition method. (C) 2013 Elsevier B.V. All rights reserved.

Abstract:By combination of a sugar group and a rhodamine group into one molecule, a bright and specific fluorescent sensor for Hg(2+) in natural water and living cells was achieved. RG1 features the high quantum efficiency for a Hg(2+)-bound sensor in natural water and sensitivity to environmentally relevant mercury in complex natural samples. The limit of detection of Hg(2+) of 1 ppb level suggests that RG1 is capable of distinguishing between the safe and toxic levels of inorganic mercury in drinking water. RG1 also establishes excellent Hg(2+)-ion specificity over alkali-, alkaline-earth metals and the first-row transition metals as well as Pb(2+) and Ag(+). Investigation on the fluorescence imaging of Hg(2+) in living cells demonstrates that RG1 might be used for monitoring Hg(2+) within biological samples.
Highly sensitive and selective detection of Pb2+ using a turn-on fluorescent aptamer DNA silver nanoclusters sensor

Abstract:A novel turn-on fluorescent biosensor has been constructed using C-PS2.M-DNA-templated silver nanoclusters (Ag NCs) with an average diameter of about 1 nm. The proposed approach presents a low-toxic, simple, sensitive, and selective detection for Pb2+. The fluorescence intensity of C-PS2.M-DNA-Ag NCs enhances significantly in the presence of Pb2+, which is attributed to the special interaction between Pb2+ and its aptamer DNA PS2.M. Pb2+ induces the aptamer to form G-quadruplex and makes two darkish DNA/Ag NCs located at the 3' and 5' terminus close, resulting in the fluorescence light-up. Moreover, Pb2+ can be detected as low as 3.0 nM within a good linear range from 5 to 50 nM (R = 0.9862). Furthermore, the application for detection of Pb2+ in real water samples further demonstrates the reliability of the sensor. Thus, this sensor system shows a potential application for monitoring Pb2+ in environmental samples.

Abstract:Abstract A novel fluorescent biosensor was constructed to detect Pb2+. Multifunctional magnetic beads modified with substrate sequence and Pb2+-specific DNAzyme were employed as recognition probes. The fluorescence of carbon dots synthesized from watermelon rind (W-CDs) was quenched by hemin. Hemin/W-CDs were employed as fluorescent signals. The presence of Pb2+ could target-trigger the biosensor, and generated G-quadruplex which could restore the fluorescence of hemin/W-CDs. The fluorescence change of the biosensor depended on Pb2+ concentration from 1 to 20 nM.

Abstract:A novel fluorescent ratiometric chemosensor based on 4-pyren-1-yl-pyrimidine (PPM) has been designed and prepared for the detection of Hg2+ in the presence of other competing metal ions in acetonitrile. The photo exhibits fluorescence color change of PPM from blue to green without and with Hg2+, which red shift of wavelength about 105nm in fluorescence emission spectra. It can serve as a highly selective chemodosimeter for Hg2+ with ratiometric and naked-eye detection. The photophysical properties of PPM confirmed a 2:1 (PPM–Hg2+) binding model and the spectral response toward Hg2+ was proved to be reversible.

Abstract:The pollution problem caused by mercury ions (Hg 2+ ) is becoming increasingly serious, and it is imperative to establish a methodology that is tailored to the precise identification of Hg 2+ . Phosphorus-doped biomass carbon quantum dots (P-BCQDs) with cyan fluorescence were synthesized via a hydrothermal approach, utilizing willow leaves as the carbon precursor and sodium pyrophosphate as a modifying agent. The morphology and optical properties of P-BCQDs were analyzed, and the results showed that P-BCQDs had a spherical structure, uniform dispersion, and particle size at 2.0±0.5 nm. Based on the principle that P-BCQDs bind to Hg 2+ to burst the fluorescence, the P-BCQDs have an optimal time of 2 min for the detection of Hg 2+ , and the optimal pH = 7, with c Hg2+ ranging from 0–20 μ mol/L. The linear relationship was achieved with the detection limit of 9 nmol/L. The spiked recovery experiments were conducted in actual water samples. The recoveries exhibited a range of 95.0% to 105.0%, accompanied by Sr ranging from 1.6% to 2.8%, which indicated that the proposed methodology is applicable for the quantification of Hg 2+ .

Abstract:Mercury ions (Hg 2+ ) are a class of heavy metal ions that can cause serious harm to the human body due to their toxicity, fluidity, and bioaccumulation. Direct contact with Hg 2+ can cause adverse effects on the human body. In addition, long-term exposure to Hg 2+ can lead to extreme toxicity of enzymes and proteins in the body, disrupting the cell cycle and thus endangering human life. It is becoming increasingly important to develop a method that is reliable, stable, and sensitive for detecting Hg 2+ . In this study, a near-infrared "turn-on" fluorescent probe based on dicyanomethylene-4 H -pyran, DCM-Hg, was designed and synthesized to determine Hg 2+ over other metal ions. Probe DCM-Hg had a detection limit (LOD) of 3.35 μM when used to detect Hg 2+ . The Hg 2+ concentrations and the fluorescence intensities at 695 nm showed a good linear correlation. Additionally, Hg 2+ in HepG2 cells was successfully visualized using the current probe.

Abstract:Novel functional oligonucleotides, especially DNAzymes with RNA-cleavage activity, have been intensively studied due to their potential applications in therapeutics and sensors. Taking advantage of the high specificity of 17E DNAzyme for Pb(2+), highly sensitive and selective fluorescent, electrochemical and colorimetric sensors have been developed for Pb2+. In this work, we report a simple, sensitive and label-free 17E DNAzyme-based sensor for Pb(2+) detection using unmodified gold nanoparticles (GNPs) based on the fact that unfolded single-stranded DNA could be adsorbed on the citrate protected GNPs while double-stranded DNA could not. By our method the substrate cleavage by the 17E DNAzyme in the presence of Pb(2+) could be monitored by color change of GNPs, thereby Pb(2+) detection was realized. The detection of Pb(2+) could be realized within 20 min, with a detection limit of 500 nM. The selectivity of our sensor has been investigated by challenging the sensing system with other divalent metal ions. Since common steps such as modification and separation could be successfully avoided, the sensor developed here could provide a simple, cost-effective yet rapid and sensitive measurement tool for Pb(2+) detection and may prove useful in the development of sensors for clinical toxicology and environmental monitoring in the future.

Abstract:Detection of lead(II) (Pb2+) ions in water is important for the protection of human health and environment. The growing demand for onsite detection still faces challenges for sensitive and easy-to-use methods. In this work, a novel surface plasmon resonance (SPR) biosensor based on GR-5 DNAzyme and gold nanoparticles (AuNPs) was developed. Thiolated DNAzyme was immobilized on the gold surface of the sensor chip followed by anchoring the substrate-functionalized AuNPs through the DNAzyme-substrate hybridization. The coupling between the localized surface plasmon (LSP) of AuNPs and the surface plasmon polaritons (SPP) on the gold sensor surface was used to improve the sensitivity. The substrate cleavage in the presence of Pb2+ ions was catalyzed by DNAzyme, leading to the removal of AuNPs and the diminished LSP-SPP coupling. The optimal detection limit was 80 pM for the sensor fabricated with 1 μM DNAzyme, corresponding to two or three orders of magnitude lower than the toxicity levels of Pb2+ in drinking water defined by WHO and USEPA. By tuning the surface coverage of DNAzyme, the sensitivity and dynamic range could be controlled. This sensor also featured high selectivity to Pb2+ ions and simple detection procedure. Successful detection of Pb2+ ions in groundwater indicates that this method has the prospect in the onsite detection of Pb2+ ions in water. Given the variety of AuNPs and metal-specific DNAzymes, this detection strategy would lead to the development of more sensitive and versatile heavy metal sensors.

Abstract:A label-free and enzyme-free colorimetric sensor for rapid detection of Pb2+ is reported, which is based on the strategy of DNAzyme-mediated RNA cleavage combined with an annealing-accelerated DNA hybridization chain reaction (HCR). As a trigger DNA, the substrate strand (STM) of DNAzyme can initiate HCR effectively. However, when it is cleaved by DNAzyme in the presence of Pb2+, the separation of DNA functional domains leads to a serious decrease in HCR efficiency. As a result, the difference in Pb2+ concentration converts into the difference of DNA assembly, which eventually leads to the color change of colloidal gold nanoparticles (AuNPs). In this work, a DNA strand (cGR5) completely complementary to the catalytic strand (GR5) of DNAzyme is used to improve the dissociation of STM to enhance the HCR efficiency. In addition, the simple operation of DNA annealing is first used to accelerate the HCR process, enabling the Pb2+ detection to be completed in about 30 min. As advantages of high sensitivity, good selectivity, strong anti-interference ability, and good practical performance are achieved, it is anticipated that the cheap and simple colorimetric sensor will be helpful for on-site detection of environmental and food samples.

Abstract:A highly sensitive colorimetric/fluorescent dual-mode sensor based on hybridization chain reaction (HCR) combining multifunctional Au NPs is presented for the detection of Hg2+ in aqueous solution. In Hg2+ absent solution, the surface of Au NPs was covered by hairpin auxiliary DNAs and a single strand DNA (ssDNA), which prevented Au NPs from salt-induced aggregation. At the same time, the fluorescence intensity of the dye-labeled hairpin probes was significantly quenched by Au NPs. In the presence of Hg2+, T-Hg2+-T coordination chemistry between helper DNA and the hairpin probes was induced which triggered the formation of extended double-stranded DNA (dsDNA) polymers via HCR. The formed dsDNA polymers were stiffer which couldn’t attach to Au NPs, resulting in a red-to-blue color change along with salt-induced aggregation of Au NPs for colorimetric sensing. Meanwhile, the fluorescence of dye-labeled DNA turns on. Due to the HCR amplification effect, a highly sensitive detection of Hg2+ was achieved with detection limit of 0.1nM. Colorimetry is suitable for the analysis in salt solution with concentration lower than 100mM. It serves as an intuitive method that Hg2+ down to 1.0nM could be identified by naked eyes. For high-concentration salt solutions such as industrial effluent, fluorescent sensing acts as a better choice. Both the color and fluorescence changes of the proposed sensor exhibited high selectivity against other metal ions. Lake water was collected and analyzed using the dual-mode sensor, the results confirmed the practicability of the proposed approach.
