Abstract:In this report, we present a new method for visual detection of Pb2+. Gold nanoparticles (Au-NPs) were synthesized in one step at room temperature, using gallic acid (GA) as reducer and stabilizer. Pb2+ is added during the gold nanoparticle formation. Analysis of Pb2+ is conducted by a dual strategy, namely, colorimetry and spectrometry. During Au-NPs synthesis, addition of Pb2+ would lead to formation of Pb-GA complex, which can induce the aggregation of newly-formed small unstable gold nanoclusters. Consequently, colorimetric detection of trace Pb2+ can be realized. As the Pb2+ concentration increases, the color turns from red-wine to purple, and finally blue. This method offers a sensitive linear correlation between the shift of the absorption band (Δλ) and logarithm of Pb2+ concentration ranging from 5.0 × 10−8 to 1.0 × 10−6 M with a linear fit coefficient of 0.998, and a high selectivity for Pb2+ detection with a low detection limit down to 2.5 × 10−8 M.

Abstract:We demonstrate the electrochemical etching of gold in a solution of hydrofluoric acid and dimethylformamide (DMF) to fabricate nanoporous gold (NpAu) films. Compared to de-alloyed NpAu, the electrochemically etched NpAu exhibits controllable pore size and etching depth by adjusting the etching conditions. We propose an etching mechanism where the organic electrolyte DMF protects the nascent gold pore walls by slowing the dissolution of gold oxide. At the same time, localized reactions occurring at the gold electrode surface focus the etching current for etching into depth. We further modify the as-prepared NpAu surface with single walled carbon nanotubes (SWCNTs) and use the nanotube-decorated surface as an electrochemical sensor to detect bisphenol A, an endocrine disruptor chemical. A 100 nM limit of detection is achieved by combining the catalytic capacity of SWCNTs with the specific surface area of a NpAu electrode.

Abstract:The sensitivity for analytes of interest is vital for environment protection and food safety. Here, we propose an extremely sensitive assay toward Pb2+ by using gold nanostars (GNSs) as probes based on the catalytic activity of Pb on etching gold atoms after being reduced in the presence of 2-mercaptoethanol (2-ME) and sodium thiosulfate. GNSs were prepared by using 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid as both the reducing and capping agents, enabling high stability and sensitivity for quantitation of Pb2+. Upon increasing Pb2+ concentration over the range of 0-10 μM, GNS solution color changed from greenish-blue to blue to purple to red, and eventually to colorless. The color change can be distinguished by naked eye at the Pb2+ concentration as low as 200 pM. Through monitoring longitudinal localized surface plasmon of GNSs, Pb2+ could be detected with a limit of detection of 1.5 pM, and the working range is 2 pM-1 μM. The ultra-high sensitivity of our assay stems from the high catalysis of Pb on etching gold on tips and branches in the presence of 2-ME and sodium thiosulfate, leading to the shape deformation to spherical gold nanoparticle and the corresponding significant changes in their optical properties. The assay provides high selectivity of Pb2+ over the tested interfering metal ions like Cu2+. With high sensitivity and selectivity, the assay was efficiently validated by analyzing water samples and monitoring the migration of Pb2+ from the tested container to water.

Abstract:A novel electrochemical sensing system for Pb(2+) has been developed based on atypical icosahedral gold nanoparticle (AI-AuNPs) modification and the improved rolling circle amplification (RCA). The newly synthesized AI-AuNPs which were applied to modify the gold electrode surface greatly enhanced the sensitivity and expanded the detection range due to its large specific surface area and a large density of irrational kinks and step atoms on the particle surface. Additionally, the improved RCA introduced a manipulated circular template DNA which can be hybridized with the primer obtained from the DNAzymes catalysis reaction in the presence of Pb(2+) as a cofactor to simplify the RCA procedure, and the largely elongated RCA products at the same time efficiently blocked the electron transfer at the electrode surface, thus significantly increased the electrochemical signal. The design of the label free electrochemical impendence sensing strategy was evidenced to be a highly sensitive and selective method for the lead ions monitoring compared with the previous reports and the detection range was 1 pM-1 μΜ with a detection limit of 290 fM (3δ rule). Furthermore, the proposed detection system was also environmental-friendly and inexpensive, which can be successfully used in real serum sample detection.

Abstract:In this paper, we report a novel colorimetric strategy for the detection of small molecules by using Pb(2+) ion as an example. In this strategy, DNAzyme duplex modified gold nanoparticles (GNPs) are designed to be unable to interact with graphene oxide (GO). However, in the presence of Pb(2+), the substrate strand of the DNAzyme is cleaved at its cleavage site, resulting in the disassembly of the DNAzyme duplex modified GNPs into three parts, i.e., the 3'- and 5'-fragments of substrate strand and the DNAzyme strand modified GNPs. By taking advantage of the efficient cross-linking effect of ssDNA-GNPs to GO, colorimetric sensor for the detection of the metal ion can be fabricated with a detection limit of 100 pM, which is much lower than the previous reports. This colorimetric method has also been used for the determination of Pb(2+) in the tap water of the local city and the water from a reservoir with satisfactory results, so it may have potential applications in the future.

Abstract:Novel functional oligonucleotides, especially DNAzymes with RNA-cleavage activity, have been intensively studied due to their potential applications in therapeutics and sensors. Taking advantage of the high specificity of 17E DNAzyme for Pb(2+), highly sensitive and selective fluorescent, electrochemical and colorimetric sensors have been developed for Pb2+. In this work, we report a simple, sensitive and label-free 17E DNAzyme-based sensor for Pb(2+) detection using unmodified gold nanoparticles (GNPs) based on the fact that unfolded single-stranded DNA could be adsorbed on the citrate protected GNPs while double-stranded DNA could not. By our method the substrate cleavage by the 17E DNAzyme in the presence of Pb(2+) could be monitored by color change of GNPs, thereby Pb(2+) detection was realized. The detection of Pb(2+) could be realized within 20 min, with a detection limit of 500 nM. The selectivity of our sensor has been investigated by challenging the sensing system with other divalent metal ions. Since common steps such as modification and separation could be successfully avoided, the sensor developed here could provide a simple, cost-effective yet rapid and sensitive measurement tool for Pb(2+) detection and may prove useful in the development of sensors for clinical toxicology and environmental monitoring in the future.
Catalytic Deposition of Pb on Regenerated Gold Nanofilm Surface and Its Application in Selective Determination of Pb2+

Abstract:We report a simple method of catalytic deposition of Pb on a gold nanofilm substrate, which was in situ prepared and used as nanocrystal seeds. Due to the unique properties of gold nanocrystal seeds, Pb could be catalytically deposited on the surface of the gold nanofilm. Compared with the deposition of Pb on bare gold electrode, a larger amount of Pb was deposited on the gold nanofilm and the electrical response was amplified significantly. The catalytic deposition of Pb on the gold nanofilm was characterized by scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and electrochemical methods. A stable and quasi-reversible redox couple was obtained in neutral solution and studied in detail. The surface of the gold nanofilm could be easily regenerated in 0.1 mol L(-1) nitric acid solution. Since the redox peaks of Pb could be effectively separated from those of other metals such as Cu, Cd, and Zn, a selective determination of Pb2+ was achieved. Linear sweep voltammetry (LSV) was used for the determination of Pb2+. The peak currents of Pb varies linearly with the concentration of Pb2+ in aqueous solution ranging from 1.0 to 10.0 micromol L(-1) (R=0.999), with a detection limit of 0.1 micromol L(-1). It is expected that the gold nanofilm will facilitate the appearance of heavy metal ion sensors with good performance.

Abstract:The photoacoustic signal generated by laser-induced nanobubbles (PA-LINB) proved to be a sensitive tool to monitor the aggregation of gold nanoparticles. Here, a simple and label-free photoacoustic method for the rapid detection of Pb2+ in the aqueous phase was developed. Due to the high affinity of Pb2+ ions to glutathione, the presence of Pb2+ led to the aggregation of glutathione-conjugated gold nanoparticles (GSH-GNPs). Hence, by measuring the variation of the PA-LINB signal after the aggregation of GSH-GNPs, Pb2+ can be quantified. A low detection limit for Pb2+ (42 nM) and a wide linear working range (~42–1000 nM) were achieved. Furthermore, the proposed method showed good selectivity against other metal ions.

Abstract:The detection of the lead heavy metal (Pb) in water is crucial in many chemical processes, as it is associated with serious health hazards. Here, we report the selective and precise colorimetric detection of Pb2+ ions in water, exploiting the aggregation and self-assembly mechanisms of glutathione (GSH)-functionalized gold nanoparticles (GNPs). The carboxyl functional groups are able to create coordination complexes with Pb2+, inducing aggregation amongst the GSH-GNPs in the presence of Pb2+ due to the chelation of the GSH ligands. The resulting aggregation amongst the GSH-GNPs in the presence of Pb2+ increases the aggregate size depending on the available Pb2+ ions, affecting the plasmonic coupling. This causes a substantial shift in the plasmon wavelength to a longer wavelength side with increasing Pb2+ concentration, resulting in a red-to-blue colorimetric or visual change, enabling the instant determination of lead content in water.

Abstract:We have developed a new nanoparticle (NP)-based chemiluminescent (CL) method for the sensitive detection of DNA hybridization. The "sandwich-type'' DNA technique comprises NH2-functionalized capture DNA immobilized on magnetic beads that can hybridize with one end of the target DNA. The other end of the target is recognized by a reporter DNA probe labeled with Au NPs on its 3-terminus. The hybridization events were monitored by a simple and direct luminol-AgNO3 CL reaction with a detection limit of 0.5 pM through a non-stripping Au NPs catalysis. Moreover, the CL reaction of non-stripping Au NPs-luminol-AgNO3 offers the advantages of being simpler and faster as compared to previous stripping Ag NPs and CuS methods. This simple and easy-to-use technique holds great promise for clinical applications of DNA hybridization.

Abstract:Nanomaterials have been widely used in energy and sensing applications because of their unique chemical and physical properties, especially their surface reactions. Measuring the local reactions of individual nanomaterials, however, has been an experimental challenge. Here we report on plasmonic imaging of surface electrochemical reactions of individual gold nanowires (AuNWs). We coated a gold thin film (plasmonic sensing layer) with a dielectric layer (Cytop) with refractive index close to that of water, and then a graphene layer for electrical contact. This design removed the interference from the sensing layer while preserving sharp surface plasmon resonance, which allowed us to obtain cyclic voltammograms of surface electrochemistry of individual AuNWs for the first time. We also investigated the difference in the electrochemical reactions of AuNWs and Au surfaces, and local distribution of electrochemical activities within a single AuNW.

Abstract:Lead pollution has remained a significant global concern for several decades due to its detrimental effects on the brain, heart, kidneys, lungs, and immune system across all age groups. Addressing the demand for detecting trace amounts of lead in food samples, we have developed a novel biosensor based on fluorescence resonance energy transfer (FRET) from fluorescein R6G to gold nanoclusters (AuNCs-CCY). By utilizing polypeptides as a template, we successfully synthesized AuNCs-CCY with an excitation spectrum that overlaps with the emission spectrum of R6G. Exploiting the fact that Pb 2+ induces the aggregation of gold nanoclusters, leading to the separation of R6G from AuNCs-CCY and subsequent fluorescence recovery, we achieved the quantitative detection of Pb 2+ . Within the concentration range of 0.002–0.20 μM, a linear relationship was observed between the fluorescence enhancement value (F-F 0 ) and Pb 2+ concentration, characterized by the linear equation y = 2398.69x + 87.87 (R 2 = 0.996). The limit of detection (LOD) for Pb 2+ was determined to be 0.00079 μM (3σ/K). The recovery rate ranged from 96 % to 104 %, with a relative standard deviation (RSD) below 10 %. These findings demonstrate the potential application value of our biosensor, which offers a promising approach to address the urgent need for sensitive detection of heavy metal ions, specifically Pb 2+ , in food samples.

Abstract:Gold nanoparticles, abbreviated as AuNPs, have been widely used in catalysis, optics, electronics, and biotechnology due to their unique performance brought by large specific surface area. In recent years, with the improvement of health awareness, the use of gold nanoparticles to detect heavy metal ions has become a research hotspot. The detection is based on colorimetric techniques, which means the metal ions are detected through the color change in solutions. This color change is visible or could be distinguished by ultraviolet-vissible spectrometers. Numerous studies have been published on gold nanoparticles, especially for its synthesis methods. This work introduced the chemical, physical, and biological preparation methods of gold nanoparticles. Understanding mechanisms for detecting heavy metal ions using AuNPs is also very important. Aggregating mechanism, anti-aggregation mechanism, and etching mechanism were also introduced. In addition, applications of using AuNPs to detect heavy metal ions were also introduced. This work will help promote the further development of this technology.

Abstract:High-spatial resolution fluorescence imaging on a high-density gold nanoelectrode array was achieved by anchoring fluorescent molecules to limit diffusion. Meanwhile, an ionic liquid as the electrolyte with a refractive index matching with the optical system could effectively improve the sensitivity of fluorescence imaging. Moreover, spatially resolved imaging of the electrochemical reactions on a single nanoelectrode modified with a single Pt nanoparticle was achieved.

Abstract:In this study, we developed a highly sensitive automatic counting method for gold nanomaterials at the single particle level, which can serve as a general sensing platform based on counting of gold nanomaterials. This method substantially improved the sensitivity and accuracy for AuNP counting by adopting the color image processing based on the distinctive localized plasmonic light-scattering of gold nanomaterials. The 60-nm AuNPs, with concentrations down to 4 fM, can be detected with our method. As a universal counting approach for gold nanomaterials, such as gold nanospheres, nanorods, and aggregates from particles under detectable size, this quantification method should be versatile to a breadth of applications.

Abstract:Lead poisoning in adults can affect the peripheral and central nervous systems, the kidneys, and blood pressure. Thus, the development of environment-friendly and simple methods for Pb2+ detection is of great importance. Herein, a label-free colorimetric method has been developed for the detection of Pb2+ based on the conformational switch from single-stranded DNA to G-quadruplex. The electrostatic interactions between DNA probe and gold nanorods (GNRs) induce GNRs to space closely. However, the electrostatic interaction is not strong enough to change the suspension state of GNRs. In the presence of Pb2+, the formation of G-quadruplexes increases the surface charge density around DNA, which is expected to strengthen the electrostatic interaction between the GNRs and the DNA. Therefore, the longitudinal absorption of GNRs decreased because the stronger interaction induced aggregation of GNRs. Importantly, the decrease in longitudinal absorption is proportional to concentration of Pb2+. By monitoring the change of absorbance, Pb2+ can be detected at a level of 3 nM with a linear range from 5 nM to 1 μM. The overall test only takes a few minutes and very little interference is observed from other metal ions. The major advantages of this method are its low cost, convenience, simplicity, sensitivity, and specificity.

Abstract:To minimize the plumbic poisoning, the construction of powerful portable chip for ultrasensitive Pb2+ assay plays a significant role in clinical diagnosis and environmental monitoring. Herein, inspired by communicating vessels, a microfluidic electrochemical sensing chip array was proposed for ultrasensitive Pb2+ detection of multiple samples. An editable nanoporous gold nanolayer with a feature of all-round conductivity and abundant active sites favoring biological ligand attachment was employed as electrochemical sensing substrate. corona-nanocomposites (CeO2@Au NPs) was designed and utilized as a differential pulse voltammetry response enhancer of the o-phenylenediamine-hydrogen peroxide electrochemical sensing system. In addition, the proposed chip contains ten detection units and no complex components (such as pump or valve), which makes it capable of processing multiple samples in batch for the in-situ detection with low cost. Under the optimized experimental conditions, the Pb2+ detection limit of 3.1 pM was achieved with good stability and reliability, which was benefited from the excellent sensing substrate and appropriate CeO2@Au NPs-trigger amplification strategy. The results indicated that the integration of novel microfluidic chip design and nanomaterials catalytic strategy provides a new paradigm for the field of in-situ analysis, and shows promise for potential applications in many analytic fields.

Detection of Ferrous Ion by Etching-Based Multi-Colorimetric Sensing of Gold Nanobipyramids

Abstract:Colorimetric sensing methods based on non-spherically symmetric gold (Au) nanoparticles have become a powerful tool in the field of biomedical detection due to their intriguing plasmonic properties. In this study, Au nanobipyramids (Au NBPs) were used as colorimetric sensing probes to detect ferrous ions (Fe2+) through tip etching. The quick etching of Au NBPs along the longitudinal direction by superoxide radicals generated by the reaction of Fe2+ and H2O2 led local surface plasmon resonance (LSPR) to blue shift and produced vivid color change that could be used for visual inspection. Under the optimal reaction conditions, the peak shift of the Au NBPs and the logarithm of the concentrations of Fe2+ had a linear relationship in the range of 10 nM to 10 μM, with a very low detection limit of 1.29 nM. During the etching process, a different end shape of the Au nanoparticles results in a different process for the morphology transition, which makes the degree of spectral change and detection sensitivity significantly different. In the presence of trace amounts of Fe2+ (<1000 nM), the detection sensitivity of Au NBPs with sharp ends which rely on aspect ratio and truncation is nine times higher than that of gold nanorods with round ends which only rely on aspect ratio. Although the color change of larger-sized Au NBPs was not clear during detection, the LSPR peak shift was more severe. Therefore, the system provides different modes for detecting Fe2+ according to Au NBPs with different sizes and characteristics.
