Abstract:A new fluorescent sensor for Zn2+ based on 2-methylquinoline has been designed and synthesized. The sensor (1) demonstrates a Zn2+-specific emission enhancement linearly by a factor of 14 with a 1: 1 binding ratio even in the presence of a wide range of biological cations. This can be correlated to the efficient Zn2+ binding of 1 and to the participation of relevant amine functions in the metal coordination which promotes a good chelation-enhanced fluorescence (CHEF) effect and prevents the photoinduced electron transfer (PET) effect. Although the addition of Cd2+ also induced an emission enhancement to a certain extent, this enhancement was less pronounced as compared to that of Zn2+, which was further demonstrated by DFT calculations. Abstract:A novel turn-on fluorescence probe L has been designed that exhibits high selectivity and sensitivity with a detection limit of 9.53 × 10−8 mol/L for the quantification of Zn2+. 1H-NMR spectroscopy and single crystal X-ray diffraction analysis revealed the unsymmetrical nature of the structure of the Schiff base probe L. An emission titration experiment in the presence of different molar fractions of Zn2+ was used to perform a Job’s plot analysis. The results showed that the stoichiometric ratio of the complex formed by L and Zn2+ was 1:1. Moreover, the molecular structure of the mononuclear Cu complex reveals one ligand L coordinates with one Cu atom in the asymmetric unit. On adding CuCl2 to the ZnCl2/L system, a Cu-Zn complex was formed and a strong quenching behavior was observed, which inferred that the Cu2+ displaced Zn2+ to coordinate with the imine nitrogen atoms and hydroxyl oxygen atoms of probe L.

Abstract:A hydrophilic bis(1,2,3-triazolyl)fluorene derivative was synthesized as a multi-photon-absorbing, zinc-ion-sensing fluorescent probe. The fluorescence response was approximately five-fold greater in presence of Zn(2+), resulting in a large binding constant (1 × 10(9)) for a 1:2 ligand to zinc complex. A four-fold increase in the two-photon absorption cross section was achieved upon binding Zn(2+). In vitro two-photon fluorescence microscopy imaging revealed a significant fluorescence increase upon introduction of Zn(2+) into HeLa cells and reversible Zn(2+) binding, demonstrating the potential of this probe for zinc ion sensing.

Abstract:Zn has vital importance to the environment as well as human health. To regulate its dietary ingestion, it is important to monitor its presence in our food and environment, evoking more accurate and more sensitive ways to quantitatively determine its content in the environment. We herein present a squaraine-based chemosensor, namely DMPSQ, which has turn-on fluorescence response toward Zn2+ ion with good linearity, high selectivity and sensitivity down to 80ppb. Theoretical calculations were also conducted to provide insight to the sensing mechanism and the spectroscopic properties of the compound.

Abstract:Fluorescent turn-on receptors are extensively employed for the detection of Zn ions contamination in the environment due to its simplicity, convenience and portability. However, developing highly sensitive and cell-imageable fluorescent turn-on probe for the recognition of Zn ions in living organisms remains a significant challenge. Herein, we have successfully synthesized a novel Schiff base probe (H2L) with a significant fluorescence turn-on response (Zn ions) by one-step synthetic method. In this work, H2L exhibited high sensitivity to Zn2+ ions upon interaction with various common metal ions in HEPES buffer solution. Its detection limit is 1.87 × 10-7 M, which is lower than the requirement of Environmental Protection Agency (EPA) and World Health Organization (WHO) guidelines. The fluorescence titration and Job's plot analysis suggested a 1:1 binding ratio between the probe and Zn ion, and the single-crystal structures obtained further confirmed this inference. In addition, the fluorescent sensor demonstrated recyclability, maintaining its fluorescence intensity for up to 6 cycles without significant decrease, which holds promise for future investigations on reversible fluorescent chemosensors. Notably, fluorescence imaging experiments demonstrated that H2L could be successfully used for the detection of Zn2+ in live cells.

Abstract:A new fluorescent sensor, 2-(2-oxo-2-(quinolin-8-ylamino)ethoxy)-N-(pyridine-2-ylmethyl) benzamide (L), composed of a quinoline group as the fluorogenic unit and a pyridin-2-ylmethanamine as the binding unit for metal ions has been synthesized. The sensor shows excellent selectivity and sensitivity with a fluorescence enhancement to Zn(2+) over other cations in acetonitrile aqueous solution. The X-ray crystal structure analysis reveals that sensor L coordinates to Zn(2+)via a 1 : 1 binding mode but to Cd(2+)via a 2 : 1 binding mode, which lead to a different spatial arrangement of the fluorogenic unit in these complexes. In addition, density functional theory calculations on L, and the Zn(2+)/L and Cd(2+)/L complexes also imply that the different structures of L significantly affect the molecular orbital energy levels and electron transition, which would result in the spectral changes to distinguish Zn(2+) from Cd(2+). The absorption study results may also suggest the Cd(2+) in the complex can be displaced by Zn(2+). Furthermore, the fluorescence imaging of Zn(2+) in living cells was obtained.

Abstract:Zinc ion, one of the most important transition metal ions in living organisms, plays a crucial role in the homeostasis of the organism. The disorder of zinc is associated with many major diseases. It is highly desirable to develop selective and sensitive methods for the real-time detection of zinc ions. In this work, double-emitting fluorescent carbon dots (CDs) are prepared by a solvothermal method using glutathione, L-aspartic acid, and formamide as the raw materials. The carbon dots specifically recognize zine ions and produce a decrease in fluorescence intensity at 684 nm and an increase at 649 nm, leading to a ratiometric fluorescent sensor for zinc detection. Through surface modification and spectral analysis, the surface groups including carboxyl, carbonyl, hydroxyl, and amino groups, and C=N in heterocycles of CDs are revealed to synergistically coordinate Zn2+, inducing the structural changes in the emission site. The CDs can afford a low limit of detection of ~5 nM for Zn2+ detection with good linearity in the range of 0.02–5 μM, showing good selectivity as well. The results from real samples including fetal bovine serum, milk powder, and zinc gluconate oral solution indicated the good applicability of the CDs in the determination of Zn2+.

Abstract:A novel and simple fluorescent chemosensor, DH, was synthesized. As designed, DH displayed excellent selectivity exhibiting a "turn-on" response for Zn2+ over other relevant metal ions based on PET mechanism in 100% aqueous media, and the binding constant of Zn2+ binding to DH was calculated to be 7.27 x 10(8) M-2. Importantly, DH exhibited high sensitivity as a Zn2+ fluorescent chemosensor with a limit of detection (LOD) of 19.25 nM, which is lower than WHO guidelines (76.0 mu M). Moreover, the 2:1 stoichiometry between the chemosensor DH and Zn2+ was verified by Job's plot, fluorescent titration and high resolution mass spectroscopy. In addition, the recognition process of DH toward Zn2+ was chemically reversible and recyclable by adding Na(2)EDTA. Moreover, due to its good biocompatibility and low cytotoxicity, DH was successfully applied to the fluorescence imaging of Zn2+ and Na(2)EDTA in living HK2 cells.

Abstract:A fluorescent heteroditopic indicator for the zinc(II) ion possesses two different zinc(II) binding sites. The sequential coordination of zinc(II) at the two sites can be transmitted into distinct fluorescence changes. In the heteroditopic ligand system that our group developed, the formations of mono- and dizinc(II) complexes along an increasing gradient of zinc(II) concentration lead to fluorescence enhancement and an emission bathochromic shift, respectively. The extents of these two changes determine the sensitivity and, ultimately, the effectiveness of the heteroditopic indicator in quantifying zinc(II) ion over a large concentration range. In this work, a strategy to increase the degree of fluorescence enhancement upon the formation of the monozinc(II) complex of a heteroditopic ligand under simulated physiological conditions is demonstrated. Fluorination of the pyridyl groups in the pentadentate N,N,N'-tris(pyridylmethyl)ethyleneamino group reduces the apparent pK(a) value of the high-affinity site, which increases the degree of fluorescence enhancement as the monozinc(II) complex is forming. However, fluorination impairs the coordination strength of the high-affinity zinc(II) binding site, which in the triply fluorinated ligand reduces the binding strength to the level of the low-affinity 2,2'-bipyridyl. The potential of the reported ligands in imaging zinc(II) ion in living cells was evaluated. The subcellular localization properties of two ligands in five organelles were characterized. Both benefits and deficiencies of these ligands were revealed, which provides directions for the near future in this line of research.

Abstract:Zinc ion (Zn2+) is a necessary transition metal ion in the human body and plays a vital role in biological systems. Therefore, it is meaningful for rapid, sensitive, and specific detection of Zn2+. Herein, a “switch-on” fluorescence sensing platform for the detection of Zn2+ was successfully established. In this work, we report an optical nanoprobe with amino functional groups based on porphyrin metal-organic frameworks (PCN-NH2) through a simple treatment method. The modification of the salicylaldehyde (SA) through the C=N acts as a quencher of PCN-NH2 and the ligand of Zn2+ can effectively inhibit the fluorescence emission of PCN-NH2. The complex of porphyrin-based metal-organic framework and salicylaldehyde (PCN/SA) acts as the fluorescence sensing probe. The presence of Zn2+ results in the new emission peak at 448 nm due to the formation of a complex between Zn2+ and SA. The corresponding fluorescence signal changes were used to accurately detect the concentration of Zn2+. The fluorescence sensing platform shows advantages, including rapid response, high sensitivity, and excellent specificity with a linear detection range of Zn2+ between 0.1 and 250 μM, and a low detection limit (LOD) of 0.07 μM. And the feasibility of the method was verified for the quantification of Zn2+ in cell lysate, environmental water, and plasma.Graphical abstract

Abstract:A readily available Schiff base quinoline derivative, quinoline-2-carboxaldehyde (pyridine-2-carbonyl)-hydrazine (QCPCH), has been synthesized via a one-step condensation reaction. It is used as a turn-on fluorescent probe for detecting Zn2+ with high selectivity, sensitivity, and rapid response time of less than 3 s. In addition, the probe QCPCH shows a low detection limit of 72 nM and a large linear detection range (0–180 μM). The mechanism of probe QCPCH for sensing Zn2+ is proposed to be based on the combined effects of chelation-induced enhanced fluorescence (CHEF) and inhibiting photoinduced electron transfer (PET); this is further demonstrated by the Job's plot analysis, Benesi-Hildebrand study, FT-IR spectrum, and 1H NMR titration. Furthermore, the QCPCH is successfully applied to determine Zn2+ in real water samples, and is made into test strips for fast, convenient, quantitative and qualitative detection of Zn2+.

Abstract:A novel fluorescent sensor, 7-hydroxy-4-methylcoumarin-8-carbaldehyde-(fluorescein) hydrazone was designed and synthesized for selective recognition of Zn2+ in HEPES buffer medium of PH 7.4. This reagent could be used as a probe for Zn2+ by monitoring changes in the absorption and the fluorescence spectral patterns. More importantly, this sensor displays an extreme selectivity, sensitivity and color change for Zn2+ over other earth- and transition metal ions, which was mainly due to the spirolactam ring-opening power of Zn2+. Upon the addition of Zn2+, an overall emission change of 33-fold was observed and the detection limit was low as 6.54 ppb. Photoinduced electron transfer process, coupled with the intramolecular charge transfer process, are proposed to explain the observed spectral response.

Abstract:A series of salicylaldehyde-2-pyridinehydrazone derivatives were found to exhibit interesting “turn-on” fluorescence response toward Zn2+ in water at neutral pH, which could be prepared readily from commercially available materials through a one-step reaction in ethanol at room temperature. Among these compounds, 1 showed a 22-fold fluorescence enhancement in 99% water/DMSO (v/v) at pH 7.0 with a linear range of 0.10–1.00μmol/L and detection limit of 30nmol/L Zn2+. Especially, 1 was not affected by Cd2+ which usually induced a comparable fluorescence response to that of Zn2+ in other reported works. Moreover, live cell imaging showed that 1 could be useful for monitoring Zn2+ within biological samples.

Abstract:Heteroleptic Cu(N^N)(P^P) complexes have garnered significant attention owing to their various structures and remarkable photophysical properties. Four novel heteroleptic copper(I) complexes (C1•2BF4ˉ, C2•3BF4ˉ, C3•3PF6ˉ and C4•3PF6ˉ) of the type {(Cu(P^P))3(μ-HATN)} synthesized by ligand conformation-directed self-assembly methodology were reported herein. These copper(I) complexes with different coordination configuration could be obtained through a straightforward change in the rigidity and volume in different P^P ligand scaffolds. Furthermore, these copper(I) complexes exhibited exceptional visible light absorption, which were contributed to metal-to-ligand charge transfer and intra-ligand charge transfer. Moreover, these emissive heteroleptic Cu(I) complexes all displayed remarkable sensitivity and selectivity towards zinc ions in “turn-on” mode (intensity enhanced ca.60 times). The detection limitation of complex C1•2BF4ˉ reached to 5.10 × 10−9 M, which was lower than those reported fluorescent sensors. The possible sensing mechanism was investigated using ESI-Ms, NMR and DFT calculation. The reversible “on-off” fluorescent sensing behavior can be restored by treating the solution alternately with EDTA tetrasodium salt and Zn2+ salt.

Abstract:A turn-on fluorescence probe for zinc ion has been synthesized, which contains a symmetric pseudo salen moiety as zinc recognition site. The multidentate salen ligand chelates with zinc ions, and generates the rigidity of the zinc complex, which subsequently results in a dramatic fluorescence enhancement of the probe. Mass spectral and spectroscopic results reveal that the reaction proceeds rapidly in a 1 to 1 stoichiometric manner to form a clean zinc complex. We have demonstrated that the fluorescence probe not only has high selectivity for zinc ion over other biological relevant ions but also shows high sensitivity for zinc ion with a detection limit of 83 nM. A fluorescence test paper has been developed by incorporating the probe on a polyamide fiber paper. The fluorescence color of the test paper became bright green-yellow under UV lamp when water sample containing zinc ions was dropped on it, suggesting its potential application in the measurement of zinc ions.

Abstract:By combining a sulfonated β-naphtol as receptor, a hydrophilic fluorescence probe with extreme sensitivity (7 ppb) and selectivity for zinc ion has been designed and synthesized in a simple and efficient way. Based on the inhibition of internal charge transfer (ICT) process, This “turn-on” type fluorescent sensor molecule features high fluorescence increase (13 fold) at 508nm as well as a red-shift from 371 to 430nm over most competitive heavy and transition metal (HTM) ions in water (pH 7.4). Furthermore, cell imaging experiments show that this fluorescence probe might be used for monitoring Zn2+ within biological samples. The role of this sensor molecule in living cells was further confirmed with the treatment of N,N,N′,N′-Tetrakis(2-pyridylmethyl)ethylenediamine. And the impact of water-solubility and liposolubility of such fluorescence probe on biological application has been deeply discussed.
